The role of matrix modifier and for the determination of cadmium in blood by graphite furnace atomic absorption spectrometry (흑연로 원자흡수분광법에 의한 혈중 카드뮴 분석시 매트릭스 개선제(matrix modifier)의 역할)

  • The low concentration of cadmium in the whole-blood was determined by graphite furnace atomic absorption spectrometry(GFAAS) after the sample was diluted five-fold by 1% Triton X-100, 2% $(NH_4)_2HPO_4$ as matrix modifier and pyrocoated graphite tube with L'vov platform was tried remove the interferences of blood matrix and reduce the loss of volatility of cadmium at higher ashing temperature($600^{\circ}C$). The criteria for evaluating the accuracy and precision of this analysis was confirmed by analysis of interlaboratory comparison(Japan) and NIST SRM No. 909(Cd in Serum). The limit of the determination for cadium was 0.1ng/ml and the relative standard deviation(RSD) at 1.0ng/ml level was about 10% for the GFAAS.

The Effect of Ecklonia cava Extracts on Bone Turnover Markers in Ovariectomized Rats (갱년기 유도 흰쥐에서 감태 추출물이 골 대사 지표물질의 변화에 미치는 영향)

  • Menopause is often associated with the incidence of several chronic diseases including osteoporosis, cardiovascular disease, and obesity. Hormone replacement therapy (HRT) is an effective regimen that has been found to prevent these diseases in postmenopausal women. However, HRT is accompanied by an increased risk of unfavorable outcomes. Therefore, this study was conducted to evaluate the effects of Ecklonia cava, a kind of seaweed, extract on bone turnover markers in symptomatic menopausal women. For this study, the following four groups of 9-week-old Sprague-Dawley rats were evaluated over 6 weeks: normal rats (SHAM), ovariectomized rats (OVX-CON) and ovariectomized rats that were treated with Ecklonia cava extracts. The optimum extraction temperature and solvent of Ecklonia cava were found to be $80^{\circ}C$ and 80% ethanol. We measured the osteocalcin and CTx content, enzyme ALP activity in serum and collagen content in the cartilage, bone, skin and lungs. We found that the levels of indicators of bone metabolism such as ALP, osteocalcin and CTx were lower in rats in the Ecklonia cava extract group than the OVX-CON group. In addition, the collagen contents in the bone, cartilage, skin and lungs decreased in response to ovariectomy, but the levels of collagen were greater in the bone of rats that were treated with Ecklonia cava extract than in the bone of rats in the OVX-CON group. According to these results, we were able to know the effects of Ecklonia cava extract on bone aging in ovariectomized rats. Consequently, we expect Ecklonia cava extract to have an effect on bone aging in postmenopausal women.

  • 목적 : Elastin 섬유는 피부, 폐, 동맥, 정맥 등 조직에서 발견된다. Elastin 섬유의 손실은 대동맥, 동맥, 세동맥의 탄력 등과 연관되어 폐기종, 혈액순환 부전 및 폐동맥 고혈압을 악화시킬 수 있다. 본 연구는 당귀약침액의 elastase 효소활성에 대한 효과와 항산화 효과에 대하여 연구하고자 계획되었다. 방법 : 당귀약침액의 elastase 억제효과와 DPPH, 및 NO 소거능을 측정하여 항산화 효과를 측정하였다. 결과 : 당귀약침액에서 통계적으로 유의한 elastase 활성 억제효과를 관찰할 수 있었다. 또한 통계적으로 강한 DPPH free radical 소거 효과가 관찰되었다. 그리고 NO 소거효과를 관찰할 수 있었으며, 그 효능은 pH 6.0에서 가장 강한 효능을 보였다. 결론 : 본 연구 결과에 의하면, 당귀약침액에는 유효한 elastase 활성 억제효과와 강한 항산화 효과가 있어, 폐기종 및 폐동맥 고혈압 치료에 유효한 작용이 있을 것으로 사료된다.

  • 골수나 유산물질에 대한 세포유전학적 검사에 있어 통상적인 염색체검사는 검사에 적합한 중기핵상을 얻기 어려워 실패하는 경우가 많다. 이러한 경우에 진단이나 치료에 도움을 줄 수 있는 방법으로 유식세포분리기를 사용하여 단일 세포내 DNA량에 따른 aneuploidy를 추적할 수 있는 가를 확인하기 위해 본 실험을 실시하였다. 79 (혈액 30, 골수 37, 유산물 12)예에서 염색체 검사와 유식세포 분리검사를 동시에 실시하여 각각의 결과를 비교한 결과 79.7% (63/79)의 일치율을 얻었다. 그러나 염색체의 손실이 없는 전좌와 역위의 경우는 물론 작은 조각의 염색체 부분이 늘어나거나 줄어든 경우에 있어서는 유식세포분리방법에 의해서 추적되지 못하였지만, 염색체 검사의 결과를 얻는데 실패한 경우에는 유식세포분리방법이 DNA량의 변화에 대한 정보를 얻을 수 있다는 것을 확인할 수 있었다. 따라서 본 연구결과는 세포유전학적 검사에서 유식세포분리방법이 염색체 검사보다 신속하며 염색체검사가 불가능한 시료에서도 DNA양에 따른 aneuploidy의 추적이 가능하다는 것을 시사한다.

