Anti-inflammatory effect and useful contents of saccharification extract powder using hot water extract from log cultivation Lentinula edodes by different UV irradiation (UV 조사시간에 따른 원목표고당화물의 유용성분 및 항염증 효과)

  • The grade and price of Lentinula edodes largely differs in preference depending on the product area and seasonal factors. The product amount of autumn L. edodes was higher than that of spring L. edodes, but high quality, which is divided into "Hwago" is low in preference. Mostly, the autumn L. edodes is obtained as powder; hence, it is necessary to develop a processing method to utilize its flavor and aroma at an affordable price. Additionally, we estimated the content of 𝛽-glucan, ergosterol, vitamin D2, reducing sugars, and free amino acids and evaluated the anti-inflammatory activity of saccharification powder of log-cultivated L. edodes. In the saccharification powders obtained via 7 min of UV irradiation of log-cultivated L. edodes, 𝛽-glucan and vitamin D2 contents were found to be the highest, whereas ergosterol content was found to be the lowest. The content of reducing sugars ranged from 62.4 mg/L to 68.2 mg/L. The free amino acids were higher in these saccharification powders than in the control. Subsequently, RAW 264.7 cells were treated with different concentrations (10, 50, 100, 200, 300, and 500 ㎍/mL) of the saccharification powders of log-cultivated L. edodes obtained via different UV irradiation time applications. The cells showed good viability; the anti-inflammatory effect was found to be the highest at 7 min UV irradiation. Therefore, 7 min of UV irradiation was determined to be the optimum condition for manufacturing saccharification powders of log-cultivated L. edodes. Hence, saccharification powders of log-cultivated L. edodes may be used as a raw material for natural sweeteners, food additives, and in the food industry.

  • 본 논문은 GIS를 이용하여 원자력 발전소 건설을 위한 최적의 부지 선택 방안을 연구한 것이다. 근래 고도 경제 성장에 따라 전력 사용량이 날로 증가되고 있으며 이에 따라 발전소의 추가 건설이 시급한 실정이다. 그러나 발전소 건설에 필요한 적지가 희소한 실정이며 정부의 국토이용 계획, 각종 개발 제한 사항에 의해 갈수록 제약을 받게 되어 부지 선정에 많은 어려움이 예상된다. 본 논문에서는 기존의 원자력 발전소 입지선정 절차를 근간으로하여 부지평가에 영향을 주리라 예상되는 지질, 골재원, 수송조건, 부지표고, 용수원, 해상조건, 접안조건 및 주변환경, 인구 분포 등의 부지평가 요소들을 중심으로 하고, GIS를 이용한 부지평가 항목의 분석을 통하여 원자력 발전소 부지의 최적 후보지를 결정하는 개념적 방안을 제시하였다.

Cultural characteristics of unrecorded species Lentinula spp. in Korea (국내 미기록 표고속 종들(Lentinula spp.)의 배양적 특성)

  • The culture condition of unrecorded species L. aciculospora, L. boryana and L. raphanica was investigated with recorded species of L. edodes as the control group in order to analyze diversity and examed relations of the species belong to Lentinula. The optimal temperature and media for the mycelial growth of L. aciculospora and L. boryana were $22^{\circ}C$ and PDA, MCM medium. L. raphanica was $28^{\circ}C$ and ME1 medium. Each of L. aciculospora, L. boryana, L. raphanica is pH 6, pH 7, pH 5 in the optimal pH respectvely. The optimal carbon and nitrogen source of L. aciculospora were glucose and malt extract. That of L. boryana was glucose and urea that of L. raphanica was sucrose and potassium nitrate. The optimal vitamin of L. aciculospora was Myo-inositol. That of L. boryana and L. raphanica were Riboflabin.

Quality Characteristics of Gangjeong Containing Various Levels of Lentinus edodes (표고버섯을 첨가하여 제조한 강정의 품질특성)

  • Gangjeong was prepared by adding Lentinus edodes to improve quality of the gangjeong and to give some functional properties. Contents of the crude protein and ash increased with increasing amount of L. edodes, while carbohydrate contents decreased. As the ratio of mushroom in gangjeong increased, lightness and yellowness in Hunter's color value tended to decrease. In terms of texture, 20% and 30% contents of L. edodes made no significant differences in gumminess, brittleness, cohesiveness, and springiness compared to control, whereas hardness showed no difference to control. Gangjeong with 30% L. edodes obtained the highest scores in quality and sensory characteristics.

  • This study was performed to determine optimal extraction conditions of fermented Polygonum multiflorum root by Lentinula edodes (JMI10079) Pegler mycelials using response surface methodology. The independent factors were extraction temperature (X1: $40-100^{\circ}C$), extraction time (X2: 2-10 hrs.), and the ratio of water to sample (X3: 33-100 mg/mL). Their effects were assessed on dependent variables of the extract properties, which included soluble solid contents (Y1), $^{\circ}Brix$ of sample extract (Y2), total polyphenol content (Y3), total flavonoid content (Y4), 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulphonic acid) radical cation scavenging activity (Y5) and 2,2-diphenyl-1-picrylhydrazyl radical scavenging activity (Y6). The experimental data obtained were fit to a second-order polynomial equation. The optimal extraction conditions for fermented P. multiflorum root were: X1: $91.22^{\circ}C$, X2: 7.72 hrs, and X3: 39.71 mg/mL.

