Biological Control of Vegetable Insect Pests with Entomopathogenic Nematodes (곤충병원선충을 이용한 채소해충의 생물적 방제)

  • Entomopathogenic nematodes, Steinernema carpocapsae, was able to invade and kill the several lepidopteran pests including the beet armyworm, Spodeptera exigua Hubner, which was the most effective target host. The beet armyworms treated with the effective nematode concentrations were died within 48 hrs. The lethal effect of the nematode was varied among the developmental stages of the host. The fifth instar larvae of the beet armyworm was more vulnerable to the nematode than the third instar larvae. Pupae was, however, refractory to the nematode. All three bioessays (topical application, filter paper test, and soil treatment) showed the positive correlation between the number of the treated nematodes and the mortality of the host. Topical application was the most effective and fast-acting method so that it gave the lethal effect 2 days earlier than did filter paper test at the same number of the treated nematodes. Soil treatment required higher number of the nematodes to get the effective lethality than did filter paper test. The fifth instar larvae of the beet armyworm expressed the specific hemolymph proteins of 5 to 10 kDa in response to nematode infection.

Biological Control Efficacy of an Entomopathogenic Nematode, Heterorhabditis megidis, Against Housefly, Musca domestica, and Flower Beetle, Gametis jucunda (메기디스 곤충병원선충(Heterorhabditis megidis)을 이용한 집파리와 풀색꽃무지의 생물적 방제 효과)

  • An endemic entomopathogenic nematode, Heterorhabditis megidis, was evaluated by its control efficacies against housefly, Musca domestica, and flower beetle, Gametis jucunda. In Petri-dish assay, the pathogenicity of H. megidis showed 456.4 infective juveniles/larva (IJs/larva) in median lethality (LC$_{50}$) against the second instar larvae of M. domestica and 238.9 IJs/larva against the second instar larvae of G. jucunda. This was contrasted with those of the other well-known entomopathogenic nematode, Steinernema carpocapsae, which showed 115.9 IJs/larva against M. domestica and 388.6 IJs/larva against G. jucunda. In field experiment, H. megidis were applied per square meter of pork farm with 1,000,000 IJs of H. megidis or apple orchard with 370,000 IJs, which were infested with M. domestica or G. jucunda, respectively. H. megidis showed 56.9% and 57.3% of control efficacies against M. domestica and G. jucunda, respectively. These results suggest a promising control technique in the field using H. megidis against M. domestica and G. jucunda.a.

Toxicity Evaluation of Chemicals using Asian Toad Embryos, Bufo gargarizans (두꺼비 배아를 활용한 화학물질의 독성평가 연구)

  • In this experiment, embryos of Asian toad, Bufo gargarizans, were investigated to evaluate toxicity of chemicals along FETAX(Frog Embryo Teratogenesis Assay-Xenopus) protocol. Asian toad, Bufo gargarizans, embryos incubated and investigation of Zn and Benomyl effect by probit analysis. As a result, depends on the concentrations of Zn and Benomyl, mortality and malformation rates were increases and larval body length were decreased. The teratogenic concentration($EC_{50}$) of Zn and Benomyl were 2.3, $1.0mg/{\ell}$, respectively and the embryo lethal concentration($LC_{50}$) Zn and Benomyl were 10.3, 6.9, respectively. The teratogenic index(TI) were 4.4 in Zn and 6.7 in Benomyl, thus showed teratogenicity in embryonic development of B. gargarizans. These results reveal that Zn and Benomyl in this experiment suppressed the development of embryos at relatively low concentration. Much of B. gargarizans embryos can be secured, and easy to incubate. In addition, mortality, malformation ratios, malformation patterns and growth rates are similar to the results from the other assay systems. Therefore, the B. gargarizans embryo teratogenesis assay system could be a useful tool to evaluate toxicity of pollutants in environment.

Toxicity Test of Biodiesel and Biodiesel-derived Neopentyl Polyol Ester Lubricant Oil Base Using Earthworm (지렁이를 이용한 바이오디젤과 바이오디젤 유래 Neopentyl Polyol Ester 윤활유 베이스의 독성테스트)

  • Toxicity test for biodiesel (BD), biodiesel-derived neopentyl polyol ester (NPE) lubricant oil base, lubricant oil for diesel engine oil (LODE) and petroleum diesel (PD) was carried out using earthworm, Eisenia fefida. According to the method by OECD 207, the $LC_{50}$ values of BD and NPE were estimated as 2,450 and 1,528 mg/kg, respectively, which indicate that these compounds are classified as slightly toxic compounds. The $LC_{50}$ values of LODE and PD were estimated as 500 and 603mg/kg, respectively, showing that theses compounds are evaluated as moderately toxic compounds.

