Trial for Drug Susceptibility Testing of Mycobacterium tuberculosis with Live and Dead Cell Differentiation (세포 염색 방법을 이용한 결핵균 감수성 검사법)

  • Background : The resurgence of tuberculosis and outbreaks of multidrug resistant (MDR) tuberculosis have increased the emphasis for the development of new susceptibility testing of the Mycobacterium tuberculosis for the effective treatment and control of the disease. Conventional drug susceptibility testings, such as those using egg-based or agar-based media have some limits, such as the time required and difficulties in determining critical inhibitory concentrations, but these are still being used in many diagnostic laboratories because of no better lternatives, considering cost and accuracy. To overcome these limits, a rapid and simple method for new susceptibility testing, using live and dead assays, was applied for a bacterial cell viability assay to distinguish dead from live bacterial cells based on two-color fluorescence. Materials and Methods Strains : Forty strains were used in this study, 20 susceptible to all antituberculosis drugs and the other 20 resistant to the four first line antituberculosis drugs isoniazid, rifampicin, streptomycin and ethambutol. Antibiotics : The four antibiotics were dissolved in 7H9 broth to make the following solutions: $0.1{\mu}g\;isoniazid(INH)/m{\ell}$, $0.4{\mu}g\;rifampicin(RMP)/m{\ell}$, $4.0{\mu}g\;streptomycin(SM)/m{\ell}$ and $4.0{\mu}g\;ethambutol(EMB)/m{\ell}$. Results : Live and dead Mycobacterium tuberculosis cells fluoresced green and red with the acridin (Syto 9) and propidium treatments, respectively. These results are very well accorded with conventional drug susceptibility testing by proportional method on Lowensen-Jensen media (L-J) containing 4 drugs (INH, RMP, EMB and SM), showing a 93.7 % accordance rate in susceptible strains and 95% in resistant strains. Conclusion : The results of the drug susceptibility testing using the live and dead bacterial cell assay showed high accordance rates compared with the conventional proportion method on L-J. This finding suggests that the live and dead bacterial cell assay can be used as an alternative to conventional drug susceptibility testing for M. tuberculosis strains.

Fluorescence Characteristic Analysis for Fiber Detection in Sectional Image of Fiber Reinforced Cementitious Composite (섬유 보강 시멘트계 복합재료의 단면 이미지에서 섬유 검출을 위한 섬유 형광 특성 분석)

  • It is important to detect fibers in the sectional image of fiber reinforced cementitious composites (FRCC), since the fiber distribution is a crucial factor to predict or evaluate the mechanical performance of FRCC. In this paper, we investigated the fluorescence characteristics of Polyvinyl Alcohol (PVA) fibers, Polyethylene Terephthalate (PET) fibers, Polyethylene (PE) fibers, and Polypropylene (PP) fibers used in Engineered Cementitious Composites (ECC), which is a special kind of FRCC that incorporates synthetic fibers and exhibits extremely ductile behavior in uniaxial tension, to detect each fiber according to its type. Furthermore, optimum excitation and emission wavelengths were proposed on the basis of maximum difference of Relative Fluorescence Intensity (RFI) between two types of fibers used in the hybrid ECC. Optimum threshold values to discriminate two types of fibers using statistical tools were also proposed. Finally, images of four types of fibers obtained using a fluorescence microscope are compared.

Synthesis and Photoluminescence Properties of Blue Phosphor Y1-xBO3:Cex3+ (청색 형광체 Y1-xBO3:Cex3+의 합성과 발광 특성)

  • $Y_{1-x}BO_3:Ce_x^{3+}$ ceramic phosphors were synthesized with changing the concentration of $Ce^{3+}$ ion by using a solid-state reaction method. The crystal structure, surface morphology, and optical properties of the blue phosphors were investigated by using X-ray diffractometer (XRD), scanning electron microscopy, and photoluminescence and photoluminescence excitation spectrophotometry, respectively. The XRD results showed that the main peak of the phosphor powders occurs at (401)와 ($31\bar{2}$) planes. As for the optical properties, the excitation spectrum occurred at 243 nm and the value of blue emission intensity peaking at 469 nm reached the maximum when the concentration of $Ce^{3+}$ ion was 0.10 mol.

