In this study, the antibacterial and phagocytosis regulation effects of Hordeum vulgare extract and pine needle extract on S. mutans, the causative bacteria of dental caries, were investigated. Ethanol extracts of domestic Hordeum vulgare powder and pine needle powder were used, and the antibacterial and phagocytic ability against S. mutans was confirmed according to the concentration of the extracts. As a result, S. mutans colony formation did not show a significant difference in the Hordeum vulgare extract but was significantly decreased in the pine needle extract. As a result of confirming the phagocytic ability of THP-1 cells for S. mutans, there was no significant difference in the Hordeum vulgare extract, but the phagocytic ability of immune cells was improved in the pine needle extract. Therefore, it suggests that pine needle extract can be used as a material for preventing dental caries.
This study was executed to evaluate the effects of natural herbal medicine extract on the antimicrobial activity and the artificial plaque formation. Schisandra chinensis, Sophora flavescens, Dryopteris crassirhizoma and Aristolochia contorta were extracted by 80% MeOH respectively and each concentration(5mg, 10mg, 20mg) of herbal medicine extract was tested to show the antimicrobial activity against S. mutans. To show the inhibition effect of herbal medicine extract on the artificial dental plaque formation each extract of 0.4% concentration was added into M 17 broth and inoculated with $5.0{\times}10^6$ of S. mutans. After 24 hour incubation each weight of artificial dental plaque was measured and compared with each other. Results obtained are as follows: 1. Dryopteris crassirhizoma showed very intense antimicrobial effect but Schisandra chinensis, Sophora flavescens and Aristolochia contorta showed a little difference against control. 2. Regarding artificial plaque formation inhibiting activity by each herbal extract at 0.4% concentration, Dryopteris crassirhizoma showed the perfect inhibition effect. Schisandra chinensis and Aristolochia contorta followed in order but Sophora flavescens showed the worst value. In conclusion, Dryopteris crassirhizoma showed strong antimicrobial effect against S. mutans and significant inhibiting effect on the artificial dental plaque formation.
The antagonistic microorganisms against Streptococcus sanguis, S. salivarius and S. mutans causing the dental caries of oral diseases were isolated from Korean traditional soy sauce. Twenty five strains were isolated by pairing culture, paper disc culture and dual culture methods. The isolate NG 06 strain was observed with various cultural and physiological test, and $Biolog^{(R)}$ Bacterial Identification System. The strain was identified as Bacillus racemilacticus. The isolate NG 16 strain was confirmed to Gram-positive, rods, endospore production, utilization of melibiose, casein hydrolysis and starch hydrolysis. Also the second strain NG 16 was identified as $\beta$. amyloliquefaciens.
This study was designed to investigate the using status and awareness of pit and fissure sealant, and how it affects on secondary caries when performed on above of initial caries. After classifying 446 occlusal surface into four groups, performed pit and fissure sealant, induced artificial caries, used DIAGNOdent (Kavo) to measure degree of secondary caries. Also, distributed a questionnaire on dental clinic in metropolitan area to find out using status and awareness of pit and fissure sealant, the following results were obtained. 1) On inspection and percussion, Group 4 corresponding to the enamel caries showed the highest secondary caries after sealant and was statistically significant difference in the order of initial group, stained group, sound group (p<0.05). 2) Inspection showed the highest percentage on tooth fissure caries diagnostic methods before sealant. 3) 56.6% didn't know about DIAGNOdent, 91.6% didn't have it. 4) In clinically, the most cause of secondary caries after sealant was a broken sealant, making caries on the downward. Based on the results of above study, degree of caries under sealant could affect on secondary caries, needs publicity about the use and necessity of objective fluorescence device.

The aims of this study were to compare the status of dental caries between mental retarded persons(MR) and normal persons and investigated the association among dental caries and oral factors(plaque index, S. mutans, Lactobacillus, buffering capacity, salivary total IgA and anti-S. mutans IgA titers). DMFS index and plaque index were significantly greater in the MR person group than in the normal person group. The concentration of S. mutans-specific IgA was significantly greater in the normal person group than in the MR person group, but that of salivary total IgA was not show the significant difference. Salivary buffering capacity was significantly greater in the normal person group than in the MR person group, but the counts of salivary S. mutans and Lactobacillus were not significantly different. By age group(I: 9-11Y, II: 12-14Y, III: 15-18Y), DMFS index and plaque index were significantly greater in the MR person group than in the normal person group at III. The S. mutans counts and Lactobacillus counts were significantly greater in the normal person group than in the MR person group at I, but those were contrary at II, III. There was a high correlation among caries index and buffering capacity, level of S. mutans and Lactobacillus, plaque index at III.

