Production of Salidroside from Callus Culture of Rhodiola sachalinensis A. Bor (홍경천(Rhodiola sachalinensis A. Bor)의 캘러스 배양에서 Salidroside생산)

  • Callus of Rhodiola sachalinensis A. Bor were induced from leaf explant on l/2MS solid supplemented with combination of auxin (2.4-D, NAA: 0.1∼2mg/L) and cytokinin (BA: 0.1∼0.2mg/L). The effects of various medium, culture conditions and phytohormones on the growth of callus were investigated. MS, WPM, B$_{5}$ medium and diluted or concentrated media (1/2X, 2X, 3X) were used to investigate the growth of callus on each media. Among these, the highest growth was observed when cultured in in 2B$_{5}$ medium containing 0.5mg/L NAA and 1mg/L BA, 3% sucrose and 49.6 mM KNO$_3$ as nitrogen source, and 2.16mM NaH$_2$PO$_4$ as phoshate source at $25^{\circ}C$ in the dark. The calli cultured with 5% sucrose produced high salidroside content (0.41% on the basis of dry wt) than normal root (0.17%).

Studies on the Conditions of Extracellular Phytase Production, by Aspergillus niger (Aspergillus niger에 의한 균본외 Phytase 생산조건에 관한 연구)

  • The distribution of acid phosphatase activity was investigated with 141 microorganisms from the type culture collection of Chong Kun Dang laboratory and the 41 strains isolated from natural sources. The phytase activity was detected mainly with fungal strains. A fungus isolated from soil and identified as Aspergillus niger had shown the highest phytase activity. The environmental conditions for the enzyme formation by the isolate and some properties of the enzyme were also studied. The results obtained were as follows: (1) The highest phytase production was observed when the fungus was cultivated at 28$^{\circ}C$ for 5 days in the corn starch based medium using the cells incubated at 34$^{\circ}C$ for 3 days as a seed. (2) The optimal initial pH of the culture medium was found to around 2 for the formation of phytase. (3) Sucrose was proved to be one of the most effective carbon sources tested for the enzyme production. (4) As an inorganic nitrogen source, potassium nitrate was found to give a good result in the production of phytase. (5) Synthesis of phytase was significantly increased by the supplement with 0.2 % corn steep liquor to the basal medium as an organic nitrogen source. (6) At the concentration of 40-80 mg inorganic phosphate per liter of the culture medium, the enzyme formation revealed the highest level. But as the phosphate was increased above this optimum concentration the phytase activity was drastically decreased although the cell density showed to be still increasing

The Effects of Steam Explosion Treatment on Composting Process with Rice and Barley Straw (볏짚 및 보리짚의 퇴비화를 위한 폭쇄의 효과)

  • This study was conducted to evaluate the effect of steam explosion treatment on composting with rice straw and barley straw. The composting was performed under the condition of $25^{\circ}C$, water content 70%, C/N ratio 30, and aeration of 100ml/min. The content of cellulose, lignin, and total carbon of compost were increased by steam explosion. During composting process total nitrogen of the compost from rice straw and steam exploded rice straw increased by $2{\sim}2.5$ percent, whereas that from steam exploded barley straw increased by 1.7 percent. However total nitrogen of compost from barley straw decreased by 0.41 percent. Therefore, barley straw is a kind of difficulty material for composting, but the composting was proceeded easily by steam explosion treatment, compared to raw barley straw.

  • The purpose of this study is to investigate the effect of hydrochemical variation of groundwater on the gas tigtness in an unlined oil storage cavern. The groundwater chemistry is greatly influenced by the seawater mixing, the water curtain and the dissolution of grounting cements. The chemical composition of groundwater greatly varies ac-cording to both the location of monitoring wells and the sampling period. Most of groundwater shows alkaline pH and high electrical conductivity. The chemical types of groundwater show the dominant order as follows : Na-Cl type > Ca-Cl type > $Ca-HCO_3(CO_3)$ type. Thermodynamic equilibrium state between chemical composition of groundwater and major minerals indicates that carbonate minerals except clay minerals can be precipitated as a secondary mineral. It means that the secondary precipitates can not greatly exerts the clogging effect into fracture aperture in rock mass around oil storage cavern. The content of total organic carbon (TOC) shows a slightly increasing trend from initial stage to late stage. The $EpCO_2$ was computed so as to assess the gas contribution on the $CO_2$ in groundwater. The $EpCO_2$ of 0$\~$41.3 indicates that the contribution of oil gas on $CO_2$ pressure in groundwater system can be neglected.