  • The Development of CCD has contributed to great advancement in mapping technology with giving benefits to research community of photogrammetry. The purpose of this paper is to find the best selection of interpolation method for creating a terrain model form close range digital photogrammetry. T-test as a kind of statistical analysis was conducted to analyze the similarity of hydraulic model with close range digital photogrammetry and trigonometric leveling. Also, many interpolation methods such as inverse distance, kriging, nearest neighbor and TIN about the hydraulic model interpolation were conducted to compare the results for computer to display actual terrain an optimum interpolation of the digital elevation model form close range digital photogrammetry. The results revealed that kriging and TIN interpolation were efficient methods to judge the hazard interpolation law by analyzing geometric similarity of hydraulic model against hydraulic model application.

  • Effects Of organic compound, photosensitizer and $K^+$ ion influx. On the light-induced ATPase of mitochondria in L. edodes purified by linear sucrose density gradient centrifugation were studied. The mitochondrial ATPase activity was investigated by various wavelength illumination at dark state. The mitochondrial ATPase was activated 139% and 128% by 10m mol dithiothreitol and 0.1m mol quinacrine, respectively. This enzyme also was activated 36% by 0.1m mol phenazine methosulfate as photosensitizer. But, 100 mg oligomycin and 1m mol phlorizin inhibited activity of enzyme to 48% and 45%, respectively. Its optimum wavelength was 690 nm on the effect of $K^+$ ion influx, its optimum pH and temperature were found to be 7.2 and $55^{\circ}C$.

Effects of Various Thermal Treatments on Physicochemical and Nutritional Properties of Shiitake Mushrooms (열처리 방법에 따른 표고버섯의 이화학적 특성 및 영양학적 분석)

  • In the food industry, thermal treatment is an important process for extending shelf-life of foods. However, heating process affects the physicochemical, nutritional, and microbial properties of foodstuff such as color, texture, pH, and proximate compositions. This study was conducted to select an optimal thermal treatment by observing physicochemical, nutritional, and microbial effects of shiitake mushrooms with different thermal treatment methods. Shiitake mushrooms were washed and sliced equally ($5cm{\times}0.5cm{\times}0.5cm$) and then heat-treated by three methods. Samples were heated in $100^{\circ}C$ boiling water, steamed for 10 min, or pan fried at $130^{\circ}C$ for 4 min. Total color difference values showed significantly increasing tendency with treatment time. For pH values, boiling water-treated mushrooms showed increasing tendency according to increased thermal treatment. For the results of hardness, boiling water or pan frying-treated mushrooms showed reduced tendency within 1 min. In the case of steam-treated mushrooms, hardness values were maintained for 1 min. Organic acid contents of steam-treated sample showed the lowest value among treatments. For microbial counts, steam-treated samples for 3 min showed the lowest value. Consequently, the results of this study suggest that steam treatment could be the optimal thermal treatment to minimize quality loss of shiitake mushrooms.

Optimization of PRISM Parameters and Digital Elevation Model Resolution for Estimating the Spatial Distribution of Precipitation in South Korea (남한 강수량 분포 추정을 위한 PRISM 매개변수 및 수치표고모형 최적화)

  • The demand for a climatological dataset with a regular spaced grid is increasing in diverse fields such as ecological and hydrological modeling as well as regional climate impact studies. PRISM(Precipitation-Elevation Regressions on Independent Slopes Model) is a useful method to estimate high-altitude precipitation. However, it is not well discussed over the optimization of PRISM parameters and DEM(Digital Elevation Model) resolution in South Korea. This study developed the PRISM and then optimized parameters of the model and DEM resolution for producing a gridded annual average precipitation data of South Korea with 1km spatial resolution during the period 2000-2005. SCE-UA (Shuffled Complex Evolution-University of Arizona) method employed for the optimization. In addition, sensitivity analysis investigates the change in the model output with respect to the parameter and the DEM spatial resolution variations. The study result shows that maximum radius within which station search will be conducted is 67km. Minimum radius within which all stations are included is 31km. Minimum number of stations required for cell precipitation and elevation regression calculation is four. Optimizing DEM resolution is $1{\times}1km$. This study also shows that the PRISM output very sensitive to DEM spatial resolution variations. This study contributes to improving the accuracy of PRISM technique as it applies to South Korea.

Extracellular Wood-degradative Enzymes from Lentinus edodes JA01 (표고 균주(菌株) JA01에서 분비되는 세포외(細胞外) 목재성분(木材成分) 분해효소(分解酵素)에 관하여)

  • The aim of this study was to investigate physiological chracteristics of Lentinus edodes in Korea. We tried to detect properties of the several wood-degradative enzymes and investigate patterns of the enzyme production. A specific carbon source was used in the enzyme induction media for each enzymes, and the crude extract was used for the enzyme solution. With these enzyme solution, we investigated optimum temperature and pH conditions of their reactions. Moreover we investigated transition patterns of the enzyme production of the several wood-degrad­ative enzmes from Complex and Saw dust media for the purpose of studying the mechanisms of the wood component degradation by this fungus. It was assumed that the order of the wood com­ponent degradation was cellulose, xylan, and then pectic substances, and that the synergistic effects of these substances also influenced the degradation of wood components.
