Tempeh Fermentation from a Mixture of Soybean and Sorghum Grain (대두-수수 혼합곡물의 템페발효)

  • Tempeh-type fermented products were prepared from soybean, sorghum or mixture of soybean and sorghum(1 : 1) with the traditional Indonesian inoculum(LARU : mixed cultures of Rhizopus oligosporus). Fermentation increased protein and fiber contents in the soybean tempeh(ST) and tempeh of soybean-sorghum mixture(SSM). Fat content was slightly higher in sorghum tempeh(SGT) and SSM than that of control. During the fermentation, pH, soluble solid and soluble nitrogen were increased, while no significant change was found in the total solids. The trypsin inhibitor activity(TIA) and phytic acid content decreased after 32 hrs fermentation. It is suggested that Rhizopus oligosporus is capable of hydrolyzing trypsin inhibitor and phytic acid of the substrate. Thiamine and niacin contents increased in all samples as compared with the unfermented control. In amino acid level, there were some decreased in total amino acids after 32 hrs fermentation in three types of tempeh. While the concentrations of lysine, valine. tyrosine and alanine in ST, SGT and SSM were increased those of serine and glutamic acid were decreased in compare to the unfermented control.

Riboflavin Status Influences the Biosynthesis of Flavin Peptides and Related Enzyme Activities in Rat Liver Mitochondria (리보플라빈 결핍이 쥐간의 미토콘드리아의 플라빈 펩티드와 관련된 효소 활성에 미치는 영향)

  • The effeds of riboflavin defidency on the biosynthesis of flavin pepddes and levels of flavoenzymes and catecholamines have been investigated. The percentage of 14C. riboflavin radioactivity formed in mitochondria appeared to increase up to 2 weeks but started to decline at 3 weeks. A significant increase of radioactivity incorporation into mitochondria and into trypsin-digestable plus trypsin-non-digestibie flavin peptides was detected in riboflavin-deficient animals. More than 35% of incorporation was observed at the end of the first week and 160% higher incorporation was observed in fiavin peptide after the second week. Activities of MAO and succinate dehydrogenase were affected markedly by riboflavin status whereas those of acetyichoilnesterase were not affected. Riboflavin defidency also brought about marked reductions in levels of epineplrrine and norepinephrine. it is concluded that the levels of flavin peptides, MAO and succinate dehydrogenase, and catecholamines were affected significanily by the availability of riboflavin and in particular the duration of its depiction.

Biochemical Characterizations of Phenylalanine Ammonia-Lyase and its Mutants to Develop an Enzymatic Therapy for Phenylketonuria (페닐케톤뇨증의 효소치료 개발을 위한 phenylalanine ammonia-lyase 및 유전자 변이형의 생화학적 특성)

  • Enzyme substitution with recombinant phenylalanine ammonia-lyase (PAL, EC 4.3.1.5) is currently being explored for treatment of phenylketonuria (PKU), an autosomal recessive genetic disorder with mutations of the gene encoding phenylalanine-4-hydroxylase (EC 1.14.16.1). However, oral administration of PAL is limited because of proteolytic digestion in the gastrointestinal tract. The aim of this study was to determine the biochemical properties of PAL and delinate the susceptibility of wild-type PAL to pancreatic proteolysis by exploring several mutants, and to develop therapeutic drugs with PAL for PKU. The specific activity of PAL was assayed and its optimal pH, temperature stability, and intestinal protease susceptibility were investigated. Its $V_{max}$ values for phenylalanine and tyrosine were 1.77 and $0.47{\mu}mol$/ min/mg protein, respectively, and its $K_m$ values were $4.77{\times}10^{-4}$ and $4.37{\times}10^{-4}\;M$, respectively. PAL showed an optimal pH at 8.5, corresponding to the average pH range of the small intestine. It showed no loss of activity at $-80^{\circ}C$ for 5 months and possessed 93.4% of its activity under $4^{\circ}C$ for 4 wks. PAL was susceptible to chymotrypsin digestion and, to a lesser extent, to trypsin, elastase, carboxypeptidase A, and B. The trypsin and chymotrypsin cleaving sites were mutated to investigate protection from pancreatic digestion and the specific activities of these mutants were evaluated. The six mutants displayed low specific activities compared to the wild-type, suggesting that the primary trypsin and chymotrypsin cleaving sites may be essential for catalytic reaction. The PAL mutants could therefore be applied as a pretreatment modality without susceptibility to proteolytic attack, however, additional modification for enhancing enzymatic activity is needed to reduce the Phe levels effectively.

