In-situ Precipitation of Arsenic and Copper in Soil by Microbiological Sulfate Reduction (미생물학적 황산염 환원에 의한 토양 내 비소와 구리의 원위치 침전)

  • Microbiological sulfate reduction is the transformation of sulfate to sulfide catalyzed by the activity of sulfate-reducing bacteria using sulfate as an electron acceptor. Low solubility of metal sulfides leads to precipitation of the sulfides in solution. The effects of microbiological sulfate reduction on in-situ precipitation of arsenic and copper were investigated for the heavy metal-contaminated soil around the Songcheon Au-Ag mine site. Total concentrations of As, Cu, and Pb were 1,311 mg/kg, 146 mg/kg, and 294 mg/kg, respectively, after aqua regia digestion. In batch-type experiments, indigenous sulfate-reducing bacteria rapidly decreased sulfate concentration and redox potential and led to substantial removal of dissolved As and Cu from solution. Optimal concentrations of carbon source and sulfate for effective microbial sulfate reduction were 0.2~0.5% (w/v) and 100~200 mg/L, respectively. More than 98% of injected As and Cu were removed in the effluents from both microbial and chemical columns designed for metal sulfides to be precipitated. However, after the injection of oxygen-rich solution, the microbial column showed the enhanced long-term stability of in-situ precipitated metals when compared with the chemical column which showed immediate increase in dissolved As and Cu due to oxidative dissolution of the sulfides. Black precipitates formed in the microbial column during the experiments and were identified as iron sulfide and copper sulfide. Arsenic was observed to be adsorbed on surface of iron sulfide precipitate.

Effects of Makgeolli and Makgeolli precipitate on Hepatotoxicity and Serum Lipid Content in Rats (막걸리와 막걸리 침전물이 간 독성 및 혈청 지질함량에 미치는 영향)

  • This study was conducted to investigate the effect of Makgeolli and Makgeolli precipitate on hepatotoxity and the serum lipid content in rats. First, we investigated the effect of Makgeolli and ethanol on the progress of alcoholic fatty liver. The effect of Makgeolli precipitate on carbon tetrachloride ($CCl_4$)-induced hepatotoxicity in the rats was then studied. Indicators of the health status of the experimental period, the body weight gain in ethanol-treated group tended to be lower than those in the control and the Makgeolli-treated groups. The weight of the liver tissue decreased significantly following the administration of ethanol. However, this was not seen following the administration of Makgeolli. The activities of serum aspartate transaminase (AST) and alanine transaminase (ALT) were decreased in the Makgeolli group compared to the ethanol group. Serum cholesterol concentrations increased in the ethanol group, but decreased in the Makgeolli-treated group to an equal volume of the ethanol-treated group. The serum HDL-cholesterol content was significantly higher in the Makgeolli group than in the ethanol group. Analysis of the impact of the Makgeolli precipitate on toxicity induced by $CCl_4$ in the liver showed that the $CCl_4$ treatment significantly increased the activities of serum ALT and AST. However, the levels of cholesterol and triglyceride in serum were decreased. The $CCl_4$ treatment increased the activities of AST and ALT. However, the raw Makgeolli precipitate decreased their activities. Moreover, raw Makgeolli precipitate significantly reduced the $CCl_4$-induced elevation of serum lipids more than heated Makgeolli precipitate. These results suggest that raw Makgeolli precipitate may exert a protective effect against $CCl_4$-induced liver injury by preventing lipid peroxidation.

Production of Concentrated Magnesium Solution from Seawater Using Industrial By-products (산업부산물을 이용한 해수로부터 고농도 마그네슘 용액의 제조)

  • With the purpose of concentrating the magnesium from seawater, in this study, we have conducted experiments to precipitate the magnesium using three precipitants (NaOH, cement kiln dust, and paper sludge ash), and then extract it using sulfuric acid. Results show that the precipitation and extraction performance does not depend so significantly on the kind of precipitant: for all the precipitants used, the precipitation efficiency is measured almost 100% and the extraction efficiency is in the short range of 77 - 89%. The magnesium concentration in the extract is measured 4975 - 5775 mg/L, implying that the magnesium in seawater should be 3.8 - 4.4 times concentrated through the precipitation and extraction reactions. The reaction efficiency of the industrial by-product used as a precipitant in this study is almost 100% similar to those of the other existing expensive precipitants.

