Mitochondrial DNA Diversity of Korean Ogol Chicken

  • 한국재래오골계는 천연기념물로 등록이 되어 있어 세계의 중요한 유전자원 중 하나이다. 현재 한국에서 사육되어 있는 오골계의 유전적 특성을 규명하기 위하여 미토콘드리아 DNA의 변이를 이용하여 계통 분석을 실시하였다. 총 31 마리의 한국재래오골계가 이 분석에 이용되었으며 10개의 haplotypes이 관찰되었다. NJ 방법으로 만들어진 계통도 분석을 통하여 이미 닭에서 알려진 A부터 C의 lineage를 포함하는 것으로 보아 한국 재래오골계는 아직도 높은 유전적 다형성을 유지하고 있음을 알 수 있었다. 이 연구 결과는 한국 재래오골계의 육종 및 보존 계획을 세우는데 유용하게 이용될 수 있을 것으로 사료된다.

Microbial Hygienic Status of Poultry Meats and Eggs Collected at the Public Markets in Seoul and Kyung-gi Regions in 1996 (1996년도 서울${\cdot}$경기지역에서의 시판계육과 계란에 대한 미생물학적 위생실태)

  • To determine the actual hygienic status of domestic chicken meats sold in public markets (conventional markets and department stores), microbial contamination levels (Total cells, Coliforms and Staphylococcal cells) and zoonotic pathogens (Salmonella species, Campylobacter species, Listeria species, and Staphylococcus aureus) isolation tests were conducted. Chicken meats and eggs tested were collected from the conventional markets (Si-Jang) and department-stores located in Seoul and Kyung-gi regions in 1996. In total cells and coliforms contamination tests, chicken meats sold in department stores were much lesser contamination status than those of Si-Jang, but staphylococcal cells level was much more higher than that of conventional markets. Salmonella isolation frequency was investigated as $68.8\%$, but Campylobacter jejuni and Listeria monocytogenes isolation frequency were appeared both $64.0\%\;and\;63.3\%$. In case of eggs sold in public markets, one of S. gallinarum strain $(0.7\%)$ was isolated only on the egg-shell part among the four-hundred and fourty-six. In comparison with foreign imported chicken meats, there were no big differences in microbial contamination status. On the other hand, both Salmonella and L. monocytogenes were isolated only in the chicken wings from Korea and China, but not from U.S.A. This data suggest that more hygienic control system in order to produce the safe and hygienic chicken meats and eggs is need in our country as soon as possible.

산란형질에 대한 한국 재래닭 계통간 교배종의 잡종강세 및 결합능력 추정

  • This study was conducted to obtain some genetic information for developing superior Korean native chickens. Heterosis and combining ability effects were estimated with 1,080 progenies of full diallel crosses of 3 strains in Korean Native Chicken. The traits of egg productivity were Investigated, and heterosis effect, general combining ability, specific combining ability and reciprocal effect were analyzed model I of the Griffing's method 1. The general performance were 147.0 days in age at first egg laying, 71.8 eggs in egg numbers to 270 days, 50.1g in egg weight at 270 days, 75.6 in egg shape index, 356$\mu\textrm{m}$ in egg shell thickness, and 4.27kg/$\textrm{cm}^2$ in egg shell strength, respectively. According to result, heterosis effect revealed comparatively high in age at first egg laying, egg numbers, egg weight, and egg shell strength. The effects estimated highly in general combining ability for age at first egg laying, egg numbers, egg weight, egg shape Index and egg shell thickness, and in specific combining ability were egg numbers, egg weight and egg shell thickness, and in reciprocal effects for egg numbers, egg shape index, and egg shell thickness. This result could be used for developing Korean native chicken.

