  • As for analytes which did not represent the differences of the selectivity on the stationary and mobile phase, secondary chemical equilibrium theory was applied to study pH effects on the separation of alkylphenols. Mobile phase was consisted of an aqueous sodium carbonate-bicarbonate buffer and acetonitrile. The maximum selectivity for adjacent peak pairs was predicted from those values of $k^{\prime}_{HA}/k^{\prime}_{A-}$ and ${\Delta}pK$. The optimum pH determined by this method was 11.18 pH and solvent selectivity were considered at the same time to invoke the full range of selectivity effects possible for separations. Quaternary mixture composed of methanol, acetonitrile, tetrahydrofuran and water was adjusted to optimum pH 11.18. As the statistical simplex technique of an overlapping resolution map (ORM) was used to predict the optimized solvent system. The optimum solvent, which gives complete separation of alkylphenols, was determined as follws MeOH : ACN : THF = 14.4 : 81.8 : 2.8.

  • The optimum concentrations of pH, sucrose and vitamins for the growth and tropane alkaloid production of hairy root clone DTLA9 (best growth line) were investigated. The optimum pH in growth and tropane alkaliod production of DTLA9 clone in SH (Schenk and Hildebrandt, 1972) basal medium without growth regulator were pH 6.3 and 6.5, respectively. Also, the optimum sucrose concentration in growth and tropane alkaliod production in the same medium were 3.0 and 2.8%, respectively. The optimum concentrations of ascorbic acid, D-pantothenate, nicotinic acid, pyridoxine, riboflavin, and thiamine on the growth of DTLA9 clone in SH basal medium without vitamins were 0.1 mM, 0.003 mM, 0.07 mM, 0.002 mM, 0.025 mM, and 0.01 mM, respectively. In particular, supplement of 0.1 mM ascorbic acid to SH basal medium without vitamins stimulated the tropane alkaloid production.

  • This study was designed to develop functional drink from jujube extract through a simple submerged culture of three basidiomycetes species. The optimum Brix and pH of the jujube extract for culturing the Ganoderma lucidum appeared to be 5 Brix and pH 4. Ten days of culture period produced maximum mycelium. The optimum Brix and pH of the jujube extract for culturing the Coriolus versicolor appeared to be 5 Brix and pH 5. Ten days of culture period produced maximum mycelium. The optimum Brix and pH of the jujube extract for culturing the Phellinus igniarius appeared to be 3 Brix and pH 5. For the maximum mycelial production eighteen days of culture period was required for Phellinus igniarius. The antitumor activity of the polysaccharides extracted from the fermented drinks was demonstrated through the tumor cell line experiments. The $IC_{50}$ values of the jujube drinks fermented with Ganoderma lucidum and Phellinus igniarius against stomach cancer cell line appeared to be one fourth that of the jujube drink which was not fermented with basidiomycetes.

  • Yeonsan Ogae has been known as supporting health and high efficacy of treatment. In recent days, as the efficacy of functional peptides has known, the optimization of oligo peptides production and its characteristics from Ogae legs has been performed. Response surface method was used to perform the optimizaion of enzyme hydrolysis. The range of processes was temperature ( 40, 50 and $60^{\circ}C$), pH( pH 6.0, 7.0 and 8.0 ), and enzyme( 1, 2 and 3% ). The degree of hydrolysis, amino acids, molecular weight of products were analyzed. The optimum process of enzyme hydrolysis were determined as temperature $58^{\circ}C$, pH 7.5, and enzyme concentration 3%. At optimum conditions, the degree of hydrolysis after 2 h reaction was 75-80%. The amino acid and were 168.131 mg/100 g, respectively. The molecular weight of products by using MALDI-TOF was ranged from 300 to 1,000 Da.

Optimization of Semi-Batch Process for Ethanol Production (에타놀 생산을 위한 Semi-batch 발효 공정의 최적화)

  • As flocculent strains are likely to have considerable potential for internal cell recycle, kinetic studies on glucose medium with flocculent Saccharomyces uvarum were carried out in batch and continuous culture. Using a mathematical model, the kinetic parameters at each temperature and pH were estimated in order to establish optimal conditions. It was found that an overall optimum temperature for growth and ethanol production in the range 33-35$^{\circ}C$ was desirable. With regard to the effect of pH, ethanol production by S. uvarum was found to be relatively insensitive to pH value between 4 and 6, with an optimum pH of around 5. At these optimal conditions a maximum ethanol productivity of 12 g/$\ell$/h was determined using semi-batch process together with 5. uvarum.

