This paper describes seasonal changes of total length, hepatosomatic index (HSI), fatness, egg-diameter composition, and fecundity of the longchin goby Chasmichthys dolichognathus(HILGENDORF). The specimens used were captured in the tide pool of Tongbaekseom, Pusan, Korea from February 1983 to January 1984. The age of longchin goby, tested by size frequency, was believed to be usually one year which grows to 8.0 cm in total length. The annual variations of HSI reached the maximum in the early spring when the gonad was actively growing and decreased during the spawning season from April to July. The coefficients of fatness were represented low values for the spawning periods. Frequency distribution of the egg diameter of mature ovary has three modes: one is the evident mode of the ripe eggs group, and the other two are modes of maturing and immature eggs groups. And an individual is considered as spawns one in the spawning season. Relationships between the fish size in total length (TL cm) and the number of ovarian eggs(F), the fish weight (BW g) and the number of ovarian eggs are indicated by the exponential equation respectively : F=42.585 $TL^{1.608}$, F= 524.589 $BW^{0.475}$.

Fatty acids in seed oil from plants are essential for human nutrients and have been used for industrial purpose. The growing demands of seed oil as food resources and feedstocks for industrial uses have attempted to modify fatty acid composition and to increase oil content in transgenic plants. However, production of unusual fatty acids in transgenic plants are limited, which is not synthesized the level same as original plants. This bottleneck was common for production of several unusual fatty acids in transgenic plants and suggests that there is different for substrate preference in oil metabolic pathway enzymes between host oil plants and original wild plants. Review of acyltransferases involved in acyl-editing and seed oil accumulation of oil plant and wild-plant producing unusual fatty acids will design strategies to maximize the production of unusual fatty acids in transgenic plants. In here, we identified eleven acyltransferase genes in castor based on sequence homology, which will be useful to increase hydroxy unusual fatty acids in transgenic plants.
Coleosporium neocacaliae, a needle rust fungus, was described and illustrated for the first time on the needles of Pinus koraiensis (Korean pine) in Korea. Detailed descriptions of the species were verified based on symptoms and signs, and light and scanning electron microscopic observations on aecia, aeciospores and peridial cells. The present needle rust fungus was apparently different from C. eupatorii, a solely recorded Coleosporium species on P. Koraiensis in Korea until this time, in having comparatively large sized aeciospores as $20-39{\times}14-27\;{\mu}m$ than those of latter species (size of aeciospores $15-27{\times}10-20\;{\mu}m$). Verrucose surface structure of aeciospores in light microscopy was revealed to be annulated with three to four rings and root-like base in scanning electron microscopy. Aecial stage of C. neocacaliae were formed on the middle days of April to the early days of June and collected around the central parts of Korean peninsula, including high mountains of about 1,000 m above sea level.
It has been well known that the expression of S. typhimurium cadBA operon requires at least two extracellular signals: low pH and high concentration of lysine. To better understand the nature of pH-dependent and lysine dependent signal transduction, mutants were isolated in JF2238(cadA-lacZ) by Tn10 insertion, spontaneous mutagenesis, and EMS treatment. Two mutants were isolated from JF2238, expressed as a cadA-lacZ operon fusion in various growth conditions, and analyzed to have mutations in cadC, a gene encoding a function necessary for transcriptional activation of cadBA. One isolate (cadC6) conferred pH-independent and lysine-independent cadBA expression and the other(cadC4) showed pH-independent and lysine-dependent cadBA expression. cadR::Tn10 and cadR4 mutants were expressed in the absence of exogenously added lysine. They were also resistant to thiosine and complemented by lysP clone from E. coli. Thus, in the absence of exogenous lysine, cadR is a negative regulator of cadBA expression. Cadaverine, the product of lysine decarboxylation, was shown to inhibit expression of cadA-lacZ fusion in cad $C^+$ cell.
Chinese CGMS (Cytoplasmic-Genetic Male Sterility) is being transfered to Korean cultivars which will be used as CGMS maintainers. These maintainer lines along with the Chinese CGMS lines and their maintainer lines were crossed with another group of Korean cultivars which will be used as the fertility restorers, and their yield heterosis were examined. The yield heterosis was calculated as high as 113.0-221.1 % in terms of heterosis (F$_1$/Midparent) with the grand mean of 150.5%, and, 86.1-179.8% in terms of heterobeltiosis (F$_1$/better parent) with the grand mean of 125.3%. Among the female parent groups, Chinese mainteiners were the highest following by Chinese MS lines and the Korean maintainers were the lowest. Heterobeltiosis of yield components were 92.7%, 120.0%, 103.0% and 112.0% for number of panicles/unit area, number of grains/panicle, fertility per cent and 1000 grain weight, respectively. Path-coefficients of yield components to the yield were calculated. The highest one was number of grains per panicle ranging 0.8073-0.8649 followed by the 1000 grain weight ranging 0.2000-0.5032. The grain yield ratios to the grand mean were calculated in terms of combining ability. It was highest with the Chinese maintainers followed by Chinese MS lines and The Korean maintainers were lowest. This facts bring cautions to the breeding efforts to transfer the Chinese CGMS in to Korean maintainers.
A PPIase gene of Bacillus stearothermophilus was screened from a genomic library by plaque hybridization using the A-1 primer as a probe. A PPIase positive plaque contained a 3.0kb insert of the chromosomal DNA. A 3.0kb fragment was subcloned into pUC18, resulting pPI1-40. A DNA fragment encoding the N-terminal portion of the PPIase in pPi-40 was amplified by polymerase chain reaction(PCR) method using the A-1 and B-2 primers. The amplified fragment was cloned into the Sma I site of pUC18 and recombinant plasmid was designated as pSN-18. The nucleotide sequence of 167bp fragment was determined. The deduced amino acid sequence of PPIase was completely matched with the determined N-terminal amino acid sequence of PPIase B. stearothermophilus. The translated protein sequence of PPIase B. stearothermophilus was compared with sequence from periplasmic PPIase from Escherichina coil ; homogies of 16 and 58%, respectively, were found. The clond PPIase gene was over-expressed in E. coil cell using pUC19 as an expression vector. The enzyme was partially purified by heat treatment and colum chromatochraphy on DEAE-Sepharose CL-6B. The molecular weight of the enzyme was dermined to be about 18.0 kDal by SDS-PAGE.