Automated Bacterial Cell Counting Method in a Droplet Using ImageJ (이미지 분석 프로그램을 이용한 액적 내 세포 계수 방법)

  • Precise counting of cell number stands in important position within clinical and research laboratories. Conventional methods such as hemocytometer, migration/invasion assay, or automated cell counters have limited in analytical time, cost, and accuracy., which needs an alternative way with time-efficient in-situ approach to broaden the application avenue. Here, we present simple coding-based cell counting method using image analysis tool, freely available image software (ImageJ). Firstly, we encapsulated RFP-expressing bacteria in a droplet using microfluidic device and automatically performed fluorescence image-based analysis for the quantification of cell numbers. Also, time-lapse images were captured for tracking the change of cell numbers in a droplet containing different concentrations of antibiotics. This study confirms that our approach is approximately 15 times faster and provides more accurate number of cells in a droplet than the external analysis program method. We envision that it can be used to the development of high-throughput image-based cell counting analysis.

다양한 활성제 이온이 치환 고용된 MgNb2O6 형광체의 특성

  • 최근에 산화물 형광체는 황화물 형광체에 비해 높은 화학적 안정성을 나타내기 때문에 백색 발광 다이오드, 전계방출 디스플레이와 플라즈마 디스플레이 패널에 그 응용성을 넓히고 있다. 마그네슘 니오베이트(magnesium niobate, MgNb2O6)는 우수한 유전 특성(상대 유전상수=18.4)을 나타내기 때문에 마이크로파 유전체로 응용 가능하며, 단일상 릴랙서 페라브스카이트(relaxor perovskite) Pb(Mg1/3Nb2/3)O3을 합성하기 위한 전구체 (precursor)로 널리 사용되고 있으며, 나이오븀산염 이온에서 다양한 색상을 방출하는 활성제 이온으로 효율적인 에너지 전달이 일어남으로써 Sm3+, Dy3+, Eu3+와 같은 희토류 이온의 좋은 모체 격자로 개발할 수 있다. 본 연구에서는 마그네슘 니오베이트 MgNb2O6 모체 결정에 다양한 활성제 이온, 즉 Eu3+, Sm3+, Dy3+, Tb3+를 선택적으로 주입하여 발광 효율이 높은 천연색 형광체를 합성하고자 한다. 특히, 모체 결정에 주입되는 활성제 이온 주위의 국소적인 환경이 반전 대칭에서 변형되는 척도를 조사하여 활성제의 주 발광 파장의 세기가 최대가 되는 최적의 조건을 결정하고자 한다. Mg1-1.5xNb2O6:REx3+ 형광체 분말 시료는 초기 물질 MgO, Nb2O5와 희토류 이온을 화학 반응식에 맞게 정밀 저울로 측량하여 플라스틱 용기에 ZrO2 볼과 함께 넣고, 소정의 에탄올을 채운 뒤 밀봉하고서, 300 rpm의 속도로 20시간 볼밀 (ball-mill) 작업을 수행하였다. 그 후, 체(sieve)로 ZrO2 볼을 걸러낸 다음에 혼합된 용액을 각 비커에 담아서 $40^{\circ}C$의 건조기에서 24시간 건조하였고, 건조된 시료를 막자 사발에 넣고 잘게 갈고 80 ${\mu}m$의 체로 걸러낸 후에, 알루미나 도가니에 활성제 이온별로 각각 담아, 전기로에 장입하여 매분당 $5^{\circ}C$의 비율로 온도를 상승시켜 $350^{\circ}C$에서 5시간 동안 하소 공정을 실시한 후에, 온도를 계속 일정한 율로 증가시켜 $1,200^{\circ}C$에서 5시간 동안 소성하여 합성하였다. 합성된 형광체 분말의 결정 구조는 $Cu-K{\alpha}$ 복사선(파장: 1.5406)을 사용하여 X-선회절장치로 측정하였으며, 형광체의 표면 형상은 전계형 주사전자현미경으로 관측하였다. 흡광와 발광스펙트럼은 제논 램프를 광원으로 갖는 형광 광도계를 사용하여 측정하였다. 모체 결정에 활성제 이온 Eu3+, Sm3+, Dy3+, Tb3+가 도핑된 형광체 분말은 각각 적색, 주황색, 황색, 녹색 발광이 관측되었다. 각 발광 스펙트럼과 결정 입자의 크기와 형상 사이의 상호 관계를 조사하였다. 실험 결과로부터, 각 형광체의 발광 파장은 활성제 이온의 종류 와 서로 밀접하게 관련되어 있으며, 형광체 시료 합성시 활성제 이온의 농도를 선택적으로 조절함으로써 발광의 세기를 제어할 수 있음을 확인하였다.