Online Game Lifestyle in Different Cultural Context: Korea and Japan (문화적 맥락과 온라인 게임 라이프스타일: 한국과 일본의 비교)

  • 본 연구는 온라인 게임공간에서 게이머들이 어떻게 서로 다른 가치관과 행동 패턴을 드러내는지를 확인하고 이를 통해 온라인 게임세계의 문화를 이해하고 분석하기 위해 실시되었다. 전 세계에서 가장 많은 게이머들이 활동하고 있고, 한국과 일본에서 동시에 서비스되고 있는 온라인 게임인 〈리니지〉의 게이머들을 대상으로 온라인 설문조사를 실시하여 '라이프 스타일' 분석틀을 이용해서 이들이 게임공간에서 보이는 행동을 분류하였다. 글 결과 한국과 일본의 리니지 사용자들에게서 거의 유사한 세 가지 라이프스타일 패턴이 추출되었으며 한국과 일본에서 라이프스타일에 따른 행동 패턴이 일관적으로 나타나고 있음을 확인하였다. 도한 한국과 일본의 문화적 차이는 각 라이프스타일 프로파일의 극단성과 분포비율에 의해 반영되는 것으로 확인되었다. 일본의 탈사회적 게이머는 한국의 그것에 비해 더 극단적인 반면 전체 응답자 중에서 차지하는 비율이 적었고 게임 세계에서의 사회 경제적 위치도 낮았으며 한국에서는 비교적 적은 비율을 차지하던 싱글 플레이어 유형이 일본에서는 가장 많은 비중을 차지하는 것으로 나타났다. 또한 일본의 게이머들은 라이프스타일에 상관없이 대부분이 게임 공동체에 가입하는 반면 한국의 게이머들은 라이프스타일에 따라 공동체 참여율이 달랐지만 전체적인 참여율은 일본에 비해 높았다. 이러한 차이는 일본 게임세계에서 공동체 참여는 기본적인 의무인 반면 한국에서는 현실적인 이익과 직결된 활동임을 시사한다. 또한 게임몰입수준은 한국보다 일본의 리니지 게이머들이 높았다. 아시아 각 국에 한국에서 개발한 동일한 온라인 게임이 확산되어 있는 현황을 고려할 때, 온라인 게이머의 라이프스타일 척도는 한국과 일본뿐만 아니라 대만과 중국, 홍콩, 그리고 미국과 같은 리니지 게임이 서비스되는 모든 국가의 문화적 차이를 이해하고 과학적으로 분석할 수 있는 방법이 될 수 있을 것으로 기대된다. 의미를 되새기는 것으로 짧은 연구를 시작하겠다. 등은 활성 값이 70% 이상으로 퇴적물 독성이 상대적으로 낮았다. 이중나선 DNA 함량은 28.4 % - 49%로 대조군에 비해서 감소가 크다. 대부분의 정점이 대조군의 30% 내외로 정점 간의 차이는 크지는 않다. 그러나 다른 측정자료와 같이 정점 22에서 18%로 최소치를 나타내고, 정점 2, 12에서 20% 내외의 값을 보인다. 종합적으로 볼 때 오염물질의 유입이 크고, 광양제철 인근 정점 들이 모두 다른 정점에 비해서 낮아서, 퇴적물 독성이 높은 정점으로 조사되었다.hiwo의 광합성 능력은 낮은 농도들에서는 대조구와 유사하였으나, 5 $\mu\textrm{g}$/l의 높은 농도에서는 초기에 매우 낮은 광합성 능력을 보이다가 시간이 경과하면서 대조군보다 더 높은 경향을 나타냈다. 이러한 결과는 식물플랑크톤이 benso[a]pyrene의 낮은 농도에서 노출될 때는 이 물질을 탄소원으로 사용할 가능성이 있음을 시사한다. 본 연구의 결과들은 연안해역에 benso[a]pyrene과 같은 지속성 유기오염물질이 유입되었을 때 내정여부에 따라 식물플랑크톤 군집내 종 천이와 일차생산력에 크게 영향을 미칠 수 있음을 시사한다.TEX>5.2개)였으며, 등급별 회수율은 각각 GI(8.5%), GII(13.4%), GIII(43.9%), GIV(34.2%)로 나타났다.ments of that period left both in Japan and Korea. Study of CO2 Adsorption Characteristics on Acid Treated and LiOH Impregnated Activated Carbons (산 처리 및 LiOH 첨착 활성탄에서 이산화탄소의 흡착 특성에 대한 연구)

  • Adsorption characteristics of $CO_2$ on activated carbons were evaluated using dynamic adsorption method in a fixed bed with acid treatment, LiOH impregnation and water vapor supply. Physical and chemical properties of the activated carbons were measured using SEM, EDS, nitrogen adsorption, FTIR and XRD. Nitric acid treatment led to the decrease in BET surface area and the increase in oxygen content of virgin activated carbon, and it produced a new functional group that included nitrogen. For the reduction of BET surface area by LiOH impregnation, the nitric acid treated activated carbon (NAC) was less than the virgin activated carbon (AC). Large particles of LiOH were present on the carbon surface when the content of LiOH was over 2 wt%. The adsorbed amount of $CO_2$ on activated carbon in a fixed bed increased with the acid treatment, LiOH impregnation and water vapor supply. The XRD results indicated that LiOH was converted to $Li_2CO_3$ after the adsorption of $CO_2$ on LiOH precursor.

