Effcts of Sophorae fructus on Antioxidative Activities and Lipid Levels in Rats (괴각(Sophorae fructus) 추출물이 흰쥐의 항산화 활성 및 지질농도에 미치는 영향)

  • This study was carried out to investigate the effects of Sophorae fructus extracts on antioxidative potential, free radical generation and the lipid levels in rats. Sprague-Dawley (SD) rats were divided into 2 groups of either AIN-76 diet (control group) or modified AIN-76 diet with 0.5% Sophorae fructus extracts for 4 weeks. The result were as follows: body weight gain and feed efficiency ratios were not significantly different in both groups. Antioxidative potentials were significantly increased in the group fed Sophorae fructus extracts compared to control group; however, there was no difference in free radical generation. Organ (heart, kidney, liver, spleen) weights in rats were not different in both groups. Total cholesterol level in Sophorae fructus group was significantly decreased compared to control group. Serum HDL-cholesterol level in Sophorae fructus group was significantly higher than control group. Serum phospholipid and triglyceride contents in Sophorae fructus group were no different from control group. These results suggest that supplementation of Sophorae fructus extracts may beneficially contribute to the effects of antioxidative potential and lipid levels in rats.

Antioxidant Effects of Sea Tangle Added Korean Cabbage Kimchi in Vitro and in Vivo (다시마를 첨가한 배추김치의 항산화 효과)

  • The antioxidant effect of Korean cabbage kimchi containing 20% of sea tangle (SK) was studied in the rats fed with high fat diet (HFD) for 8 weeks. The rats (n=40) were divided into four experimental groups as a high fat diet group (HFD), HFD supplemented either with Korean cabbage kimchi used as experimental control (HCK), with SK (HSK), or with J-kimchi (HJK) that was purchased at the local market. The amount of kimchi supplemented was 10%. DPPH radical scavenging activities of SK were significantly higher than those of CK. Kimchi suppressed the hepatic lipid peroxidation significantly, especially by HSK (p<0.05). Inhibition of thiobarbituric acid reactive substances (TBARS) formation in HSK was the greatest among the kimchi groups (p<0.05). The activities of $Cu{\cdot}Zn$-superoxide dismutase (SOD), Mn-SOD and catalase decreased significantly (p<0.05) by kimchi supplementation. SOD and catalase activities of HSK were found to be the lowest among the kimchi groups. The decreased enzyme activity in kimchi group might be due to the less amount of lipid peroxides produced in the rats fed kimchi diet. The lowest antioxidative enzyme activities observed in HSK were in line with those of hepatic POV and TBARS of HSK. Our findings confirmed that kimchi acted as an antioxidant in the high fat fed rats and its antioxidant effect was significantly increased by the addition of sea tangle.

Antioxidative Activity and Related Compounds of Apple Pomace (사과박의 항산화 활성 및 항산화 성분)

  • To enhance the utilization of apple pomace for the functional food resources, we analyzed the useful components and examined the antioxidant activity of apple pomace. The contents of total dietary fiber, total flavonoid, total phenolic acid and vitamine C were 55.56%, 458 mg%, 1048 mg% and 19.8 mg%, respectively. Protocatechuic acid, cinamic acid, caffeic acid, ferulic acid, syringic acid, vanillic acid and phydroxybenzoic acid were identified in the apple pomace extract by GC-MS. Phloridzin and quercetin-3-glucoside were identified in the apple pomace extract by HPLC. Ethyl acetate fraction showed the highest antioxidative ability by ferric thiocyanate method. Malondialdehyde(MDA) formation in normal rat liver tissue also showed the lowest in ethyl acetate fraction.

The Effects of Pulsed Electromagnetic Fields on Blood components, Antioxidant enzymes and Reactive Oxygen in Hyperlipidemic Rats (맥동전자장이 고지혈증 흰쥐의 혈액 성분, 항산화 효소 및 활성 산소에 미치는 영향)

  • The present study was aimed to investigate the effects of the application of pulsed electromagnetic fields on the blood components associated hyperlipidemia, antioxidant enzymes and reactive oxygen. The subjects were divided into three groups: General Diet, High fat Diet, High fat diet and pulsed electromagnetic fields. Pulsed electromagnetic field was applied with pulsed electromagnetic energy therapy equipment. The Glucose, free fatty acids, triglycerides, cholesterol and insulin was used to measure hyperlipidemia-related blood components, and the GSH, GRD, XO and MDA was used to measure antioxidant enzymes and reactive oxygen. The applied pulsed electromagnetic field has improved the concentrations of blood components and increasing GSH and GRD, that were decreased due to a high-fat diet and by reducing the increased MDA and XO to a level of general diet group. Therefore, the applied pulsed electromagnetic field can be the effective treatment for changes in the blood composition of hyperlipidemic rats, enhancement of antioxidative activity and the inhibition of reactive oxygen.

