Two different ${\alpha}$-amylase isozymes (AMY1 and AMY2) found in barley malt share up to 80% of amino acid sequence identity with each other, but their enzymatic properties differ remarkably. AMY1 shows the highest activity at low concentration of calcium ion, while AMY2 is highly active at high calcium concentration. Meanwhile, BASI (Barley ${\alpha}$-Amylase/Subtilisin Inhibitor) protein specifically inhibits only AMY2. In the present study, three separate regions in AMY genes (I, II, and III) were assigned on the basis of restriction enzyme sites and four kinds of chimeric amylases have been obtained by swapping a part of regions with each other. Each chimera gene was successfully over-expressed in Pichia pastoris. From the results of enzymatic characterization, both AMY211 and AMY122 showed the mixed or intermediate type of calcium-dependent activity between AMY1 and 2. Meanwhile, only AMY221 chimera could be significantly inhibited by BASI protein. As a result, it can be proposed that some amino acid residues in the region I and II, except region III, of barley ${\alpha}$-amylases play very important roles in calcium-dependency and interaction with BASI.
The effects of some sodium phosphates as auxiliary agents were studied on the blockade of hardness for silk degumming. In this work, four kinds of sodium phosphates were tested and the results were obtained through masking effects of metallic ions, difference of pH value and boil-off ratio. The degumming of calcium ingredient was analyzed by means of atomic absorption spectrophotometer and degumming test of cocoon shell was performed in the presence of calcium ingredient and sodium phosphates added to soap solution. In the view of the effects of sodium phosphates on calcium hardness, tetrasodium pyrophosphate(TSPP) and sodium phosphate dibasic(SPD) masked calcium ions more than sodium phosphate monobasic(SPM) and sodium hexametaphosphate(SHP). SHP and TSPP have excellent abilities of masking ferrous ions. The pH values of TSPP solution is higher than others, but lower than soap solution. The pH values were differently measured one another among the sodium phosphates but the boil-off ratio was increased in case of sodium phosphate with high pH value.

This is an experiment to complement new ways of using concentrated water discharged from the seawater desalination plant. In this study, metakaolin, which has excellent chloride ion immobilization effect, was used as the main binder, and 10% and 20% of calcium oxide were substituted with the activator. In addition, tap-water(TW) and reverse osmosis brine water(RW) were used as mixed water. As a result of the experiment, the mixture using RW showed higher compressive strength than TW. It also showed low water absorption and high density. In the mixture using RW as mixed water, a hydration reaction substance called Friedel's salt could be observed. Considering the corrosion problem of steel, RW is considered to be applicable to products such as non-reinforced concrete, brick, and curb stone. Through this study, it is thought that it is meaningful to propose a new application method other than the ocean release of RW.

Although it is not a pathological symptom, Dentinal Hypersensitivity (DH) describes pain felt by patients whose tooth roots are exposed outside of the gums and are therefore sensitive to external stimuli. DH is caused by tooth brushing or gum diseases and treatment to reduce the sensitivity can include use of materials having stimulation activity for DH or a resin material applied periodontally. This study examined the hypersensitivity treatment effects of a four-week treatment with a toothpaste containing hydroxyapatite and tricalcium phosphate (Hap-TCP toothpaste). The Hap-TCP toothpaste was made by mixing a commercially available fluorine-containing toothpaste with 10% (W/W) hydroxyapatite and 19% (W/W) tricalcium phosphate (both 99% purity based on XRD analysis). The tooth hypersensitivity treatment effect was surveyed by scoring VRS values, and showed no significant initial difference compared with the control. However, after 1 week of use, the pain reduction value was 8% in the treatment group compared to the control group. This value increased to 30% and 60% after 2 and 4 weeks, respectively. Hypersensitivity to cold stimulation, which was used as a VAS value, showed no initial significant differences compared with the control, but was significantly decreased after 1, 2, and 4 weeks in the experimental group, with more than a 3-fold difference after 4 weeks. These findings confirmed that remineralization can alleviate DH as hydroxyapatite fills dentinal tubules and calcium, phosphorus, and tricalcium phosphate ion equilibrium is established.

Concrete prevents corrosion of reinforcing bars due to its strong alkalinity. However, in the sea, strong alkali components with a pH of 12 to 13 are eluted, which adversely affects the ecological environment and growth of marine organisms. In this study, the mechanical properties and durability of the low alkali mortar were evaluated for the development of a low alkali mortar for the 3D printed artificial reefs. As a result of evaluation of strength characteristics, the α-35 mixture, which were produced with fly ash, silica fume and α-hemihydrate gypsum, satisfied the strength requirement 27 MPa in terms of compressive strength. As a result of pH measurement, it was found that mixing with alpha-type hemihydrate gypsum resulted in minimizing pH due to the the formation of calcium sulfate instead of calcium hydroxide production. As a result of the chloride ion penetration resistance test, the α-35 mixture exhibited the best performance, 3844C. As a result of measuring the length change over time, the α-35 mixture showed the shrinkage 33.5% less compared to the Plain mix.

We studied the release characteristics of the marine sediment which could facilitate sea eutrophication through some lab-scale simulation experiments. Environmental indicators such as pH, ORP(oxidation reduction potential), nitrogens, and phosphates were measured in order to calculate the corresponding release rates. $CaO_2$, an oxygen releasing compound was used to determine how it would effect on the natural process of sedimental release. COD, ammonia nitrogen, phosphorous compounds were less released under the oxic environment caused by $CaO_2$. This basic data will help developing methodology for reducing marine eutrophication which may be initiated by the sedimental release.

