Anaerobic Treatment of Leachate Solubilized from Thermal Hydrolysis of Sludge Cake (하수슬러지 케이크 열수분해 탈리액의 혐기성 분해 특성)

  • This study was performed to evaluate the feasibility of anaerobic pretreatment for the leachate solubilized from thermal hydrolysis of sewage sludge cake. Overall process for the treatment of sludge cake consists of thermal hydrolysis, crystallization of magnesium, ammonium, and phosphate (MAP) for the leachate and anaerobic digestion of supernatant from MAP crystallization. The experimental evidence showed that the optimum ratio of Mg : P for the struvite crystallization of leachate solubilized from thermal hydrolysis of sludge cake was 1.5 to 1.0 as weight basis at the pH of 9.5. With this operational condition, the removal efficiencies of ammonia nitrogen and phosphorous achieved 50% and 97%, respectively. The mesophilic batch test showed that the ultimate biodegradability of the supernatant from MAP crystallization reached 63% at S/I ratio of 0.5. The readily biodegradable fraction of 90% ($S_1$) of the MAP supernatant BVS (Biodegradable Volatile Solids, $S_0$) degraded with $k_1$ of $0.207day^{-1}$ for the initial 17 days where as the rest slowly biodegradable fraction ($S_2$) of 10% of BVS degraded with $k_2$ of $0.02day^{-1}$ for the rest of the operational period. Semi-Continuously Fed and Mixed Reactor (SCFMR) was chosen as one of the best candidates to treat the MAP supernatant because of its total solids content over 6%. Maximum average biogas production rates reached 0.45 v/v-d and TVS removal efficiency of 37~41% was achieved at an hydraulic retention time (HRT) of 20 days and its corresponding organic loading rate (OLR) of 1.43 g VS/L-d.

  • The biodegradation experiment, the TOD analysis and the element analysis for dispersant, Bunker-C and dispersant/Bunker-C oil mixtures were conducted for the purposes of evaluating the biodegradability of dispersnat/Bunker-C oil mixtures and studying the consumption of dissolved oxygen with relation to biodegradation in the seawater. The results of biodegradation experiment showed the mixtures with $1:10{\sim}5:10$ mix ratios of dispersant to 4mg/l of Bunker-C oil to be $0.34{\sim}2.06mg/l$ of $BOD_5$ and to be $1.05{\sim}5.47mg/l$ of $BOD_{20}$ in natural seawater. The results of TOD analysis showed 1mg of Bunker-C oil to be 3.16mg of TOD. The results of element analysis showed the contents of carbon and hydrogen to be $87.3\%\;and\;11.5\%$ for Bunker-C oil, respectively, but nitrogen element was not detected in Bunker-C oil. The biodegradability of dispersant/Bunker-C oil mixture shown as the ratio of $BOD_5$/TOD was increased from $3\%\;to\;11\%$ as a mix ratio of dispersant to 4mg/l of Bunker-C oil changed from 1:10 to 5:10, and the mixtures were found to belong in the organic matter group of low-biodegradability. The deoxygenation rates($K_1$) and ultimate oxygen demands($L_o$) obtained through the biodegration experiment and Thomas slope method were found to be $0.072{\sim}0.097/day$ and $1.113{\sim}6.746mg/l$ for the mixtures with $1:10{\sim}5:10$ mix ratios of dispersant to 4mg/l of Bunker-C oil, respectively. The ultimate oxygen demand of mixture was increased as a mix ratio of dispersant to Bunker-C oil changed from 1:10 to 10:5. This means that the more dispersants are applied to the sea for Bunker-C oil cleanup, the more decreases the dissolved oxygen level in the seawater.

The Investigation of Biodegradation Characteristics of Xylene by Soil Inhabited Microorganisms (토양 서식 미생물을 이용한 자일렌(xylene) 분해특성 조사)

  • The purpose of this study is to investigate the biodegradation characteristics of the xylene by BTEX-degrading bacteria, Pseudomonas putida BJ10, isolated from oil-contaminated soil and bio-degradation pathway of the xylene. The removal efficiencies of o, m, p-xylene in mineral salts medium (MSM) by P. putida BJ10 were 94, 90 and 98%, respectively for 24 hours. It shows clear difference compared with the control groups which were below 3%. The removal efficiencies of BTEX by P. putida BJ10 in gasoline-contaminated soil were 66% for 9 days. They were clearly distinguished from the control groups (control and sterilized soil) which were 32 and 8%. 3-methylcatechol and o-toluic acid were detected after 6 and 24 hours during the o-xylene biodegradation pathway. Therefore, we confirmed o-toluic acid as the final metabolite. And intermediate-products were somewhat different with previously published studies of the transformation pathway from o-xylene to 3-methylcatechol.

