Optimum environmental condition of live container for long distance transport in live abalone Haliotis discus hannai (북방전복, Haliotis discus hannai의 장거리 수송을 위한 적정 수온 및 염분 조건)

  • We investigated the survival rate by water temperature and salinity, physiological rhythm and morphological change of live abalone to get to know optimum water temperature and salinity suitable for long-distance transportation of live abalone. At $8^{\circ}C$ and above, 96-100% of survival rate was shown at all experiment groups. At $6^{\circ}C$, 66% of abalones survived in normal seawater but they showed 0% of survival rate at $30{\pm}0.5psu$ and $26{\pm}0.5psu$ of salinity at the same water temperature. There was no significant difference of oxygen consumption rate for a week between the seawater and $30{\pm}0.5psu$. Also, a positive correlation was shown between salinity and water temperature and the oxygen consumption rate was slightly higher at $30{\pm}0.5psu$ than seawater. Thinned epithelial layers and expansion of lymph sinus were observed less than $30{\pm}0.5psu$ or below $6^{\circ}C$ of temperature. This result shows that the optimum level of water temperature and salinity is considered to be $6-8^{\circ}C$ and more than $30{\pm}0.5psu$ respectively.

  • Histology and ultrastructure of the gill in the granular ark, Tegillarca granosa are described using light and transmission electron microscopy. The gill of the clam have typical structure of the filibranch type. The gill filament have several band of lateral and apical cilia. The epithelial layer surrounding the hemolymph sinus is simple and consists of epithelial cells, ciliated cells and secretory cells. The epithelial cells are usually squamous and covered with microvilli. The ciliated cells are usually columnar and can be divided into two types (A and B). Type A cells are more abundance and have lower electron density than B cells. Ultrastructure of the cilia showed that '9+2' microtubular structure of the axial filament and '$2{\times}9$' proximal centriole structure in the cross section. Secretory cells are mainly observed in the apical region of the filament and can be divided into three types of A, B and C with morphological features of the secretory granules. Type A cells of oval shaped are more abundance than other secretory cells and contains numerous secretory granules of low electron dense. Type B cells contains secretory granules of membrane-bounded and high electron dense. Secretory granules of type C cells are elliptical and fine granules surrounding the homogeneous core materials.

Structures of hemal nodes and hemolymph nodes in sheep (면양의 혈절과 혈림프절의 구조)

  • This study was performed to investigate the gross anatomical features and microscopical structures of the hemal nodes and the hemolymph nodes in the sheep. The hemal nodes and hemolymph nodes were observed mainly in periphery of the thoracic and abdominal aortae of the animals. The size of hemal nodes was generally smaller than that of the hemolymph nodes, and the shape of the both organs was spherical or ovoid. The color of the hemal nodes was red, while that of the hemolymph nodes was grary with red bands. The hemal nodes were surrounded by a relatively thick connective tissue capsule and there were extensive subcapsular sinuses distended by a great number of erythrocytes. Although a few number of lymphatic nodules and small areas of diffuse lymphatic tissues were observed in the parenchyma, no typical cortex and medulla were defined in the hemal node. Small numbers of blood vessels were found at the connective tissue capsule but lymph vessel was not observed microscopically in this organ. The hemolymph nodes were covered by a relatively thick connective tissue capsule and there was a hilus in each node. The parenchyma was divided into cortex and medulla. The cortex was composed of a few numbers of lymphatic nodules and some diffuse lymphatic tissues. The medulla comprised medullary sinuses and cords. Afferent and efferent lymph vessels were observed at the periphery of the capsule adn the hilus, respectively. The subcapsular and medullary sinuses were not extensive but filled with small numbers of erythrocytes. The stroma of hemal node and hemolymph node was composed of reticular cells and fibers, and the capsule and trabecula consisted of collagenous fibers with smooth muscle fibers.

Effects of Portulaca oleracea L. Supplementation on Reproductive Performance, Blood Profiles, Immune Response and Fecal Microflora in Multiparous Sows (모돈 사료 내 Portulaca oleracea L.의 첨가가 번식성적, 혈액성상, 면역반응 및 분 중 미생물에 미치는 영향)

  • This study was undertaken to determine the effects of dietary supplementation with herbal extracts of Portulaca oleracea L. (POL), on the reproductive performance, blood profiles, immune response, and fecal microbial population in multiparous sows. On the basis of initial body weight, a total of 20 multiparous sows (Landrace×Yorkshire; 229.2±1.15 kg) were randomly allotted to 2 treatment groups, each including 10 replicates. The dietary treatments were administered during the lactation period, and included a corn-soybean meal-based diet (control group) and diet supplemented with 0.05% POL. Throughout the experimental period, no significant differences were observed between the two diet groups for body weight, backfat thickness, and reproductive performance. Blood profile analysis revealed significantly decreased lymphocyte concentrations and IL-1β levels (p<0.05), and increased serum IgG levels (p=0.051), of sows consuming the POL supplemented diet. Furthermore, coliform counts tended to decrease in sows consuming diet supplemented with POL (p=0.063). Taken together, our results indicate that POL supplemented diets exert beneficial effects on blood profiles, immune response, and fecal microflora of multiparous sows, without any negative effect on the reproductive performance.

