lnhibitory Effect o fVarious Cruciferous Vegetable on the Growth of Human Cancer Calls (인체암세포증식에 있어 십자화과 채소의 억제효과)

  • The anticarcingenic effect of methanol extracts from such cruciferous vegetables as cabbage, red cabbage, Korean cabbage, kale, cauliflower, broccoli, radish root, leafy radish, rape leaves and shepherd’s purse on the growth of human K-562 leukemia cells, MG-63 osteosarcoma cells, HT-29 colon cancer cells and AGS gastric cancer cells were studied. All of cruciferous vegetables inhibited more than 70% of the growth of K-52 leukemia cells and more than50% fo rhe growth fo AGS gastric cancer cells. Particularly, kale, broccoli and shepherd’s purse showed inhibition rates of 93.5%, 93,5% and 96.3% on the growth of AGS gastric cancer cells, respectively. In case of HT-29 colon cancer cells, the methanol extracts of cabbage, kale and shepherd’purse exhibited 82.4%, 72.15, 79.4% and 95.6% of inhibitory effects, respectively. The cabbage, kale, cauliflower and shepherd’s purse extracts also highly suppressed the proliferation of MG-63 cells. Generally the 10 cruciferous vegetable we studied strongly decreased the growth of various human cancer cells in vitro, however, kale and shepherd’s showed the most effective vegetable among them.

  • 주치의에게는 암환자의 암 전이 여부가 초미의 관심사다. 암환자의 10명 중 9명이 전이암으로 사망하기 때문이다. 암의 전이 초기에 그 전이 여부를 방사선 진단법으로 가능하지 않다. 혈액을 채취하여 암의 전이 유무를 진단 하는 기술이 개발되고 있다. 이 혈중 암세포는 혈구세포 10억개당 1~100개 정도의 극히 미량이 존재하여 암세포 분리기술이 특별히 잘 개발되어야 한다. 최근 마이크로바이오칩 형태의 분리기술이 큰 기술적 진화를 보이고 있어 본고에 소개하고자 한다. 이 기술은 한 가지 큰 의미를 갖는다. 그것은 암환자의 암 전이 모니터링에 필요한 도구가 될 수 있기 때문이다. 전이암세포 검출 키트로 전이암세포를 계수 하여 환자에게 투약한 항암제가 적합한지에 대한 답을 의사는 얻을 수 있다. 전이암세포 진단용 마이크로바이오칩 기술이 기존의 영상진단법만큼 중요한 임상 수단이 될 것으로 전망된다.

Laser Captured Microdissection

  • 대부분의 조직은 여러 가지 세포가 모여서 이루어지기 때문에 그 중의 어떤 특정세포에서 발현하는 물질을 분석하려면 조직을 이루고 있는 각각의 세포를 분리해내야 한다. 이렇게 순수하게 세포를 분리해내는 기술 중의 하나가 Laser Captured Microdissection (LCM)이 다. LCM의 개발로 기존에 사용되던 방법에 비하여 빠르고 간편하면서, 매우 정확하게 원하는 세포를 순수 분리해서 그 세포의 분자생물학적 또는 생화학적인 분석을 할 수 있게 되었다. LCM은 현미경으로 조직절편을 관찰하면서 원하는 세포를 낮은 에너지의 laser를 사용하여 도려내는 방법으로 조직절편 이외에도 도말된 혈액이나 자궁경부 조직, 그리고 배양된 세포를 cytocentrifugation한 후에 원하는 세포를 포획 할 수도 있다. LCM을 이용한 연구는 여러 분야에서 다양하게 진행되고 있으며, 특히 같은 조직 내에 존재하는 정상세포와 전이중인 세포, 그리고 암세포를 구분해 냄으로써 암의 전이기전 및 병인 연구에 매우 큰 공헌을 하고 있다. 이렇게 분리된 세포는 RT-PCR, LOH (loss of heterozygosity), microsatellite instability, differential gene profiling, cDNA microarray, Western blot, 2D PAGE protein analysis 등의 기법을 접목하여 연구하게 된다. 본 논단을 통하여 1996년 개발된 LCM의 원리와 이제까지 LCM을 이용한 연구 성과를 살펴보고자 한다.