  • To maximize astaxanthin (3,3'-dihydroxy-$\beta\beta$'carotene-44'-dione) production by high density Haematococcus pluvialis cultures, various, media were examined Among tested media, \`Hong Kong Medium and Modified Bolds Basal Medium showed the best result for cell growth ( $2.0$\times$10^{ 6}$cells /mL) and for astaxanthin content per cell (9.7 mg astaxanthin mg/g cell), respectively, Maximum astasanthin concentration of 6.1mL was obtained at pH 7.5, $20^{\circ}C$~$25^{\circ}C$ Deficiencies of nitrogen source($NaNO_3$ and proteose-peptone) found to simulate astaxanthin formation Relatively low light inten- sity of $60\mu$E ($\m^2$s) was sutiable for vegetative cell growth while higher light intensity was required for higher astaxanthin accumulation.

Improvement of Anodic Performance by Using CTP Binder Containg Nickel (니켈을 함유한 콜타르 피치 결합제를 이용한 미생물연료전지 산화전극 성능개선)

  • The composite anodes of expanded graphite (EG) and multiwall carbon nanotube (MWCNT) for microbial fuel cells were fabricated by using coal tar pitch (CTP) binder containing nickel (Ni), and the effect of the anodes with the binders on the performance of the MFCs were examined in a batch reactor. During the start-up of the MFCs, quick increase in voltage was observed after a short lag phase time, indicating that the CTP binder is biocompatible. The biomass attatched on the anode surface was more at higher Ni content in the binder, as well as at smaller amount of CTP binder for the fabrication of the anode. The internal resistance of the MFC was smaller for the anode with more biomass. Based on the results, the ideal combination of CTP and Ni for the CTP binder for anode was 2 g and 0.2 g, respectively. The maximum power density was $731.8mW/m^2$, which was higher 23.7% than the anode with Nafion binder as control. The CTP binder containing Ni for the fabrication of anode is a good alternative in terms of performance and economics of MFCs.

  • The information related to the heavy metal pollution in the environment was obtained from studies on the effects of pH, phosphate and soil properties on the adsorption of metal ions (Cd, Cu, Ni, and Zn) by soils. Three soil materials; soil 1 with low CEC (8.2 me/100g) and low organic carbon content (0.34%); soil 2 with high CEC (36.4 me/100g) and low organic carbon content (1.8%) and soil 3 with high CEC (49.9 me/100g) and high organic carbon content (14.7%) were used. Soils were adjusted to several pH's and equilibrated with metal ion mixtures of 4 different concentrations, each having equal equivalents of each metal ion (0.63, 1.88, 3.12 and 4.38 micromoles per one gram soil with and without 10 micromoles of phosphate per one gram soil). Reported here are the results of the equilibrium study on soil I. The rest of the results on soil 2 and soil 3 will be repoted subsequeutly. Generally higher metal ion concentration solution resulted in higher final metal ion concentrations in the equilibrated solution and phosphate had minimal effect except it tended to enhance removal of cadmium and zinc from equilibrated solutions while it tended to decrease the removal of copper and nickel. In soil 1, percentages of added metal ions removed at pH 5.10 were; Cu 97, Ni 69, Cd 63, and Zn 55, while increasing pH to 6.40, they were increased to Cu 90.9, Zn 99, Ni 96, and Cd 92 per As initial metal ion concentration increased, final metal ion concentrations in the equilibrated solution showed a relationship with pH of the system as they fit to the equation $p[M^{++}]=a$ pH+b where $p[M^{++}]=-log$[metal ion concentration in Mol/liter]. The magnitude of pH and soil effects were reflected in slope (a) of the equation, and were different among metal ions and soils. Slopes (a) for metal ions in the aqueous system are all 2. In soil 1 they were; Zn 1.23, Cu 0.99, Ni 0.69 and Cd 0.59 at highest concentration. The adsorption of Cd, Ni, and Zn in soil 1 could be represented by the Iangmuir isotherm. However, construction of the Iangmuir isotherm required the correction for pH differences.

Preparation of Electrically Conductive Composites Filled with Nickel Powder and MWCNT Fillers (다중벽 탄소나노튜브와 니켈 분말을 포함하는 전도성 복합체 제조)

  • In this study, we prepared electrically conducting composites using epoxy resin of diglycidyl ether of bisphenol A (DGEBA) as a matrix, triethylenetetramine (TETA) as a hardener and nickel powder or multi-walled carbon nanotubes (MWCNTs) grafted with $-NH_2$ groups (MWCNT-$NH_2$) as electrically conducting fillers. Electrical conductivity of composite films were measured by coating on the slide glass with a doctor blade. We measured modification reactions of MWCNT and reaction of MWCNT-$NH_2$ with DGEBA epoxy resin by fourier transform infrared spectrometer (FTIR), thermogravimetric analyzer (TGA) and elemental analyzer (EA). Morphology of composites was investigated by scanning electron microscope (SEM) and sheet resistances of composites were measured by 4-point probe. We found $(9.87{\pm}1.09){\times}10^4{\Omega}/sq$ of sheet resistance for epoxy composite containing both 40 wt% nickel powder and 0.5 wt% of MWCNT-$NH_2$ as fillers, equivalent to epoxy composite containing 53.3 wt% nickel powder only as a filler.