  • Alismatis Rhizoma has been used as diuretics and antiphlogistics in the Chinese oriental medicine. A trypsin inhibitor was isolated from Alismatis Rhizoma using DEAE ion exchange column, trypsin affinity column, and FPLC chromatography, and its activity and characteristics were studied. The purifed Alismatis Rhizoma trypsin inhibitor (ARTI) was estimated to be about 22 kDa. The sequence determination on N-terminal amino acid residues and 84 amino acid residues has been completed, yet no homology has been found with trypsin inhibitors reported at NCBI. ARTI did not show inhibitory activities on chymotrypsin and elastase, however it exhibited a significant inhibitory activity on bovine trypsin, and formed a complex with rat liver 10-formyltetrahydrofolate dehydrogenase.

A Fluorometric Assay for Trypsin Inhibitor (트립신 저해단백질의 형광측정법)

  • A fluorometric method is described which permits the assay of trypsin inhibitor contained in a sample in an extremely small amount, utilizing a novel reagent generally called fluorescamine. The fluorometric assay with an enzyme kinetic approach has been found to be at least 100 times more sensitive than the well-known Kunitz's spectrophotometric method, considerably taster and less complicated, when it was demonstrated with the anti tryptic activities of very dilute extracts from soybean, red-bean and mung bean. Details of experimental procedure as well as theoretical considerations will be discussed.

Surface Modification of Nano Porous Silica Particle for Enzyme Immobilization (효소 고정화를 위안 실리카 나노세공 입자의 표면개질)

  • The objectives of this study were to develop nano-pore silica particles and to modify its surface for use as an enzyme immobilization matrix. Sol-gel reaction was used to produce silica particles of various nano pore sizes with hydroxyl groups on their surfaces. The surface was modified with aldehyde that was confirmed by fluorescence imaging. Trypsin was covalently immobilized by reductive amination. Surface density of the immobilized trypsin was ca. $350{\mu}g/m^2$, which was approximately 17- and 35-fold higher than those from the surfaces with hydroxyl and amine group, respectively. About 90% of the initial enzyme activity was maintained after the 12th use of repeated use. When compared with the commercial matrices, the nano-pore silica particle was superior in terms of immobilization yield and specific activity. This study suggests the nano porous silica particles can be used as enzyme immobilization matrix for industrial applications.

Antihypertensive Effects of Casein Hydrolysate in Spontaneously Hypertensive Rats (자연발증고혈압쥐에서 카제인 가수분해물의 혈압강하효과)

  • The aim of this study was to determine if a low-molecular weight casein hydrolysate has an anti- hypertensive effect in spontaneously hypertensive rats (SHR). Prior to the in vivo experiment, the casein hydrolysate was confirmed to be resistant to gastrointestinal digestion by confirming the retention of its potency as an inhibitor of angiotensin I-concerting enzyme after incubation with pepsin, trypsin, or chymotrypsin. The in vivo anti-hypertensive effect of the hydrolysate was determined by the tail cuff method. Following an oral administration of the hydrolysate solution, the systolic blood pressure (SBP) decreased by 12.9% (-28.9mmHg; P<0.05) at 3 h after the administration at a dose of 500mg/kg body weight. When the hydrolysate was administered as an emulsion with 30% egg yolk, its anti-hypertensive effect was even more greater at the same dose(-30.8mmHg or -15.9%; P<0.01). In a 50-day long-term trial where the casein hydrolysate was administered once a day, the SBP-lowering effect of the hydrolysate was apparent (P<0.05) from day 35 through the end. Moreover, organ weights and plasma glutamate oxaloacetate transaminase and glutamate pyruvate transaminase activities of the administered SHR were not significantly different from those of controls at the end of the long-term trial.