Expression of CD40, CD86, and HLA-DR in CD1c+ Myeloid Dendritic Cells Isolated from Peripheral Blood in Primary Adenocarcinoma of Lung (원발성 폐선암환자의 말초혈액에서 분리한 CD1c+ 골수성 수지상 세포에서의 CD40, CD86 및 HLA-DR의 발현)

  • Background: There have been several reports using animal experiments that CD1-restricted T-cells have a key role in tumor immunity. To address this issue, we studied the expression of markers for CD1c+ myeloid dendritic cells (DCs) isolated from peripheral blood in the clinical setting. Material and Method: A total of 24 patients with radiologically suspected or histologically confirmed lung cancer who underwent pulmonary resection were enrolled in this study. The patients were divided according to histology findings into three groups: primary adenocarcinoma of lung (PACL), primary squamous cell carcinoma of lung (PSqCL) and benign lung disease (BLD). We obtained 20 mL of peripheral venous blood from patients using heparin-coated syringes. Using flow-cytometry after labeling with monoclonal antibodies, data acquisition and analysis were done. Result: The ratio of CD1c+CD19- dendritic cells to CD1c+ dendritic cells were not significantly different between the three groups. CD40 (p=0.171), CD86 (p=0.037) and HLA-DR (p=0.036) were less expressed in the PACL than the BLD group. Expression of CD40 (p=0.319), CD86 (p=0.036) and HLA-DR (p=0.085) were less expressed in the PACL than the PSqCL group, but the differences were only significant for CD86. Expression of co-stimulatory markers was not different between the PSqCL and BLD groups. Expression of markers for activated DCs were dramatically lower in the PACL group than in groups with other histology (CD40 (p=0.005), CD86 (p=0.013) HLA-DR (p=0.004). Conclusion: These results suggest the possibility that CD1c+ myeloid DCs participate in control of the tumor immunity system and that low expression of markers results in lack of an immune response triggered by dendritic cells in adenocarcinoma of the lung.

  • Background: Cyclin-dependent kinase (CDK) inhibitors are family of molecules that regulate the cell cycle. The CDKN2, a CDK4 inhibitor, also called p16, has been implicated in human tumorigenesis. The CDKN2 inhibits the cyclin/CDK complexes which regulate the transition from G1 to S phase of cell cycle. There is a previous report that homozygous deletion of CDKN2 region on chromosome 9p21 was detected frequently in astrocytoma, glioma and osteosarcoma, less frequently in lung cancer, leukemia and ovarian cancer, but not detected in colon cancer and neuroblastoma. However, little is known about the relationship between CDKN2 and laryngeal cancer. Therefore this study was initiated to investigate the role of CDKN2 in human laryngeal squamous cell carcinoma development.1) Materials and methods: We used 5 human laryngeal carcinoma cell lines whether they have deletions or losses of CDKN2 gene expression by DNA-PCR or RT-PCR, respectively. We examined 8 fresh frozen human laryngeal cancer tissues to detect the loss of heterozygosity (LOH) of CDKN2. PCR was performed by using microsatellite markers of short arm of human chromosome 9 (D9S126, D9S144, D9S156, D9S161, D9S162, D9S166, D9S171, D9S200 and D9SIFNA). For informative cases, allelic loss was scored if the signal of one allele was significantly decreased in tumor DNA when compared to the same allele in normal DNA. Results: The CDKN2 DNA deletion was observed in 3 cell lines. The CDKN2 mRNA expression was observed in only one cell line, which was very weak. LOH was detected in 7 cases (87.5%). Conclusion: These results suggest that CDKN2 plays a role in the carcinogenesis of human laryngeal squamous cell carcinoma.

  • 혈액 세포 형성 과정인 조혈 작용은 콜로니 자극인자 (Colony Stimulating Factor, CSF)라 불리는 몇 종류의 당 단백질에 의해 조절된다. 이들 자극인자 중, GM-CSF는 다계통에 작용하는 조절인자로서 과립구와 거식세포의 생성을 조절한다고 알려져 있다 한편 GM-CSF는 생체내에서 백혈구의 형성을 조절하는 인자이기 때문에 골수이식을 한 환자 및 화학요법이나 방사선 치료를 받는 암환자에게서 발생하는 백혈구의 감소현상을 완화시키는 역할을 한다. 혈액의 보조화와 관련하여 의학적 효능을 나타낼 것으로 간주되는 GM-CSF를 유전자 재조합 기술로 효모에서 발현, 정제하여 물리화학적 특성 및 역가를 측정하는 것이 이 연구의 기본목적이다.