  • Trichlooethylene (TCE) has become a widespread contaminant in air, soil, and underground water due to extensive industrial used and improper disposals. Since TCE is a suspected carcinogen and constitutes public health concerns, many treatment systems have been investigated to remove this hazardous waste. One of the most premising reactor systems for the treatment of TCE is trickling biofilter (TBF), in which monooxygenase (MO), the corresponding enzyme for initiating primary substrate oxidation, fortuitously degrades TCE via cometabolism. TCE, however, is not easily treated by simple TBF. This is mainly due to the toxicities of TCE and its degradation products to microbial film in TBF. In this paper, recent progresses on the development of bioreactor system for the treatment of TCE, especially gas-phase TCE, are reviewed. The potentials of novel biofilm reactor systems were also discussed for the long-term continuous treatment of TCE.

  • As the dispersants and the dispersant/oil mixtures are degraded naturally by the microorganisms in the seawater, the consumption of dissolved oxygen may cause marine organisms to be damaged especially in the waters where the dissolved oxygen level is low due to the pollution and the restriction of seawater flow. The biodegradation experiment, the TOD analysis and the element analysis for three dispersants(SG, GL and WC) and a nonionic surfactant(OA-5) were conducted for the purposes of evaluating the biodegradability of dispersants and studying the effect of dispersants on dissolved oxygen in the seawater. The results of biodegradation experiment showed 1mg of dispersants to be equivalent to $0.403{\sim}0.595mg$ of $BOD_5$ and to $0.703{\sim}0.855mg$ of $BOD_{20}$, and 1mg of nonionic surfactant to be equivalent to 0.50mg of $BOD_5$ and to 0.97mg of $BOD_{20}$ in the natural seawater. The results of TOD analysis showed 1mg of dispersants to be $2.37{\sim}2.80mg$ of TOD and 1mg of nonionic surfactant to be 2.45mg of TOD. The results of element analysis showed carbon content and hydrogen content to be $67.6{\sim}76.5\%$ and $10.2{\sim}12.2\%$ for dispersants, and $65.3\%$ and $10.3\%$ for nonionic surfactant, respectively. No nitrogen element was detected in dispersants and a nonionic surfactant. The biodegradability of dispersants shown as the ratio of $BOD_5/TOD$ was found to be in the range of $17{\sim}21\%$, and that of nonionic surfactant was found to be about $20\%$. This means that dispersants and nonionic surfactant belong in the organic matter group of middle-biodegradabilily. The deoxygenation rates($K_1$) and ultimate oxygen demands($L_o$) obtained through the biodegration experiment and Thomas slope method were found to be $0.121{\sim}0.171/day$ and $3.155{\sim}3.810mg/l$ for 4mg/l of dispersants and to be 0.181/day and 1.911mg/l for 2mg/l of nonionic surfactant in the seawater, respectively.

  • It is essential to understand the decomposition characteristics for developing the optimum anaerobic digestion system of organic wastes. In this study, BMP (Biochemical Methane Potential) test using serum bottle was conducted to evaluate the anaerobic degradability of fish offal. 3 different groups of fish offal including waste from mackerel and hairtail handling except viscera and fish viscera were chosen for the substrates. Grinded fish offal was transferred anaerobically to serum bottle in amounts of 50 ml, 100 ml and 150 ml, respectively. BMP test was carried out in triplicate. Cumulative methane production and methane production rate depending on incubation time were evaluated. These results varied depending on substrate characteristics. The average values of ultimate methane yield ranged between $420ml{\cdot}CH_4/g{\cdot}VS$ and $490ml{\cdot}CH_4/g{\cdot}VS$, and the methane production and degradation rate of viscera were higher than those of other parts of fish offal. According to the analysis of elemental composition, average C/N ratio of fish offal used in this study was 5.2. Theoretical ultimate methane yield calculated from elemental composition was $522ml{\cdot}CH_4/g{\cdot}VS$. Biodegradability was calculated as 0.847.

