Seasonal Occurrence and Damage of Bactrocera scutellata (Diptera: Tephritidae) in Jeonbuk Province (전북지역에서 호박꽃과실파리(Bactrocera scutellata)의 발생소장과 피해)

  • The striped fruit fly, Baetrocera scutellata (Diptera: Tephritidae), is one of the most important pests in Cucurbitaceae plants. Among 6 attractants for B. scutellata used, Cue-lure was shown better effect than the others. Newly emerged adult of B. scutellata had been occurred early June of the year tested. There were two peaks of occurrences of B. scutellata, one was from mid-July to early August and the other was early September suggesting that B. scutellata has two generation a year. The larva of B. seutellata damaging the female flower and stem of the pumpkin was found for the frist time on Korea. The blossoms were damaged and fallen by B. scutellata which were 53.8% of male flower and 30.7% of female flower in the 6 pumpkins. The pumpkin flowers were damaged 51.9% by B. scutellata. Especially thc pumpkin flowers damaged were shown 72.7% (1015/1397 individuals) from August to September. The larva of B. scutellata was found from the Trichosanthes kirilowii on 24 June 2009 in Jinan-gun Jeollabuk-do.

  • 호박(Cucurbita moschta DUCH)은 한국인들이 즐겨먹는 식용자원으로서 독특한 향미와 약리작용을 갖고 있다. 또한 기후 환경에 대한 적응력이 높고 병해충에도 비교적 강하여 농약사용이 적은 무공해 식품으로 알려져 있다. 호박의 과육에는 혈전증 예방과 두뇌발달에 기여하는 $\alpha$-linoleic acid가 다량 함유되어 있고, 비타민 A의 전구체인 $\beta$-carotene이 풍부하며, 위장이 약한 사람, 회복기의 환자, 부기가 있는 산모에게 좋은 식품으로 애용되어 왔다. 또한 예로부터 호박씨는 독성이 없으므로 어린이, 임산부, 허약자의 조충, 구제약으로 또는 젖이 잘 나지 않을 때에도 많이 쓰여왔다. 그리고 호박꽃은 잎과 함께 식용으로 널리 이용되었을 뿐 아니라 벌레에 물렸거나 상처 난 곳에 으깨어 문지르면 효과가 있어 민간요법으로 사용되어 왔다. 따라서 본 연구에서는 우리나라 전역에서 쉽게 구할 수 있는 호박꽃의 생리활성능을 검색해 봄으로써 천연기능성 식품 소재로의 활용가능성을 제시해 보고자 하였다.

The Attraction Effect of Different Types of Cuelure on Striped Fruit Flies, Zeugodacus scutellata, in Jeju and Gyeongbuk Regions (제주 및 경북지역에서 cuelure 제형별 호박꽃과실파리(Zeugodacus scutellata)의 유인효과)

  • The striped fruit fly, Zeugodacus scutellata (Diptera: Tephritidae), is one of important pests in Cucurbitaceae plants. The cuelure is currently using with various forms for surveillance to the Z. scutellata male as well as Z. cucurbitae, Z. tau, and Bactrocera tryoni, which are categorized as prohibited-quarantine fruit flies in Korea. This study was conducted to verify the attraction effect of applying the cuelure in different forms in field conditions. In this study, we used cuelure by 3 different forms as solid, liquid, and wax to striped fruit flies using the Steiner trap. The trap was placed in two Jeju and one Gyeongbuk regions and the number of attracted flies by each cuelure form is monitored by every 2 weeks during 2021. Attraction effect on Z. scutellata was significant difference by different cuelure forms in two Jeju regions. Liquid and wax forms of cuelure showed high attraction number to the Z. scutellata male compared with solid form when flies were occurred with high population from July to August in Jeju. However, there was no significant difference on attraction effect at low population in Juju and Gyeongbuk by different forms. Based on field assay, liquid and wax form of cuelure are recommended for surveillance against Z. scutellata male. Furthermore, these results also strongly suggest that the attractant, liquid and wax forms of cuelure, could be effective for surveillance to quarantine fruit flies such as Z. cucurbitae, Z. tau, and B. tryoni in Korea.

Technologies Required for Development of Trap-based MAT Control Against the Striped Fruit Fly, Bactrocera scutellata (호박꽃과실파리의 트랩형 수컷박멸제 기반 기술 개발)

  • The striped fruit fly, Bactrocera scutellata, infests pumpkin flowers. Males are attracted to raspberry ketone (RK) and feed the attractant. This study was conducted to determine essential techniques to develop a male annihilation technique (MAT) of this insect pest. Effective attractants were screened in laboratory and field conditions. Both males and females were responsive to methyl eugenol (ME) in laboratory, though no flies were attracted to ME traps in field conditions. In contrast, cuelure (CL), which is a chemical derivative of RK, was effective to attract males of B. scutellata in both laboratory and field conditions. However, RK was equivalent or superior to CL when they were formulated in a form of wax dispenser. A pyrethroid insecticide along with the attractant was effective to attract and kill B. scutellata. Funnel trap was useful for MAT to confirm and count dead flies. These results indicate that MAT against B. scutellata consists of RK and bifenthrin in a wax type formulation, which is installed to a funnel type of trap. These essential factors would be useful to develop MAT applicable to control B. scutellata in fields.

