Isolation of a Bacterium with Potent Antibacterial Activity against Swine Atrophic Rhinitis Bacteria and Antibacterial Activities (돼지 위축성 비염균에 대한 항균성을 나타내는 세균의 분리 및 항균활성)

  • A bacterium(Y-5) which has potent antibacterial activity against swine atrophic rhinitis bacteria (Bordetella bronchiseptica, Pasteurella multocida) was isolated from soil and identified as a strain of Bacillus sp. Y-5 upon investigation of the morphological and physiological characteristics. The culture broth obtained from incubation of the Y-5 strain at $30^{\circ}C$ for 21 h in tryptose-bouillon agar medium (pH 6.0) showed active antibacterial activity against Bordetella bronchiseptica and the culture broth that of $30^{\circ}C$, pH 6.5, 18 h showed active antibacterial activity against Pasteurella multocida.

  • 물/유기용매의 혼합용액에 키토산을 분산시키고 이 현탁액에 글리시딜 트리알킬 암모늄염, 글리시딜 아릴염 등의 에테르 시약을 상온에서 현탁상태로 반응시켜 4급화 키토산을 제조하는 새로은 방법을 개발하였다. 이 방법으로 합성한 결과 수득율이 90%이상이었으며 후처리과정이 필요없었다. 4급화 키토산의 균에 대한 최소저지농도(MIC)를 조사한 결과 E. coli O157:H7 균주의 MIC는 0.007%-0.01%(70-100ppm)로 추정되었고 V. vulnificus 균주의 MIC는 0.005%(50ppm)정도이고 V.vulnificus 균주의 MIC는 0.007%(70ppm)이었다. 작물에 대한 항균성을 알아보기 위하여 4N-키토산의 항균 활성만이 검정되었는데, 4N-키토산은 고추 역병균인 P. capsici에 대하여 가장 높은 항균 활성을 보여 4,000 ppm에서부터 키토산 무첨가 대조구에 비하여 항균활성을 보였으며 P. capsici보다는 약하지만 잿빛곰팡이병균 B. cinerea에 대하여도 약간의 항균 활성을 보이고 있어, 16,000 ppm에서는 항균서이 뚜렷이 나타났으나 고추 탄저병균인 C. gloeosporioides에 대해서는 16,000 ppm 이라는 높은 농도에서도 항균 활성을 전혀 보이고 있지 않았다.

Antibacterial Activity of actobacillus sp. KJ-5 Isolated from Pig Feces (돼지분변에서 분리한 Lactobacillus sp. KJ-5의 항균특성)

  • A lactic aci bacteria producing antibacterial substance was isolated from pig feces. This strain was identified as a genus Lactobacillus, through its morphological, cultural and physiological characteristics. Lactobacillus sp. KJ-5 isolated showed the strong inhibitory effect on the growth of Salmonella paratyphi. The production of antibacterial substance was growth associated form during the batch culture of Lactobacillus sp. KJ-5 and the maximum production was obtained at the culture temperature of $37^{\circ}C$ as well as optimum temperature of cell growth. The antibacterial activity of the filtrate of culture broth was decreased by adjusting the pH 6.2 and was not affected by catalase treatment. The antibacterial substance was partially purified by methanol and acetone extraction, whtch exhibited three spots in the thin-layer chromatography and one of them showed an antibacterial activity, This substance also showed the maximum absorption of UV at 270nm and an antibacterial activity was completely inactivated by the treatment of proteolytic enzymes.

Quality Characteristics of Unshiu Orange and Pear Packaged with Paper Incorporated with Antimicrobial Agents (항균소재를 함유한 포장재로 포장한 밀감과 배의 저장중 품질 특성 변화)

  • In order to help the preservation of the unshiu orange and pear, antimicrobial paper incorporating grapefruit seed extract and zeolite was applied to pack fruits. Unshiu orange was packed in a box (24${\times}$24${\times}$22 cm) attached with antimicrobial paper and then stored respectively at l$0^{\circ}C$. Pears were wrapped individually before storage at l$0^{\circ}C$. During the storage, weight loss, pH, total acidity, soluble solid content, microbial load and decay were measured as quality indices. Steady pH increase in unshiu orange was observed to slightly decrease total acidity during the storage with little difference between the packaging treatments. The microbial loads of total aerobic bacteria, and yeast/mold counts were suppressed during storage by the antimicrobial paper packaging, which also contributed to reducing the decayed unshiu orange. Limited reduction of total aerobic bacteria and yeast/mold counts was observed only for initial storage period for the pears wrapped with 9 and 12% antimicrobial agent-added papers. Antimicrobial paper was useful for the reduction of microbial load in unshiu orange and pear without other quality deterioration.

