Screening of Antimutagenic Activities from Cereals and Beans Including Rice (쌀을 포함한 곡류 및 두류의 항변이원 활성의 검색)

  • We have established the quantitative method for assay antimutagenic activity from natural products using SOS chromotest technique. Establishment of the method in this study makes it possible to numerize antimutagenic activities from samples in term of the sample amount required for 50% inhibition to mutagenic activity induced by the chemical mutagen under the standard assay condition. Antimutagenic activities of rices from different cultivars as well as other cereals were assayed through this method. The results revealed that antimutagenic activities of mutant cultivar, Suwon 393(Hyangdo) and Sanghaehyanghyulla(Jado), were higher than Chuchung which mainly consumed for steamed rice, and also indicated that antimutagenic activities of cereals, such as job'tear, buckwheat, small red bean, black bean were generally higher than that of brown rice.

  • The objective of this study was to screen the antimutagenicity of yam enzymatic browning reaction product (YEBRP), mucopolysaccharide and dietary fiber from yam to the mutagen of sodium azide and 2-aminoflourene (2-AF). Antimutagenicity of YEBRP on the mutagenicity of sodium azide showed no difference compared to control without YEBRP but that of 2-AF was high In all substrate. (P<0.01) On the mutagenicity of sodium azide and 2-AF, antimutagenicity of mucopolysaccharide and dietary fiber were high (p<0.01) in $\alpha$-cellulose and hemicellulose, Antimutagenicity of u-cellulose on the mutagenicity of 2-AF was high at 5 hours reaction time but that was decreased as the reaction time increased.

  • 암을 유발시키는 원인은 생물, 물리, 화학적 요인 등 여러 가지가 있으나 근자에 들어 산업후에 따르는 환경오염의 증가는 화학적 원인에 의한 발암가능성을 증대시키고 있다. 이러한 화학적 인자의 발암 기전 중 많은 부분이 유전자에 상해를 일으킴으로써 암을 발생시킨다고 알려져 있다. 그러므로 항암실험을 수행하였던 파푸아뉴기니아산 열대식물 13과 23종 부위별 36시료에 대하여 항변이원성 검색실험을 통하여 유전독성을 가진 화학적 요인에 의한 발암 가능성에 대한 예방 효과를 알아봄으로써 발암에 대한 예방약물 개발의 기초 자료를 제공하고자 본 실험을 수행하였다.

Correlation of Lipid Soluble Compounds of Colored Rices and its Mutagenicity, Antimutagenicity and Antioxidativity (유색미 지용성 성분들의 변이원성.항변이원성.항산화 효과에 대한 상관관계)

  • The fractions extracted with methylene chloride from colored rice seeds of 19 cultivars were prepared to examine the correlationties of both antimutagenicity and antioxidiativity with physiological functionalities. The data revealed a positive correlation of the antimutagenicity with the content of $(3.{\beta},22Z)-Acetate-stigmasta-5,22-dien-3-ol$, 24-Oxocholesterol acetate, 6(E),8(E)-Heptadiene, and Eicosane. For antioxidativity, electron donating ability to DPPH radicals exhibited a positive correlation with the content of (24R,25S)-Aplysterylacetate, however, negative correlations were found between scavenging activity toward hydroxyl radicals and the content of either Tetradecanoic acid or Methyl ester-hexadecanoic acid, $(3.{\beta},24S)-Stigmast-5-en-3-ol$, respectively. In addition, a positive correlation was detected between the inhibitory effect of the fractions and the content of $(3.{\beta},24S)-Stigmast-5-en-3-ol$, however, a negative correlation with the content of 3',3'-Dimethylspiro [acridane-9,1'-indane], $(3.{\beta})-24-Methylene-9,19-cyclolanostan-3-ol$, and some other compounds was observed, respectively.

Desmutagenic Effects of Extracts Aloe with Different Solvents (알로에 용매별 추출물의 항변이원성)

  • Desmutagenic effects of water-soluble and water-insoluble fractions from Aloe vera and Aloe avoresence with different solvents against the mutagenicity of 4-nitro-o-phenylenediamine(NPD) were investigated. Desmutagenic effects of water-insoluble fractions from Aloe vera and Aloe avoresence against NPD-induced mutations in TA97 and TA98 strains were very high compared with water-soluble fractions: whereas water-soluble fractions from two kinds of Aloes showed the inhibition effects of 4∼56% water-insoluble fractions showed the activities of 54∼100% for Aloe vera and 74∼100% for Aloe avoresence. Especially, the addition of 20 $\mu$ι of water-insoluble fractions per plate showed high activities up to 90% in TA97 and TA98 strains.

