The Antioxidative Effects of Rhododendron brachycarpum Extracts (만병초(Rhododendron brachycarpum) 추출물의 항산화 효과)

  • The objective of this study was to investigate the antioxidative capacity of Rhododendron brachycarpum 95% ethanol extracts. Total antioxidant status was examined by total antioxidant capacity against ABTS radical reactions. Total antioxidant capacities of R. brachycarpum extract at the concentrations of 0.2 and 1 mg/mL were 0.33 and 2.26 mM Trolox equivalents, respectively. Superoxide scavenging activities of R. brachycarpum extract at the concentrations of 0.2 and 1 mg/mL were 45.0 and 77.0%, respectively. Oxygen radical absorbance capacities of R. brachycarpum extract at the concentrations of 5 and 100 ${\mu}g/mL$ were 40.88 and 131.00 ${\mu}M$ Trolox equivalents, respectively. Total phenolic contents of R. brachycarpum extract at the concentrations of 0.2 and 1 mg/mL were 0.37 and 1.25 mM gallic acid equivalents, respectively. R. brachycarpum extract at the concentration of 0.1 mg/mL inhibited 0.2 mM and 0.5 mM tert-butyl hydroperoxide induced cyototoxicity by 52.1 and 30.3%, respectively, in HepG2 cell culture system. Thus, strong antioxidant and cytotoxicity-inhibiting effects of R. brachycarpum extract seem to be due to, at least in part, the prevention from free radicals-induced oxidation as well as high levels in total phenolic contents.

Effects of Opuntia ficus-indica var. saboten Ripe Fruits on Protection of Neuronal PC-12 Cells and Cholinesterase Inhibition (백년초의 PC-12 신경세포 보호 및 콜린가수분해효소(cholinesterase) 저해 효과)

  • Oxidative stress caused by reactive oxygen species is ascribed to many neurodegenerative diseases like Alzheimer's disease. Phenolic antioxidants can reduce the oxidative stress. In this study, ripe fruits of Opuntia ficus-indica var. saboten (OFS) were extracted using 80% (v/v) aqueous ethanol. Total phenolic and flavonoid contents of the OFS fruits (100 g) were 409.9 mg gallic acid equivalents and 72.2 mg catechin equivalents, respectively. The OFS fruits had antioxidant capacity at 381.2, 298.2, and 3,219.9 mg vitamin C equivalents/100 g in ABTS, DPPH, and ORAC assays, respectively. The OFS fruits showed protective effects on PC-12 cells against oxidative stress in a dose-dependent manner, partly due to decrease of intracellular oxidative stress. Furthermore, the OFS fruits inhibited both acetylcholinesterase and butyrylcholinesterase. Consequently, these results suggest that the OFS fruits might be served as a source of functional materials to reduce oxidative stress in neuronal cells and to inhibit cholinesterases.

Comparative Study of Biological Activities at Different Cultivation Area and Harvesting Times for New Variety 'Sammany' of Gom-chewi (곰취 신품종 '쌈마니'의 재배지역 및 수확시기별 생리활성 비교)

