Condition of mycelial culture and inoculum volume of spawn on cultivation of Agrocybe cylindracea (버들송이의 균사배양조건 및 최적 접종량 설정)

  • Studies were made to optimize the media composition and cultural condition for mycelial growth of Agrocybe cylindracea. Sawdust spawn of media composition for optimal growth was found to be pine sawdust combination of 30% wheat bran and poplar sawdust combination of 20% corn bran were the best of the optimal combination. The optimal concentration of white sugar was 1.0~1.5%. The nitrogen sources was found to be yeast extract and soybean powder. Also, optimal concentration were $0.7g/{\ell}$ and $0.1g/{\ell}$, respectively. The mineral sources of optimal medium compositions were $MgSO_4{\cdot}7H_2O\;0.3g/{\ell}$, $KH_2PO_4\;0.5g/{\ell}$ and $K_2HPO\;1.2g/{\ell}$. Optimal amount of inoculum for cultivation of A. cylindracea were $20{\sim}25g/850m{\ell}$ and $25m{\ell}/850m{\ell}$ in the sawdust spawn and liquid spawn, respectively.

Agroforestry Strategies Reflecting Residents' Attitudes in a Semi-arid Region - Focusing on Elsentasarhai Region in Mongolia - (주민의식을 반영한 반건조지역의 산림농업 전략 - 몽골 엘센타사라이 지역을 중심으로 -)

  • This study explored agroforestry strategies to achieve ecological and economic effects simultaneously for Elsentasarhai region in Mongolia under desertification, based on attitude survey with a questionnaire, field survey on planting sites, and literature review. The agrosilvopastoral approach was suggested as a type of agroforestry practices which combined tree planting for combatting desertification and wind damage to crops, agricultural crop production for income improvement, and livestock raising, a major industry in the study region. Populus sibirica and Ulmus pumila native to desert regions were selected for tree planting, and Hippophae rhamnoides, potato, and fodder appropriate for the short growing season were chosen for income crop production, reflecting residents' attitudes and growth environments. As a strategy of land allocation to satisfy multiple effects of the agrosilvopastoral approach, the alley cropping technique was recommended which arranged alternately strips of trees as windbreaks and income crops in multiple rows. The study also explored desirable planting techniques to improve conditions of income crop production and tree growth against drought and strong winds in the alley cropping. Study results will be useful as fundamental information to implement sustainable agroforestry in Mongolia and other semi-arid regions where knowledge concerned is lacking.

Transient Expression of β-gulucuronidase (GUS) gene in Immature Ovules and Calli Derived from Cottonwood Species (Populus deltoides) by Microprojectile Bombardment (포플러의 미성숙(未成熟) 배(胚)와 캘러스에서 유전자총(遺傳子銃)에 의(依)한 GUS-gene의 일시적(一時的) 발현(發現))

  • Excised immature ovules and calli derived from the stems of cottonwood were bombarded with microprojectiles carrying plasmid DNA containing CaMV-35S promoter and ${\beta}$-glucuronidase(GUS) gene. After bombarded, the expression of GUS gene was detected by the assay of 5-bromo-4-chloro-3-indolyl-${\beta}$-gluconide(X-gluc). Transient gene expression was measured by counting the number of distinct regions of GUS activity per explant. As major parameters, the number of shots and the period of exposure to X-gluc after the bombardment were investigated for detecting GUS gene expression. In this experiment, the percents of GUS gene expression showing spots were 56.8 from immature ovules and 75.9 from micro-calli of cottonwood species. Among the treatments, two consecutive shots and 48 hour exposure produced about $25.75{\pm}2.77$(per ovule), $11.43{\pm}1.22$(per mini petridish) spots, respectively, Microprojectile particle bombardment provides a useful method to assay transient expression in both types of explants. Furthermore, our results represent that the excised ovule and/or the calli might be stably transformed by the biolistics.