  • 구제역은 소, 돼지 등 발굽이 두 개로 갈라진 가축들에게 감염을 유발하는 전염성이 매우 높은 바이러스성 질병이다. 구제역 감염 시 입 주변, 구강 내, 코, 발굽사이 등에 수포가 생기며 고열과 식욕이 저하되어 심하게 앓거나 죽게 되는데, 강한 감염 전파력을 가졌음에도 치료제가 없고, 감염확인 즉시 확산 방지를 위한 살 처분만이 이루어지고 있다. 따라서 무엇보다도 빠른 감염여부 진단이 중요하다. 현재까지 구제역을 진단하는 방법으로는 감염 된 가축의 혈액에서 구제역 항원 단백질에 대한 항체형성 여부를 확인하는 항체진단법과 수포액과 같은 체액을 채취하여 세포배양을 통한 구제역 바이러스 분리방법이 있지만 두 가지 모두 짧은 잠복기를 갖는 구제역 바이러스를 빠른 시간 내 진단하기는 어렵다. 따라서 본 연구에서는 보다 빠른 구제역 진단 키트개발을 위해 NCBI Pubmed를 이용하여 구제역바이러스가 가지는 6개의 주요 단백질을 확인하였고, NCBI BLAST를 이용하여 6개의 단백질 중 구제역 바이러스에 특이적인 항원 단백질 peptidase C28을 선정하였다. 선정된 단백질의 아미노산 서열을 이용하여 IEDB analysis resource를 통해 peptidase C28의 epitope 부위를 예측하였다. 예측 된 부위의 아미노산 서열을 NCBI BLAST에서 정상 동물과 비교하여 구제역바이러스 특이 항원 단백질 epitope peptide를 최종 선정하였다. 이를 이용한 구제역 바이러스 진단키트 제작은 보다 빠른 진단을 통해 감염 확산을 조기에 차단하고 경제적 손실과 피해를 최소화 할 수 있다.

  • A novel sensitive and non-invasive ring-type glucose probe was designed and measured. The magnitude and phase of return loss change with different concentration of the glucose solution inside the loop structure. Ring type probe is more advantageous than open ended probe to measure electrical characteristics under the skin. The maximum difference of return loss was 0.94 for the concentration difference of 0% and 20% at 285MHz and more sensitive in low frequency.

Efficacy of Tranexamic Acid during Primary Total Knee Arthroplasty: Comparative Study between Intravenous Use and Topical Use (일차 슬관절 전치환술 중 트라넥삼산의 정맥 내 투여와 국소 사용의 효과에 관한 비교 연구)

  • Purpose: This study examined the effectiveness of tranexamic acid in reducing postoperative blood loss in total knee arthroplasty by comparing the methods of administration between an intravenous group, topical group, and non-tranexamic acid group. Materials and Methods: This was a retrospective case series study of patients who underwent primary total knee arthroplasty from March 2017 to February 2019 performed by a single surgeon. The study population was divided into three groups according to the method of tranexamic acid administration (Group I: intravenous group, Group II: topical group, Group III: non-tranexamic acid group). To evaluate the effectiveness of tranexamic acids, the total amount of postoperative blood loss, postoperative hemoglobin loss, and volume of red blood cell transfusion in the three groups were compared. Results: The total amount of postoperative blood loss was lower in the tranexamic acid administered group than in the non-tranexamic acid group (1,366±866 ml). Among the administration methods, the intravenous group (987±449 ml) was significantly lower than the topical group (1,136±339 ml) (p=0.004). Postoperative hemoglobin loss was lower in the tranexamic acid group than the non-tranexamic acid group. Among the administration methods, the intravenous group was lower than the topical group. The transfusion rate was higher in the non-tranexamic acid group (5.7%) than the tranexamic administered group. The transfusion rate of the intravenous group was 1.4%, and no patient required a transfusion postoperatively in the topical group. The number of postoperative thromboembolic events, as a complication of tranexamic acid, was similar in the three groups. Conclusion: Tranexamic acid was effective in reducing postoperative blood loss after primary total knee arthroplasty compared to the non-tranexamic acid administered group. No significant difference in the complications induced by tranexamic acid was observed among the three groups.

