The Utility of TAR Vectors Used for Selective Gene Isolation by TAR Cloning. (TAR Cloning에 의한 선별적 유전자 분리에 사용되는 TAR Vectors의 유용성에 관한 연구)

  • The Transformation-Associated Recombination (TAR) cloning technique allows selective isolation of chromosomal regions and genes from complex genomes. The procedure requires knowledge of relatively small genomic sequences that reside adjacent to the chromosomal region of interest. This technique involves homologous recombination during yeast spheroplast transformation between genomic DNA and a TAR vector that has 5'and 3' gene targeting sequences. In this study, we examined the minimum size of specific hooks required for a single-copy gene isolation and compared the utility of different TAR vectors, radial and unique vectors, by cloning the same single-copy gene. The efficiency of TAR cloning of the hHPRT gene was same using hooks varying from 750 to 63 bp. The number of transformants decreased approximately 20-fold when the TAR vector contained two unique hooks versus using a radial vector, but the percentage of positive recombinants increased over 2-fold when a unique TAR vector was used. Therefore, we suggest that the two-unique TAR vector is suitable for general TAR cloning given its high selectivity, and the radial TAR vector is more suitable when genomic DNA is in limited quantity, for example, DNA isolated from pathological specimens. Moreover, we confirm the minimal length of a unique sequence in a TAR vector is approximately 60 bp for a single-copy gene isolation.

  • This paper suggests study of basic structure design and structural analysis for the twin car-ferries. The rules and methodology for the strength analysis of medium and small sized high speed vessels with a length of more than 50m and a length/width ratio of more than 12, such as car-ferries, have not been clarified yet. Therefore, in this paper, the members scantling were carried out using the KR Rule, and the car-ferry exclusive vessel standard were further applied to verify the structural strength the design. The scantling of the members is based on the Korea Classification standards, and the car-ferries standards were additionally applied to verify the structural strength of the design. Especially, the ultimate strength of hull girder was additionally carried out by using car-ferry exclusive vessel standard, and proposed to overcome ambiguity of design by existing standard. The results of this study are expected to be useful as basic data related to structural design and structural analysis of high speed twin-hull car-ferries. The present car-ferry design has a sufficient safety margin in strength point of view according to the KR rule.

Preparation and Physical Properties of Poly(ethylene-co-ethyl acrylate)/Carbon Nanotube Nanocomposites (폴리에틸렌에틸아크릴레이트/카본나노튜브 나노복합체의 제조 및 물성)

  • Multi-walled carbon nanotubes (MWCNT)-reinforced poly(ethylene-co-ethyl acrylate) (EEA) nanocomposites were prepared by both melt and solution mixing methods. The mechanical, thermal, and electrical properties were investigated as a function of type and loading of CNT. The tensile strength and modulus increased, while elongation at break decreased with increasing MWCNT content. The hollow-type MWCNT showed an improved tensile strength and elongation at break compared with a conventional MWCNT. The thermal degradation temperature was increased by around $40^{\circ}C$ with increasing the amount of MWCNT. The melt-mixed composites showed the highest volume resistivity. In the case of solution-mixed composites, the conventional MWCNT was estimated to show much lower volume resistivity than that of hollow MWCNT. The number and length of extruded CNT onto the fractured surface increased by both increasing the content of CNT and employing the tensile strain to the sample. The melt-mixed specimens showed much smaller number and shorter length of extruded CNT.

Breeding of Phalaenopsis 'SM 333' with Mini Multiple Flower Formation (소형 다화 분지성 호접란 'SM 333' 육성)

  • A new Phalaenopsis cultivar 'SM 333' was bred by Sangmiwon Orchid, Korea, which produces young plants through tissue culture techniques. The new cultivar 'SM 333', showing the phenotype of multiflora with pink color and small, multibranching-type characteristics, was derived from crossing between Phalaenopsis 'Odoriko' and 'Be Tris'. An elite individual number '02-03-33' later termed 'SM 333' was selected among about 300 individual progenies, based on an intensive selection process covering vegetative and flowering distinctiveness over more than 2 years. In year 2004-2005, the 1st and 2nd characteristic analyses were carried out through performance and uniformity tests. 'SM 333' shows flower color that is bright clean pink (RHS # RP69D) and flower shape that is formal type with 5.0 and 5.8 cm in flower height and width, respectively. 'SM 333' is regarded as raceme flower type suitable for the small casual flower market. The leaves of 'SM 333' grow horizontally and about 20.8 cm in length and 6.5 cm in width. This cultivar also possesses no genetic variation, and is amenable to fast in vitro propagation and easy growth due to its vigorous growth habit. This 'SM333' was registered (Reg. # 2916) with Korea Seed & Variety Service (KSVS) on 1st December, 2009, and the plant breeder's right is currently controlled by Sangmiwon Orchid Company, Korea.