  • An actinomycetes strain No.4 which produce the cholesterol oxidase(EC 1.1.3.6), was isolated from soil and identified as Streptomyces sp. based on taxonomic studies. The conditions of cholesterol oxidase production and enzymatic properties were investigated. The optimum composition of medium for production of the enzyme was 1% soluble starch, 2% corn steep liquor, 0.1% $KH_2PO_4$, 0.1% $NaNO_3$ and 0.05% $MgSO_4$ (pH 7.0). The optimum pH and temperature of the cholesterol oxidase were pH 6.0~7.5 and $37^{\circ}C$, respectively. The enzyme was stable in the range of pH 6.0~9.0. The isoelectric point determined by multichambered electrofocusing unit was in the range of pH 6.0~6.5.

Characterization of Acidic Nucleotidase from Aspergillus niger (Aspergillus niger의 Acidic Nucleotidase의 특성)

  • Acidic nucleotidase from Asfiergilius nlger has been partially purified by Sepharose CL-6B gel filtration and DEAE-Sephacel ion exchange chromatography. The optimum pH and temperature for the enzyme reaction with 5'-AMP or 3'-AMP as a substrate were 4.5 and 55%, respectively. However, the optimum temperature became 70% when p-nitrophenyl phosphate was used as a substrate. The enzyme was stable at acidic pH. The enzyme activity was not affected by addition of various nucleotides, nucleosides and inorganic phosphates. Ferric, aluminium, vanadate and molybdate ions inhibited the enzyme activity dramatically. In kinetic studies, $K_m$), values for 3'-AMP, 5'-AMP and p-nitrophenyl phosphate were 1.39 mM, 1.5 mM and 5.77 mM, respectively. The substrate efficiency ($V_{max}/K_m$) shows 3'-AMP is the prefered substrate for the enzyme among tested substrates.

Culture Conditions of Geobacillus kaustophilus DSM 7263 for Production of Thermophilic Extracellular Lipase (내열성 extracellular lipase 생산을 위한 Geobacillus kaustophilus DSM 7263의 배양조건)

  • A producer of thermophilic extracellular lipase, Geobacillus kaustophilus DSM 7263, was selected from various microorganisms of the Geobacillus genus. We investigated optimum conditions for mass production of G. kaustophilus lipase. Among the different natural oil media, olive oil was optimal for enzyme production. The maximum amount of enzyme production was obtained when G. kaustophilus was grown in a medium containing 0.5% olive oil as a carbon source. The pH and temperature for optimal growth were pH 8.0 and $55^{\circ}C$, respectively, while the optimum pH and temperature for lipase production were pH 6.0 and $50^{\circ}C$, respectively. In the presence of $Mg^{2+}$ and $Mn^{2+}$, lipase production was dramatically enhanced by 247% and 157%, respectively, whereas enzyme production was inhibited by $Zn^{2+}$, $Cu^{2+}$, and $Cd^{2+}$. The addition of 0.1% (v/v) triton X-100 increased lipase production and cell growth when compared to the negative control.

  • In this study, the hydrolysis conditions for the production of anti-inflammatory peptides from meat and roe hydrolysates of sandfish (Arctoscopus japonicus) were determined by measuring the nitric oxide (NO) scavenging enzymatic activity, experimental pH, temperature, enzyme concentration, and hydrolysis time. The optimal conditions determined when using meat hydrolysate were a pH value of 5.0, at a temperature of $30^{\circ}C$, 1% enzyme concentration, and 4 h hydrolysis time. The optimal conditions when using roe hydrolysate were a pH of 5.0, a temperature of $70^{\circ}C$, enzyme concentration of 3%, and hydrolysis time of 3 h. The NO scavenging activities of meat and roe hydrolysate were determined to be 18.94 and 19.81%, respectively. In summary, this study determined the optimum enzymatic hydrolysis conditions for the production of anti-inflammatory peptides from sandfish.