Efficiency of Apatite and Limestone in Removing Arsenic from Acid Rock Drainage at the Goro Abandoned Mine (인회석 및 석회석을 이용한 고로폐광산 ARD 내의 비소 저감효율 연구)

  • An active apatite drainage system has been developed at the Goro abandoned mine, comprising a grit cell, a reaction cell, and a precipitation pond. Leachate from an abandoned adit and tailing ponds is collected in a pipeline and is transported to the apatite drainage system under the influence of the hydraulic gradient. The results of a laboratory experiment performed in 2004 indicate that the reaction cell requires 38.8 ton/year of apatite and that precipitate will have to be removed from the precipitation pond every 3 months. The purpose of this study is to evaluate a laboratory test on the efficiency of limestone and apatite in removing arsenic from ARD (acid rock drainage), and to evaluate the suitability of materials for use as a precipitant for the leachate treatment disposal system. The laboratory tests show that the arsenic removal ratios of limestone and apatite are 67.4%-98.3%, and the arsenic removal ratio of apatite is inversely proportional to its grain size. The arsenic compounds are assumed to be Johnbaumnite and Ca-arsenic hydrate. Therefore, apatite and phosphorous limestone can be used as a precipitant for the removal of arsenic, although it is difficult to remove arsenic from ARD when it occurs in low concentrations.

  • 본 논문은 철강공장의 냉각수 처리 시스템에서 고로 집진수를 대상으로 한 응집교반실험을 실시한 결과, 주기적인 관 갱생작업(산세척)과 분산제의 주입이 필요하며, 음이온 고분자응집제에 양이온 고분자응집제를 첨가하여 사용할 경우에 탁도 제거측면에서 우수하였다. 급속교반 후 완속교반을 적용할 경우 급속교반 후 침전보다 탁도 제어 효과가 더 우수한 것으로 관찰되었으므로, 침전지의 처리효율을 증가시키기 위해서는 급속교반과 완속교반이 동시에 필요하였다.

Sorption of Eu(III) and Th(IV) on Bentonite Colloids Considering Their Precipitation and Colloid Formation (침전 및 콜로이드 형성을 고려한 Eu(III)와 Th(IV)의 벤토나이트 콜로이드에 대한 수착)

  • In this study, a sorption experiment of multivalent nuclides such as Eu(III) and Th(IV) relatively stable for redox reactions was carried out for bentonite colloids which had been prepared from the domestic Gyeongju bentonite. The amounts of the nuclides lost by an attachment to bottle walls, by a precipitation, and by a colloid formation were estimated by performing blank tests for the sorption experiments. Sorption coefficients, $K_d's$, reflecting the mass losses were obtained and investigated for the sorption of Eu(III) and Th(IV) onto the bentonite colloids. The net sorption coefficients $K_d's$ considering all the three mass losses were measured as about $10^6-10^7\;mL/g$ and $7{\times}10^6-10^7\;mL/g$ for Eu(III) and Th(IV), respectively, depending on pH. In particular, a precipitation occurred mainly at a pH greater than 5 for Eu(III) and a precipitation and colloid formation significantly occurred at a pH greater than 3 for Th(IV). The precipitation and colloid formation of the multivalent nuclides of Eu(III) and Th(IV) therefore should be considered when $K_d's$ are rightly obtained over the pH range where their precipitation and colloid formation become significant at a given concentration.

  • The effect of ferrous/ferric molar ratio and precipitants on the formation of nano size magnetite particle was investigated by coprecipitation method. Ferrous sulfate and ferric sulfate were used as iron sources and sodium hydroxide and ammonium hydroxide was used as a precipitant. Single phase magnetite was synthesized with all of experiment conditions (ferrous/ferric molar ratios and precipitants). Particle size was smaller, and particle size distribution was narrower when NaOH was used than $NH_4OH$ was used. The crystallinity and particle size was increased and narrower particle size distribution with increasing molar ratio ferrous/ferric sulfate with the same precipitant. Super paramagnetism could be obtained at all of experiment conditions. The highest saturation magnetization (72 emu/g) was obtained when the ferrous/ferric molar ratio was 2.5 and precipitant was used $NH_4OH$.

