Optimization of Microwave-Assisted Pretreatment Conditions for Enzyme-free Hydrolysis of Lipid Extracted Microalgae (탈지미세조류의 무효소 당화를 위한 마이크로파 전처리 조건 최적화)

  • The purpose of this study was to effectively produce the biosugar from cell wall of lipid extracted microalgae (LEA) by using microwave-assisted pretreatment without enzymatic hydrolysis process. Response surface methodology (RSM) was applied to optimization of microwave-assisted pretreatment conditions for the production of biosugar based on enzyme-free process from LEA. Microwave power (198~702 W), extraction time (39~241 sec), and sulfuric acid (0~1.0 mol) were used as independent variables for central composite design (CCD) in order to predict optimum pretreatment conditions. It was noted that the pretreatment variables that affect the production of glucose (C6) and xylose (C5) significantly have been identified as the microwave power and extraction time. Additionally, the increase in microwave power and time had led to an increase in biosugar production. The superimposed contour plot for maximizing dependent variables showed the maximum C6 (hexose) and C5 (pentose) yields of 92.7 and 74.5% were estimated by the predicted model under pretreatment condition of 700 w, 185.7 sec, and 0.48 mol, and the yields of C6 and C5 were confirmed as 94.2 and 71.8% by experimental validation, respectively. This study showed that microwave-assisted pretreatment under low temperature below $100^{\circ}C$ with short pretreatment time was verified to be an effective enzyme free pretreatment process for the production of biosugar from LEA compared to conventional pretreatment methods.

Study on the Degradation of Pigskin Collagen Using Irradiation Technique (방사선조사를 이용한 돈피 콜라겐의 저분자화 연구)

  • This study was intended to observe possibility of which radiation technique can be used for oligopeptide production from pigskin collagen to reduce environmental pollution in processing and simplify the processing steps. Raw pigskin was ground using chopper, and then defatted in acetone cooled at $-20^{\circ}C$ freezer. Defatted dried pigskin was irradiated at 20, 40, 60, 100, 150, 200, 250, and 300 kGy using Co-60 gamma rays irradiator. With irradiation doses, the amount of soluble proteins increased, and the viscosity and turbidity of soluble proteins decreased, which could be clue of that irradiation degrade high molecular proteins directly. pH of soluble proteins from defatted pigskin increased in the sample above 150 kGy, and low molecular weight components (below 24 kDa) in SDS-PAGE increased. From gel permeation chromatography of the hydrolysates of pigskin irradiated at 300 kGy showed the major peak of 9,000, 8,200, 860, and 170 Da.

A Study on the Bioavailability of Calcium in Eggshell Powder Fortified Ramen in the Growing Rats (성장기 쥐에서 난각분 강화라면의 칼슘 이용성에 관한 연구)

  • This study has investigated the bioavailability of calcium in eggshell powder (ESP) fortified ramen. Wean-ling SD rats were maintained for 2 weeks on 0.05$\%$ low calcium diet then assigned randomly to one of 5 groups. Animals were provided for 4 weeks one of 3 different experimental diets containing 0.15$\%$ calcium as $CaCo_{3}$, NFDM, or ESP fortified ramen. Two control groups were fed either 0.05$\%$ calcium or general ramen containing 0.08$\%$ calcium. The body weight, diet intake, food efficiency ratio (FER), bone growth, calcium contents of bones, and apparent absorption were measured. Experimental results show that regardless of calcium sources 0.15$\%$ calcium groups increased body weight, weight and length, calcium content, and the strength of two bones (tibia and femur) significantly compared to 0.05$\%$ calcium group. The apparent absorption rate of calcium also showed similar results supporting the bioavailability of ESP fortified ramen is not inferior to either NFDM or $CaCO_{3}cdot$ The results indicate that ESP fortified ramen is a proper mediate for calcium fortification in Korea.

유청을 이용한 발효유주 제조중 Kefir Grain의 미생물학적 특성

  • 모짜렐라 치즈 제조 후 부산물로 생산된 유청에 가당을 하고 케휘어 그레인, 포도주 효모를 접종하여 새로운 발효유주 제조공정을 개발하고자 하였다. 그 결과 가당원료로서 물엿을 첨가한 유청 용액의 적정산도가 가장 높게 나타났으며, 첨가한 당의 종류별당 이용율은 배양초기에는 비교적 차이가 큰 편이었지만 배양시간이 경과할수록 차이가 감소하였다. 가당 유청 용액중에서 복합균주의 생육특성은 환원탈지유와는 달리 젖산구균의 생육이 우세하였다. 치즈 유청을 살균 후 당을 첨가하고 케휘어 배양액과 포도주 효모배양액을 접종하여 $20^{\circ}C$의 저온에서 알코올 발효를 실시하였을 때, 배양 1일부터 발포현상이 나타났으며 4일간 배양 후 1.2% 알코올을 함유하는 발효배양액을 얻을 수 있었다. 발효액을 2회에 걸쳐 증류농축기에서 $75^{\circ}C$에서 25분간 감압 하에서 알코올을 추출하여 20% 알코올 원액을 얻을 수 있었다.