Studies on the Post-hatching Development of Sertoli and Leydig Cells in the Testis of Korean Native Chickens (한국재래 닭 고환 지지세포와 간질세포의 부화 후 발달에 관한 연구)

  • Morphometric changes in testicular Sertoli and Leydig cells from hatching to adulthood were studied using Korean native chickens of 1, 2, 4, 6, 8, 10, 12, 14, 16, 18, 21, 24, 28, 32, 44, 52 and 64 weeks (n=13 chickens per group) of age. The objective of this study was to understand the developmental phase of the Sertoli and Leydig cells with age. Testis of chickens was fixed by whole body perfusion using a fixative containing $2.5\%$ glutaraldehyde in cacodylate buffer, processed and embedded in Epon-araldite. Using 1 Um sections stained with methylene blue-azure II, qualitative and quantitative (stereological) morphological studies were performed. The average volume of a testis of 1 week old Korean native chickens was determined as $0.148\;cm^3$ and the parameter increased linearly from 1 week to 21 weeks days $(28.86\;cm^3)$, and did not change from 21 weeks to 64 weeks. The volume density of the seminiferous tubules increased with age from $32.6\%$ at week 1 to $92.89\%$ at week 64. The volume density of the interstitium represents $67.4\%$ of the testicular parenchyma at week 1. This proportion progressively diminished during development to reach a value of $7.11\%$ at week 64. The volume density of the Leydig cells decreased almost linearly from 1 week $(4.9\%)$ to 14 weeks $(1.7\%)$ and remained unchanged thereafter. In contrast, the Sertoli cells occupied a volume density of $3.4\%$ at week 1, increased progressively up to 18 weeks of age $(10.79\%)$ and remained unchanged thereafter. The absolute volume of the Leydig and Sertoli cells per testis increased significantly from week 1 to week 21 but did not change significantly from week 24 to week 64. The number of Leydig cells per testis increased almost linearly from 1 week to 21 weeks, remained high and unchanged with advancing age. The number of Sertoli cells per testis increased gradually with age from 1 week to 14 weeks and remained unchanged thereafter.

Genotype Analysis of the Major Histocompatibility Complex Region in Korean Native Chicken (한국 재래닭의 MHC 영역 유전자형 분석)

  • The chicken major histocompatibility complex (MHC) is known to be associated with disease resistance and susceptibility to several pathogens. The microsatellite marker LEI0258 is physically located between the BG and BF of MHC region and variations near this marker have been well documented. In this report, the LEI0258 marker was used to find specific alleles for the Korean native chicken. The MHC haplotype was analyzed by PCR screening and sequencing of LEI0258 region in four different breeds including black Korean native chicken, brown Korean native chicken, Cornish and Rhode island red. The serologically same MHC haplotypes showed the differences in repeat numbers, a few indels or single nucleotide polymorphisms by sequencing analysis. Even though we could not identify specific alleles for Korean native chickens, the genotypes analyzed in these breeds can give valuable information for the relationships with disease resistance and establishment of breeding strategies for the Korean native chicken.

Effects of Dietary Energy, Protein on Growth and Blood Composition in Cross-bred with Korean Native Chicks (사료 단백질 및 에너지 수준이 재래닭의 성장과 혈액의 성상에 미치는 영향)

