Studies on Carbohydrate Accumulation in Aspergillus niger During the Differentiation (검정곰팡이의 분화(分化)에 따른 총탄수화물(總炭水化物) 및 트레하로즈량(量)의 변동(變勳)에 대한 연구(硏究))

  • The changes of total carbohydrates and trehalose levels during differentiation of A. niger were studied. Aspergillus niger was cultivated in Czapeck-Dox medium by the method of surface culture and shake culture. Total carbohydrates and trehalose were fractionated and determined by the method of Trevelyan and Harrison (1956). Total carbohydrates and trehalose accumulated in A. niger during sporulation. The influences of nutritional starvation on the levels of cellular carbohydrates in A. niger, which was cultivated in each nitrogen, phosphate and glucose limited Czapeck-Dox medium, were studied. The levels of total carbohydrates and trehalose in A. niger cultivated in nitrogen limited medium increased much than those cultured in full medium. The total carborates and trehalose levels in A. niger cultivated in phosphate limited medium increased, but the increases were less than those cultured in nitrogen limitted medium. In glucose limited medium, any changes of total carbohydrates and trehalose levels were not found. It is considered that the biochemical mechanisms responsible for the changes of total carbohydrates and trehalose levels may be related with differentiation of Aspergillus niger.

Effects of Nitrogen Fertilization and Clipping Interval on Mineral and Water-soluble Carbohydrate Contents in Korean Lawngrass (Zoysia japonica steud) (질소시용과 예초간격이 한국잔디(Zoysia japonica Steud.)의 주요영양성분 및 가용성 탄수화물함량에 미치는 영향)

  • 질소비료 3수준(0,350,700kg N-ha-1year-1) 및 예초간격 3수준( 10,20,30일 간격)하에서 이 요인들이 식물체 각부위(엽, 엽초를 포함한 차, 포복경 및 근)내, N,P,k,Ca,Mg 및 가용성 탄수화물함량에 여하히 영행을 미치는가게 대해서 1983년 6월부터 10일 까지 대전 배재대학실험포장에서 조성한지 3년 지난 한국잔디를 대상으로 실험하였던 바 결과는 다음과 같다. 1. 질소 증시한 결과 식물체 각부위내 N함량은 증가하였다. 특히 N성분은 엽부위에 다량으로 집적 되어 있어 저장양분으로서의 역할을 하지못하는 것으로 사료되었다. 2. 질소비료에 대한 P성분의 반응은 식물체부위에 따라 변화가 심하게 나타났는데 엽부위로서는 P함량이 감소한 반면, 경 및 포복경에서는 예초간격이 30일에서 10일로 단축되었을 때 한하여 증가하는 양상 을 보였다. 3. K 함량은 질소를 증랭시용할때 엽과 경부위에 특히 많이 축적되었으며 근부위에서는 질소시용이 오히려 K함량을 감소시키는 요인으로 작용하였다. 그러나 N과 K함량간에는 지상부위와 포복경에서 정의 상관관계가 인정되었다. 4. Ca 함량은 엽과 경부위에서 예초간격을 짧게 하였을때 질소시용에 따른 부의 반응을 보였다.5. Mg 함량은 질소나 예초간격에 거의 영향을 받지 않았다. 6. 가용성 탄수화물함량은 질소시용량이 증가함에 따라 비례적으로 감소하였다. 그러나 예초간격의 영향은 경부위를 제외한 각부위에서 나타나지 않았다. 7. 영양성분에 대한 예초간격의 효과는 질소만큼 크지 않았으나 질소시용과 함께 나타난 유의적 변화는 부위별로 각각 다르게 표현되었다. 즉 N함량은 예초간격을 단축하였을 때 엽 및 경부위에서만 증가되었고 Ca 함량은 30일간격으로 연장할때만 증가되는 경향이었다. 한편 K 및 Mg함량은 예초간격에 의해 영향을 받지 않거나 변동이 심한 상황으로 나타났다.

