Acetylcholine-1ike Effects of Mori Folium Water Fraction on Blood Pressure and Intestinal Movement in Rats (혈압 및 장관 운동에 대한 상엽 수층분획의 아세틸콜린 유사효과)

  • 1. 혈압 및 맥박 실험 Acetylcholine과 상엽 수층분획을 단독으로 정맥에 투여한 경우 각각에서 농도 의존적으로 일시적인 혈압 강하 효과가 나타났고 빈맥 현상이 관찰되었다. Nitric Oxide 합성효소 억제제인 N$^{G}$ -Nitro-L-Arginine Methyl Ester(10 mg/kg I. v)를 전처리한 경우 위 두 약물에 의한 혈압강하효과는 공히 증가되어졌다. 또한 두 약물에 의한 혈압강하 효과는 Atropine Sulfate(1 mg/kg i.v) 전처리로 완전히 차단되었다. Cholinesterase 억제제인 Physostigmine (0.05 mg/kg i.v) 전처리는 상엽의 혈압강하 효과에 아무런 영향을 나타내지 못하였다. 2. 장관 실험 Acetylcholine과 상엽 수층분획을 organ bath에 첨가한 경우 각각에서 농도 의존적으로 장관 수축력을 증가시켰다. 혈압반응에서와 같이 장관실험에서도 두 약물에 의한 장관 수축력의 증가는 Atropine Sulfate(1$\times$$10^{-5}$ M)의 존재하에서는 완전히 차단되어졌다. 이상의 결과로부터 상엽 수층분획은 Acetylcholine과 유사한 작용을 지닌 물질을 함유하는 것으로 사료된다.

Effect of Saponin and Non-saponin of Panax Ginseng on the Blood Pressure in the Renovascular Hypertensive Rats (신성고혈압백서에서 혈압에 미치는 고려홍삼사포닌과 비사포닌의 효과)

  • The effect of saponin and non-saponin of Panax Red Ginseng on the blood pressure and nitric oxide production were investigated in the conscious free moving one-kidney, one-clip Goldbaltt hypertensive (lK, 1C-GBH) rats. Mean blood pressure in the control and lK, 1C-GBH rats was decreased by the administration of ginseng saponin (100 mg/kg, i.v.). The hypotensive effect induced by ginseng saponin was reached maximum at 2-4 minutes and was slowly recovered to the initial level of blood pressure. Also ginseng saponin induced reflex tachycardia in the conscious both rats. Contrast to the response induced by ginseng saponin, hypotensive effect induced by non-saponin of panax ginseng is minimal. Plasma nitric oxide concentration was increased by the treatment of ginseng saponin (100 mg/kg, i.p for 5 days) in both rats. It has been shown by western blotting that the expression level of the protein for endothelial nitric oxide synthase (eNOS) in the aorta of rats was not increased by the treatment of ginseng saponin (100 mg/kg, i.p). However, eNOS activity in aortic homogenates of both rats were increased by the treatment of ginseng saponins. From the above results, the hypotensive effect of saponin was greater than that of non-saponin of Panax Red Ginseng. The lowering effect of blood pressure by ginseng saponin may be due to the increase of plasma nitric oxide concentration via the increase of endothelial nitric oxide synthase activity in the renovascular hypertensive and control rats.

Antihypertensive Effects of Casein Hydrolysate in Spontaneously Hypertensive Rats (자연발증고혈압쥐에서 카제인 가수분해물의 혈압강하효과)

  • The aim of this study was to determine if a low-molecular weight casein hydrolysate has an anti- hypertensive effect in spontaneously hypertensive rats (SHR). Prior to the in vivo experiment, the casein hydrolysate was confirmed to be resistant to gastrointestinal digestion by confirming the retention of its potency as an inhibitor of angiotensin I-concerting enzyme after incubation with pepsin, trypsin, or chymotrypsin. The in vivo anti-hypertensive effect of the hydrolysate was determined by the tail cuff method. Following an oral administration of the hydrolysate solution, the systolic blood pressure (SBP) decreased by 12.9% (-28.9mmHg; P<0.05) at 3 h after the administration at a dose of 500mg/kg body weight. When the hydrolysate was administered as an emulsion with 30% egg yolk, its anti-hypertensive effect was even more greater at the same dose(-30.8mmHg or -15.9%; P<0.01). In a 50-day long-term trial where the casein hydrolysate was administered once a day, the SBP-lowering effect of the hydrolysate was apparent (P<0.05) from day 35 through the end. Moreover, organ weights and plasma glutamate oxaloacetate transaminase and glutamate pyruvate transaminase activities of the administered SHR were not significantly different from those of controls at the end of the long-term trial.