Geophysical and Geological Investigation for Selecting a Dinosaur Museum Site in the Dinosaur Egg Fossil Area, Gojeong-ri, Hwasung, Gyeonggi Province (경기도 화성 고정리 공룡알 화석지 공룡생태박물관 부지선정을 위한 지구물리 및 지질조사)

  • In this study, we investigated the geologic structure of the basement and overlying sediments of the construction site of the dinosaur egg fossil museum in Hwasung, Gyeonggi Province through refraction seismology, drilling, and downward seismic velocity measurements in the drill holes. The construction site ($350{\times}750\;m^2$) is located in the reclaimed area south of Sihwa Lake, Gojeong-ri. About 6,950 m of seismic refraction data consisting of 11 lines were acquired using a sledge hammer source. Drilling to the basement was performed at five sites. Sediment samples from drilling were analysed for grain-size distribution and age dating. At two drill holes, seismic velocity was measured with depth using a hammer as a seismic source. The geological structure of the study area consists of, from top to bottom, a tidal flat layer (5 ~ 12 m thick), a weathered soil layer (2 ~ 8 m thick), and the basement. The basement is interpreted as Cretaceous sedimentary rocks that tend to be shallow eastward. The volume of the tidal flat sediments and weathered soil in the study area is estimated as $1.4{\times}10^6\;m^3$, weighing $3.5{\times}10^6$ tons. The rate of sea level rise since 8,000 yrs BP is estimated to be 0.1 ~ 0.15 cm/yr.

The Relationship Analysis between the Epicenter and Lineaments in the Odaesan Area using Satellite Images and Shaded Relief Maps (위성영상과 음영기복도를 이용한 오대산 지역 진앙의 위치와 선구조선의 관계 분석)

  • The purpose of this paper is to analyze the relationship between the location of the epicenter of a medium-sized earthquake(magnitude 4.8) that occurred on January 20, 2007 in the Odaesan area with lineament features using a shaded relief map(1/25,000 scale) and satellite images from LANDSAT-8 and KOMPSAT-2. Previous studies have analyzed lineament features in tectonic settings primarily by examining two-dimensional satellite images and shaded relief maps. These methods, however, limit the application of the visual interpretation of relief features long considered as the major component of lineament extraction. To overcome some existing limitations of two-dimensional images, this study examined three-dimensional images, produced from a Digital Elevation Model and drainage network map, for lineament extraction. This approach reduces mapping errors introduced by visual interpretation. In addition, spline interpolation was conducted to produce density maps of lineament frequency, intersection, and length required to estimate the density of lineament at the epicenter of the earthquake. An algorithm was developed to compute the Value of the Relative Density(VRD) representing the relative density of lineament from the map. The VRD is the lineament density of each map grid divided by the maximum density value from the map. As such, it is a quantified value that indicates the concentration level of the lineament density across the area impacted by the earthquake. Using this algorithm, the VRD calculated at the earthquake epicenter using the lineament's frequency, intersection, and length density maps ranged from approximately 0.60(min) to 0.90(max). However, because there were differences in mapped images such as those for solar altitude and azimuth, the mean of VRD was used rather than those categorized by the images. The results show that the average frequency of VRD was approximately 0.85, which was 21% higher than the intersection and length of VRD, demonstrating the close relationship that exists between lineament and the epicenter. Therefore, it is concluded that the density map analysis described in this study, based on lineament extraction, is valid and can be used as a primary data analysis tool for earthquake research in the future.

Conservation Treatment on the Bamboo Sunblind from the No. 1 Catchment Site in Baesanseongji, Busan (부산 배산성지 1호 집수지 출토 대나무 발 수습 및 보존처리)