  • Two experiments were conducted to establish the basic data of feeding system in cross-bred Korean native chicks. A total of seven hundred twenty chicks were replaced the 36 floor pen for the first five weeks with $3{\times}3$ factorial design in Experiment 1. Four hundred eighty of five weeks old chicks were raised from six to ten weeks of age with $3{\times}2$ factorial design in Experiment 2. Dietary ME and CP were formulated to contain 3,000, 3,050, 3,100 kcal/kg and 21, 22, 23%, respectively in Experiment 1 and 3,050, 3,100, 3,150 kcal/kg and 18, 19% in Experiment 2. Weight gain, feed intake were measured and calculated the feed conversion. Blood were collected and analyzed at the end of experiments. In Experiment 1, weight gain showed significantly higher in 3,050, 3,100 kcal/kg treatments than 3,000 kcal/kg treatment (P<0.05), but was not different in CP treatments. Feed intake was statistically high in 3,000 kcal/kg treatment compared with 3,050 and 3,100 kcal/kg ones (P<0.05), and more increased in 21% CP treatments compared to that of 22 and 23 CP treatment (P<0.05). Feed conversion of birds fed 3,050 and 3,100 kcal/kg diet showed much lower than 3,000 kcal/kg treatments (P<0.05). FCR was signicantly improved (P<0.05) in chicks fed diets containing 21 and 22% CP as compared to that fed 20% CP. Blood protein, glucose, and total cholesterol tended to increase in high energy and diet treatments. Blood HDL was increased as dietary energy increased, whereas LDL increased in low CP treatments. In Experiment 2, weight gain was not consistent between treatments, but more increased in 18% CP treatments compared to that of 19% CP treatment from six to ten weeks old in cross bred chicks (P<0.05). Feed intake was similar to the result of weight gain, but more increased in 19% CP treatment than 18% CP treatment (P<0.05). There were no statistically difference in FCR, but seemed to improve as dietary ME increased. Blood total protein and glucose increased as dietary CP was high, but triglyceride and HDL increased in high versus low ME (P<0.05). The results of these experiments suggested that optimum dietary ME and CP, were 3,050, 3,150kcal/kg and 22, 19% for the first five weeks and second one, respectively.

Comparison of Physicochemical Characteristics of the Meat in Four Lines of Korean Native Chickens (4계통 재래종 닭고기의 이화학적 특성 비교)

  • This study was conducted to compare carcass yield and meat quality among four lines of Korean native chickens (Yeonsan Ogye, Hyunin Black, Hoengseong Yakdak and Hwangbong) and White Leghorn as control. Chickens (N=23, male) were grown under same conditions and slaughtered at 56 weeks old to observe the physicochemical differences in breast and leg meats. The live and carcass weights of Hwangbong were significantly higher than other chickens (p<0.05). Four lines of Korean native chickens, regardless of the part, had higher shear force value than White Leghorn (p<0.05). In addition, the breast meat of Korean native chickens had higher cooking loss and lower water holding capacity than that of White Leghorn. The cooked meat of Hwangbong particularly were more chewy, gummy and cohesive with lower tenderness than other chickens (p<0.05). Oleic acid content which is related to meat flavor was significantly higher in the breast meat of Hoengseong Yakdak (39.6%). The meat of Korean native chickens had lower n6/n3 ratio, in which the breast and leg meat of Hoengseong Yakdak contained the lowest ratio (p<0.05). The meat of Hwangbong and Hoengseong Yakdak were more acceptable than the others according to sensory test (p<0.05).

한국 재래닭의 발생.발육단계별 telomere와 telomerase activity 분석

  • This study was carried out to analyze the amount of telomeres and telomerase activity of several chicken cells. Telomere quantity and telomerase activity were analyzed during organ development, growth and aging in embryonic and adults chicken. Analyzed cells were whole embryos and the cells from brain, heart, liver, kidney, lymphocytes and germinal tissues in Korean Native Chicken. The amount of telomeric DNA was analyzed by quantitative fluorescence in situ hybridization (Q-FISH) techniques using a chicken telomere repeat probe. Telomerase activity was performed by Telomeric Repeat Amplification Protocol (TRAP) assay. In results, telomerase activity was highly detectable in early embryonic cells, germinal cells and kidney cells. Whereas the cells from brain, heart, and liver had gradually down-regulated pattern of telomerase activity. Analyzing the telomere quantities on chicken cells, the amount of telomeric DNA of most chicken cells gradually decreased as growth. From these results, the amount of telomeric DNA was directly affected by telomerase activity. Consequently the telomere quantity and telomerase activity are closely relate to cell differentiation and tissue specificity during developmental and growing stages.