  • 본 연구는 꾸준한 신체활동이 고혈압을 강하시키는데 어떠한 효과가 있는지를 확인하는데 그 목적이 있다. 실험대상은 연령과 성별이 같은 SHR로 실험군과 통제군으로 무작위샘플로 분류하였다. 실험실적응 및 운동적응 기간을 거친 후 운동기간별 혈압의 변화를 관찰하였다. 통계분석은 SPSS 10.0을 사용하였다. 실험결과 집단 간 혈압의 차이가 있었다(p<.05). 실험집단간, 집단내 기간별 상호작용은 유의하게 나타났다(p<.05). 결론적으로 주지적인 신체활동은 SHR의 혈압을 감소시키는 효과가 있다.

Effect of Soybean Hydrolysate on Hypertension in Spontaneously Hypertensive Rats (대두 가수분해물에서 분리한 UF-peptide가 in vivo에서 자발성 고혈압 흰쥐의 혈압 강하에 미치는 영향)

  • Effect of soybean hydrolysate prepared by ultrafiltration(UF-peptide) was evaluated regarding its action on blood pressure, selected biochemical changes in spontaneously hypertensive rats(SHR). Male SHR were divided into three groups and fed diet with 0, 3, 10% UF-peptide for 6 weeks. Development of hypertension was determined by tail cuff method during the experimental period. After the experimental period, angiotension I converting enzyme(ACE) activity in serum and vascular tissue, and lipids level in serum were measured. Spontaneous elevation of blood pressure was suppressed by the UF-peptide supplementation after two weeks of feeding. After five weeks of feeding the development of hypertension was significantly inhibited in the UF-peptide administered groups compared to the control. In addition, the UF-peptide was effective for lowering serum lipids level such as triglyceride and total cholesterol, as well as ameliorating some pathological changes in SHR.

조구등의 혈압강하작용과 성분연구

  • 혈압강하작용은 oxyindole계 분획에서 제일 강력하였으며, 다음이 hetero-yohimbine계 분획이며, 극성 alkaloid계 분획이 제일 약하였다. 특히, oxyindole계 및 hetero-yohimbine계는 epinephrine의 승압작용을 길항하지 아니하였으므로 이들의 혈압강하작용은 혈관의 수축작용을 길항하는 기전과는 무관함을 알 수 있었다. 반면에 화학적으로는 hetero-yohimbine계 분획에서 hirsuteine, corynantheine 및 hirsutine을 분리하였고 극성 alkaloid 분획에서는 3$\alpha$-dihydrocadambine을 분리 동정하였으며 전자 3종의 alkaloid는 열(10$0^{\circ}C$)에 안정하고 후자는 불안정함을 확인하였다.

  • ABSTRACT - To examine antihypertensive components of Ganoderma lucidum in Korea, two kinds of fruiting bodies (J and K) were used for extraction with water and the extracts were purified by ethanol precipitation and dialysis. Three fractions, i.e., the aqueous total extract(A), the ethanol supernatant(B} and the purified precipitate(C), were compared for antihypertensive activity in anesthetized spontaneously hypertensive rats (SUR). Although fractions A and B showed the activity, fraction C did not. Particularly, fraction B of sample K produced 44.3% reduction in diastolic blood pressure and 30.6% reduction in heart rate after i.v. administration of a dose of 10 mg/kg. Direct effects of this fraction B to the heart were observed in the isolated blood perfused heart preparation of the dog. It induced positive chronotropic and inotropic responses dose-dependently in the case of sample J. In the case of sample K, marked chronotropic and inotropic effects on atrial muscle but not on ventricular muscle were induced. In both samples, coronary blood flow (CBF) was dose-relatedly increased.reased.