Neutralization of Acid Rock Drainage from the Dongrae Pyrophyllite Deposit: A Study on Behavior of Heavy Metals (동래 납석광산 산성 광석배수의 중화실험: 중금속의 거동 특성 규명)

  • In this study, we have investigated the geochemical behavior and fate of heavy metals in acid rock drainage (ARD). The ARD was collected from the area of the former Dongrae pyrophyllite mine. The Dongrae Creek waters were strongly acidic (pH : 2.3~4.2) and contained high concentrations of $SO_4$, Al, Fe, Mn, Pb, Cu, Zn, and Cd, due to the influence of ARD generated from weathering of pyrite-rich pyrophyllite ores. However, the water quality gradually improved as the water flows downstream. In view of the change of mole fractions of dissolved Fe, Al and Mn, the generated ARD was initially both Fe- and AA-ich but progressively evolved to more Al-rich toward the confluence with the uncontaminated Suyoung River. As the AR3 (pH 2.3) mixed with the uncontaminated waters (pH 6.5), the pH increased up to 4.2, which caused precipitation of $SO_4$-rich Fe hydroxysulfate as a red-colored, massive ferricrete precipitate throughout the Dongrae Creek. Accompanying the precipitation of ferricrete, the Dongrae Creek water progressively changed to more Al-rich toward downstream sites. At the mouth of the Dongrae Creek, it (pH 3.4) mixed with the Suyoung River (pH 6.9), where pH increased to 5.7, causing precipitation of Al hydroxysulfate (white precipitates). Neutralization of the ARD-contaminated waters in the laboratory caused the successive formation of Fe precipitates at pH<3.5 and Al precipitates at higher pH (4~6). Manganese compounds were precipitated at pH>6. The removal of trace metals was dependent on the precipitation of these compounds, which acted as sorbents. The pHs for 50% sorption ($pH_{50}$) in Fe-rich and Al-rich waters were respectively 3.2 and 4.5 for Pb, 4.5 and 5.8 for Cu, 5.2 and 7.4 for Cd, and 5.8 and 7.0 for Zn. This indicates that the trace metals were sorbed preferentially with increasing pH in the general order of Pb, Cu, Cd, and Zn and that the sorption of trace metals in Al-rich water occurred at higher pH than those in Fe-rich water. The results of this study demonstrated that the partitioning of trace metals in ARD is not only a function of pH, but also depends on the chemical composition of the water.

An Experimental Study on Flocculation and Settling of Fine-grained Suspended Sediments (부유물질의 응접작용 및 침전특성에 관한 실험적 연구)

  • A laboratory flume experiment, using turbulence-generating acryl tank and natural sediments, was conducted to investigate the effects of salinity, concentration of suspended sediment, turbulence and clay minerals on the flocculation and settling of fine-grained suspended sediments. While experiments were run, a sequence of water samples were taken near the bottom of the tank to analyze the variations of size distribution and relative contents of clay minerals. The results of the salinity experiment indicate that median settling velocity ($W_{50}$) increases linearly with salinity. Different settling processes of suspended sediments under variable concentrations appear to be predictable, depending upon the range of the suspension concentration. At concentrations less than 200 mg/l, $W_{50}$ is rarely varied with concentration probably because of the individual--grain settling mode. In the range of 200 to 13,000 mg/l show $W_{50}$ and concentration a good relationship following an empirical formula: $W_{50}=0.45C^{0.44}$. This relationship, however, no longer holds in concentrations exceeding 13,000 mg/l; instead, a more or less reverse one is shown. This result suggests an effect of hindered settling. The turbulence effect is somewhat different from that of concentration. Turbulence accelerates the flocculation and settling susepended sediments at low concentration (200 mg/l), whereas at high concentration turbulence breaks floes down and impedes the settling. Size distribution of suspended sediments sampled near the bottom of the tank tend to be more negatively skewed and leptokurtic in turbulent conditions compared to those in static conditions. The clay mineral analysis from the sequential water samples shows that over time the content of smectite decreases most rapidly with illite remaining concentrated in suspension. This means that smectite, among other clay minerals, plays the most effective role in the flocculation of fine-grained sediment in saline water.

Extraction and Separation of Protein-bound Polysaccharide by Lentinus edodes (표고버섯 배양액으로부터 단백다당류의 추출 및 정제 방법)

  • The extraction and separation methods of protein-bound polysaccharides from the mycelium and culture broth of L. edodes were investigated. The use 2% solution of surface active agent, Triton X-100 was effective for extraction of the protein-bound polysaccharide from the mycelium. The extraction of the protein-bound polysaccharides from mycelium with hot water was achieved by 4 hours extraction at 10$0^{\circ}C$. For the separation and partial purification of the protein bound polysaccharides the column chromatography using DEAE-Cellulose, DEAE-Sephadex and Sephadex proved to be effective.
