A Study on the Cementation Reaction of Cadmium by Zinc Powders from Leaching Solution of Waste Nickel-Cadmium Batteries (폐니켈-카드뮴 전지 침출액으로부터 아연 분말을 이용한 카드뮴의 치환반응에 대한 연구)

  • Cementation is one of economical and efficient recycling method precipitating the metal ion in solution by adding another active metal. In this study for optimizing cadmium recovery efficiency, it was performed as a function of the effect of pH, temperature, particle size, and input amount of zinc in 0.1 M $CdSO_4$ solution and Ni-Cd battery leaching solutions, respectively. The particle size of zinc and temperature were key factors for Cd cementation and it was confirmed that the input amount of 2.6 of Zn/Cd ratio using granular-type zinc was optimal condition for selective Cd recovery efficiency at $25^{\circ}C$.

Removal Efficiency of Settleable Solids in Seawater Aquaculture Farm Wastewater (하이드로싸이클론을 이용한 해수 양식장 침전 고형물의 제거 효율 평가)

  • Flow-through aquaculture systems generate large amounts of wastewater containing compounds such as solids that can settle near aquafarms and cause eutrophication. The settled solids are often reintroduced into flow-through systems, and aquatic animals can be affected by the solids and pathogens associated with these solids. For a sustainable aquaculture operation, adequate wastewater treatment is required. Hydrocyclones are one of the most promising technologies for the removal of solids in aquaculture wastewater. In this study, a model for performance prediction of hydrocyclones was investigated under three different operating conditions: water temperature, solids concentration, and water inlet velocity. The synthetic solids solution was prepared using settled solids from abalone aquaculture farms. The daily solids removal rates of the tested hydrocyclones ranged from 0.18 to 26.0 g solids-m-3-day-1, and removal efficiency ranged from 5.1 to 34.4%. The inlet water velocity had the greatest effect on solids removal and hydrocyclone efficiencies. The following multiregression model equation was derived from the daily solids removal rate (g solids-m-3-day-1) results for water temperature (T, ℃), solids concentration (SS, mg-L-1), and tangential inlet water velocity (TIV, m-sec-1): daily solids removal rate: f(z)=4.465+0.809TIV-0.375T+0.217SS (r2=0.976).

A Study on Alumina Nanoparticle Dispersion for Improving Injectivity and Storativity of CO2 in Depleted Gas Reservoirs (고갈 가스전에서 CO2 주입성 및 저장성 향상을 위한 알루미나 나노입자의 분산 특성 연구)

  • In this study, the Al2O3 nanofluid was synthesized as an additive for improving the injection efficiency and storage capacity of carbon dioxide (CO2) in a depleted sandstone reservoir or deep saline aquifer. As the base fluid, deionized water (DIW) and saline prepared by referring to the composition of API Brine were used, and the fluid was synthesized by using Al2O3 nanofluid with CTAB (cetyltrimethyl-ammonium bromide), a cationic surfactant. After that, the dispersion stability was evaluated by using visual observation, dynamic light scattering (DLS), transmission electron microscope (TEM), and miscibility test. As a result, it was presented that stable nanofluid without agglomeration and precipitation after reaction with 70,000 ppm of brine could be synthesized when the nanoparticle concentration was 0.05 wt% or less.

Prediction of Coagulation/Flocculation Treatment Efficiency of Dissolved Organic Matter (DOM) Using Multiple DOM Characteristics (다중 유기물 특성 지표를 활용한 용존 유기물질 응집/침전 제거효율 예측)

