Cross Infectivity of Nuclear Polyhedrosis Viruses to the Common Armyworm, Pseudaletia separata (멸강나방에 대한 곤충핵다각체병바이러스의 교류감염성)

  • This studies were carried out to selected high pathogenic nuclear polyhedrosis viruses(NPVs) against Pseudaletia(=Leucania) separata for the introduction of microbiol control of the insect NPV in Korea. Among 21 NPVs, Sesamia inferens and 4 P. separata NPV strains were highly pathogenic against P. separata when fed orchard grass leaves smeared virus suspension on the 2nd instar larvae. Three NPV strains (LsNPV-F, LsNPV-G, LsNPV-Y) were more susceptible to the younger instar than the older instar P. separata larvae when fed artificial diet mixed with the virus to the insect.

  • A nuclear polyhedrosis virus isolated from the oriental tobacco budworm larvae, Helicoverpa assulta (Guenee) was characterized by electron microscopy, SDS-PAGE, restriction endonuclease analysis and cross infectivity. The shape of a polyhedron was $1.0\mu\textrm{m}$ in average with icosahdral outline, and the virus particle was $65nm\times300nm$ in average with rod-shape. The nuclear polyhedrosis virus was contained a single nucleocapsid within a viral envelope embedded in a polyhedron. The polyhedral protein was composed of a single polypeptide with a M.W. of 31 Kd. The genome size of the virus by restriction endonuclease analysis was about 120 Kb. Among several nuclear polyhedrosis viruses, the nuclear polyhedrosis virus from Helicoverpa assulta (HaNPV) and Autographa california nuclear polyhedrosis virus (AcNPV) were infected the oriental tobacco budworm larvae.

Enhanced Effectiveness of Spodoptera litura Nucleopolyhedrovirus with Organic Acids and Functional Matters (유기산, 기능성물질 혼합에 의한 담배거세미나방 핵다각체병바이러스의 병원성 증진효과)

  • This experiment was conducted to improve activity of Spodoptera litura Nucleopoly-hedrovirus (SINPV) combined with organic and functional matters. In combination of SINPV mixed with organic matters, LT$_{50}$ values of SINPV 1.0${\times}$10$^{5}$ PIBs/$m\ell$ combined with boric acid of 2,000 ppm were 4.5 days. It was 1.5 days shorter than SINPV 1.0${\times}$10$^{5}$ PIBs/$m\ell$ alone. The body weight of larva infected with SINPV 1.0${\times}$10$^{5}$ PIBs/$m\ell$ combined with boric acid of 2,000ppm was not increased, and S. litura was completely dead in 7 days after treatment. It suggested that addition of boric acid in SINPV application enhanced the pathogenicity against S. litura larvae. In laboratory experiment of combination of SINPV with functional matters, LT$_{50}$values of SINPV 1.0${\times}$10$^4$ PIBs/$m\ell$ alone were 7.3 days, but those of SINPV 1.0${\times}$10$^4$ PIBs/$m\ell$ with electrolyzed oxidizing water, pyroligneous liquor or kitosan were 10.4, 9.3 and 11.2 days, respectively.   • Viral insecticides were formulated with Spodoptera litura nuclear polyhedrosis virus and different U.V. protectants based on white carbon, molasses, and white carbon and molasses mixture to use as microbial control agents. Effect of rainfall on the attachment of formulated viruses to leaves was no different between the treated and the non-treated experiment. Persistence of the formulations was lated 5 days on the surface-sprayed leaves and 12 days on the under-sprayed leaves which was showing 60% mortality. Total mortality of the viral insecticides was more than 97% with no differences among them. Field evaluation of three viral insecticides in soybean field was very successful then carried out in Chinju, a southern part of Korea. Mortality by the formulation in the field during 14 days was more than 93%, but the formulations contained molasses showed phytotoxicity on soybean leaves. Spray effect of the viral insecticides was begun to appear from 7 days later than that of chemical insecticide.

  • Mass production of Spodoptera litura nuclear polyhedrosis virus (SINPV) was carried out on massively reared host insects. The yield of SINPV was maximal with $6.7{\;}\times{\;}10^9$ PIBs per larva on the 8th day post inoculation, when 5th instar larvae were inoculated with $1.1{\;}{\times}{\;}10^7$ PIBs per ml, and 2 g of artificial diet was sufficient for food consumption of a larva. The moribund larvae were more suitable for handling and mass production of virus than the completely dead larvae. The larvae, when treated with methoprene ($Manta^{\circledR}$), prolonged their larval period and consequently became bigger to result in higher yield(about 15%) of virus.