Inhibitive Effects of Meju Extracts Made with a Single Inoculum of the Fungi Isolated from the Traditional Meju on the Human Leukemia Cell Line (전통 메주에서 분리된 단독균으로 제조한 메주추출물의 혈액암세포에 대한 저해효과)

  • In order to study the antitumoral effect of meju extracts, which was made with a single inoculum of the microorganism, the cytotoxicity effects on several human leukemia cells such as promyelocytic leukemia cell (HL60), histiocytic lymphoma cell (U937) and acute T-cell leukemia Jurkat cell, and lymphocyte were analyzed by MTT assay. Twenty one microbes, mainly fungal genera, were isolated from Korean traditional mejus of different regions. From those collected isolates, meju was manufactured and extracted with 80% methanol, respectively. Meju methanol extracts exhibited low activites in cytotoxicity tests on HL60 cell, but high antitumoral effects of meju methanol extracts were shown on U937 and Jurkat cells. Meju methanol extracts made with a genera of Mucor, Absidia and Aspergillus showed prominant cytotoxic activities, especially. However all these extracts had no inhibitory effects on the cell growth of lymphocyte under the same conditions.

Effects of Plum Fruits Extracts at Different Growth Stages on Quinone Reductase Induction and Growth Inhibition on Cancer Cells (생육시기별 피자두 추출물의 암세포 증식 억제 효과 및 Quinone Reductase 유도 활성에 미치는 영향)

  • The plum (Prunus salicina L., cv. ‘Soldam’) fruits were harvested at different growth stages, and then extracted using 80% methanol, respectively. The methanol extracts of plum were investigated for their growth inhibition on 6 kinds of human cancer cells using MTT assay and for their activity to induce quinone reductase (QR) in murine Hepa1c1c7 cells. Among various methanol extracts of plum, the plum 1~4 (immature fruit), which thin out 10~25 days before final harvest, showed higher anticarcinogenic activity against 5 kinds of cancer cells than plum 5~9 (intermediate-mature and mature fruit). Especially, plum 1 and 2 were exhibited the strongest growth inhibiting activities to AGS, HepG2 and MDA cancer cells. Also the plum extracts induced the activity of QR, an anticarcinogenic marker enzyme, in Hepa1c1c7 cells while the induction of QR activities by adding plum extracts were shown to be a little difference depending on growth stages. These results suggested that methanol extracts of immature plum can be considered as an effective natural cancer chemoprevention materials.

  • The current population of elderly is increasing and the with an extended average lifespan, the frequency of cancerous occurrences have also increased, with these increases and the increase in radiotherapy for cancer patients, recognitions of harm and importance have become known. This article was known tumor treatment of patients with hematopoietic disorder by doing a comparative study on the changes in blood cells caused by the acute effects of trace dose to high dose of radiation exposed to mice. According to the sensitizer injection may give rise to harm to the components of peripheral blood. This material needs to be considered when for treating tumor patients and the risks of hematopoietic harm and believe that radiation therapy will be reasonable.

  • 깊은 생체 조직에서 전자기 에너지의 일부가 피부로 전달되는데, 이때 생체 내부에서 피부로 전달되는 전자기 에너지의 세기는, 주파수 대역과 전자기파를 흡수, 반사, 투과시키는 인체의 매질에 따라 다르다. Microwave Radiometry는 인체 내부 조직에서 방출되는 1-6 GHz 대역의 전자기 에너지 일부를 피부 표면에서 측정하여 일정한 체적내의 인체 내부 온도 평균온도를 추정하는 방법이다. 이러한 Radiometry로 암이나 종양 등의 이상 조직을 진단하는 의학적 가설은, 암이 진행시 악성 종양의 세포의 신진대사가 정상 세포보다 활발하게 되고 또한 종양 세포 주위로 혈액의 유입이 증가하게되어, 주위의 정상 세포 보다 열을 보다 더 방출하는 데 있다. 이때 발생된 열은 일정한 주파수 대역의 마이크로파 에너지를 방출하게 되고, 이에 Radiometry로 인체에 무해하고(passive), 비침습적(non-invasive), 방사능의 영향이 없는 (non-ionizing) 방법으로 인체 내부에서 전달되는 전자기 에너지 강도를 측정하여 종양 부위와 주위 정상부위의 온도 차이를 추정 의학진단에 응용할 수 있다. 본 논문에서는 이러한 Microwave Radiometry의 의학적 응용과 생리학적 특성을 고려한 인체모델용 팬텀에 대하여 살펴본다.