Changes of Enzyme Activity, Trypsin Inhibitor, Tannin and Phytic Acid during Heat Treatment of Soybean (콩의 열처리 중 효소, 트립신 저해제, 탄닌, 피트산의 함량 변화)

  • This study was performed to investigate the change of lipoxygenases and urease activities, trypsin inhibitor, tannin and phytic acid contents during heat treatment of Jinpum soybean. The lipoxygenase-1 and urease possessed their activities after heating at $60^{\circ}C$ for 100 min, but their activities disappeared rapidly at $80^{\circ}C$ and $100^{\circ}C$ for 20 and 10 min. There were no lipoxygenase-2 and -3 activities in Jinpum soybean with and without heating. Trypsin inhibitor was lost 91.9%, 78,1% and 58.6% of the activity after heat treatment at $100^{\circ}C$ for 50 min, at $80^{\circ}C$ for 100 min and at $60^{\circ}C$ for 100 min, respectively. The tannin content was increased by heat treatment. The content of and phytic acid was increase after heating at $60^{\circ}C$ for 100 min, unchanged at $80^{\circ}C$ for 100 min and decreased at $100^{\circ}C$ for 100 min.

Physiological activities of Pyrus pyrifolia Nakai, malt, Ziziphus jujuba Mill., Leonurus japonicus Houtt., Scutellaria baicalensis, and their mixtures (배, 맥아, 대추, 익모초, 황금 및 이들 혼합물의 생리활성)

  • This study investigated the anti-inflammatory, trypsin activity, and antioxidant effects of 11 kinds of plant extracts to discover materials for developing optimal mixtures that improve inflammation and help digestion. Ziziphus jujuba Mill. (ZJ), Leonurus japonicus Houtt. (LJ), Scutellaria baicalensis (SB), Platycodon grandiflorum, and Aster scaber extracts had excellent anti-inflammatory effects by reducing excessive nitric oxide (NO) and tumor necrosis factor-α content in lipopolysaccharide (LPS)-stimulated RAW264.7 macrophages. The malt (MA), Pyrus pyrifolia Nakai (PP), Raphanus sativus L., Platycodon grandiflorum extracts among the 11 kinds of plant extracts had high trypsin activity. The antioxidant activity of the plant extracts was examined by the DPPH radical scavenging activity, and the SB, PE, JU, and MA extracts had high antioxidant activity. Therefore, PP, MA, ZJ, LJ, and SB were selected to develop optimal mixtures that improve inflammation and help digestion. The extract of plant mixture containing PP, MA, ZJ, LJ, and SB in the ratio 1:1:2:1:2 (w/w) significantly inhibited NO production than the extract of PP, MA, ZJ, LJ, and SB, respectively, in LPS-stimulated RAW 264.7 macrophages. The DPPH radical scavenging activity of the mixture extract was significantly higher than the extract of PP, MA, ZJ, and LJ, respectively.

  • 이번 연구는 아플라톡신 $B_1(AFB_2)$의 독성이 오리의 생산성, 체내 기관, 간 효소 활성도, 외관상 소화율, 영양소 소화율에 미치는 영향을 알아보기 위한 것이다. 1일령의 육용오리 90마리를 3개의 처리군으로 나눠 10마리씩 펜에서 사육하였다. 그룹1은 일반 사료를 급여하였고, 그룹 2와 3은 각각 아플라톡신 $20{\mu}g/kg$, $40{\mu}g/kg$이 포함된 오염된 쌀을 섞어 6주 동안 급여하였다. 그 결과 아플라톡신에 오염된 사료를 섭취한 그룹의 증체량과 사료 섭취량이 감소하였고, 사료요구률(feed to gain ratio), 간, 신장, 췌장의 무게가 높은 것으로 나타났다. 알라닌 아미노전이효소(ALT, serum alanine aminotransferase)와 혈중 아스파라진산 아미노전이효소(AST, aspartate aminotransferase)의 활성도도 아플라톡신 오염 그룹에서 유의성을 보이며 높았다. 아플라톡신 오염 그룹의 오리들의 십이지장에서 채취한 단백질 분해효소, 키모트립신, 트립신(이자액에서 분비되는 단백질 분해효소), 전분 가수 분해효소 등 소화효소의 활성도가 증가한 반면, 조단백질의 외관상 소화율은 유의성있게 낮은 것으로 나타났다. 이는 아플라톡신에 오염된 사료로가 오리의 생산성과 영양소의 외관상 소화율을 감소시키고 십이지장 내용물의 소화효소활성을 변화시킨다고 볼 수 있다.