  • Landfilling is most widely used as the final disposal tool of solid wastes. Solid wastes landfilled are stabilized by microbial degradation which is affected by several factors such as moisture, oxygen, pH, alkalinity, sulphate, nutrient, inhibitor, hydrogen, and temperature. Especially moisture plays a major role in microbial degradation. In this study, the effects of moisture on the degradation of municipal solids waste (MSW) were investigated. Four lysimeters with four different levels of moisture content i.e., 20, 30, 40, and 50% were operated; lysimeters were packed with MSW, and anaerobically operated. Anaerobic lysimeters with higher moisture content produced more $CO_2$ and landfill gases (LFG). It means that the moisture has a positive effect on the microbial degradation.

  • Microorganisms which can degrade organic compounds such as proteins, lipids, and cellulose in food wastewater, were isolated from food wastewater, livestock wastewater, earthworm, and etc. Among these, eleven strains which showed higher degrading activities against three organic compounds, were finally isolated, characterized, and identified. Nine strains were found to be Bacillus species, and other two were to be Enterobacter sp. and Pantoea agglomerans. The strains FWB-5 (Bacillus pumilus), FWB-6 (B. lichenisformis) and OD-4 (Pantoea agglomerans), isolated from food wastewater and livestock wastewater, respectively, showed higher three enzyme activities to organic compounds, especially to cellulose, compared to other strains. The optimal growth conditions for the great enzyme activities were at $37^{\circ}C$ with pH 7.0 for FWB-5 and OD-4 strains, whereas, these were at $25^{\circ}C$ with pH 7.0 for FWB-6 strain.

  • Mastitis is the most costly disease results in lost milk production, decreased milk quality, milk discard, early culling of cows, drug costs and labor costs in dairy cow. Until now, a antibiotic administration at the end of lactation, dry cow therapy has been known the most effective and widely used mastitis control method. However, dry cow therapy do not control a new infection in the late dry and prepartum period because dry cow products have only persistent activity in the early dry period. Therefore, this study was conducted to evaluate clinical effect of sustained released biodegradable cephalexin microsphere using PLGA in bovine mastitis control during dry period. PLGA has been approved as controlled drug release system because of non-toxic, non-tissue reactive and bioerodible characteristics. This study revealed that cephalexin microsphere had a spherical shape with characteristic porous structure on the surface. Also, in vitro drug release studies are clearly observed that the release rate of cephalexin from PLGA microsphere decrease during the first 21 days after initial burst and then increase again between 3 and 4 weeks showing pulsatile releasing pattern. On the other hand, as tried in field the new infection rate, cure rate and mean SCC after parturition in cephalexin microsphere infused group were significantly differenced as compared to the control group. Accordingly, a sustained release of cephalexin from a biodegradable microsphere could make dry cow therapy more efficiently by preventing a new infection and decreasing the number of existing infection of mammary gland during dry period.

Comparison of Azo-dye Removal Based on the Enzymatic Differences in T. versicolor and P. chrysosporium (T. versicolor와 P. chrysosporium의 효소발현 특성에 따른 Azo계 염료(Orange II) 제거 특성 비교)

  • Stepwise reductions of glucose and Orange II concentration were observed from the experiment of both white-rot fungi such as T. versicolor and P. chrysosporium. As a result, typical removal patterns in those dual substrate system were categorized through several distinctive steps: initial lag period, primary and secondary carbon consumption periods. Also, based on the total removal amounts of Orange II, COD and Color during the experimental period, similar removal extent were observed from both species experiments, within the maximal error range of 5%. However, it was refereed that the internal steps of Orange II removal on enzymatic level should be different between two species: Enzyme Lac showed good affinity for Orange II removal in T. versicolor, however in P. chrysosporium enzyme LiP represented more close affinity to the similar experimental condition. Thus, even though the superficial removal amount of calcitrant Orange II at different fungal species was merely similar, removal pathway of enzymatic levels and intermediates produced during the fungal decomposition would be different.
