A field study on early stabilization of waste landfill using air injection and leachate recirculation (공기주입과 침출수 재순환 방법을 이용한 폐기물 매립지 조기안정화에 관한 현장 실험 연구)

  • Field study was conducted for 5 months to investigate the effect of leachate recirculation on aerobic landfill stabilization at active landfilling site. The area of field experiment was $24{\times}24m$ and 9 vertical air injection wells with screen ranging 3~9 m were installed. Aerobic landfill operation for 5 months increased average internal landfill temperature to $70^{\circ}C$ and 8 % of landfill height was settled down. $94m^3$ of leachate was recirculated for 1 month to increase moisture content of landfill to favor microbial degradation of organic matter, which resulted in temporary increase of groundwater level and anaerobic environment. But leachate recirculation triggered increase of internal landfill temperature of neighboring monitoring well. Because excessive leachate recirculation decreased internal landfill temperature by cooling effect, internal landfill temperature should be checked to avoid abrupt decrease of temperature during leachate recirculation. Also, to prevent anaerobic environment, intermittent leachate recirculation was recommended.

  • Clostridium is a genus of Gram-positive, rod shape, spore-forming obligate anaerobe. Recently, Clostridium has been attracted as a host for bio-based chemical production, due to its diversity of substrate utilization and the production ability for metabolites which can be used as a building block for chemical production. Especially, butanol produced from Clostridium has been considered as an alternative fuel. As a transportation fuel, butanol has a higher energy density and lower hygroscopicity compared to ethanol, the first generation biofuel. Recently, metabolic engineering of Clostridium has been massively conducted for butanol production. In this study, the metabolic engineering strategy of Clostridium for butanol production has been reviewed with a brief perspective.

Preservation of Kimchi by ${\gamma}-Ray$ Irradiation (감마선 조사에 의한 김치저장에 관한 연구)

  • To improve the storage method for kimchi, optimal ripening kimchi was irradiated with doses of 1, 3, 5kGy Co-60gamma radiation, followed by the microbiological, physicochemical and senosory evaluations during storage at $5^{\circ}C$. 1. Total aerobic count increased in the beginning of storage and then decreased slowly as the number of total lactobacilli (anaerobe) increased. The above, total aerobic and lactobacilli were reduced by 1 to 3 log cycles with irradiation and at the 90th day after storage the number of total lactobacilli remained $1.30{\times}10^{8}$ per ml in 3 kGy irradiated group. Irradiation treatment at 3 kGy sterilized coliforms and molds contaminating the sample as the level of $2.0{\times}10^{4}$ per ml and $5.4{\times}10^{2}$ per ml respectively and no apparent growth was observed in both control and 1 kGy irradiated groups after 20 days of storage. The population of yeast, $3.5{\times}10^{3}$ per ml initially, increased steadily during kimchi storage and at 90 days of storage the number was shown to be $5.6{\times}10^{4}$ per ml and $6.5{\times}10^{2}$ per ml in control and 3 kGy irradiated groups, respectively. 2. In the physicochemical changes during kimchi storage, pH, acidity and volatile acid of non-irradiated control at the 45th day after storage were 4.0, 0.7% and 0.066%, while those of 3 kGy irradiated group were 4.2, 0.59 and 0.06% at the 90th day of storage, respectively. The reducing sugar content of all stored samples changed inversely total acidity content, indicating irradiation delayed the changes of them. The amount of ascorbic acid decreased gradually with the storage time and irradiation dose increase. Textural parameters of 3 kGy irradiated group were superior to those of other groups at the latter stage of storage. 3. Sensory evaluations showed that 3 kGy irradiation was the optimum dose level to extend the shelf-life of kimchi more than two months as compared to control.

Microflora and enzyme activity of conventional Meju, and isolation of useful mould (재래식 메주의 미생물군, 효소역가 및 유용 균주의 분리)