Ultrastructural characteristics of blacktip grouper, Epinephelus fasciatus spermatozoa were investigated using transmission and scanning electron microscopy. The spermatozoa of E. fasciatus consisted of a spherical head part, a midpiece with cytoplasmic canal entrance and a flagellum with lateral fins. Internal ultrastructurally, the nucleus contains high electron dense chromatin having granular particles and has no acrosome. The centriolar complex lies outside of the nuclear fossa and it is connected by the osmophilic filaments. Also the osmophilic filaments connect between the centriolar complex and the nuclear membrane. The midpiece contains eight to nine spherical mitochondria, cytoplasmic canal and necklaces. The flagellum has a typical 9+2 axonemal structure. The lateral fins contain vesicles and a typical 9+2 axonemal structure. Consequently this study contributes to comparative grouper spermatology and provide useful systematic taxonomic characters.
Multiple myeloma (MM) is the leading cause of death among hematologic neoplasms. Recently, microRNA has been reported to be useful in the diagnosis of multiple myeloma. This study examined whether miR-221 could be used as a diagnostic marker for multiple myeloma. The study was performed on 20 patients with multiple myeloma without any other hematological diseases. MicroRNA extraction was performed using formalin-fixed paraffin-embedded (FFPE) tissues obtained from the bone marrow of patients with multiple myeloma. miR-15a, miR-16, miR-21, miR-181a, and miR-221 were selected as the microRNA target genes for multiple myeloma. The significance of microRNA was based on a fold change of <1.5. To quantify the fold changes, data normalized to the human gene, SNORD43, were used as the values of the patient group. Fold change values greater than 1.5 were defined as "overexpression", whereas values less than -1.5 were defined as "underexpression". Of note, 65.0% (13/20) of samples showed significant "overexpression" in the levels of miR-221 expression and plasma cells with a group of more and less than 30% in MM patients did not show any significance of plasma cell (P<0.05). The results of other studies showing a correlation between the expression of miR-221 and MM in Caucasians were confirmed. These results suggest that miR-221 may be a useful indicator for diagnosing patients with MM. In conclusion, miR-221 is useful in the diagnosis and determining the prognosis of multiple myeloma in Koreans.

In this study, PAT protein of genetically modified maize was prepared from the recombinant E. coli strain BL21 (DE3), and mice were immunized with the recombinant PAT protein. After cell fusion and cloning, two hybridoma cells (PATmAb-7 and PATmAb-12) were chosen since the monoclonal antibodies (Mabs) produced by them were confirmed to be specific to PAT protein in the indirect enzyme-linked immunsorbent assay (ELISA) and western blot tests. There were no cross-reactions of either Mabs to other GM proteins or to the extracts of non-GM maize. The ELISA based on the PATmAb-7 can sensitively detect 0.3 ng/g PAT protein in corn. These results indicate that the developed Mabs can be used as bio-receptors in the development of immunosensors and biosensors for the rapid and simple detection of GM corn adulterated in foods.

Aromatic hydrocarbons are toxic environmental pollutants that are detrimental to the ecosystem and human health. Among them, chlorotoluene and nitrotoluene are toxic to hydrobios and irritate the skin, eyes, and respiratory organs of humans. We herein report the development of recombinant microbial biosensors for cheap and rapid monitoring of chlorotoluene and nitrotoluene compounds. Plasmids were constructed by inserting the xylR regulatory gene for BTEX (benzene, toluene, ethylbenzene, and xylene) degradation into upstream of Po' (the DmpR activator promoter Po with the deletion of its own upstream activating sequences) or Pu (the cognate promoter of XylR)::lacZ (the ${\beta}$-galactosidase gene) and transformed into Escherichia coli $DH5{\alpha}$. In the presence of inducers, the biosensor cells immobilized in agarose developed a red color in 1-2 h due to the hydrolysis of chlorophenol red ${\beta}$-D-galactopyranoside (CPRG), a substrate of ${\beta}$-galactosidase that was expressed by the inducers. Among BTEX, high responses were specifically observed with o-, m-, p-chlorotoluene ($0.1{\mu}M-100 mM$) and o-, m-, p-nitrotoluene (0.1 mM-100 mM). Po' demonstrated higher responses than those with Pu. The biosensors immobilized in agarose showed good stability after 21 days' storage at $4^{\circ}C$, and responses in untreated wastewater spiked with chlorotoluene and nitrotoluene, suggesting they can be used to detect compounds in wastewater.
