Parameters Affecting Nitrite Accumulation in Submerged Biofilm Reactor (생물막 반응기에서 아질산성 질소의 축척에 미치는 영향인자)

  • The objective of this study was to assess parameters affecting nitrite accumulation, which offers advantages in terms of less aeration energy and carbon consumption for denitrification. The influence of the alkalinity to $NH_4{^+}-N$ concentration ratio, pH, FA(free ammonia) concentration and temperature on nitrite accumulation was investigated. The experiment was performed with supernatant from dewatering process of anaerobic digested sludge using a submerged biofilm reactor. The influent contains high strength of ammonium nitrogen and the alkalinity was insufficient for complete nitrification. An increased nitrite accumulation was observed with increase in alkalinity to $NH_4{^+}-N$ concentration ratio. The increase in alkalinity to $NH_4{^+}-N$ concentration ratio has been a maior reason for the high pH value and FA concentration in the reactor. It can be considered that selective inhibition of Nitrobacter can be causes of nitrite accumulation. The nitrite accumulation increased with increment of temperature at fixed alkalinity to $NH_4{^+}-N$ concentration ratio.

  • Hydrogen($H_2$) as a clean, and renewable energy carrier will be served an important role in the future energy economy. Several biological $H_2$ production processes are known and currently under development, ranging from direct bio-photolysis of water by green algae, indirect bio-photolysis by cyanobacteria including the separated two stage photolysis using the combination of green algae and photosynthetic microorganisms or green algae alone, dark anaerobic fermentation by fermentative bacteria, photo-fermentation by purple bacteria, and water gas shift reaction by photosynthetic or fermentative bacteria. In this paper, biological $H_2$ production processes, that are being explored in fundamental and applied research, are reviewed.

Nitrogen Removal in Fluidized Bed and Hybrid Reactor using Porous Media (다공성 담체를 이용한 유동상 및 하이브리드 반응기에서의 질소제거)

  • A fluidized bed reactor containing porous media has been known to be effective for nitrogen and organic matters removal in wastewater. The porous media which attached microbes plays important roles in simultaneous nitrification/denitrification (SND) due to coexistence of oxic, anaerobic and anoxic zone. For SND reaction, oxygen and organic substrates should be effectively diffused from wastewater into the intra-carrier zone. However, the overgrowth heterotrophic microbes at the surface of porous media may restrict from substrates diffusion. From these viewpoints, the existence and effect of heterotrophic bacteria at surface of porous media might be the key point for nitrogen removal. A porous media-membrane hybrid process was found to have improved nitrogen removal efficiency, due to stimulated denitrification as well as nitrification. Microelectrode studies revealed that although intra-media denitrification rate in a conventional fluidized bed was limited by organic carbon, this limitation was reduced in the hybrid process, resulting in the increased denitrification rate from 0.5 to $4.2\; mgNO_3-N/L/hr$.

Continuous Hydrogen Gas production by Immobilized Anaerobic Microorganisms (고정화 혐기성 미생물에 의한 연속적인 수소 생산)

  • Hydrogen producing acidogenic microorganisms were self-immobilized using organic-inorganic hybrid polymer within 5 minutes. During the continuous tratment of synthetic wastewater at a hydraulic retention time of 20 hours, at 37$^{\circ}C$, pH 5.0, the self-immobillized granules were maintained in a stirred tank reactor. The black colored granules gradually became milky. Image analysis showed that the mean diameter of the milky colored granules ranged from 1.5 to 20. mm. The maximum bio-gas procuction rate was 380 ml/L/hy and the concentration of H$_2$was around 50%, while no methane was detected. Granular ECP was extracted and its content was measured to elucidate the role of the organic-inorganic hybrid polymer. Further increases of granule concentration are expected to increase the hydrogen production rate.

Anaerobic Digestion Efficiency of Remainder from Bacterial Cellulose Production Process using Food Wastes (음식 폐기물을 이용한 박테리아 셀룰로오스 생산 공정 잔류물의 혐기성 소화효율)

  • This study was performed to examine the availability of anaerobic digestion of the remainders caused by bacterial cellulose production process using food wastes. They maybe to be considered as others second pollution sources. Thus, this study was targeted to minimize content of organic material and to obtain more energy in those remnants using two-phase UASB reactor. The working volume of first hydrolysis fermentor was 35 L (total 55 L) and the second methane fermentor was 40 L (total 50 L). The organic loading rate of hydrolysis fermentor was 3 g-VS/L${\cdot}$day and 25,000 ppm of $COD_{cr}$ for methane fermentor. The hydraulic retention time was 18 days for hydrolysis reactor and 33 days for methane reactor. The hydrolysis reactor and methane reactor were performed at 35, 40$^{\circ}C$ respectively. For the efficient stable performance, the composition of organic wastes at each stage was as follow; Food waste with bacterial culture remnants (1 : 1), bacterial cellulose remnants, bacterial cellulose culture remnants with food wastes saccharified solids (1 : 1). When the anaerobic digestion was performed stably at each stage, the COD removal efficiency was 88, 90, 91 % respectively. At this time, methane production rate was 0.26, 0.34, $0.32m^3\;CH_4/kg-COD_{remove}$. As well as the values of anaerobic digestion at third stage were more higher than values of anaerobic digestion using food wastes. It is clearly to say that the food wastes zero-emission system constructed in our lab is more efficient way to treat and reclaim food wastes.