  • To obtain useful mould strain in soybean fermentation industry, we collected conventional Meju all over the Korea and tested existance of aflatoxin in Meju collected. Also, we measured distribution of microorganism and enzyme activity of Meju and isolated Aspergillus oryzae O4-5 as a industrially useful mould. The results obtained were summarized as follows: 1. Aflatoxin was not detected by EZ-Screen Test Kit and HPLC analysis in various Meju sample collected all over the Korea from 1994.12 to 1995.2. 2. Colony forming units(CFU) of mould, yeasts, aerobe, and anaerobe were $1.3{\times}10^4-2.8{\times}10^6$, $1{\times}10^2-1.5{\times}0^6$, $2.0{\times}10^7-8.0{\times}10^8$ and $3.0{\times}10^6-7.3{\times}10^8$ per gram of Meju, respectively, indicating that CFU inter Meju samples were varied with big difference. 3. The activities of ${\alpha}$-amylase and gluco-amylase were 5-80 units and 2-34 units per g of Meju, respectively. It was shown that enzyme activity was varied depending on where the Meju was collected. 4. The activities of acidic, neutral and alkaline protease were 5-33 units, 5-302 units and 5-363 units per g of Meju, respectively. Acidic protease of Improved Meju made by D-Company was higher than that of conventional Meju as 66 units per g of Meju. 5. CNU O4-5 strain selected as a noble strain could produce amylase and protease in high level, and identified as a strain that belongs to Aspergillus oryzae. 6. The activities of acidic and neutral protease of Aspergillus oryzae CNU O4-5 strain isolated were about 10% higher than those of Aspergillus oryzae JM which has been used in soybean fermentation industry. Amylase activity of CNU O4-5 strain was similar to Aspergillus oryzae JM.

Effect of Chitosan on Microbial Community in Soils Planted with Cucumber under Protected Cultivation (오이 시설재배에서 키토산 처리가 토양 미생물상에 미치는 효과)

  • Soil microbial community and soil physiological parameters were investigated by analyzing phospholipid fatty acids extracted from the soils amended with chitosan powder and solution in a cucumber greenhouse. The soils were sampled at 90, 160, 200 days after treatment. Identified fatty acids were analyzed with principal component (PC) analysis. Chitosan powder soils and chitosan solution soils were separated from non-treated control soils by PC1 and PC2 90 days after treatment, respectively. And chitosan powder soils were separated from non-treated control soils by PC2 160 days after treatment. The ratio of fungi to bacteria increased significantly in chitosan solution-amended soils compared with the control soils 90 days after treatment. Microbial groups and physiological parameters were investigated 160 days after treatment: vesicular-arbuscular mycorrhizal fungi (VAM) significantly increased in soils amended with chitosan powder compared with other soils, the ratio of gram negative bacteria to gram positive bacteria and cyclo-fatty acids to precursors were significantly higher and lower in soils amended with chitosan solution and chitosan powder compared with control soils, respectively, and the ratio of fungi to bacteria were significantly lower in control soils compared with chitosan-treated soils. The chitosan powder increased the ratio of aerobic to anaerobic bacteria and lowered the ratio of saturated to unsaturated fatty acids compared with chitosan solution 200 days after soil application. In conclusion, chitosan powder changed the soil microbial community and the effects maintained up to 160 days after soil application. The effect of physiological parameters on the soil microbial community started to appear 160 days after and continued up to 200 days after soil application of chitosan.

Optimum Management Plan of Swine Wastewater Treatment Plant for the Removal of High-concentration Nitrogen (고농도 질소제거를 위한 축산폐수 처리시설 적정관리 방안)

  • The amount of swine wastewater reaches about $197,000m^3$ per day at live-stock houses in the whole country. A half of the swine wastewater resources are too small to be restricted legally. This untreated wastewater causes the eutrophication in the water bodies. In case of swine wastewater treatment, the solid-liquid separation must be performed because feces(solid phase) and urine(liquid phase) have large differences in nitrogen and phosphorus concentration. It is necessary to assess exactly the concentration of the pollutants in swine wastewater for planning the wastewater treatment facilities. A full-scale operation was carried out in K city and the plant is consists of conventional plant, the supplementary flocculation basin of chemical treatment process and $anaerobic{\cdot}aerobic$ basin for nitrogen removal. The improved full-scale swine wastewater treatment plant removed the $1,500{\sim}3,000mg/l$ of total-nitrogen(T-N) to 120mg/l of T-N and $131{\sim}156mg/l$ of total-phosphorus(T-P) to $0.15{\sim}1.00mg/l$ of T-N. Accordingly, as a results of operational improvement, the removal efficiencies of T-N and T-P were over $92{\sim}96%$, 99%, respectively. The continuous supply of organic carbon sources and the state of pH played important roles for the harmonious metabolism in anaerobic basin and the pH value of anaerobic basin maintained at about 9.0 for the period of the study.