  • 쌈용과 장아찌로 많이 이용하며 다양한 효능을 가진 곰취(Ligularia fischeri)의 신품종 중에 항산화 활성이 높은 것으로 밝혀진 '쌈마니' 품종에 대하여 좀 더 구제적인 항산화 활성을 알아보기 위하여 재배지역 및 수확시기별 항산화 성분 및 생리활성 정도를 비교 분석하여 지역별 적정 수확시기를 구명하고자 본 연구를 수행하였다. 시료는 2018년 4월 18일부터 2018년 7월 9일 까지 20일 간격으로 4회에 걸쳐 고랭지인 고령지농업연구소 대관령시험포장과 평난지인 강릉시험포장에서 수확하였으며 모두 동결 건조하여 분말형태로 갈아 실험에 사용하였다. 분석결과 '쌈마니' 품종의 지역 및 수확시기별 수율은 유사하였으며 총페놀 함량은 평난지에서는 5월 9일이 264.7mg GAE/g로 가장 높았고 그다음이 5월 29일, 6월 19일, 4월 18일 순이었다. 대체적으로 5월에 높았으며 고랭지에서도 초기 생산보다는 중기 생산기인 6월 18일에 총페놀 함량이 274.4mg GAE/g로 가장 높았고 그다음이 7월 9일, 5월 28일, 5월 10일 순이었다. 총플라보노이드 함량도 같은 경향으로 평난지에서는 5월 9일에 76.0mg QE/g로 가장 높았으며 그다음이 5월 29일, 6월 19일, 4월 18일 순이었으며 고랭지에서는 6월 18일에 66.5mg QE/g으로 가장 높았고 그다음이 7월 9일, 5월 28일, 5월 10일 순이었다. 재배지역 및 수확시기별 DPPH 활성산소 제거능에서도 평난지에서는 5월 9일과 5월 29일에 효과적이었으며 고랭지에서는 6월 18일에 효과가 있는 것으로 나타나 너무 이른 시기나 늦어지면 활성이 떨어지는 것으로 생각된다. 또한 환원력에 있어서도 평난지에서는 $1,000{\mu}g/m{\ell}$의 농도에서 5월 9일, 5월 29일, 6월 19일에 높은 경향을 나타냈으며 고랭지에서는 6월 18일에 높게 나타났다. 이러한 결과로 볼 때, 쌈마니 품종은 평난지에서는 5월, 고랭지에서는 6월에 수확하는 것이 항산화 활성이 높은 것으로 사료되었다.

Antioxidant Activity of Ulmus davidiana var. japonica N. and Hemipteleae davidii P. (느릅나무와 시무나무의 항산화 활성)

  • Cortex of Ulmus davidiana var. japonica N. and Hemipteleae davidii P. have been used without classification according to countries as oriental medicine for treatment of various disorders. This study was conducted to elucidate the difference in antioxidant activity and total phenol content of these medicinal materials. Root cortex and trunk cortex of two medicinal plants were extracted with 80% ethanol $(at\;85^{\circ}C)$ and water (at room temperature), respectively. All of the extracts at $200{\sim}5\;{\mu}g/ml$, showed more effective scavenging activity on superoxide radical than ascorbic acid, and 80% ethanol and water extracts of Ulmus root cortex (URC) was more effective. Scavenging activities on DPPH radical of 80% ethanol extracts from URC and Ulmus trunk cortex (UTC) at 10 ${\mu}g/ml$, (41.4, 35.6%) were higher than those of the other six extracts and ${\alpha}-tocopherol\;(2.3{\sim}24.0%)$. Inhibitory activities on human low density lipoprotein (LDL) oxidation of 80% ethanol extracts from UTC and URC (84.3, 81.7%) at $10\;{\mu}g/ml\;(31.3{\sim}78.2%)$ were higher than those of the other six extracts and ${\alpha}-tocopherol$. However, antioxidant activities on linoleic acid peroxidation of all of the extracts were relatively lower than that ${\alpha}-tocopherol$. Total phenol content of 80% ethanol extract from URC was the highest value as 50.8% and that of water extract from HRC was the lowest as 5.6% among the extracts. From these results, it is suggested that Ulmus root cortex was the most effective in antioxidant activity.

Physiological Activities According to Cultivars and Parts of Ulsan Pear (울산지역 생산 배의 품종 및 부위별 생리활성)

  • This study was performed to confirm physiological activities according to cultivars and parts of Ulsan pear (wonhwang, pungsu, whangkeum, whasan and shingo). Total contents of phenolic compounds from peel, pulpy substance and core were 699.3-800.6, 51.5-112.5 and 254.0-401.5 mg/100 g as tannic acid equivalent, respectively. There were difference contents by cultivars, and peel and core of shingo and pulpy substance of wonhwang showed high contents, respectively. Total flavonoid contents of peel, pulpy substance and core were 125.2-164.2, 25.9-35.9 and 45.1-60.0 mg/100g, respectively and those of shingo cultivar showed comparatively high. Electron donating ability was in the order of peel (66.1-90.7%), core (48.5-82.8%) and pulpy substance (24.9-58.2%), and whasan cultivar showed comparatively low. Nitrite scavenging activity was in the order of peel (58.2-100.8%), core (59.5-86.2%), pulpy substance (39.9-82.5%). There were little difference by cultivars of core but peel and pulpy substance of shingo cultivar showed comparatively low nitrite scavenging activity. And as the concentration of each extract increased, nitrite scavenging activity increased. Xanthine oxidase inhibition rate was in the order of peel (14.1-75.4%), core (5.3-71.8%), pulpy substance (2.2-67.5%). And there little difference by cultivars and which was increased as the concentration of each extract increased.