Production of Methane from Anaerobic Fermentation of Marine Macro-algae (해조류의 혐기성 발효를 이용한 메탄 생산)

  • Methane was produced from the anaerobic digestion of marine macro-algae. Elemental analysis was first performed to estimate the theoretical methane production of three macro-algae (Undaria pinnatifida, Laminaria japonica, Hizikia fusiformis). Three algae were found to contain C 34 ~ 36%, H 5%, O 37 ~ 43%, N 2 ~ 4%, S 0.4 ~ 0.7%, and ash 14~21%, and the theoretical methane content was in the range of 56 ~ 60%, which can produce 442 ~ 568 mL $CH_4$ per g of volatile solid (VS). Using the biological methane potential (BMP) test, we found that L. japonica resulted in the highest yield of methane (52%). Moreover, various operational conditions, such as algae amount, pH, salinity, particle size, and pre-treatment, were investigated in order to find an optimal condition of anaerobic digestion. At pH 8.0, the autoclaved L. japonica (5g VS/200 mL), when used without washing salt, produced 268.5 mL/g VS which is 65% of the theoretical methane productions. Furthermore, using a CSTR (with the working volume of 7 L out of the total volume of 10 L), we have successfully operated the reactor for 65 days and obtained maximum methane production rate of 1.4 L/day with purity of 70%.

Bioconversion of D,L-ATC to L-cysteine Using Whole Cells (D,L-ATC의 L-cysteine으로의 생물학적 전환반응에서의 균체이용 기술)

  • In the conversion of D.L-2-amino-$\Delta^2$-thiazoline-4-carboxylic acid(D,L-ATC) to L-cysteine using Pseudomonas sp. CU6. the effects of surfactants on whole cells and the stabilities of cellfree enzyme solution and continuous reactor packed with immobilized whole cells were investigated. The enzymatic reaction was little accomplished by whole cells without adding surfactants, whereas it was well carried out with SDS or Triton X-loo comparable to the case using cell-free enzyme solution. Enzyme activity of the cell-free solution was lost by 50% after 7 hours of storage at $30^{\circ}C$, but not at all under an anaerobic condition by sparging nitrogen gas. On the other hand. effect of nitrogen gas did not appear in a continuous reactor using immobilized whole cells, and hydroxylamine, an inhibitor of L-cysteine desulfhydrase, lowered the enzyme stability.

  • In this study, Anaerobic sewage sludge in a batch reactor operating at $35^{\circ}C$ was used as the seed to investigate the effect of pretreatments of waste activated sludge and to evaluate its hydrogen production potential by anaerobic fermentation. Various pretreatments including physical, chemical and biological means were conducted to utilize for substrate. As a result, SCODcr of alkali and mechanical treatment was 15 and 12 times enhanced, compared with a supernatant of activated sludge. And SCODcr was 2 time increase after re-treatment with biological hydrolysis. Those were shown that sequential hybridized treatment of sludge by chemical & biological methods is most efficient process for sludge treatment. The pre-treatment activated sludge was tested to conform hydrogen production potential in batch experiments. When buffer solution was added to the activated sludge, hydrogen production potential increased as compare with no addition.

A Study on the Biogas Production and VSS Concentration in Organic Wastewater Treatment Using a Downflow Anaerobic Packed Bed Reactor with HRT Change (하향유식 혐기성 고정상 생물반응기에서 유기성 폐수의 HRT변화에 따른 Biogas 생성 및 VSS 농도에 관한 연구)

  • Characteristics of a downflow anaerobic packed bed reactor with raschig ring ceramics as a packing were measured and discussed for the basic evaluation of the process. A synthesized glucose substrate wastewater were used as a feed and process characteristics such as pH, biogas production, composition of produced gas, COD removal and VSS were measured with the hydraulic retention time (HRT) changing from 0.25 to 2 days. As a result, this type of reactor was applicable in continuous operation within the given HRT range and the transient period approaching the steady state was about 20 days. The content of methane in produced gas increase with HRT was always high above 50% enough to use as energy source. The COD removal efficiency increased gradually as HRT increased. The axial profile of VSS concentration in the reactor usually showed the maximum at the lower region and the minimum at the middle. The VSS concentration at the upper region and the exit appeared similarly. However, at 0.25 day of HRT, the VSS concentration of effluent became higher than that of the upper region. Therefore the optimum HRT of this reactor occurred about 0.5 day, at which the production of methane began to be just stabilized and loss of VSS and COD removal were resonable.

  • The lab-scale anaerobic continuous reactor which was filled with the sludge of anaerobic digestion from Suyoung wastewater treatment plant was operated by feeding of various concentrations and flow rates. This experiment indicated that more than 6,870 mgCOD/L of substrate concentration was required to promote good metabolism and growth of anaerobic biomass. And increasing loading rate slowly was also required in order to treat substrate of higher concentration and higher loading rate. The substrate concentration of about 10,000 mgCOD/L was adequate to generate biogas efficiently. The pH was sharply decreased at the onset of higher leading rate, but the pH was restabilized soon at 8. During the experiment, the amount of the attached biomass was kept constant.