Seasonal Changes of Nodule Activity, Carbohydrates and Nitrogen and their Inter-relationships in Alfalfa (알팔파근류의 질소고정활성과 체내탄수화물 및 질소함량의 연중변화와 이들의 상호관계)

  • Alfalfa field was established to investigate seasonal changes of nodule activity and contents of carbohydrates, and nitrogen, and also to examine their relationships in alfalfa (Medicago sativa L.). Periodical sampling of alfalfa in cutting and uncutting plots was collected to measure growth of plants, development and activity of nodule, and content of carbohydrates and nitrogen in the third year following year of establishment. Nodule activity of alfalfa root appeared to early April, increased to a maximum in beginning of June (flowering stage), and then decreased and generally remained low from late-July to mid-August, and again increased from early September, and then decreased and generally remained low for the rest of growing season. After flowering, nodule weight tended to decrease slowly throughout the growing season. Until flowering stage, increase of dry weight was closely related with that of nodule activity. But after flowering the curve pattern of dry weight did not fit to that of nodule activity due to decrease of supply of assimilate to nodule, drought, and high temperature. Total nonstructural carbohydrates in roots were closely correlated with nodule weight and nodule activity. While, nitrogen contents in leaves were closely correlated with nodule weight and nodule activity. Also cutting on July prevented unnessesary losses of respiration during summer to provide rapid recovery of nodule activity.

A Study on Calorie and Proximate Components of Traditional Korea Gruel (한국 전통죽의 영양소에 관한 연구 - 열량 및 일반성분을 중심으로 -)

  • This study was conducted to analyze 15 traditional Korean gruels for nutrient density using the Index of Nutrient Quality (INQ). The calorie of the gruels ranged from 148.8 (Daechujuk) to 294.1 kcal (Jatjuk) per serving size. The calorie in 'Jatjuk' was the highest, covering 33.3% of Dietary Reference Intakes for Koreans (KDRI) per meal. The carbohydrate content was between 23.0 g (Dakjuk) and 52.1 g (Patjuk) per serving size. The average content of carbohydrate in gruels was $34.7{\pm}9.1\;g per serving size which covers 24.4% of KDRI per meal. Among the 15 gruels, Patjuk contained the highest carbohydrate content (52.1 g per serving size, 36.7% of KDRI per meal). The highest nutrient density (INQ) of carbohydrate in gruels was Dachujuk (INQ 1.4). The protein content of the gruels was between 2.3 (Huinjuk) and 22.3 g (Dakjuk) per serving size. The average content of protein in gruels was 25.6% of KDRI per meal. Dakjuk contained the highest protein content ($8.2{\pm}4.9\;g$ per serving size) and followed by Dakjuk (INQ 2.5), Guljuk (INQ 1.5) and Kongjuk (INQ 1.3) in nutrient density (INQ) of protein. The fat content of the gruels were between 0.3 (Daechujuk) and 17.8 g (Jatjuk) per serving size. The average content of fat in gruels was $17.8{\pm}0.12\;g$ per serving size which showed 21.1% of KDRI per meal. Among the gruels, Jatjuk contained the highest fat ($17.8{\pm}0.12\;g$ per serving size, 77.4% of KDRI per meal). In order of the high nutrient density (INQ) of protein, Jatjuk (INQ 2.4) was followed by Heugimjajuk (INQ 1.5) and Kongjuk (INQ 1.5).

인공산성비 처리에 대한 은행나무와 곰솔의 생장반응

  • 은행나무와 곰솔의 유식물의 산성비에 대한 생장에 미치는 영향을 알아보기 위하여 pH 5.6을 대조구로 하여 pH 4.0, 3.2와 2.4의 인공산성비를 3개월간 처리하여 생장량, 엽록소 함량과 탄수화물의 함량을 조사하였다. 은행나무와 곰솔의 유식물은 pH2.4에서 생장이 가장 억제되었으며 반면에 pH 3.2에서는 생장이 촉진되었다. 전체적인 생장량은 뿌리에 비해 지상부의 생장에서 산성비의 영향을 더 받았다. 엽록소 함량은 산성비의 산도에 따라 중요한 차이가 없었으며 은행나무 잎의 탄수화물의 함량은 포도당과 설탕의 함량이 pH 4.0에서 가장 낮았고 전분의 함량은 pH 3.2에서 가장 낮았다.

  • 치석다침착자의 치석의 화학성분함량을 구명하기 위하여 치태제거후 6개월에 치태침착도가 심한 사람과 경한 사람을 분류하여 전악으로부터 치태를 채취하였다. 그리고 채취된 치태 세포성분획과 비세포성 부분으로 분리하여 각 분획에서 탄수화물, 단백질, 지질, Ca, Mg, K 및 P의 함량을 화학적으로 분석측정한 결과는 다음과 같다. 1. 치석다침착자의 치태 건조중량당 10%의 탄수화물, 43%의 단백질, 18%의 지질, 4.3%의 Ca, 0.15%의 Mg, 0.29%의 K과 2.87%의 P으로 구성되어 있다. 2. 탄수화물, 지질, K은 비세포 성분획에서 단백질, Mg, 세포성분획에서 많이 차지하였다. 3. Ca, Mg, P의 함량은 치석다침착자의 치태에서 K의 함량은 치석경침착자의 치태에서 높았다. 4. Ca/P비는 비세포성분획보다 세포성 분획에서 훨씬 낮았다.