Antioxidant effects of Geagibokrounghwan in experimental mouse fed cholesterol (고(高)Cholesterol 식이 생쥐에 미치는 계지복령환의 항산화효과(抗酸化效果))

  • This study was made to investigate the antioxidative effects of Geagibokrounghwan on the hepatic and renal lesion induced by cholesterol in mouse. The normal group was fed basal diet and water ; control groups were fed basal diet containing 0.5% of cholesterol ; test groups were fed the Geagibokrounghwan extract($10m{\ell}/kg$) after fed basal diet containing 0.5% of cholesterol for 6 weeks. In the liver and kidney of control group, lipid peroxidation(LPO) was significantly increased, however, the activities of superoxidation dismutase(SOD) and catalase and the amount of glutathion(GSH) were significantly decreased. In the liver and kidney of test group, lipid peroxidation(LPO) was decreased significantly as compared with control group. Contrary to this, the activities of superoxide dismutase(SOD), catalase and the amount of glutathion(GSH) were significantly increased. These results indicate that Geagibokrounghwan revealed the antioxidant effects, which may reduce the hepatic and renal damage induced by cholesterol in mouse.

Effects of Fatty Acids and Vitamin E Supplementation on Antioxidant Systems in the Liver and Serum of the Second Generation Rat (지방산 및 비타민 E 보충 식이가 제2세대 흰쥐 간조직과 혈청의 항산화체계에 미치는 영향)

  • Effects of dietary fatty acids and vitamin E on antioxidant system were studied in rat liver and serum. Sources of dietary fat (10 wt%) were safflower oil (SO) poor in $\omega$3 fatty acid and mixed oil (MO) with computer-adjusted fatty acid ratios (AA/DHA=1.4, $\omega$6/$\omega$3=6.3, P/M/S=1.0/l.5/1) with (ME) and without (MO) vitamin E (500 mg/kg diet). Rats were fed the three kinds of diet from 3~4 wks prior to the conception. At the age of 3 and 9 wks of the second generation rat, antioxidant vitamins and glutathione peroxidase (GSH-Px), superoxide dismutase (SOD) activities were measured in the liver and serum. The concentrations of $\beta$-carotene were lower in ME than in MO and SO in the liver at the age of 3 wks. It seemed that vitamin E has an inhibitory action on the uptake of $\beta$-carotene or acts as a preferred antioxidant to $\beta$-carotene. The concentrations of lycopene were lower in SO than in MO in the liver at the age of 3 wks. The concentrations of cryptoxanthin showed no significant changes within groups. The activities of GSH-Px tended to increase in ME compared to MO and the ratios of SOD/GSH-Px tended to decrease in ME compared to MO in the liver at the age of 3 weeks. The activities of antioxidant enzyme at the age of 3 weeks and 9 weeks were similar. This suggested that the activity level of antioxidant enzymes reached to the adult level at the age of 3 weeks which is the end point of lactation period.

Effects of Water Extract from Orostachys japonicus on Lipid and Antioxidant Metabolism in the Liver of Obese Mice (와송이 고지방 식이로 유도된 비만 쥐의 간내 지질 및 항산화 대사에 미치는 영향)

  • The purpose of this study was to investigate the changes in the liver lipid and antioxidant metabolism in obese rats following ingestion of Orostachys japonicus (OJ) hot water extract. The rats were fed a high fat diet for 8 weeks to induce obesity. In the experimental group, 2.5% or 5% OJ hot water extract was added to the diets for 6 weeks. To determine the changes in the liver lipid and antioxidant metabolism in obese rats, the livers were collected and weighed, lipid content was measured, and antioxidant enzyme activity was determined. A significant decrease in liver weight was observed in the group fed with OJ hot water extracts and decreased lipid accumulation was observed in the morphological observation of liver tissue. Total cholesterol and triglyceride contents were also decreased. Both OJ extracts significantly decreased 2,2-diphenyl-1-picrylhydrazyl radical and intracellular reactive oxygen species levels. Further, the levels of the antioxidant enzymes superoxide dismutase and catalase were significantly increased in rats treated with OJ extracts. These results suggest that the intake of OJ hot water extracts may improve liver lipid metabolism by activating antioxidant enzymes.

Effects of Isoflavone-Rich Bean Sprout on the Lipid Metabolism of the Ethanol-Treated Rats (고 Isoflavone 콩나물이 만성 에탄올 투여 흰쥐의 지질대사에 미치는 영향)

  • The present study evaluated the effects of isoflavone-rich bean sprout on the lipid metabolism in ethanol-treated rats. Fifty male Sprague-Dawley rats were randomly divided into five groups: normal control, alcohol control, low soybean sprout+ethanol (low SS), high soybean sprout+ethanol (high SS) and isoflavone extract+ethanol (IE). They were fed experimental diets based on Lieber-DeCarli liquid diet for 40 days. Body weight, food intake and feed efficiency ratio (FER) of ethanol-treated groups were significantly suppressed compared with that of the normal control group. Among the ethanol-treated groups, high SS group showed in significant increase in the body weight, food intake and FER. Supplementation of isoflavone-rich soybean sprout powder or isoflavone extract significantly decreased plasma triglyceride (TG), total cholesterol (TC), atherogenic index (AI) and increased the ratio of HDL-cholesterol to TC. Supplements also significantly decreased total lipid, TG and TC in liver tissue compared with that of alcohol control. There was a significant decrease in hepatic lipid peroxidation products in IE group compared with other ethanol-treated groups. This results suggest that supplementation of isoflavone-rich bean sprout powder may exert beneficial effects on lipid metabolism in chronically alcohol-treated animals by improving lipid profiles in plasma and liver tissues.