Monitoring Technique of Pumpkin Fruit Flies Using Terpinyl Acetate-Protein Diet Lure and Development of Its Spraying Formulation for The Fly Control (Terpinyl acetate-단백질먹이 유인제를 이용한 호박과실파리류 연중발생 모니터링 기술 및 살포용 방제 제형 개발)

  • Two tephritid fruit flies are infesting pumpkins in Korea. Both are classified into genus of Zeugodacus. The striped fruit fly, Z. scutellata, males are attracted to a lure called Cuelure (CL), which has been used to monitor the occurrence of this fruit fly in the crop field. In contrast, no effective male lure was not developed to monitor the pumpkin fruit fly, Z. depressa. Protein diet lure has been used to attract females of most fruit flies. The addition of terpinyl acetate (TA) was effective to increase the attractiveness of Z. depressa. This study aimed to monitor the occurrence of Z. depressa in pumpkin field with TA-protein diet lure. To validate the efficiency of TA-protein diet lure, Z. scutellata was monitored in a year of 2019 using CL and TA-protein diet lures, and the yearly monitoring data were compared. The occurrence patterns derived from both lures were similar except late season after October. The extended catches of TA-protein diet lure might be explained by the adult diapause induction of Z. scutellata at late September. Monitoring Z. depressa with TA-protein diet lure gave two peaks at mid July and August-September, in which more than 80% catches were females. Based on the attractiveness of TA-protein diet lure, its wettable powder with an addition of spinosad insecticide was formulated and sprayed to pumpkin crops. After 7 days post-spray, the control efficacy recorded more than 70%. However, the control efficacies decreased as the time progressed after the spray. These results demonstrated the application of TA-protein diet lure for monitoring occurrence of Z. depressa in pumpkin-cultivating field conditions. The wettable powder containing spinosad can be applied to develop a new control agent against two pumpkin fruit flies.

Development of Female Annihilation Technique Against Pumpkin Fruit Flies Using Protein-based Terpinyl Acetate (단백질 기반 Terpinyl Acetate를 이용한 호박과실파리류 암컷 유살 기술 개발)

  • Bactrocera depressa and B. scutellata are serious insect pests to pumpkin in Korea. Adult stage which exposes to aerial condition is the optimal target for control because the immature stages of both species hide within host fruit, flowers or under ground. This study aimed to develop an attract-to-kill technique of their females. A commercially available attract-to-kill agent was not effective to attract these two pumpkin fruit flies. This study developed an attractant containing protein-based terpinyl acetate (TA). This TA-protein lure was effective to attract two pumpkin fruit flies. It attracted both sexes with female-biased frequency (60-70%). This TA-protein lure would be used for control both pumpkin fruit flies and may be used for monitoring field populations of B. depressa, in which specific attractant is not known.

Technique to Generate Sterile Males of Striped Fruit Flies, Zeugodacus scutellata, using Electron Beam Irradiation and their Application to Genetic Control (전자빔 조사를 이용한 호박꽃과실파리의 불임 수컷 제조 기술과 유전방제 응용)

  • The striped fruit fly, Zeugodacus scutellata, is endemic in Korea, but it has been regarded as one of the serious quarantine pests throughout the world. Sterile insect release technique (SIT) has been used to eradicate quarantine fruit flies. This study developed a technique to generate sterile males and applied SIT to control Z. scutellata. First of all, the reproductive systems of Z. scutellata adults were examined with fluorescent microscope. Polytrophic ovaries comprises of around 100 follicles with developing oocytes. Each follicle contains an oocyte with several nurse cells and are surrounded with follicular epithelium. Oocyte development began at 10 days after adult emergence (DAE) and formed chorionated oocytes after 20 DAE. On the other hand, male testes were well developed just after adult emergence. The vas deferens was filled with motile sperms. To generate sterile males, different doses (0~1,000 Gy) doses of electron beam were irradiated to 3~5 days old pupae of Z. scutellata. When male pupae were irradiated with electron beam at 200 Gy, they developed and mated with females without any significant difference compared to untreated males. Although the untreated females mated with the 200 Gy-irradiated males laid eggs, no eggs did not hatch. The 200 Gy-irradiated males were then applied to untreated male and female flies in a density ratio of 1:9 (untreated males : treated males). The laid eggs suffered significant infertility. These results suggest that electron beam-irradiated pupae at 200 Gy resulted in male sterility and the resulting males would be applied to SIT.