  • The chemical composition and molecular weight characteristics of dissolved organic matter (DOM) exert a profound influence on the efficiency of organic matter removal in water treatment systems, acting as efficiency predictive indicators. This research evaluated the primary chemical and molecular weight properties of DOM derived from diverse sources, including rivers, lakes, and biomasses, and assessed their relationship with the efficiency of coagulation/flocculation treatments. Dissolved organic carbon (DOC) removal efficiency through coagulation/flocculation exhibited significant correlations with DOM's hydrophobic distribution, the ratio of humic-like to protein-like fluorescence, and the molecular weight associated with humic substances (HS). These findings suggest that the DOC removal rate in coagulation/flocculation processes is enhanced by a higher presence of HS in DOM, an increased influence of externally sourced DOM, and more presence of high molecular weight compounds. The results of this study further posit that the efficacy of water treatment processes can be more accurately predicted when considering multiple DOM characteristics rather than relying on a singular trait. Based on major results from this study, a predictive model for DOC removal efficiency by coagulation/flocculation was formulated as: 24.3 - 7.83 × (fluorescence index) + 0.089 × (hydrophilic distribution) + 0.102 × (HS molecular weight). This proposed model, coupled with supplementary monitoring of influent organic matter, has the potential to enhance the design and predictive accuracy for coagulation/flocculation treatments targeting DOC removal in future applications.

Efficiency of Poultry Manure Biochar for Stabilization of Metals in Contaminated Soil (계분 바이오차를 이용한 토양 중금속 안정화 효율 평가)

  • Stabilization of heavy metals such as Pb, Cd, Zn, and Cu was evaluated in contaminated soil treated with poultry manure (PM) as well as its biochars pyrolyzed at $300^{\circ}C$ (PBC300) and $700^{\circ}C$ (PBC700) at the application rates of 2.5, 5.0, and 10.0 wt% along with the control, prior to 21-days incubation. After incubation, soil pH was increased from 6.94 (control) to 7.51, 7.24, and 7.88 in soils treated with PM 10 wt%, PBC300 10 wt%, and PBC700 10 wt% treatments, respectively, mainly due to alkalinity of treatments. In the soil treated with PM, the concentrations of the toxicity characteristic leaching procedure (TCLP)-extractable Pb, Cd, Zn, and Cu were increased by up to 408, 77, 24, and 955%, respectively, compared to the control. These increases may possibly be associated with an increased dissolved organic carbon concentration by the PM addition. However, in the soil treated with PBC700, TCLP-extractable Pb, Cd, Zn, and Cu concentrations were reduced by up to 23, 38, 52, and 36%, respectively, compared to the control. Thermodynamic modelling using the visual MINTEQ was done to predict the precipitations of $Pb(OH)_2$, $Cu(OH)_2$ and P-containing minerals, such as chloropyromorphite [$Pb_5(PO_4)_3Cl$] and hydroxypyromorphite [$Pb_5(PO_4)_3OH$], in the PBC700 10 wt% treated soil. The SEM-elemental dot mapping analysis further confirmed the presence of Pb-phosphate species via dot mapping of PBC700 treated soil. These results indicate that the reduction of Pb concentration in the PBC700 treated soil is related to the formations of chloropyromorphite and hydroxypyromorphite which have very low solubility.

The Optimal Condition of Performing MTT Assay for the Determination of Radiation Sensitivity (방사선 감수성 측정법으로서 MTT 법 시행 시의 최적 조건에 대한 연구)

  • Purpose : The measurement of radiation survival using a clonogenic assay, the established standard, can be difficult and time consuming. In this study, We have used the MTT assay, based on the reduction of a tetrazolium salt to a purple formazan precipitate by living cells, as a substitution for clonogenic assay and have examined the optimal condition for performing this assay in determination of radiation sensitivity. Materials and Methods : Four human cancer cell lines - PCI-1, SNU-1066, NCI-H630 and RKO cells have been used. For each cell line, a clonogenic assay and a MTT assay using Premix WST-1 solution, which is one of the tetrazolium salts and does not require washing or solubilization of the precipitate were carried out after irradiation of 0, 2, 4, 6, 8, 10 Gy. For clonogenic assay, cells in $25\;cm^2$ flasks were irradiated after overnight incubation and the resultant colonies containing more than 50 cells were scored after culturing the cells for $10\~14$ days. For MTT assay, the relationship between absorbance and cell number, optimal seeding cell number, and optimal timing of assay was determined. Then, MTT assay was performed when the irradiated cells had regained exponential growth or when the non-irradiated cells had undergone four or more doubling times. Results : There was minimal variation in the values gained from these two methods with the standard deviation generally less than $5\%$, and there were no statistically significant differences between two methods according to t-test in low radiation dose (below 6 Gy). The regression analyses showed high linear correlation with the $R^2$ value of $0.975\~0.992$ between data from the two different methods. The optimal cell numbers for MTT assay were found to be dependent on plating efficiency of used cell line. Less than 300 cells/well were appropriate for cells with high plating efficiency (more than $30\%$). For cells with low plating efficiency (less than $30\%$), 500 cells/well or more were appropriate for assay. The optimal time for MTT assay was after 6 doubling times for the results compatible with those of clonogenic assay, at least after 4 doubling times was required for valid results. In consideration of practical limits of assay (12 days, in this study) cells with doubling time more than 3 days were inappropriate for application. Conclusion : In conclusion, it is found that MTT assay can successfully replace clonogenic assay of tested cancer cell lines after irradiation only if MTT assay was undertaken with optimal assay conditions that included plating efficiency of each cell line and doubling time at least.