  • 흰쥐 정자의 성숙과정중에 일어나는 부정소 상피세포, 내강 및 정자 사이의 상호관계를 알아보기 위하여 각 실험군 별로 웅성호르몬 투여에 따른 탄수화물 관기의 함량 변화를 측정하였고, tunicamycin 투여 후 hexosamine의 할량 변화를 살펴봄으로써 glycosylation되는 장소를 확인하였다. 부정소두와 부정소이 상피세포내의 hexose 함량은 거세 후 5일군부터 유의성있게 감소하였고, testosterone 투여시는 각각 5일 , 10일군부터 유의하게 증가하였으며 , hexosamine과 sialic acid의 경우도 통계적으로 유의하게 변화하는 시기는 달랐으나 hexose와 유사하게 증가와 감소하는 경향이 나타났으므로 이러한 하나하나의 탄수화물 관기의 함량 변화는 웅성호르몬에 의존적임을 알 수 있었다. Tunicamycin을 투여하여 hexosamine의 함량 변화를 측정하였을 때 투여 기간에 따라 상피세포와 내강액에서 유의하게 감소하였으나, 정자에서는 일관성있게 감소되지 않았으므로 정자가 직접 hexosamine또는 여러 당단백질 합성에 관여하는 것은 아니며 부정소상피세포나 내장에 의하여 영향받을 수 있다는 가능성을 확인하였다.

  • Microstructure of abaxial leaf surface and carbohydrate content of propagules grown in different culture conditions such as heterotrophic, mixotrophic and autotrophic carbon source were investigated. In the leaves of propagules which were grown in the green house, autotrophic and mixotrophic conditions, wax layer was observed, but in the leaves of the heterotrophic propagules, it was not observed. Size and number of stomata of the leaves in the heterotrophic condition was larger and more numerous than that of autotrophic propagules. Especially, stomata of the leaves in the autotrophic condition was similar to the leaves of plant grown in green house. Carbohydrate content was higher in photoautotrophic condition than that in mixotrophic and heterotrophic culture. Also, Free sugar content showed higher in photoautotrophic propagules than that in mixotrophic and heterotrophic culture. In all the culture conditions, content of glucose were higher than that of other free sugars.

  • Dietary fiber content and carbohydrate digestibility of dried soymilk residue (DSR) and tofu containing DSR were evaluated. Insoluble dietary fiber (IDF) content was 37.4 and 49.8% (%, moisture free basis) for common soymilk residue and DSR, respectively. Both soymilk residues contained 12.5% of soluble dietary fiber (SDF, dry basis). Tofu containing DSR, which is partially substituted with DSR corresponding to 10% weight of soybean used, had higher dietary fiber content (30% more for RDF and 45% more for SDF) than tofu manufactured in traditional manner. Carbohydrate digestibility was much lower in all tofu products ranging from 11% to 21%, and there was a negative correlation( r = -0.9243) between carbohydrate digestibility and total dietary fiber content.

  • To establish a culture system with enhanced cellular nutrition, we investigated the effects of light quality (blue, 450 nm; yellow, 590 nm; and red, 630 nm) of a light-emitting diode (LED) on the biochemical composition of Tetraselmis suecica and T. tetrathele. The protein content of both species was higher (42-69%) than the content of other biochemical substances under all wavelengths. Carbohydrate, protein, and lipid contents were higher under the yellow wavelength, which showed a low growth rate, than those under other wavelengths. The contents of all biochemical substances were low under the red wavelength, which showed a high growth rate. These results indicated that protein synthesis occurs in response to decreased cell division rate, while lipid and carbohydrate synthesis occurs owing to altered chemical composition and enzymatic activity. Therefore, we suggested a two-phase LED culture system, which emitted red LED during the early-middle exponential phase and yellow LED during the late exponential and stationary phases, to increase the yield of useful biochemical substances of T. suecica and T. tetrathele.