Effect of Debinding and Sintering Conditions on the Tensile Properties of Water-atomized STS 316 L Parts by Powder Injection Molding (수분무 STS 316L 분말사출성형체의 탈지 및 소결공정에 따른 인장 특성)

  • The purpose of the present study is to investigate the influence of thermal debinding and sintering conditions on the sintering behavior and mechanical properties of PIMed 316L stainless steel. The water atomized powders were mixed with multi-component wax-base binder system, injection molded into flat tensile specimens. Binder was removed by solvent immersion method followed by thermal debinding, which was carried out in air and hydrogen atmospheres. Sintering was done in hydrogen for 1 hour at temperatures ranging from 1000℃ to 1350℃ The weight loss, residual carbon and oxygen contents were monitored at each stage of debinding and sintering processes. Tensile properties of the sintered specimen varied depending on the densification and the characteristics of the grain boundaries, which includes the pore morphology and residual oxides at the boundaries. The sinter density, tensile strength (UTS), and elongation to fracture of the optimized specimen were 95%, 540 MPa, and 53%, respectively.

전처리 조건에 따른 구리박막 표면에서의 특성변화

  • 최근 IT산업의 급속한 발달로 모바일 제품과 반도체 및 IC 패키지 등의 전자제품의 소형화, 경량화 및 고성능화되어 가고 있다. 따라서 반도체 공정에서 단위소자의 고속화를 구현하기 위한 금속배선공정에 사용되는 금속재료가 최근에 최소 선폭을 갖는 디바이스에서는 구리를 배선 재료로 전환하고, 향후에는 모든 디바이스가 구리를 주요 배선재료로 사용할 것으로 예측되고 있다. 반도체 소자 공정 중 시료 표면 위에 형성되는 오염물은 파티클, 유기오염물, 금속 불순물 그리고 자연 산화막으로 나눌 수 있다. 구리 표면에 생성되는 부식생성물의 종류에는 CuO, $Cu_2O$, $Cu(OH)_2$, $CuCO_3{\cdot}Cu(OH)_2$와 같은 생성물들이 있다. 이러한 부식생성물이 구리박막 표면에 형성이 되면 성장된 구리박막의 특성을 저하시키게 된다. 이러한 다양한 오염물들을 제거하기 위해서 여러 가지 전처리 공정에 대한 연구가 보고되고 있다. 본 연구에서, 스퍼터 방식으로 구리를 증착한 웨이퍼 (Cu/Ti/Si) 를 대기 중에 노출시켜 자연 산화막을 성장시키고, 이 산화막과 대기로부터 흡착된 불순물을 제거하기 위해 계면 활성제인 TS-40A와 $NH_4OH$ 수용액을 사용하여 이들 수용액이 구리 표면층에 미치는 영향에 대해 조사 분석하였다. 사용된 TS-40A는 알칼리 탈지제로서 웨이퍼 표면의 유기물을 제거하는 역할을 하며, $NH_4OH$는 구리를 제거하는 부식액으로 산업현장에서 널리 사용되고 있다. 다양한 표면 전처리 조건에 따른 구리박막 표면의 형상 및 미시적 특성변화를 SEM과 AFM을 이용하여 관찰하였고, 표면의 화학구조 및 성분 변화를 관찰하기 위해 XPS를 측정하였으며, 전기적 특성변화를 관찰하기 위해 4-point prove를 사용하여 박막의 면저항을 측정하였다.

Process Development for Production of Antioxidants from Lipid Extracted Microalgae Using Ultrasonic-assisted Extraction (탈지미세조류로부터 초음파추출을 이용한 항산화 물질 생산 공정 최적화)

  • Ultrasound-assisted extraction (UAE) has attracted growing interest, as it is an effective method for the rapid extraction of bioactive compounds from plants with a high extraction efficiency comparable to the conventional extraction. In this study, UAE was used for the extraction of polyphenols from lipid extracted microalgae (Tetraselmis KCTC 12236BP) and the effects of five extraction variables on the total phenolic compounds (TPC) were studied. For the optimization of extraction parameters, particle size, solid-to-liquid (L/S) ratio, ethanol concentration, extraction temperature and extraction time have been examined as independent variables. All variables exhibited the significant effects on the extraction of TPC and extraction temperature showed the most significant effect among five variables. The optimal extraction conditions were the extraction using mixed particle, S/L ratio of 10%, ethanol concentration of 60%, extraction temperature of $100^{\circ}C$ and extraction time of 30 min, which gave the 8.7 mg GAE/g DW for TPC. Compared with conventional hot-water extraction, TPC extraction under UAE was increased by up to 1.8 fold with same extraction condition. This study showed that UAE under low temperature and short extraction time was proven to be an effective extraction process for TPC production from LEA compared to conventional hot-water extraction process.