Production and Characterization of Extracellular Polysaccharide Produced by Pseudomonas sp. GP32 (Pseudomonas sp. GP32에 의해 생산된 세포 외 다당류의 생산 및 특성)

  • A strain GP32 which produces a highly viscous extracellular polysaccharide was conducted with soil samples and identified as Pseudomonas species. The culture flask conditions for the production of extracellular polysaccharide by Pseudomonas sp. GP32 were investigated. The most suitable carbon and nitrogen source for extracellular polysaccharide production were galactose and (NH4)2SO4. The optimum carbon/nitrogen ratio for the production of extracellular polysaccharide was around 50. The optimum pH and temperature for extracellular polysaccharide production was 7.5 and 32℃, respectively. In batch fermentation using a jar fermentor, the highest extracellular polysaccharide content (15.7 g/l) was obtained after 70 hr of cultivation. The extracellular polysaccharide produced by Pseudomonas sp. GP32 (designated Biopol32) was purified by ethanol precipitation, cetylpyridinium chloride (CPC) precipitation, and gel permeation chromatography. Biopol32, which has an estimated molecular weight of over 3×107 datons, is a novel polysaccharide derived from sugar components consisting of galactose, glucose, gulcouronic acid and galactouronic acid in an approximate molar ratio of 1.85 : 3.24 : 1.00 : 1.42. The solution of Biopol32 showed non-Newtonian characteristics. The viscosity of Biopol32 exhibited appeared to be higher at all concentration compared to that of zooglan from Zoogloea ramigera. An analysis of the flocculating efficiency of Biopol32 in industry wastewater (food, textile, and paper wastewater) revealed chemical oxygen demand (COD) reduction rates 58.4-67.3% and suspended solid (SS) removal rates 82.6-91.3%. Based on these results, Biopol32 is a possible candidate for industrial applications such as wastewater treatment.

Removal of Dissolved Organic Matter by Ozone-biological Activated Carbon process (오존처리와 생물활성탄 공정에 의한 상수원수 중의 용존유기물 제거)

  • The removal yield of dissolved organic matter in drinking water by biological activated carbon (BAC) process was investigated. The tested processes wer raw water-AC process (BAC1), raw water-ozonation-BAC process (BAC2), and raw water-ozonation-coagulation/sedimentation-BAC process (BAC3). The amounts of organic matter was measured as dissolved organic carbon (DOC), ulta-violet radiation at 254 nm wavelength ($UV_{254}$), total nitrogen (T-N), ammonia nitrogen (NH_3$-N), and total phosphate (T-P). As a results, 30.7% DOC was removed by BAC2 process, which showed higher removal efficiency than BAC1 or BAC3 processes. The removal yield of $UV_{254}$ in BAC1, BAC2, and BAC3 processes were observed as 45.3%, 44.6%, 58.4%, respectively. And the removal yield of ammonia nitrogen were 66%, 81%, 29% in each BAC processes. The optimal empty bed contact time (EBCT) of BAC processes was estimated as 10 minute. This study has shown that BAC process combined with ozone treatment was efficient for removing dissolved organic matter in water.