  • The traditional ripening method of clay was analyzed. An advanced refining method of clay using enrichment cultures of iron r reducing bacteria was developed. After the traditional ripening, the whiteness of the clay was increased due to removal of | iron impurities by inhabitant dissilmaltien with iron reducing bacteria. Other characteristics of the refined clay such as v viscosity, plasticity, and strength were also improved by iron reducing bacteria. An advanced method of clay refinement with a anaerobic enrichment cultivation of iron reducing bacteria supplemented with an extra carbon source such as glucose was s suggested. When the clay was treated by the advanced method. the refinement time could be reduced to 1/6 of that r required by the traditional method. The physical properties of the refined clay by the advanced method were better than t those of the traditionally refined clay.

  • The purpose of the study is to collect basis data for to establish standard administrative processes of liquid fertilizer treatment. From this survey we could make out the key point of each step through a case of effective liquid manure treatment system in pig house. It is divided into six step; 1. piggery slurry management step, 2. Solid-liquid separation step, 3. liquid fertilizer treatment (aeration) step, 4. liquid fertilizer treatment (microorganism, recirculation and internal return) step, 5. liquid fertilizer treatment (completion) step, 6. land application step. From now on, standardization process of liquid manure treatment technologies need to be develop based on the six steps process.

Effects of Fermented Diet Using Probiotics from Pine Needle Microbes on Growth Performance, Blood Characteristics, Carcass Traits and Economy of Pigs (솔잎 토착미생물을 이용한 발효사료 급여가 돼지의 성장, 혈액성상, 도체형질 및 경제성에 미치는 영향)

  • This study was conducted to investigate the effect of probiotic from pine needle microbes on growth performance, blood characteristics, carcass traits and economy of pigs. One hundred eighty pigs (L${\times}$Y${\times}$D, average body weight=75 kg) were grouped and housed with 15 animals per pen for 3 replications per each treatment. The ingredients and pine needle microbes mixed and fermented on anaerobic condition for 10 days to make the fermented diet. The basal diet was substituted with 0 (CON), 3 (T1), 5 (T2) and 10% (T3) of fermented diet, the pigs were fed for 42 days and slaughtered at the end of the trial to measure the carcass traits. Crude protein concentration of fermented diet on 10 day was higher (p<0.05) than that of on 0 day, while crude fiber concentration and pH were lower (p<0.05). The supplementation of fermented diet did not affect on growth performance and feed intake. Hemoglobin concentration of whole blood in T3 was higher (p<0.05) than that of CON, and hematocrit concentrations in CON and T1 were higher (p<0.05) than that of T2 and T3. The concentration of platelet was higher (p<0.05) in T1 than in T2 and T3. The CON had higher (p<0.05) total cholesterol, LDL-cholesterol and triglycerides concentrations than the treatments, whereas lower (p<0.05) HDL-cholesterol concentration. Carcass weight and dressing in T1 and T3 were higher (p<0.05) than those of CON and T2. The backfat thickness was higher (p<0.05) in CON and T2 than in T3. The carcass grade and economy improved by supplementation of fermented diet compare to CON. In conclusion, the supplementation of fermented diet using probiotics from pine needle microbes could improve health and meat quality and reduce the feed cost of pigs.

Effects of Yeast Addition as Starter on Fermentation of Kimchi (효모 Starter 의 첨가가 김치발효에 미치는 영향)

  • The purpose of this study is to investigate the effects of yeast addition as starter on kimchi fermentation. The strains used as starter were Saccharomyces sp. YK-17 and Saccharomyces fermentati YK-19 isolated from kimchi, grew under anaerobic condition and low temperature, which showed the acid and base resistances. Chemical and microfloral changes, as well as the sensory properties of starter added kimchi during fermentation were compared with the control fermented without starter. The acidity of kimchi juice was lower and pH was higher in starter added kimchi than the control. Particularly addition of S. fermentati YK-19 prolonged the optimally fermented period (pH 4.0, acidity $0.6{\sim}0.8%$) up to more than 63%. The content of lactic acid, the major non-volatile organic acid in kimchi, was increased rapidly followed by S. sp. YK-17 and S. fermentati YK-19 group. The microfloral changes were found a little different among the samples. Among the microorganisms, Leuconostoc sp. and Lactobacillus sp. showed highest change, and Streptococcus sp. and Pediococcus sp. showed ralatively low change. The growth of Lactobacillus sp. which was the main acidifing microorganism was inhibited by starter addition, particularly by S. fermentati YK-19. The sensory characteristics of acidic and moldy flavor were significantly reduced by the addition, while fresh flavor was increased in starter added group.
