Characterization of an antifungal compound isolated from an antagonistic fungus Aspergillus terreus against phytopathogenic fungi (식물병원균 생육을 저해하는 Aspergillus terreus로부터 분리한 향균물질의 특성)

  • An antagonistic fungus inhibiting growth of various phytopathogenic fungi was isolated from greenhouse soils and identified. Mophological features of fruiting structures on potato dextrose agar and colorless globose to ovate heavy walled hyaline cells from the vegetative mycelium grown on MY20 agar indicate that this antagonist is Aspergillus terreus. The antagonistic activity is due to the production of antifungal compounds. An antifungal compound was purified from its culture filtrate using chloroform extraction, column chromatography, and thin layer chromatography. The purified antibiotic was effective to various phytopathogenic fungi and identified as butyrolactone I. $ED_{50}$ values measured by petri-plate assay through effective dosage(ED)-probit analysis were 9.7, 13.7, 23.3, 42.6 and 102.7 ppm on Botrytis cinerea, Rhizoctonia solani, Pythium ultimum, Phytophthora capsici, and Fusarium oxysporum, respectively.

  • The volatile essential oil isolated from Houttuynia cordata were separated to 11 fractions by Prep-HPLC, of these, a fraction(Fr. 6) which carried the characteristic Houttuynia cordata flavor(fishy) contained 2-undecanone, ${\beta}-myrcene$, ${\beta}-ocimene$, 1-decanol and decanoyl acetaldehyde, as identified by GC-MS. From this observation, it may be inferred that 2-undecanone and decanoyl acetaldehyde could be the compounds which play a crucial role in flavoring of Houttuynia cordata. In test of antibacterial activity of eleven fractions of volatile essential oil from H.C., the growths of nine Gram negative bacteria were inhibited obviously when treated with and Fr. 6 including 2-undecanone, ${\beta}-myrcene$, ${\beta}-ocimene$, 1-decanol and decanoyl acetaldehyde, and Fr. 5 including decanal, endobornylacetate, fenchene and decanoic acid, respectively.

Inhibitory Effect of Lactic Acid Bacteria and its Metabolites on the Growth of Staphylococcus aureus (젖산균과 그 대사산물이 Staphylococcus aureus의 생육에 미치는 억제효과)

  • Lactic acid bacteria(LAB) and its metabolites were tested for their inhibitory effect on the growth of Staphylococcus aureus in vitro. When S. aureus and LAB were incubated simultaneously in MRS broth, the growth of S. aureus began to be suppressed after 12 hour of incubation, completely inactivated within 24 hour of incubation by L. lactis and 48 hour by L. casei, L. acidophilus, and Sc. thermophilus. The pH values of media incubated by S. aureus and LAB were about 4.5 at 12 hour of incubation and 3.5 at 48 hour. The metabolites of the four LAB all exerted antibacterial activity on the growth of S. aureus in TS broth, but is got lost the antibacterial activity by heating ($100^{\circ}C,\;20\;min$). On treating metabolites with catalase, only L. lactis were lost its antibacterial activity. Organic acids (acetate, lactate) showed more active inhibition than inorganic acid (HCl) at pH 3.5, 4.5 but there is no significant differance at pH 5.5, 6.5.

Antimicrobial activity of honeybee venom against fish pathogenic bacteria (국내산 봉독의 어류병원성 세균에 대한 항균활성)

  • In this study, we examined antimicrobial activity of the bee venom isolated from honeybee (Apis mellifera L.) against fish pathogenic bacteria, Edwardsiella tarda, Vibrio ichthyoenteri and Streptococcus iniae of cultured olive flounder, Paralichthys olvaceus. The bee venom exhibited significant antimicrobial activity against the both Gram-negative bacteria, E. tarda and V. ichthyoenteri and Gram-positive bacteria, S. iniae. Minimum inhibitory concentration (MIC) and minimun bactericidal concentration (MBC) of the bee venom were 17.6 ${\mu}g$/ml 34.6 ${\mu}g$/ml against E. tarda., and 1.76 ${\mu}g$/ml, 6.8 ${\mu}g$/ml against V. ichthyoenteri, respectively. MIC and MBC of the bee venom were 3.49 ${\mu}g$/ml, 11 ${\mu}g$/ml, respectively against S. iniae. The postantibiotic effect (PAE) of the bee venom was 5 hr, 6 hr, and 7 hr against E. tarda, V ichthyoenteri, and S. iniae, respectively. In addition, its antimicrobial activity was stable under various pH conditions. According to these results, the bee venom showed the excellent antimicrobial activity against the tested pathogenic bacteria.