Application of Oral Fluid Sample to Monitor Porcine circovirus-2 Infection in Pig Farms (구강액을 이용한 양돈장의 Porcine circovirus-2 감염에 대한 모니터링)

  • Porcine circovirus-2 (PCV2) has been implicated in many clinical diseases/syndromes that are now referred to as PCV-associated diseases (PCVAD). Due to significant economic losses caused by PCVAD, many swine operations have launched extensive monitoring programs for PCV2. Traditional serum sampling is, however, rather expensive and laborious, hampering effective large scale pathogen surveillance. A field-based longitudinal study was conducted to assess the utility of pen-based oral fluid sample as an alternative to serum for herd PCV2 testing. Six pens (25 pigs/pen) at each of 3 different sites were used in the study. One oral fluid and 5 random serum samples per pen were collected at 3, 5, 8, 12, and 16 weeks of age, and the sera were pooled by pen for testing. All samples were tested for PCV2 by real-time PCR and for antibodies by indirect fluorescent antibody test (for both anti-PCV2 IgG and IgA) and 3 ELISA assays (blocking ELISA, indirect ELISA, and IgG/IgM sandwich ELISA). PCV2 DNA was detected in oral fluid samples sporadically until 8 weeks and in all pens at 16 weeks. PCV2-specific IgG was detected in oral fluid samples at 3 weeks and persisted until 5 to 8 weeks in all sites. Anti-PCV2 IgG and IgA were detectable in oral fluid samples collected at 16 weeks from all of the pens at 1 site. The detection of PCV2 and anti-PCV2 antibody in oral fluid samples correlated positively with results on pooled sera, suggesting that oral fluids can be a cost-effective alternative to serum for herd monitoring of PCV2 infection.

Effects of Nicarbazin and Hot Temperature on Water Balance, Acid-Base Balance, and Body Temperature in Adult Roosters (고온하의 수탉 성계에계 Nicarbazin의 급여가 수분 출납, 혈액의 산-염기 평형 및 체온에 미치는 영향)

  • An experiment was conducted to study the effects of nicarbazin supplementation (125 ppm) on the water balance, blood acid-base balance, and rectal temperature of 16 SCWL adult roosters in normal($21^{\circ}C$) and hot(35~$36^{\circ}C$) temperatures. There were 4 d of preliminary period, followed by 2 d of sampling and data collection period for each temperature treatment. The amounts of DM intake and excretion were not affected by heat stress but by rucarbazln supplementation. The DM metabolizability of diets, however, was decreased by nicarbazin. The amounts of water intake and evaporative water loss were increased in heat stressed birds. Nicarbazin increased further the amounts of water intake and evaporative water loss in hot temperature. Nicarbazin also decreased the blood p$CO_2$ and increased blood pH of heat stressed birds. The rectal temperature of the bird was increased in hot temperature, and the nicarbazin supplementation worsened this effect. Results indicate that heat stress raises the body temperature of the chicken. In order to suppress the rise of body temperature, the birds enhance the evaporative water loss through panting. The nicarbazin supplementation to heat stressed birds, however, increases the body temperature to such an extent that panting alone may not be able to return the body temperature back to normal.

Usefulness of Clinical Scoring System in the Diagnosis of Group A Streptococcal Pharyngitis (인두염 환아에서 A군 연구균 검출의 임상적 점수제의 유용성)

  • Purpose : Proper diagnosis of group A streptococcal pharyngitis that may cause chronic diseases in childhood is not easy because its signs and symptoms would be nonspecific. Because results of classical throat culture delays for one to two days, we'd like to determine whether early antibiotics would be introduced with according to the clinical score system. This study was undertaken to evaluate of clinical usefulness of scoring system based on the clinical and laboratory findings. Methods : From Jan. 1998 to Dec. 2000, 10 clinical items based on modified 9 items by Breese in 1977 were checked in patients with pharyngitis who visited on outpatients clinic of pediatrics, Kyunghee University Hospital. We compared the results of throat culture with the points of clinical score system. Results : Out of 45 cases, the positive culture for Group A Streptococcus was 20 and negative culture was 25. When we applied more than 30 points of score, which correspond to 70 percentile of study population, the sensitivity and specificity were 35.0% and 96.0%, respectively. Conclusion : Although sensitivity was relatively low this scoring system, but the high specificity may be useful diagnostic tool in the areas where the rate of isolation of Group A Streptococcus is low.