Transformation of Populus alba $\times$Populus glandulosa Using Phosphinothricin Acetyltransferase Gene (Phosphinothricin acetyltransferase 유전자를 이용한 현사시의 형질전환)

  • This study was conducted to produce herbicide resistant plants by transferring phosphinothricin acetyltransferase (PAT) gene into Populus alba $\times$ Populus glandulosa No .3 using Agrobacterium tumefaciens MP 90/PAT. Leaf segments from in vitro grown shoots of hybrid poplar No. 3 were soaked in a AB medium containing Agrobacterium tumefaciens MP 90/PAT for 10 min and cocultivated for 2 days on MS medium containing 1.0 mg/L 2,4-D and 0.2mg/L kinetin (CIM). Putative transformed calli could be selected after cocultivation of leaf segments on CIM supplemented with 50mg/L kanamycin and 500mg/L cefotaxime for 3 weeks. The selected calli were cultured on CIM supplemented with 50 mg/L kanamycin and 500 mg/L cefotaxime for 5~8 weeks before transfer to WPM containing 1.0mg/L zeatin, 0.1mg/L BAP, 50 mg/L kanamycin and 500mg/L cefotaxime for shoot regeneration. Shoots were regenerated from the callus after 4 week cultivation, and the regenerants were grown on the same medium for 7~l0 weeks. The plants rooted on 1/2 WPM containing 0.2 mg/L IBA and 50 mg/L kanamycin. To confirm the gene insertion into plants, GUS activity was detected by histochemical assay in the transformed plants. Finally, the presence of both NPT II and PAT genes from the transgenic plants were confirmed by PCR amplification with the gene specific primers and subsequent PCR-Southern blot with DIG-labeled PAT gene probe. After acclimatization in pots for 4 weeks, the plants were sprayed by 3 mL/L of Basta to test resistance to the herbicide. The transgenic plants remained green, whereas all the control plants died after one week.

Analysis of risk management system of GM crops in China for the development of global GM crops (글로벌 GM 작물 실용화를 위한 중국의 GM 작물 안전관리제도 분석)

  • We analysed the current status of development of GM crops and national biosafety framework including legislation-related agricultural GMO in China to provide the policy for the development of global GM crops in Korea. In China, several GM crops including cotton, petunia, tomato, sweet pepper, poplar, and papaya have been approved for commercialization and they have been cultivated at more than 4 million ha. In addition, GM rice and GM maize have also obtained approval for productive testing in 2009. China will be the first country to approve GM rice for commercialization. Prior to commercialization in China, all GM crops must be approved by government authority for biosafety assessment specified by national legislation including restricted field testing, enlarged field testing, productive testing and safety certificate. According to China's legislation, agricultural GMOs have been classified by research and testing, production and processing. All GMOs must go through 3 steps of field testing (restricted, enlarged and productive). Prior to conducting each field testing, it has to be approved by government authority. It is assumed that at least one to two years will be taken for each step of field testing (total 4 to 8 years to obtain the final safety certificate) along with a large amount of budget.

Construction of a Full-length cDNA Library from Korean Stewartia (Stewartia koreana Nakai) and Characterization of EST Dataset (노각나무(Stewartia koreana Nakai)의 cDNA library 제작 및 EST 분석)

  • In this study, we report the generation and analysis of 1,392 expressed sequence tags (ESTs) from Korean Stewartia (Stewartia koreana Nakai). A cDNA library was generated from the young leaf tissue and a total of 1,392 cDNA were partially sequenced. EST and unigene sequence quality were determined by computational filtering, manual review, and BLAST analyses. Finally, 1,301 ESTs were acquired after the removal of the vector sequence and filtering over a minimum length 100 nucleotides. A total of 893 unigene, consisting of 150 contigs and 743 singletons, was identified after assembling. Also, we identified 95 new microsatellite-containing sequences from the unigenes and classified the structure according to their repeat unit. According to homology search with BLASTX against the NCBI database, 65% of ESTs were homologous with known function and 11.6% of ESTs were matched with putative or unknown function. The remaining 23.2% of ESTs showed no significant similarity to any protein sequences found in the public database. Annotation based searches against multiple databases including wine grape and populus sequences helped to identify putative functions of ESTs and unigenes. Gene ontology (GO) classification showed that the most abundant GO terms were transport, nucleotide binding, plastid, in terms biological process, molecular function and cellular component, respectively. The sequence data will be used to characterize potential roles of new genes in Stewartia and provided for the useful tools as a genetic resource.