  • This experiment was conducted to investigate pathogenicity of Spodoptera litura nucleopolyhedrovirus (SINPV) with different temperatures to mass production. In laboratory, LC$_{50}$ values of SINPV were 5.534$\times$10$^3$ PIBs/ml and 4.l0$\times$10$^2$PIBs/ml in 1st instar larvae at 2$0^{\circ}C$ and 28$^{\circ}C$, respectively, but those were increased at 32$^{\circ}C$. LC$_{50}$ at 3rd and 5th instar larvae were showed the lowest value at 28$^{\circ}C$. LT$_{50}$ values of SINPV in 1.0$\times$10$^{5.7}$ PIBs/ml were determined at various temperature conditions (20-32$^{\circ}C$). The result showed that treatments at 28$^{\circ}C$ and 32$^{\circ}C$ shortened LT$_{50}$ values, but treatments at 2$0^{\circ}C$ and 24$^{\circ}C$ was relatively longer. Also LT$_{50}$ values was shortened in high concentration and young larva. LT$_{50}$ values are dependent on the temperature, viral concentration and larval instar.

  • Pathogenicity of Spodoptera exigua nuclear polyhedrosis virus (SeMNPV) against the host insect and 8 species of lepidopterous insects and cross infection of baculoviruses to third instar of S. exigua larvae were studied to determine as a biocontrol agent for S. exigua. The median lethal concentrations($LC_{50}$)of the SeMNPV to egg mass was $2.855\times10^5$ PIBs/ml and higher than that to the larvae of S. exigua. Mortality of the SeMNPV in third ins tar larvae was more increased than that in first and fifth instar of S. exigua larvae by 1.16 and 4.11 times, respectively. The median lethal times($LT_{50}$) to $1.56\times10^6$ PIBs/ml was in the range of 4.25 to 5.04 days. Infectivity of the SeMNPV against eight species of lepidopterous insects was showed only in the host insect, S. exigua. Autographa cali/ornica MNPV, Mamestra barassicae MNPV, and Trichoplusia ni MNPV were cross-infected to third instar of S. exigua larvae among ten of baculoviruses tested.

  • Three viral insecticides were differently formulated with a nuclear polyhedrosis virus isolated from Spdodoptera litura by addition of feeding attractant, anti-precipitate of polyhedra, spreading agent, and UV-protectants. Sucrose was effective for attraction of larval feeding to increase the mortality and for protection of polyhedra from inactivation by sunlight when added 1% to 5% of sucrose solution to the formulations. Contents of additives to the formulations were 0.5% in polyvinyl alcohol to prohibit the precipitation of polyhedra and 0.1% in Triton X-100 to spread and wet the formulations to the plant. Inactivation of the virus under sunlight was decreased when added 800g of white carbon to 100 L of water in the white carbon formulation and 30% of molasses to the molasses's. In the formulation of white carbon and molasses mixtures, activation of the virus was increased when mixtured 500g of the former with 10% of the latter. Three formulations were persisted their pathogenicity more than 95% of mortality at 3 days p.i. Encapsulation of the polyhedral surface was more distinctively coated with the carbon and showed more effective in the residual effects of the white carbon than others, but the molasses more attractive for larval feeding.

  • Biochemical characteristics of Spodoptera uigua nuclear polyhedrosis virus (SeNPV) isolated in Jinju were studied. SeNPV contained a number of nucleocapsids within a viral envelope embeded in polyhedra. The polyhedral protein of SeNPV was composed of a single polypeptide with a M. W. of 30kd. Double-immunodiffusion test showed that the polyhedral protein of SeNPV had common antigenic determinants with SINPV and BmNPV. Virion proteins of SeNPV were resolved into 49 polypeptides by silver staining after SDS-PAGE. The approximate genome size of SeNPV by restriction endonuclease analysis was 1l0kb.

Effects of Field Application of Spodoptera litura Nucleopolyhedrovirus to Control S. litura in Chrysanthemum (국화 포장에서 담배거세미나방 방제를 위한 핵다각체병바이러스 살포효과)

  • This experiment was conducted to investigate control effect of Spodoptera litura using nucleopolyhedrovirus in chrysanthemum seedlings of open field and plastic house. Values of LT$_{50}$ of treatment 1.0$\times$10$^{6.8}$ PIBs/ml were 6.2-5.1 days in open field and 6.9-5.4 days in plastic house. Values of LT$_{50}$ and LT$_{95}$ were shorter in open field than in plastic house. Cumulative mortality was 100% in 1.0$\times$10$^{7.8}$ PIBs/ml and also higher in open field than in plastic house. In chrysanthemum field, organic synthetic insecticide (endosulfan EC) killed S. litura larvae in 2 days after application. Motality of S. litura larva inoculated with S. litura nucleopolyhedrovirus (SINPV) 1.0$\times$10$^{6.8}$ PIBs/ml was found out from 4 days after application and maintained during 14 days. Protection values of with SINPV 1.0$\times$10$^{8}$ PIBs/ml after 16 days were 94.0% and 89.4% in open field and plastic house, respectively, and those of endosulfan 350 ppm were 91.4% and 88.6%, respectively.