Dental caries results from localized demineralization of tooth enamel by acids of bacterial origin produced from the fermentation of dietary sugars. A group of related oral bacteria, collectively known as mutans streptococci, are implicated as the primary etiological agents of human caries. Within this group, Streptococcus mutans has been known as a causative agent for dental caries. As well as acid production yielding the demineralization of tooth enamel, adherence and colonization of S. mutans to the teeth are also important for their virulence Cell-surface fibrillar proteins, which mediate adherence to the salivary pellicle are virulence components of mutans streptococci, and primary candidates for a human caries vaccine. Here we report that the AgI/II gene from S. mutans GS-5 were cloned by PCR amplification of the bacterial chromosomal DNA and the integrity of cloned genes were confirmed by nucleotide sequencing. Sequence analyses showed the sequence alignment of 280 nucleotides between the cloned AgI/II and the reported sequence of S. mutans GS-5 showed the perfect match The cloned genes which signal nucleotide was truncated, were transferred into bacterial expression vector and the recombinant proteins were purified as His-tag fusion proteins In order to generate polyclonal antibodies against the recombinant proteins, AgI/II mr, some $100{\mu}g$ of the proteins was injected into mice three times. It can be used for an effective vaccine production to prevent dental caries caused by pathogenic S. mutans.

The aim of this study is to develop and establish rapid, convenient, and accurate method of analyzing salivary IgA against S. mutans semi-quantitatively. Relative salivary IgA titer was calculated as maximum dilution fold of S. mutans protein that was not detected by salivary antibody after measuring relative intensity of the immune blot bands by densitometry. Analyses were performed in caries-experienced and non-experienced children. Mean IgA titer of non-experienced group shows higher level than that of caries-experienced without statistical significance due to high individual variety of antibody titer in non-experienced group: $2^{6.278{\pm}2.260}$ in non-experienced group and $2^{5.730{\pm}0.499}$ in caries-experienced group(p=0.464). Those results suggest that naturally induced salivary IgA antibodies against S. mutans were present in all subjects, but high titer of antibodies were not achieved in caries-experienced group. On the contrary, antibody titer in non-experienced group shows marked individual variations suggesting that antibody production is multifactorial. In conclusion, immune-slot blot method developed in this study would be useful and applicable in semi-quantitative analysis of antibodies.

AgI/II of Streptococcus mutans(S. mutans) is an important virulence factor that contributes to the pathogenesis of S. mutans-induced dental caries. In oral cavity, salivary IgA antibodies act as safeguards against enormous challenges from oral bacteria. IgA antibodies inhibit adherence of cariogenic microorganisms to hard surfaces. Analysis of salivary IgA against AgI/II can be very useful diagnostic and powerful communication tools to the dental caries The purpose of this study was to investigate correlation between salivary AgI/II specific IgA and incidence of dental caries among children and young adults. Subjects consisted of 28 children and 18 adults. They were assigned to four groups : Group I deft index $\leq$3), Group II(deft index $\geq$4), Group III(DMFT index $\leq$3), Group IV(DMFT index $\geq$4) and they was divided two groups into caries resistant group and caries susceptible group. The study group were examined caries activity and their salivary IgA was evaluated by enzyme-linked immunosorbent assay. The results are as follows : 1. There was a positive correlation between the number of S. mutans and caries activity. 2. The titer of salivary IgA against the AgI/II was significantly higher in caries resistant group than caries susceptible group(p<0.01). 3. The titer of salivary IgA against the AgI/II in Group III was significantly higher than Group II(p<0.05).
The purpose of this study is to investigate anticariotic activity of the ethyl acetate soluble extract of Sophora flavescens Ait. for the prevention of dental caries and glucosyltransferase activity caused by Streptococcus mutans. The fraction 5-4-3 showed strong growth inhibition activity against Streptococcus mutans (MIC, 3.13 $\mu\textrm{g}$/ml). The glucosyltransferase activity of the active fraction 5-4-3 inhibited the formation of glucan and showed 77% of the antiproliferative effect at 100 $\mu\textrm{g}$/ml (P<0.05). Two flavanones, (2S)-2'-methoxy kurarinone (1) and (+)-kurarinone (2), were isolated from the active fraction 5-4-3 of the ethyl acetate soluble extract of S. flavescens Ait. Their structures were elucidated using spectroscopic methods.