The Nuclear DNA Content and Histological Characteristics of Triploid Poplars Grown In Vitro (현사시나무 3배체의 핵 DNA 함량 및 조직학적 특성)

  • Herein we analyzed the nuclear DNA content and the histological characteristics of the triploid of the 'Hyunsasi' (Populus alba ${\times}$ P. glandulosa $F_1$) which were developed for biomass production and molecular breeding research. The flow cytometric analysis showed that the nuclear DNA content of the 3 triploids were 1.6 times greater than those of the diploid. In terms of histological characteristics, the cross-section area of the stem of 'Line-18' was 1.6 times larger than that of the diploid. The area of pith, and cortex and phloem of the stem of 'Line-18' was also 1.6 and 2.0 times larger than that of the diploid, respectively. Moreover, the length and area of guard cell of 'Line-18' was 1.2 times larger than that of the diploid. These results helps to understand the cytological characteristics of the triploid poplar clones, and further investigations in the growth rate and wood properties of the triploids growing in the field will determine whether the triploid poplars are good candidates for molecular breeding programs and for the improvement of industrial biomass productivity.

Studies on the Mycelial Cultivation and the Rhizomorph Production of Armillaria mellea (Armillaria mellea의 균사배양 및 균사속 생산에 관한 연구)

  • Five strains of Armillaria mellea were collected from the forest of Chonbuk province and isolated from the tissue of fruit bodies. Nutritional and environmental characteristics of mycelial cultivation and rhizomorph production of Armillaria mellea isolated were determined in sawdust media, woody inocula and soils. The sawdust media of Styphnolobium japonicum, Culhamia simplex, Populus monilifera and Populua davidiana were proper for mycelial growth. The ranges of optimum pH, temperature and moisture content for mycelial growth were in the range of $4.5{\sim}5.0$, ${\sim}25^{\circ}C$ and $65{\sim}70%$, respectively. Among the various additives and inorganic salts added, 10% rice bran and 3% $CaCO_3$ were effective to mycelial growth. The woody inocula of Styphnolobium japonicum, Culhamia simplex, Quercus acutissima and Quercus veriabilis were proper for rhizomorph production. The ranges of optimum pH, moisture content and temperature for rhizomorph production were in the range of $4.5{\sim}4.9,$ $45{\sim}55%\;(w/w)$ and $20{\sim}24^{\circ}C$, respectively. Distribution of rhizomorphs in soil was varied with depth, but the main concentration occurred in the range of $7.5{\sim}12.5\;cm$. They were rarely found below 25.0 cm.

Annual Removal of Soil Nutrient by Stem Harvest in a Willow (Salix spp.) Plantation (버드나무(Salix spp.) 조림지내 벌채에 의한 년간 토양양분 수탈)

  • Nutrient removal during stem harvest was evaluated in a one-year rotation willow bioenergy plantation. For the stem nutrient content, the stem biomass and stem nutrient concentration were collected in the winter of 1987-1993 from the established willow plantation at Tully, New York, U.S.A. in 1987. Five willow clones and one hybrid poplar clone were planted. Half of the plots were fertilized annually with $336kg\;ha^{-1}$ N, $112kg\;ha^{-1}$ P, and $224kg\;ha^{-1}$ K. All trees were harvested annually. Mean annual nutrient removals of N, P, K, Ca, and Mg by annual stem harvesting over seven years were respectively 30-70, 4-10, 14-40, 19-59 and $3-5kg\;ha^{-1}$. Fertilized plants exported higher quantities of nutrients than non-fertilized ones. Nontheless, quantities of nutrients exported were well below the quantities supplied by fertilization suggesting that nutrients removal by stem harvesting is not likely to cause a decrease in soil fertility. However, in non-fertilized plots, the amount of nutrients removed could result in decrease of nutrient availability and soil fertility over the long-term. An evaluation of the clones revealed that clone SV1 is the most nutrient efficient.

Growth characteristics and productivity of oyster mushroom with the addition of Artemisia annua (개똥쑥 첨가에 따른 느타리의 생육 특성 및 수량성)

  • This study sought to determine the optimal mixing ratio of Korean cultivated Artemisia annua for production of functional oyster mushrooms. After 23 days of cultivation, mycelial growth was 12.7 cm in medium supplemented with 5% A. annua and 12.5 cm in control. Mycelial growth progressively slowed with the addition of A. annua, with barely any growth (2.1 cm) in the presence of 70% A. annua. Mycelial density was high density without significant difference between treatments. The pileus diameter was greater in the presence of A. annua than in the control, but the pileus thickness was only slightly higher compared to the control. The stipes thickness was greatest for 15% A. annua, and the length of stipes was longest at 10% A. annua, but was lower than the value of the control. The L value of the fruiting zone was highest 10% firewood, and the L value of freshwater was highest at 5% A. annua. The fruiting body yield was highest to (122 g/850 ml) in medium supplemented with 5% A. annua but markedly decreased at higher levels of A. annua.