Studies on Toxicological Evaluation of Pesticides(Fungicide, Insecticide, Herbicide) using Tree Frog Embryos, Hyla japonica (청개구리 배아를 활용한 농약류(살균제, 살충제, 제초제)의 독성평가 연구)

  • This study used the probit analysis to evaluate the toxicity of three chemicals - benomyl (Germicide), carbofuran (insecticide), and thiobencarb (herbicide) - with the FETAX (Frog Embryo Teratogenesis Assay-Xenopus) protocol using the incubated embryos of tree frog, Hyla japonica. The results showed that the larval body length decreased while the mortality and malformation rates increased as the concentrations of benomyl, carbofuran, and thiobencarb increased. The teratogenic concentration ($EC_{50}$) of benomyl, carbofuran, and thiobencarb were 1.00, 0.58, 4.75 mg/L, respectively, indicating that the malformation of larvae was the most sensitive to carbofuran. The embryo lethal concentration ($LC_{50}$) was 7.04, 28.71, and 16.12mg/L, respectively, indicating that benomyl showed the lowest embryo lethal concentration. The teratogenic index (TI) was 7.04 in Benomyl, 49.50 in Carbofuran, and 3.39 in Thiobencarb, indicating that the TI values were above 1.5, which is the criterion of teratogenicity, for all three chemicals. All three pesticides examined by this study were considered to be the most teratogenic substances, and the carbofuran was the most potent teratogen.

  • 카바메이트계 농약인 카보푸란은 어류에 대한 독성이 매우 높으며, 낮은 농도에서 어류의 척추기형이나 행동이상을 유발한다. 이러한 카보푸란의 독성기전을 밝히기 위한 일환으로 신경물질대사와 관련이 깊은 acetylcholine esterase(AChE)와 monoamine oxidase(MAO)에 미치는 농약의 효과를 송사리(Oryzias latipes; Medaka fish)를 이용하여 평가하였다. Medaka fish에 대한 카보푸란의 반수치사농도(LC$_{50}$)는 2.5 ppm이었으며, 1 ppb 카보푸란에 24시간 노출된 경우, AChE 효소활성이 머리와 몸통부위에서 각각 30, 20%씩 감소되었다. 한편, MAO 효소활성은 카보푸란의 농도가 증가함에 따라 머리부위에서는 감소한 반면, 몸통부위에서는 증가하는 경향을 보였다. 특히 카보푸란의 농도가 1 ppb에서도 송사리의 MAO 효소활성이 영향을 받는 것으로 나타나, 카보푸란에 의한 송사리의 행동이상은 AChE 활성 뿐 아니라 MAO활성의 변화에 의한 복합적인 효과일 가능성이 높다.

Acute Toxicity of Kuwait Crude Oil (WSF) to Mysid, Neomysis awatschensis (Neomysis awatschensis에 미치는 원유의 급성독성)

  • Acute toxicity of crude oil (WSF) on the mortality and respiration rates of Neomysis awatschensis was examined. This experiment was conducted by static and short-term bioassay procedure. In lethal test, the test animals were exposured to 8 different concentrations to determine $LC_{50}$ value (median lethal concentration). The concentrations of total hydrocarbon of 96-hr $LC_{50}$ value at $14^{\circ}C\;and\;20^{\circ}C$ were 1.01 ppm and 0.78 ppm, respectively. $LT_{50}$ (the median lethal time) also was determined. The $LT_{50}$ of 0.56 ppm was found within 100 hours, while the $LT_{50}$ of 5.6 ppm was 21 hours at $14^{\circ}C$. At $20^{\circ}C$, the $LT_{50}$ values of 0.56 ppm and 5.60 ppm were 95 hours and 17 hours, respectively. There was little difference between two temperature experiments. The effect of WSF on respiration rate was more sensitive than that on mortality, but no considerable difference was shown between different concentrations in this experiment. The results of these experiments indicated that relatively low concentration of dissolved crude oil fraction can impact on small crustacean in the marine ecosystem.