  • In the present study, safe management and value improvement of bamboo sunblind, which is an item of cultural heritage, were performed by adopting stable conservation treatment methods. The bamboo sunblind used in the present study was excavated from No. 1 catchment site in Baesanseongji, Busan. It was determined that the main material used to make the sunblind was bamboo, and herbal plants were used to weave the bamboo using lacquer as an adhesive agent. All contaminants and soil adhered to the sunblind was removed. Thereafter, the sunblind, which was recovered in the form of blocks, was washed separately after fixing it to a temporary plaster frame and to avoid the blocks from breaking during washing. Then, polyethylene glycol (PEG) impregnation was utilized for the reinforcement treatment. Based on the preliminary test results, the shape of the sunblind was fixed using a stainless-steel frame to prevent physical damage that may occur during the drying process. Thereafter, the bamboo sunblind was vacuum freeze-dried. PEG 20% (in ethyl alcohol) was applied as a surface treatment agent for stabilization the sunblind. After the surface treatment, the bamboo sunblind were joined together to fit the maximum width, and the rectangular shape of the sunblind was restored-as best as possible-while filling in the missing parts by maximizing the use of unknown members such as in the disturbed layers below bamboo sunblind surface. The conservation treatment was completed by fixing the bamboo sunblind into the fabricated frame.

Development of Tomographic SASW Method to Evaluate Two-Dimensional Variability of Shear Stiffness (지반 및 구조물의 이차원적 전단강성 평가를 위한 토모그래픽 SASW 기법의 개발)

  • The SASW (Spectral-Analysis-of-Surface-Waves) method, which evaluates the stiffness structure of the subsurface and structures nonintrusively and nondestructively, has been successfully used in the civil engineering applications. However, the SASW method assumes that the subsurface or structures consist of horizontal multi-layers, so that the method has some difficulty in continuously evaluating the integrity of a tunnel lining and a pavement system. This difficulty prevents the SASW method from being used to generate a tomographic image of stiffness for the subsurface or structures. Recently, the GPR technique which has the advantage of continuously evaluating integrity of the subsurface and structures has been popular. This advantage of GPR technique initiated the efforts to make the SASW method, which is superior to GPR and other nondestructive testing methods due to its capability of evaluating stiffness and modulus, be able to do continuous evaluation of stiffness structure, and the efforts finally lead to the development of \ulcornerTomographic SASW Technique.\ulcorner Tomographic SASW technique is a variation of the SASW method, and can generate a tomographic image of stiffness structure along the measurement line. The tomographic SASW technique was applied to the investigation of lateral variability of a sand box placed by the raining method for the purpose of verifying its effectiveness. Tomographic SASW measurements on the sand box revealed that the investigated sand box has different shear stiffness along the measurement line, which gave a clue of how to make a better raining device.

Growth $Al_2$O$_3$/ZrO$_2$eutectic fibers by the micro-pulling down method and its mechanical properties (Micro-pulling down법을 이용한 $Al_2$O$_3$/ZrO$_2$eutectic fiber의 제조 및 기계적 특성)

  • $Al_2O_3/ZrO_2$eutectic fibers were grown by micro-pulling down technique and investigated their microstructure as a function of solidification rate. $Al_2O_3/ZrO_2$eutectic fibers 0.2~2 mm in diameter and 500 mm in length have been grown with a pulling rate of 0.1~15 mm/min. The eutectic microstructures changed as a function of fulling rate from rod-shaped to cellular shape containing some thin lamellar pattern via uniform lamellar structure. Typical lamellar thickness decreased from about 380 nm to 110 nm as the pulling rate increased from 1 mm/min to 15 mm/min. The interlamellar spacing fitted with the inverse-square-root dependence on pulling rate according to $\lambda$= $1{\times}v^{-1/2}$, where $\lambda$ has the dimension in $\mu\textrm{m}$ and v is $\mu\textrm{m}$/s. Hardness value reached 13.1 GPa at 15 mm/min of pulling rate and tensile strength 900 MPa at 10 mm/min were also increased as the interlamellar spacing decreased.

  • The crystal structure Tris(ethylenediamine)nickel(II)Dichromate has been determined by X-ray crystallography. Crystal data: a=8.268(2), b=13.865(2), c=14.921(2)Å, γ=102.04(2)°, V=1672.9(5)Å3, Z=4, Monocline, P21/b (space group No.=14), Dcalc=1.806 gcm-3, μ=24.05 cm-0.1. The intensity data were collected with Mo-Kα radiation(λ=0.7107Å) on an automatic four-circle diffractometer with a graphite monochromator. The structure was solved by Patterson method and refined by full matrix least-square methods using unit weights. The final R and S values were R=0.045, Rw=0.051, Rall=0.059 and S=2.171for 2248 observed reflections. The two carbon atoms of a ring of Ni(en)-ion were split into crossed four atoms. In consideration of α- and β-angles of two rings of a disordered ethylenediamine of Nien3-ion and the hydrogen bonds between Ni(en)3-cation and Cr2O7-anion, the configuration of Ni(en)3-ion is assumed to be disordered with Λδδδ and Λδδλ.