기능성 해조음료를 개발하기 위하여 미역, 톳 및 다시마를 저온하에서 마쇄, 원심분리, 한회여과를 거쳐 생해조 세포액을 얻었으며, 각 해조 생세포액의 성분 분석과 색차 측정 및 관능검사를 통하여 품질평한 결과 다음과 같았다. 원심분리 후 한외 여과에 의한 생세포액의 수율은 생해조에 대하여 6.8~56.5% 였으며, 톳과 미역의 수율은 높았으나 다시마는 수율이 낮았다. 해조액을 한회여과한 다음 동결건조한 시료의 회분 합량은 28~60%였으며, 조단백 함량은 12.7%~6.7%정도 였는데 미역과 톳은 다시마 보다 각 성분의 함량이 많았다. 알긴산과 chlrophyll 및 carotenoid 은 한회여과막에 의하여 대부분 제거되었으나 톳의 polyphenol은 30K 한외여과막에 의해서도 효과적으로 제거되지 않았다. 다시마와 톳 생세포액의 주요 아미노산은 glutamic acid와 aspartic acid 였으며, 미역의 경우는 alanine, aspartic acid, valine이었다. 해조 생세포액중의 무기 질은 칼륨의 함량이 특히 높았으며 양이온의 경우 나트륨, 마그네슘, 칼슘, 철, 아연, 망간, 구리순으로 함유 되어있었다. 음이온으로는 $Cl^{-}$,$SO_{4}^{2-}$,$HPO_{4}^{2-}$,$NO_{3}^{-}$의 함량이 높았으며, I와 Br의 함량은 다시마가 톳과 미역에 비하여 높았다.
The effect of Vanadate on the isometric contraction, membrane potential and intracellular calcium ion activities of rabbit myocardial cells were investigated, using calcium selective microelectrode, filled with neutral calcium ion carrier, ETH-1001. The resting tension, the membrane potential and the intracellular calcium ion activities were recorded in normal Tyrode solution and compared with those in the contracture induced by 10 mM Vanadate. The following results were obtained: 1. The dose-response relationship between the contraction of Vanadate and twitch tension showed near-maximum response in 5mM with no corresponding changes in action potential. 2. The resting tension increased up to the amplitude of a control twitch in 10mM Vanadate with resting membrane potential, hyperpolarized. 3. Increase in intracellular calcium ion activities proceeded the contracture by 10mM Vanadate which were restored to the control level in accordance with a decrease of intracellular calcium ion activities. 4. The amplitude of contractures by 10mM Vanadate were 90-120% of the control twitch tension in which the intracellular calcium ion activities were increased about 70 times from p Ca, 7.1 in the control to p Ca, 5.8 in contractures.
BACKGROUND: Gypsum($CaSO_4{\cdot}2H_2O$) is known as an ideal amendment to improve soil quality of the reclaimed coastal land. Since gypsum has very high concentration of electron acceptor like ${SO_4}^{2-}$, its application might be effective on reducing $CH_4$ emission during rice cultivation, but its effect has not been studied well. METHODS AND RESULTS: The effect of gypsum on $CH_4$ emission and rice growth characteristics was studied by pot test, which was packed by reclaimed paddy soils collected from Galsa, Hadong, Gyeongnam province. Chemical-grade gypsum was applied in two soils having EC 2.25 and 9.48 dS/m at rates of 0, 0.5, 1.0 and 2.0%(wt/wt). $CH_4$ emission was characterized a week interval by closed chamber method during rice cultivation. $CH_4$ emission rate was significantly decreased with increasing salt accumulation and gypsum application levels. With increasing gypsum application, dissolved ${SO_4}^{2-}$ concentration in the leachate water was significantly increased, which might have suppressed $CH_4$ production in soil. Total $CH_4$ flux was dramatically decreased with increasing gypsum application. In contrast, rice yield was increased with increasing gypsum application and then achieved maximum productivity at 1.0% gypsum application in two soils. CONCLUSION(s): Gypsum is a very good soil amendment to suppress $CH_4$ emission in reclaimed coastal paddy soils, and improve rice productivity and soil properties. The optimum application level of gypsum is assumed at ca. 1% to improve soil productivity with reducing effectively $CH_4$ emission during rice cultivation.

Due to the attractive benefits with regard to bone health, digestion, and hydration, carbonated water consumption have rapidly grown over the past few years. However, the acidic drink has latent potential for enamel erosion. The most experimental studies about the enamel erosion have focused on the carbonated beverages with sugar and artificial sweeteners. Here, we determined the enamel erosion potential by commercially available carbonated waters with bovine teeth. The erosion was verified by pH value, calcium concentration, and scanning electron microscope. Then plaque accumulation by bacterial adhesion was determined on the enamel erosion surface to measure roughness. In the present study, we observed that the increased calcium content after being immersed in carbonated waters result from the overall enamel erosion. There were no significant differences between general carbonated waters and mineral waters for erosive capacity. Therefore, commercially available carbonated waters are potentially erosive. In addition, oral bacteria strongly adhered to the erosive enamel surfaces thereby facilitating the development of dental plaque. Thus, it is urgently necessary to provide food safety information on the carbonated water as acidic drink to prevent the enamel erosion.