Biological Control of Beet Armyworm, Spodoptera exigua(Lepidoptera: Noctuidae) with Entomopathogenic Nematodes(Steinernematid and Heterorhabditid) in Greenhouse (시설재배지에서 곤충병원성 선충, Steinernematid와 Heterorhabditid를 이용한 파밤나방(Spodoptera exigua)의 생물적 방제)

  • Five strains of Korean entomopathogenic nematodes(EPN), steinernematids and heterorhabditids(Steinernama carpocapsae GSN1, Steinernema sp. GSNUS-10, Steinernama sp. GSNUS-14, Heterorhabditis bacteriophora Hamyang, Heterorhabditis sp. GSNUH-1) were evaluated and tried in petri dish, pot, and vegetable greenhouses for environmentally friendly control of beet armyworm, Spodoptera exigua. $LC_{50}$ values of five EPN strains against beet armyworm was different depending on nematode strain and beet armyworm instar. $LC_{50}$ value of Steinernema carpocapse GSN1(GSN1) was 3.8-5.1 infective juveniles(Ijs) in 2nd to 4th instars of beet armyworm. Pathogenicity of five EPN strains against beet armyworm different in nematode strain, concentration, application times, and vegetable species in pot and greenhouse. Steinernema spp. was more effective than Heterorhabditis spp. against beet armyworm. Two or three times of applications of EPN were found to be effective regardless of nematode strain and concentration in pot and greenhouse. ENP showed different reactions on vegetable species. Efficacy of EPN was higher on Chinese cabbage than that on cabbage and kale. GSN1 was one of the most effective nematodes and 100,000 infective juveniles per $m^2$(720,000 Ijs/$7.2m^2=1{\times}10^9$Ijs/ha) resulted in higher mortality in greenhouse.

Susceptibility of the Alfalfa Weevil, Hypera postica (Coleoptera: Curculionidae) to Korean Entomopathogenic Nematodes in Laboratory Assays (한국산 곤충병원성 선충에 대한 알팔파바구미의 감수성 실내 검정)

  • Laboratory tests were conducted to determine the susceptibility of the alfalfa weevil, Hypera postica to Korean entomopathogenic nematodes, Steinernema carpocapsae GSN1 strain (ScG), S. glaseri Dongrae strain (SgD), Heterorhabditis bacteriophora Hamyang strain (HbH), and Heterorhabditis sp. Gyeongsan strain (HeG) at the petri dish assay. The larval mortality of H. postica was significantly different depending on nematode species and treatment concentration. SgD and HeG strain were more effective against H. postica larva than ScG and HbH strain. When SgD and HeG strain were treated with the rate of >20 infective juveniles (ijs) per alfalfa weevil larva, mortality was ca. 77.5-100% at the late instars of H. postica in 3 days. The number of established nematode was significantly different depending on nematode species, whereas number of progeny was not significantly different. The mean number of established us of SgD strain in a host was the highest at 80 ijs by 30.2. The highest progeny number of HeG strain was 2,671.5 with 80 ijs. All nematode strains were not parasitic entirely to H. postica adults. These results show that alfalfa weevil late larva is highly susceptible to Korean entomopathogenic nematodes in the laboratory.

Effects of fungicide tebuconazole on the embryonic development of Korean domestic frogs (Bufo gargarizans, Hyla japonica, and Pelophylax nigromaculatus) (살균제 Tebuconazole이 한국산 개구리류(두꺼비, 청개구리, 참개구리) 배아 발달에 미치는 영향)

  • In this experiment, we investigated the toxicity of tebuconazole (fungicide) using domestic frog embryos, along the FETAX (Frog Embryo Teratogenesis Assay-Xenopus) protocol. Bufo gargarizans, Hyla japonica, and Pelophylax nigromaculatus embryos were incubated, and investigation of the tebuconazole effect was performed by the probit analysis. As a result, depending on the concentrations of tebuconazole, the mortality and malformation rates were increased and larval body length was decreased. The teratogenic concentrations (EC50) of tebuconazole were 34.4mg L-1, 10.6mg L-1, and 14.9mg L-1, respectively, and the embryo lethal concentrations(LC50) of tebuconazole were 74.7 mg L-1, 38.5 mg L-1, and 39.1 mg L-1, respectively. The teratogenic index (TI) valuesof tebuconazole were 2.19, 3.58, and 2.65; thus, it showed teratogenicity in embryonic development of these three frogs. These results revealed that in this experiment, tebuconazole suppressed the development of embryos at a relatively low concentration. In addition, mortality, malformation ratios, malformation patterns, and growth rates were similar to the results from the other assay systems. Therefore, tebuconazole was thought to have an effect on the embryo development of domestic frogs. In future, it will be necessary to identify species specificity in order to the clarify the causes of differences in mortality, malformation rate, and malformation patterns depending on the species.