Removal of Red Tide Organisms -1. flocculation of Red Tide Organisms by Using IOSP- (적조생물의 구제 -1. IOSP에 의한 적조생물의 응집제거-)

  • This study was to examine the physicochemical characteristics of coagulation reaction between ignited oyster shell powder (IOSP) and red tide organisms (RTO), and its feasibility, in developing a technology for the removal of RTO bloom in coastal sea,IOSP was made from oyster shell and its physicochemical characteristics were examined for particle size distribution, surface characteristic by scanning electron microscope, zeta potential, and alkalinity and pH variations in sea water. Two kinds of RTO that were used in this study, Cylindrotheca closterium and Skeletonema costatum, were sampled in Masan bay and were cultured in laboratory. Coagulation experiments were conducted using various c(Incentrations of IOSP, RTO, and a jar tester. The supernatant and RTO culture solution were analyzed for pH, alkalinity, RTO cell number, IOSP showed positive zeta potentials of $11.1{\~}50.1\;mV\;at\;pH\;6.2{\~}12.7$, A positive zeta potential of IOSP slowly decreased with decreasing pNa 4,0 to 2,0. When pNa reached zero, the zeta potential approached zero, When a pMg value was decreased, the positive zeta potential of IOSP increased until pMg 3.0 and decreased below pMg 3.0. IOSP showed 4.8 mV of positive zeta potential while RTO showed -9.2 mV of negative zeta potential in sea water. A positive-negative EDL (electrical double-layer) interaction occurred between $Mg(OH)_2$ adsorption layer of IOSP and RTO in sea water so that EDL attractive force always worked between them. Hence, their coagulation reaction occurred at primary minimum on which an extreme attractive force acted because of charge neutralization by $Mg(OH)_2$ adsorption layer of IOSP. As a result, the coagulation reaction was rapidly processed and was irreversible according to DLVO (Deriaguin-Landau-Verwey-Overbeek) theory. Removal rates of RTO were exponentially increased with increasing both IOSP concentration and G-value. The removal rates were steeply increased until 50 mg/l of IOSP and reached $100{\%}\;at\;400\;mg/l$ of IOSP. Removal rates of RTO were $70.5,\;70.5,\;81.7,\;85.3{\%}$ for G-values of $1,\;6,\;29,\;139\;sec^(-1)$at IOSP 100 mg/l, respectively. This indicated that mixing (i.e., collision among particles) was very important for a coagulation reaction.

The Effect of Strong Acid and Ionic Material Addition in the Microwave-assisted Solubilization of Waste Activated Sludge (Microwave를 이용한 폐활성슬러지의 가용화 반응에서 강산과 이온성 물질의 첨가가 미치는 영향)

  • The study of waste activated sludge (WAS) solubilization has been increased for sludge volume reduction and enhancing the efficiency of anaerobic digestion. Microwave (MW)-assisted solubilization is an effective method for the solubilization of WAS because this method can lead to thermal, nonthermal effect and ionic conduction by dielectric heating. In this study, the solubilization of WAS by MW heating and conductive heating (CH) was compared and to enhance the MW-assisted solubilization of WAS at low MW output power, chemical agents were applied such as $H_2SO_4$ as the strong acid and $CaCl_2$, NaCl as the ionic materials. Compared to the COD solubilization of WAS by CH, that by MW heating was approximately 1.4, 6.2 times higher at $50^{\circ}C$, $100^{\circ}C$, respectively and the highest COD solubilization of WAS was 10.0% in this study of low MW output power condition. At the same MW output power and reaction time in chemically agents assisted experiments, the COD solubilization of WAS were increased up to 18.1% and 12.7% with the addition of $H_2SO_4$ and NaCl, however, that with the addition of $CaCl_2$ was 10.7%. This result might be due to the fact that the precipitation reaction occurred by calcium ion ($Ca^{2+}$) and phosphate ion (${PO_4}^{3-}$) produced in WAS after MW-assisted solubilization. In this study, $H_2SO_4$ turned out to be the optimal agent for the enhancement of MW efficiency, the addition of 0.2 M $H_2SO_4$ was the most effective condition for MW-assisted WAS solubilization.