Effects of 20-hydroxyecdysoneon Adult Development of Diapausing Fall webworm (Hyphantria cunea Drury) Pupae (휴면중인 미국흰불나방 번데기의 성충발육에 미치는 20-hyderoxyecdysone의 영향)

  • Studies were carried out to elucidate the efficacy of exogeneous 20-hydroxyecdysone treatment on terminating pupal diapause in the fall webworm. Hyphantria cunea Drury. And the difference was also investigated between normal adult development and post-diapause development after 20-hydroxyecdysone treatment. In the diapause termination rate of pupae treated with 20-hydroxyecdysone after storage for various periods at 16L:8D and $25\pm$$1^{\circ}C$, at the highest rate was observed from the group stored for the longest period and the lowest rate from those stored for 1.5 months. The time needed for adult emergence was inversely proportional to the chilling (at $0\pm$$1^{\circ}C$) period, and the longer its exposure period at low temperature, the higher its sensitivity to 20-hydroxyecdysone treatment. Pupal diapause duration was almost the same, regardless of storage period in the total darkness or at the photoperiod of 16L:8D, and also they successfully emerged to adult even without any experience at low temperature. The oxygen consumption rate in normally developing pupae showed nearly a typical U-form. But, that of diapausing pupae treated with 20-hydroxyecdysone slowly increased and jumped 14 days after the treatment. Pupal diapause began before formation of adult tissues, an a timing of adult tissue formation coincided with ascending timing of the metabolic rate in both normally developing pupae and diapausing pupae treated with 20-hydroxyecdysone. The diapausing pupae treated with 20-hydroxyecdysone was similar to normally developing pupae in band patterns of proteins from haemolymph or fat body.

Influence of denitrified biofloc water on the survival rate and physiological characteristics of Pacific white shrimp juveniles, Litopenaeus vannamei (바이오플락 탈질수가 어린 흰다리새우, Litopenaeus vannamei의 생존율 및 생리특성에 미치는 영향)

  • This study investigates the effect of denitrified biofloc water on changes in the water quality parameters and the physiological characteristics of shrimps. Biofloc rearing water contains a large number of microorganisms and can rapidly stabilize the water quality and energy saving if reusable due to high water temperatures. Rearing water contain floating bacteria with both aerobic and anaerobic bacteria. Therefore, when the carbon source is added in limited air supply, the anaerobic state is activated and the denitrification process is possible. In this study, the denitrification water had the following properties: ammonia (6.9 mg L-1), nitrite (0.3 mg L-1), nitrate concentration (9.2 mg L-1), high pH (8.42) and alkalinity (590 mg L-1). The experimental group consisted of seawater (SW, control), a mixture of Seawater and denitrified biofloc water (DNW) in the ratio of 3:1, 1:1 and DNW only. All experiments were done in triplicate. As a result, the survival rate never changed even when 100% of the denitrification water was utilized. However, a body fluid analysis showed that creatine and BUN were increased due to index of stress and the tissue damage resulting from the high denitrified water content. Body fluid ions (Na+, K+, and Cl-) significantly decreased as the denitrified water content increased. It was recommended that the denitrification water be mixed with a certain ratio (less than 50%) in the future as it may affect the osmotic pressure control in shrimps.

The Antimicrobial Activity of Bacterial-challenged Black Soldier Fly, Hermetia illucens (세균에 의해 면역이 유도된 동애등에의 항균활성)

  • In the larvae of the black soldier fly, Hermetia illucens, innate immunity mechanisms are activated in response to various pathogens and stimulants, resulting in the expression of antimicrobial peptides (AMPs). To induce the mass production of AMPs, H. illucens fifth instar larvae were immunized with five different kinds of bacteria. We isolated from the hemolymph of the H. illucens larvae after bacterial challenge, and their antimicrobial activities against Gram-positive bacteria (Staphylococcus aureus) and Gram-negative bacteria (Escherichia coli) were measured using the inhibition zone assay. Among these five different kinds of bacteria, the hemolymph of Bacillus subtilis-challenged H. illucens larvae showed the strongest antimicrobial activity against both Gram-positive bacteria and Gram-negative bacteria. The antimicrobial activity of the hemolymph of $1{\times}10^9cfu/ml$ B. subtilis-challenged H. illucens peaks at 24 hr at 48 hr post-infection and gradually declines with time. Moreover, the immunized hemolymph also showed strong antimicrobial activity against various poultry pathogens such as S. enteritidis, S. typhimurium, and S. pullorum. These results suggest that the expression of AMP genes in B. subtilis-challenged H. illucens is up-regulated by innate immune responses, and that B. subtilis-challenged H. illucens overexpressing AMPs may be useful as a feed additive in livestock diets to reduce the need for antibiotics.