A Study on the Electron Beam Welding Characteristics of Automatic Transmission Parts (자동변속기 부품의 전자빔 용접특성에 관한 고찰)

  • 최근 국내외에서 자동차 연비와 성능 향상을 위하여 고부가가치 자동변속기의 개발이 적극적으로 추진되고 있다. 특히, 유럽(ZF)과 일본(JATCO, AISIN)을 비롯한 변속기 부품의 기술선진국에서는 전자빔 용접을 적용한 고부가가치 자동변속기 제품이 개발되어 다양한 고급차 모델에 적용되고 있으며 2010년까지 8속 자동변속기를 출시할 예정으로 개발을 추진하고 있다. 이러한 자동변속기를 개발하기 위하여 부품의 성형 및 가공정밀도 관리가 중요하며, 최종 조립되는 부품의 전처리(연질화처리, 탈지, 이물질 제거등)와 용접시 용접변형을 최소화하기 위한 공정조건의 엄격한 관리가 요구되고 있다. 변속기 부품으로 사용되는 소재는 성형성 확보를 위한 연질재(SPCC)와 강도를 확보하기 위한 고강도강(SAPH400, SPFH590)으로 서로 다른 두께의 재질이 조립되어 최종단계에서 용접공정에 의해 제작되고 있다. 특히, 연질재는 복잡한 형상으로 성형한 다음, 가공을 거쳐 경화를 위한 연질화처리가 요구되기 때문에 건전한 용접을 하기 위해서는 용접면에 접한 연질화 처리부는 연질화 처리재가 잔존하지 않도록 완전한 제거할 필요가 있다. 만약 전처리 공정에서 충분한 세척과 가공품의 정도관리가 확보되지 않으면 전자빔 용접시 고온에서 이물질 증발과 잔류, 스패터 발생등에 의한 용접부 결함발생이나 용접변형으로 인하여 제품불량이 발생하게 된다. 따라서 본 연구에서는 최종 조립공정인 용접작업시 발생되는 결함을 방지하고 이 두께 및 이종소재의 전자빔 용접비드 형상(비드 폭, 용입깊이)을 최적화 함으로서 용접부 변형을 최소화하고 충분한 강도를 확보하고자 전자빔 용접전류와 속도에 따른 비드형상을 비교검토 하고자 하였다. 그리고 실차적용시 신뢰성을 확보하기 위하여 전자빔 용접한 자동차 변속기 부품의 비틀림 시험을 실시하였으며 전자빔 용접 전, 후의 변형량을 측정하여 변속기 부품으로서의 적용성을 검증하고자 하였다.

Pre-treatment condition and Curing method for Fabrication of Al 7075/CFRP Laminates (Al 7075/CFRP 적층 복합재료 제조를 위한 전처리 조건과 경화방법 연구)

  • A study has been made to establish an optimum condition in the surface treatment and curing method that is important for the fabrication of Al 7075/CFRP laminates. PAA(Phosphoric Acid Anodizing) provided a good adhesive strength and FPL(Sulfuric / Sodium Dichromate Acid Etching) had a similar adhesive strength with PAA. On the other hand, the poor adhesive strength was shown on vapor degrease and CAA(Chromic Acid Anodizing). By using the atomic force microscope(AFM), it was found that the PAA oxide surface obviously had a greater degree of microroughness as compared to vapor degrease, CAA and FPL treated surfaces. These results support the concept of a mechanical interlocking of the adhesive with-in the oxide pores as the predominant adhesion mechanism. In curing methods, the adhesive strength of co-curing method was higher than that of secondary curing method. With respect to stability of specimen shape, the secondary curing method was better than co-curing method. DMA(Dynamic Mechanical Analysis) test revealed $T_g$ in curing times over 60 min is nearly same, so it is estimated they will have similar degree of curing and joint durability in using FM300M adhesive film.

A Study on the Process Capability Analysis of MIM Product (금속분말 사출성형 제품의 공정능력분석에 관한 연구)

  • Metal Injection Molding (MIM) is attractive because it produces consistent, complex-geometry components for high-volume, high-strength, and high-performance applications. Also MIM using in optical communication field, display field, and semi-conductor field is a cost-effective alternative to metal machining or investment casting parts. It offers tremendous single-step parts consolidation potential and design flexibility. The objective of this paper is to study the suitability of design, flow analysis, debinding and sinterin processes, and capability analysis. The suitable injection conditions were 0.5~1.5 second filling time, 11.0~12.5 MPa injection pressure derived from flow analysis. The gravity of the product is measured after debinding an sintering. The maximum and minimum gravity levels are 7.5939 and 7.5097. the average and standard deviation are 7.5579 and 0.0122; when converted into density, the figure stands at 98.154%. According to an analysis of overall capacity, PPM total, which refers to defect per million opportunities(DPMO), stands at 166,066.3 Z.Bench-the sum of defect rates exceeding the actual lowest and highest limits-is 0.97, which translates into the good quality rate of around 88.4% and the sigma level of 2.47.