Characterization of Antibacterial Substance - Producing Bacillus subtilis Isolated from Traditional Doenjang (전통 된장으로부터 분리한 향균물질 생산 Bacillus subtilis의 특성)

  • A bacterium which has high enzymatic activities such as amylase, cellulase and protease was isolated from Korean traditional soybean food, doenjang. The isolated bacterium was identified to Bacillus subtilis HS25 by the test of morphological and biochemical properties according to Bergey's Manual of Systematic Bacteriology and API 50 CHL kit, and by the 16S rDNA sequence. The isolated B. subtilis HS25 had a potent antibacterial activity against food born causative or pathogenic bacteria. B. subtilis HS25 is endospore forming cell and contained flagella and abundant viscous material at the out layer of cell wall. It was rod type bacterium $(0.5{\sim}0.8{\times}3{\sim}5{\mu}m)$ having biochemical characteristics such as gram staining(+), catalase(+), oxidase(-) and hydrolysis of esculin(+). The optimal medium compositions for production of antibacterial substance in the B. subtilis HS25 were 1% of soluble starch, 0.5% of yeast extract, 0.5% of peptone and 0.05% of MgCl$_2{\cdot}6H_{2}O$. The optimum temperature and pH of the growth of the B. subtilis HS25 was 35$^{\circ}C$ and pH 7.5, respectively. The antibacterial activity was more high in neutral to a little alkaline pH (6.5-10.5) than in acidic pH. The optimal shaking speed to grow and to produce antibacterial substance of the B. subtilis HS25 was 160${\sim}$200 rpm. The optimal culture time for antibacterial activities of the bacterium were shown to be in the range of 12-36 hr.

Antimicrobial Activity of Extracts from Citrus Seeds (감귤류 종자 추출물의 향균활성)

  • To develope natural food preservatives antimicrobial effect of the natural products against food-related bacteria and yeast The purpose of this study was evaluate antimicrobial effect of the citrus seeds. antimicrobial activities of methanol extracts from the citrus seeds investigate against Escherichia coli O26, Staphylococcus aureus 6358, Saccharomyces cerevisiae IBM 4274, Bacillus licheuiformis 9945a and Alcoligenes faecalis. Citrus seeds is containing to moisture 4∼6.5%, curd protein 11∼15%, curd lipid 32∼46%, curd carbohydrate 22∼45 % and ash 2∼4 %, that is containing to flavornoid 12∼48mg% and phenolic compound 22∼53mg%. Solidity content of the methanol extract from the citrus seeds was 0.8∼1.2%. Almost all of the methanol extracts from citrus seeds exhibited growth inhibiting activities for most of microorganisms tested. The methanol extracts from Citrus grandis, C. sunki, C. sulcata showed the growth inhibitory effects against Escherichia coli O26. The methanol extracts from C. obovoidea, C. sulcata, C. aurantium showed the growth inhibitory effects against Staphylococcus aureus 6358. The methanol extracts from C. obovoidea, C. sulcata, C. tangerina showed the growth inhibitory effects against Saccharomyces cerevisiae IBM 4274. The methanol extracts from C. obovoidea, C sunki, C. sulcata, C. tangerinan, C. natsudaidai, C. iyo, C. aurantium showed the growth inhibitory effects against Bacillus licheuiformis 9945a. The methanol extracts from C. obovoidea, C sunki, C. sulcata, C. aurantium showed the growth inhibitory effects against Alcoligenes faecalis. Among this especially, Showed growth inhibiting activity of the methanol extracts from Citrus sulcata that about microorganisms investigated. If apply searching suitable application method about such the citrus seeds antimicrobial activity, role as good antimicorbial material in storage or cooking of food, processing is expected.