Fine Structure and Histopathological Changes Exposed to Acute High Salinity of the Gill of Japanese Clam, Corbicula japonica (일본재첩, Corbicula japonica 아가미의 정상구조와 고염분 급성노출에 의한 조직병리학적 변화)

  • This study was performed to observe ultrastructure of the gill and to ascertain the effect of salinity on histopathological and ultrastructural changes in the gill of the Japanese clam, Corbicula japonica. Experimental period was 7 days. Experimental groups consisted of control, 5, 10, 20 psu. $LC_{50}$ (96 h.) by the probit was 19.55 psu. Mortality was significantly different from the control (p < 0.05). Inner demibranch of the gill of C. japonica was wider 1.37 times than outer demibranch (p < 0.001). The filament zone on the plica can be distinguished by the six epithelial celll cell; frontal ciliated epithelium ($7{\mu}m$), latero-frontal ciliated epithelium ($5{\mu}m$), postlatero-frontal epithelim ($3{\times}8{\mu}m$), and lateral ciliated epithelium ($5{\mu}m$) in the frontal zone, endothelial cellin the intermediate zone, and abfrontal cell in the abfrontal zone. It had one type of secretory cell that was filled with fibrous substances of low electron density. The gill of C. japonica exposed to 5 psu for 7 days was observed partially disappearance of the cilia, and glycogen granule in the filament. In the 10 psu, gill appeared partially modification of epithelial cell and destruction of the glycocalyx. Gill exposed to 20 psu was extended nuclus of the ciliated epithelial cell, destruction of the organelles, and observed glycogen granules infiltration and numerous vacuoles. Moreover, more than 50% filaments were observed that come out chitinous rod from disappearance of epithelial cell in the filament. Therefore, the destruction of the cilia and epithelial cell induce physiological activity and it may be leading directly to death.

Mode of Action of Antimicrobial Peptides Identified from Insects (곤충 유래 항균 펩타이드의 작용 기작)

  • Insects represent the largest class within the animal kingdom in terms of species number. Humans had been utilized insect in the broad area, including food, agriculture, industry, pharmaceuticals and so on. At present, insects are emerging as a leading group for identifying and extracting novel bioactive substances due to enormous number and a high nutritional value. Insects rely on a suite of systemic response to resist infection such as immune cells, hemocytes, activation of enzymes cascades, and antimicrobial peptide/protein. Among the substances, antimicrobial peptides (AMPs) are main components of potent antimircrobial innate defense system into the insect hemolymph. AMPs raise influential candidate as avenue to resolve the development of antibiotic-resistant microbial organism. Insect AMPs are classified into four main classes: cecropins, insect defensins, glycine/proline-rich peptides. Insect AMPs have been purified, over 150. In this review, AMPs derived from several insects were summarized including honey bee, dung beetle, butterfly and longicorn beetle. These peptides almost exhibited potent antimicrobial activities against human microbial pathogens without causing remarkable hemolysis to erythrocytes excluding melittin, and their mode of action(s) are based on disruption of the plasma membrane or fungal apoptosis. Therefore, study of insect AMPs is expected to be useful for designing novel therapeutic antimicrobial applications.

I. Laboratoy Rearing of the Aphidophagous Ladybeetle, Harmonia axyridis; Yolk Protein Production and Fecundity of the Summer Adult Female (무당벌레(Harmonis axyridis)의 사육 I;여름 성충의 난황단백질 생성 및 산란력)

  • The adult of the aphidophagous ladybeetle, Harmonia axyridis, was reared by feeding an artificial diet (chicken's 1iver:sugar = 5:2, v/w). The adult ladybeetle used was from pupae collected in the field in midJuly. Yolk protein production, fecundity and longevity of the adult female were checked under 3OoC, 16L:8D. Yolk proteins (Vg3 and Vg4) were detected from the hemolymph of the female 5 days after the emergence. One hundred pairs of female and male were reared separately. First oviposition started from 7 days afier the emergence and laid an average of 163.9k144.0 eggs during lifetime. Seventeen females did not lay eggs at all. Longevity of the female was 60.6k16.9 days. The application of a JH analog, methoprene (2.5 pg), caused the occurrence of yolk proteins in the hemolymph 1 day earlier than that of the control. Weekly application of 2.5 pg of methoprene induced 83.3% from the 30 females to lay eggs within 15 days after the emergence, while the control had 51% only. Methoprene made all females lay eggs, causing the increase in the fecundity. Though the longevity of the female was not effected by the weekly application of methoprene, one time excess dose (12.5 pg) caused high in mortality and low in fecundity.