The Influence of over Foraging, and Pollinating Activities on Tomato Fruits by a Korean Native Bumblebee, Bombus ignitus S, (Hymenoptera : Apidae) in Cherry-tomato Houses (시설방울토마토에서 호박벌(Bombus ignitus)의 방화활동 및 과도한 방화가 토마토에 미치는 영향)

  • This study examined the foraging activities and the Influence of excessive foraging activity by B. ignitus workers on the quality of cherry-tomato fruits in cherry-tomato green-house. The peak time of pollination by B. ignitus worker was recorded at 8:00 and the average of foraging time on each flower was 11.7 secs. The total pollinating time of B. ignitus was 41 mins and 37 secs and its total number of visiting flowers was 195.2. The average time of staying on flowers was 8.8 ${\pm}$5.4 secs when two bumblebees were allowed to pollinate per Pyong (3.954 square yards) in the green house of tomatoes for 24 hours, but it was two times shorter (4.0${\pm}$3.1 secs) compared with that when one bumble-bee was allowed to pollinate per Pyong for one hour. The stamen color of tomato flowers visited by the bumblebee for one hour was brown while the stamen color for 24 hours was dark brown due to the frequent visiting. The average fruit bearing rate of the 4th-6th clusters of tomatoes visited for one hour was 48.9%, which was not significantly different from those allowed to visite for 24 hours. However, the number of seeds of flowers visited for 24 hours was 64.0, which was more than 55.3 seeds of flowers visited for one hour. The weight of fruit was 25.4 g and 24.4 g, respectively. The sweetness was not different significantly between one and 24 hours of visit (P > 0.05). The result revealed that end pro-duct of cherry-tomato was not influenced by over foraging activity of B. ignitus workers on tomato flowers.

Fungicidal Activity of 46 Plant Extracts against Rice Leaf Blast, Rice Sheath Blight, Tomato Late Blight, Cucumber Gray Mold, Barley Powdery Mildew and Wheat Leaf Rust (46종 식물추출물의 식물병 방제효과)

  • Ethanol extracts from 46 plants were tested for their fungicidal activity against six plant diseases consisting of Maynaporthe grisea, Rhizoctonia solani, Botrytis cinerea, Phytophthora infestans, Puccinia recondita, and Erysiphe graminis in the greenhouse studies. Strong activity at 5 and 10 mg/pot was produced from the extracts of Helianthus annuus flowers and Zea mays leaves against P. grisea. In a test with B. cineara, extracts of H. annuus leaves, H. annuus flowers, Chrysanthmum coronarium var. spatiosum, Cucurbita moschata seeds, Lycopersicon esculentum, Z. mays, and Z. mays leaves had strong activities at 5 mg/pot. In a test with P. recondita, strong activity was obtained from the extracts of Capsicum frutescens, C. moschata seeds, H. annuus seeds, L. esculentum, and Malva veticillata at 5 mg/pot. Against E. graminis, extracts of Cucumis sativus, H. annuus seeds, Salanum tuberosum, Z. mays, and Z. mays leaves produced strong activities at 10 mg/pot. All the extracts were ineffective against P. infestans and R. solani. Among seven extracts tested, the extracts of H. annuus leaves and flowers were highly effective against all the strains of B. cinerea resistant to carbendazim, procymidone, and diethofencarb. Furthermore, potent fungicidal activity was produced from the extracts of C. coronarium var. spatiosum and C. moschata seeds against the SSR, SRR, and RSR strains of B. cinerea, and Z. mays and Z. mays leaves against SSR and RSR. Extract of L. esculentum showed very strong activity only against RRS of B. cinerea.

Antioxidant Activity and Safety Evaluation of Juice Containing Protaetia brevitarsis (흰점박이꽃무지(Protaetia brevitarsis) 함유 음료의 in vitro 항산화 관련 생리활성효능 및 안전성 검증)

  • The purpose of this study was to assess the antioxidant activity of vegetable extracts (pumpkin, aloe, and artichoke) containing Protaetia brevitarsis (PB) and the clinical and pathological changes in ICR mice after a single oral administration. The total polyphenol (TP) and 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging ability, total radical trapping antioxidant potential (TRAP), oxygen radical absorbance activity (ORAC), and single cell gel electrophoresis assay were done to measure their antioxidant activities. The effect of vegetable extracts containing PB in TP and the ORAC value was significantly higher than those without PB. In addition, all extracts had effective $DPPH{\cdot}$ scavenging and $ABTS{\cdot}+$ scavenging activities. The protective effect of vegetable extracts with/without PB on $H_2O_2$-induced DNA damage was found. In a single-dose toxicity study, mortality, body weight, physiological signs, and biochemical analysis were analyzed. Seventy mice were randomly assigned to 7 experimental groups and were administered three vegetable extracts with and without PB (2 g/kg). A full 14 days after administration, no mice mortality was observed in any group. Body weight, physiological signs, and biochemical analysis were never significantly different from those of the control group. Taken together, these findings indicate that vegetable extracts containing PB with antioxidant activities and safety could be applied as medicinal and edible resources in an industrial area.