Antioxidative and Antimutagenic Activity of Ethanolic Extracts from Giant Embroynic Rices (거대배아미 에탄올 추출물의 항산화활성 및 항변이원성)

  • 70% ethanolic extracts were prepared from the three mutant rice cultivars with giant embryo termed Shinsunchal-giant embryonic rice, Whachung-giant embryonic rice and Nampung-giant embryonic rice, and its antioxidative and antimutagenic properties were evaluated and compared. For analysing antioxidativity, various antioxidative indices, such as electron donating ability to DPPH radical, scavenging capacity to hydroxyl radicals generated by Fenton reaction, scavenging capacity to superoxide radicals generated by HPX/XOD system, inhibitory effect on autoxidation of linoleic acid and inhibitory effect on membrane lipid peroxidation derived from rabbit erythrocyte ghost, were determined. For analysing antimutagenicity, suppressive effects on mutagenesis induced by the chemical mutagen, mitomycin C, were measured using E. coli PQ 37 as a indicator cell. The results showed that for both antioxidativity and antimutagenicity the giant embryonic rices were more effective compared to the general cooking rice, Among the giant embryonic rice cultivars, Nampung-giant embryonic rice tended to be most effective, showing its scavenging activity to DPPH radical, superoxide radical and hydroxyl radical, and inhibitory activity to lipid peroxidation was 2,3-, 3,3-, 1.7-, and 2.5-fold greater than those of normal rice, respectively.

Screening of Mutagenicity and Antimutagenic Activity against Chemical Direct Mutagens of Ethanolic Extracts from Colored Rice Bran (유색미 에탄올 추출물의 변이원성 및 화학적 직접변이원에 대한 항변이원 활성 검정)

  • The cytotoxic, mutagenic and antimutagenic activities against chemical direct mutagens such as mitomycin C, 4-nitroquinoline-N-oxide, 2,4,7-trinitro-9-fluorenone of the 70% ethanol extracts of 29 colored rice varieties and chuchung as a control were examined. The results obtained using authentic alkaline phosphatase activity as a growth representative of the indicator cell E. coli PQ 37 demonstrated that the extracts of 13 kinds of colored rice varieties including Jumlalocal and Jumlalocal-1 showed strong toxic effect on the cell growth. However the extracts of DK 1, SC-5, LK 1A-2-12-1-1 and wx 139-3-64-20-3-1 seemed to have stimulatory effects on the cell growth. The mutagenicity and antimutagenicity of the colored rice varieties were screened using SOS chromotest. The mutagenic activity was detected from Jumalocal-1, IR 17491-5-4-3-3 and Jumlalocal. On the contrary, 7 samples including LK 1-3-6-12-1-1, Parnkhari 203, Jumlalocal, wx 139-3-64-20-3-1, Muthumanikam, HP 883-1-1-1-B-1-1 and Jumlalocal-1 were shown to have antimutagenic acitivities against the chemical direct mutagens used in this study.

Screening of the Mutagenicity and Antimutagenicity of the Hot Water Extracts from Medicinal Plants (한약재 열수 추출물의 변이원 및 항변이원 활성의 검색)

  • The mutagenicity, antimutagenicity, and cytotoxicity of the hot water extracts of 130 medicinal plants were examined. The results obtained using authentic alkaline phosphatase activity as the growth representative of the indicator cell, E. coli PQ37, demonstrated that the extracts of 14 medicinal plants including, Forsythia koreana Nakai, showed strong toxic effect on the cell growth, however, the extracts of 22 medicinal plants, including Phlomis umbrosa Turcz., were shown to have stimulatory effect on the cell growth. The mutagenicity and antimutagenicity of the medicinal plants were screened using SOS chromotest. The mutagenic activity was detected from 36 kinds of the samples, including Forsythia koreana Nakai and Coptis japonica Nakai. On the contrary, 12 medicinal plants, including Crataegus pinnatifida Bunge, were shown to have a significant antimutagenic activity.