  • 극한의 추위를 견디기 위한 내한성 코팅 직물의 개발은 활발히 진행되었으나, 항균성에 대해서는 활발한 개발이 이루어지지 않고 있다. 본 연구는 내한성 코팅 직물에 천연항균제를 처리하여 물성의 변화를 알아보고자 한다. 현재 시판되고 있는 내한성 코팅 수지에 해조류에서 추출한 천연항균제를 처리하여 상용성을 알아보았다. 내한성 코팅수지에 천연항균제를 처리하면, 내한성 코팅 수지의 고유 물성은 유지되면서 항균성이 나타나는 것을 알 수 있었다.

  • Recently sewage facilities mainly consisted of concrete structures are being deteriorated seriously by biochemical reaction originated from sulfur-oxidizing bacteria. To prevent biochemical corrosion of the sewage concrete, antibiotics which inhibit growth of sulfur-oxidizing bacteria have to be developed and applied necessarily. In this study, we are going to introduce technology which biochemical corrosion of sewage facilities concrete could be controlled effectively by antibiosis of antimicrobial concrete

  • Deterioration of food is in general caused by the presence of microorganisms and chemical compounds of food itself. There exists antimicrobial compound in the food, however, addition of food antiseptics, additives, or physico-chemical processing is a common practice. The safety of artificial chemical antiseptics became a serious public concern, therefore, new natural antiseptic compounds are in need to be developed. We have isolated a new natural antifungal protein (KBS-B2) from Astragal seed through ammonium sulfate precipitation and column chromatography using FPLC Mono-S and Superose 12HR. The purified protein inhibited growth of Candida albicans, and spore germination of food spoiling fungi such as Aspergillus ochraceus, Penicillium expensum, P. digitatum and Botrytis cineria. Antifungal effect of the KBS-B2 protein could be directly assayed by bioautography overlaying the fungal spores on the electrophoresed acrylamide gel. The comparison of N-terminal amino acid sequences of the KBS-B2 with known antifungal protein revealed that had 50% homology to thaumatin and zeamatin like proteins.

  • Bacterial diseases with mixed infection have recently occurred at land-based flounder farms in Korea. Thus, single antibacterial is not effective for therapy of mixed bacterial diseases of fish because of their different causative bacteria. The purpose of the present study was to obtain basic data for positive usefulness of a combination of antibacterials used for synergism to mixed bacterial diseases of fish. Snergistic interaction in combination of antibacterials was determined by in vitro antimicrobial activity against selected fish-pathogenic bacteria, Vibrio anguillarum, Edwardsiella tarda, Streptococcus sp., Staphylococcus epidermidis, on the basis of Checkerboard assay using fractional inhibitory concentration (FIC) indices. Synergistic interactions were observed in combinations of (oxytetracycline HCL+lincomycin), (tetracycline HCL+florfenicol), (oxytetracycline HCL+florfenicol) against V. anguillarum, (sodium nifurstyrenate+florfenicol), (tetracycline HCL+florfenicol), (sodium nifurstyrenate+oxolinic acid), (oxytetracycline HCL+florfenicol) against E. tarda, (ciprofloxacine+oleandomycin), (oxytetracycline HCL+oleandomycin), (tetracycline HCL+oleandomycin), (oxytetracycline HCL+lincomycin), (oxytetracycline HCL+spiramycin), (oxytetracycline HCL+erythromycin), (doxycycline HCL+oleandomycin), (tetracycline HCL+spiramycin) against Streptococcus sp., and (ciprofloxacine+erythromycin), (florfenicol+erythromycin), (doxycycline HCL+oleandomycin), (ciprofloxacine+oleandomycin) against S. epidermidis.

  • A new bacteriocin produced by Bacillus subtilis cx1, was partially purified and characterized. The bactericoin from B. subtilis cx1 was stable in the range of pH 2.5-9.5. B. subtilis csx1 retained its antimicrobial activity to long-term exposure at $-20^{\circ}C$ and $-70^{\circ}C$. However, B. subtilis cx1 was inactivated completely within 15 min over $60^{\circ}C$ and lost 50% of its antimicrobial activity within 15 min at $50^{\circ}C$, B. subtilis cx1 was inactivated by protease, trypsin, proteinase K and carboxypeptidase, which indi-cates its protein nature. Direct detection of the antimicrobial activity on Tricine -SDS-PAGE suggested an apparent molecular mass of about 9,500 dalton.

Antibacterial Effects of Leek and Garlic Juice and Powder in a Mixed Strains System (복합균주에 대한 부추와 마늘 생즙 및 가루성분의 항균특성)

  • The aim of this study was to evaluate the antimicrobial activities of leek and garlic extract juice and powder against Escherichia coli, Salmonella enteritidis, and Staphylococcus aureus in a single strain and a mixed strains system. Garlic juice and powder showed higher antimicrobial activity against Sta. aureus than that of E. coli or S. enteritidis. The antimicrobial activities of the leek and garlic powders decreased with increasing temperature and time, but stabilized at various pHs. The antimicrobial effects of mixtures of various ratios between the leek and garlic extracts increased with an increase in garlic extract content. No synergistic effects by the leek and garlic mixtures were observed. In a mixed strains system, the antimicrobial effects of leek and garlic powder were similar to those in a single strain system.