Preparation and Low-Voltage Luminescent Properties of $SrTiO_3$:Al, Pr Red Phosphor (저전압용 $SrTiO_3$ : Al, Pr 적색 형광체 합성 및 발광특성)

  • The $SrTi0_3$:Al, Pr red phosphors were prepared by solid state reaction method. Phosphor preparation parameters such as sintering temperature and time were optimized for the photoluminescence(PU intensity and the cathodoluminescence(CL) intensity. Powder samples showed the characteristic X-ray diffraction patterns of the perovskite structure and the average particle size of 3~5/$\mu\textrm{m}$ for particle size distribution(PSD) analysis. Also, scanning electron microscopy for the powder samples showed that the particles are reasonably crystallized with spherical shape. Especially, higher low voltage CL properties of $SrTi0_3$:Al, Pr phosphors than commercial $Y_2O_3$:Eu phosphors are expected to be applied for a low voltage field emission display(FED).

사파이어 기판에 증착한 MgMoO4:Eu3+ 박막의 광학 특성

  • 본 연구에서는 라디오파 마그네트론 스퍼터링 방법을 사용하여 증착 온도를 변화시켜면서 Eu 이온이 도핑된 MgMoO4 적색 형광체 박막을 사파이어 기판 상부에 성장하였다. 타겟은 고상반응법을 사용하여 직접 제작하였다. 형광체 박막의 구조, 표면, 광학적 특성은 X-선 회절장치, 주사전자현미경, 투과도 및 광여기발광 측정장치를 사용하여 측정하였다, 증착 온도는 100, 200, 300, $400^{\circ}C$이었으며, 증착 후 $870^{\circ}C$에서 열처리 공정을 실행하였다. 이와 더불어, $400^{\circ}C$에서 증착한 박막을 다양한 온도 $770-920^{\circ}C$에서 열처리를 수행하여 각각의 특성을 분석하였다. 증착 온도 $200^{\circ}C$에서 성장한 박막의 경우에 614 nm에 피크를 갖는 주 적색 발광 피크가 관측되었으며, 열처리 온도를 달리한 박막의 경우에는 $920^{\circ}C$에서 가장 강한 발광 피크가 나타났다. UV-VIS 분광광도계를 사용하여 박막의 투과도와 흡광도를 측정하였으며, Tauc의 모델을 사용하여 밴드갭 에너지를 계산하였다. 증착 온도 변화에 따라 성장된 박막의 투과도는 평균 82% 이상 이었으며 밴드갭 에너지는 4.1 eV이었다. 박막의 결정 구조는 단사정계임을 확인하였다. 특히, 결정 입자, 발광 피크의 세기와 투과도의 상관 관계를 조사하였다.

Fluorescence photon counting rate as a function of dye concentration: Effect of dead time of photon detector (색소 농도에 따른 형광 광자의 계수율 : 광자 검출기의 dead time 효과)

  • A single molecule detection system, which consists of confocal fluorescence microscope and single photon counter, has been used to observe the dye concentration dependence of photon counting rate. With increasing concentration, a saturation effect of counting is observed and demonstrated on the basis of the dead time of photon detector. The equations presented here show the relations between the counting rate and some parameters such as probe volume, quantum efficiency of detector, and fluorescence photon number entered onto detector. The signal-to-noise ratio is also discussed briefly.

Fabrication of computer-interfaced photon counter for single molecule detection in solution (용액내 단일 분자 검출을 위한 컴퓨터 인터페이싱 광자계수기의 제작)

  • In order to acquire the fluorescence burst signals emitted from single dye molecules that pass through a detection space defined by a confocal microscope, a computer-interfaced photon counter has been fabricated. The maximum count rate is about 80 MHz, which is limited by the counter devices used. Using both the operating computer program written by BASIC and the 486 PC computer, the minimum bin-width of 25 $mutextrm{s}$ has been achieved. The characteristics of fluorescence burst signals emitted from JA22 molecules at about 1$\times$$10^{-11}$m mol/L in ethylene glycol are discussed briefly.

DETECTION OF OCCLUSAL CARIES USING LASER FLUORESCENCE (레이저 형광법의 교합면 우식증 탐지 효과)

  • The purpose of this study was to evaluate the diagnostic validity of an incipient occlusal caries using argon laser fluorescence. Extracted human premolars and molars with enamel carious lesion of occlusal surface were assessed using visual examination, visual examination with probing, argon laser fluorescence and histologic depth of carious lesion. The results in each of all the three detection methods were compared to the assessment of histologic depth of carious lesion using polarized microscope. The results from the present study can be summarized as follows; 1. There was highly correlation between the histologic depth of occlusal caries and all three detection methods(P<0.01). 2. The reproducibility(kappa value) of the visual examination, visual examination with probing and argon laser fluorescence between the histologic depth of occlusal caries was 0.189, 0.128, 0.472. The highest correlation was seen between detection of occlusal caries by argon laser fluorescence and histologic scores by polarized microscope. The results from this study indicated that argon laser fluorescence considered to be accurate and reliable method in detecting occlusal caries.