  • New cephalosporin antibiotics, 7-[(3,4-dihydro-6-methoxycarbonyl-2,2-dimethyl-2H-1,4-thiazin-3-yl)acetamido]-3-heterocyclothiomethyl-3-cephem-4-carboxylic acid derivatives(2a∼2d) were synthesized. Antibacterial activities of these new cephalosporin derivatives and the relationship between their structures and their activities were examined. Among them,7-[(3,4-dihydro-6-methoxycarbonyl-2,2-dimethyl-2H-1,4-thiazin-3-yl)acetamido]-3-[(2-methyl-1,3,4-thiadiazol-5-yl)thiomethyl]-3-cephem-4-carboxylic acid exhibited the broad antibacterial activities against Gram(+) and Gram(-) bacteria.

  • The antimicrobial and antioxidative activities were investigated to confirm the utility of wood vinegar. Antimicrobial activity was performed by paper disc method and liquid culture. The growth inhibition was observed in all microbial species at a dose of as low as 25 $\mu$m of wood vinegar by paper disc method. Also, in liquid culture, S. cerevisiae and P. aeruginosa were more inhibited the growth than others in the concentration of $2\%$ (v/v). For measuring of antioxidative activity, wood vinegar was fractionated with acidic, phenolic, basic and neutral fraction; and their antioxidant activities were measured by the radical scavenging effect on DPPH radical. In four fractions, phenolic fraction showed high antioxidative activity.

Changes in Antimicrobial Activity of Hydrolyzate from Mustard Seed (Brassica juncea) (겨자 가수분해물의 항균성 변화)

  • 겨자의 가수분해 시간에 따른 향균력을 비교하고, 각종 미생물에 대한 겨자 물 추출물의 최소 저해 농도 및 최소 저해 투여량을 측정한 결과 다음과 같았다. 겨자의 자가분해 시간에 따른 물 추출물의 향균성은 처음에 아주 약하게 나타났던 것이 12시간 방치 후 크게 증가하였다. 최대 항균활성은 24시간 방치 후 나타났으며 이후 거의 변하지 않았다. 겨자를 첨가한 물의 산도는 시간이 지남에 따라 증가하였고, 겨자의 농도가 클수록 그 정도가 크게 나타났다. $37^{\circ}C에서$ 24시간 자가분해한 겨자 물 추출물의 MID는 Bacillus subtilils가 $70\mu1/disc로$ 다른 실험 균주에 비하여 낮았으며, 젖산균이 $95\mu1/disc로서$ 높게 나타났다. MIC는 Bacillus subtilils, Staphylococcus aureus, Salmonella typhimurium 및 Vibrio parahaemolyticus 균이 $70\mu1/ml로$ 다른 실험 균주에 비하여 낮게 나타났으며, 젖산균 곰팡이에서 $90\mu1/ml로$ 높게 나타났다. 자가분해 시간에 따른 겨자물 추출물의 중류성분은 자가분해 시간이 지남에 다라 점차 증가하는 경향이었고, isothiocyanate 류의 함량은 24시간 때에 최대치를 나타내었다.

  • 8-Acyl-2-hydroxyphenazine-5,10-dioxides and 8-acyl-2-aminophenazine-5,10-dioxides were synthesized by the reaction of hydroquinone and 4-aminophenol with 6-acylbenzofuroxans which had been obtained from acetanilide and n-acyl chlorides bearing butanoyl, hexanoyl and octanoyl groups. The antimicrobial activities of these phenazine dioxide derivatives were investigated in terms of minimum inhibitory concentration by the common twofold dilution technique. It was observed that the antimicrobial activity of the phenazine dioxide derivatives bearing octanoyl group was stronger than that of those bearing butanoyl and hexanoyl groups in gram positive microorgamisms, but it was observed that the antimicrobial activity and the number of the carbon atom of acyl groups did not have any relation in gram negative microorganisms. When the activity of xanthine oxidase which is the key enzyme in the generation of superoxide anion radical ($O_2^-$), was measured in the presence of phenazine dioxide derivatives, the inhibitory action of the enzyme activity of 8-acyl-2-hydroxyphenazine-5,10-dioxides was increased in accordance with the number of the carbon atom of acyl groups.