Comparison of Stress Response between Korean Native Chickens and Single Comb White Leghorns subjected to a High Stocking Density (닭의 품종 간 스트레스 반응 정도 비교 분석)

  • With Single Comb White Leghorn (WL) and Korean Native Chicken (KNC) breeds, we compared the stress response with chicken breeds that were subjected to a high stocking density. Stress response was analyzed by the quantity of telomeric DNA, the rate of DNA damage and the expression levels of heat shock proteins (HSPs) and hydroxyl-3-methyl-glutaryl coenzyme A reductase (HMGCR) genes on tissues and blood. The telomere length and telomere shortening rates were analyzed by quantitative fluorescence in situ hybridization on the nuclei of lymphocytes and tissues. The DNA damage rate of lymphocytes was quantified by the comet assay. The expression levels of HSP70, HSP90-${\alpha}$, HSP90-${\beta}$ and HMGCR genes were measured by quantitative real-time polymerase chain reaction in lymphocytes. There was no significant difference between KNC and WL in body weight, weight gain, telomere shortening rate and DNA damage rate. However, the growth rate significantly decreased in chickens raised under high stocking density conditions, as compared to the control group. The telomere-shortening rate, DNA damage and HSPs expression of the lymphocytes were significantly higher in the high stocking density group than the control. The stress condition and breeds had a significant effect on the expressions of HSP70, HSP90-${\alpha}$ and HSP90-${\beta}$ in lymphocytes, except HMGCR. The stress response of WL was higher than that of KNC, as analyzed to the expression of HSP70 and HSP90-${\alpha}$. Therefore, we concluded that the chickens which were exposed to a high stocking density had increased the individual physiological stress response regardless of breeds, and White Leghorns are more susceptible to stress condition than Korean Native Chickens.

  • Changes in the chicken testis from hatching to adulthood were studied in Korean native chickens of 1, 2, 4, 6, 8, 10, 12, 14, 16, 18, 21, 24, 28, 32, 44, 52 and 64 weeks (n=13 chickens per group) of age. The present study was to investigate in more detail the post-hatching development of testis in Korean native chickens. Testes of chickens were fixed by whole body perfusion using a fixative containing 2.5% glutaraldehyde in cacodylate buffer, processed and embedded in Epon-araldite. Using $1{\mu}m$ sections stained with methylene blue-azure II, qualitative and quantitative(stereological) morphological studies were performed. Sperm production was measured by routine technique. The average volume of a testis of 1 week old Korean native chickens was determined as 0.015 g and the parameter increased linearly from 1 week to 21 weeks days (28.9 g), and did not change from 21 weeks to 64 weeks. The volume density of the seminiferous tubules increased with age from 32.6% at week 1 to 92.89% at week 64. The volume density of the interstitium represents 67.4% of the testicular parenchyma at week 1. This proportion progressively diminished during development to reach a value of 7.11% at week 64. Total sperm production per testis increased significantly from 18 weeks to 28 weeks and remained unchanged. Sperm production per 1 g testis increased significantly from 18 weeks to 28 weeks, did not change significantly from 28 weeks to 52 weeks, and declined significantly at 64 weeks of age. The average diameter of the seminiferous tubules gradually increased with age from 1 week $(42.4{\mu}m)$ to 21 weeks $(412.8{\mu}m)$. The length of the seminiferous tubules was 0.34 m at 1 week, increased significantly in subsequent age groups and reached 72.2 m by weeks 64. The stage of germ cell development in seminiferous tubules was classified as 1) spermatogonia $(1\sim8\;weeks)$, 2) spermatogonia and spermatocytes $(10\sim12\;weeks)$, 3) spermatogonia, spermatocytes and round spermatids $(14\sim16\;weeks)$, and 4) speramatogonia, spermatocytes, spermatids and spermatozoa $(18\sim64\;weeks)$. These results clarified the pattern of changes in the testicular development in Korean native chickens from hatching to adulthood as 1) neonatal-prepubertal $(1\sim12\;weeks)$, 2) puberty$(14\sim18\;weeks)$, and adult$(21\sim64\;weeks)$.