Antioxidant activity of Bamboo powder and its immunoreactivity in the pig (대나무 분말의 항산화력과 돼지의 면역 활성에 미치는 영향)

  • The present study was designed to explore the antioxidant effect of Bamboo powder and its immunoreactivity in pigs. We investigated the functional properties of Bamboo extracts by means of measuring the contents of total polyphenols and flavonoid as well as determining ABST, DPPH radical scavenging activity, and hydroxyl radical scavenging activity and anticancer activity. The total phenolic compound and flavonoids contents of Bamboo extracts were 171.25 mg/g and 127.5 mg/g, respectively. The DPPH radical, hydroxyl radical, ABST radical scavenging activity of Bamboo extracts were 17.3%, 12.5% and 21.5%, respectively. Evidenced by MTT and cell cycle assay, Bamboo dose-dependently inhibited the cell proliferation and induced G0/G1-phase arrest in CHO cells at concentrations of 100, 250, and 500 ${\mu}g/ml$ Bamboo extracts. More than 80% of apoptotic cells were observed by staining with annexin V in 500 ${\mu}g/ml$ Bamboo-treated CHO cells, indicating that Bamboo had potent anticancer activities. Next, to investigate the effect of Bamboo on cytokine, immunoglobulin concentration, and blood compositions, flatting pigs were fed with Bamboo powder for 38 days. Flatting pigs were divided into 4 groups; basal diet (control), basal diet supplemented with 1% Bamboo powder (T1), 2% Bamboo powder (T2), and 3% Bamboo powder (T3). The level of hemoglobin increased in the all Bamboo-fed groups compared with the normal control group. In particular, platelet levels in the all Bamboo-treated groups increased by approximately 90% compared with the levels from pig on a normal control. Serum levels of immunoglobulins (IgG, IgA) in the pigs fed Bamboo powder were modestly increased, and the interferon-${\gamma}$ level also was strongly increased in 2% or 3% Bamboo-fed groups compared with the levels in control groups. Together, these results demonstrated that Bamboo extracts had an effective capacity of scavenging for ABTS, DPPH, and hydroxyl radicals and showed correlation with potent phenol and flavonoid contents, thus suggesting its antioxidant potential. Moreover, administration of Bamboo in 2~3% improved blood parameters and platelets, and especially immunity-related ones such as IgG, IgA, and interferon-${\gamma}$, leading to be potential feed additives in flatting pigs.

Growth Inhibition and Induction of Apoptosis in Human Bladder Cancer Cells Induced by Fermented Citrus Kombucha (감귤 콤부차 발효액의 인체 방광암세포에 대한 성장억제와 Apoptosis에 미치는 영향)

  • Kombucha is a slightly sour beverage fermented by symbiotic micro-organisms, including bacteria and yeasts. In this study, we examined the biological activities of citrus Kombucha (CK) produced by addition of citrus extract to original Kombucha (K). After fermentation for 10 days, radical scavenging activity examined by ABTS and DPPH assays increased by approximately 20% compared to that of K. Moreover, content of total phenolic compounds significantly increased by 60% compared to that of K. Cell proliferation assays utilizing MTT showed that CK treatment significantly inhibited growth of bladder cancer cells, T-24 and 5637, in a dose-dependent manner with $IC_{50}$ values of 4 and 7 mg/mL, respectively. Annexin V staining showed that CK treatment led to apoptosis of cells in a dose-dependent manner. T-24 cells were more sensitive to CK treatment than 5637 cells, as 8 mg/mL of CK resulted in 97% apoptosis of T-24 cells. Western blotting showed that CK treatment led to up-regulation of apoptotic proteins, including caspases-3, -8, -9, and PARP, in bladder cells not in K-treated cells. Taken together, these results demonstrate that CK may be developed as a functional beverage.

Antioxidant and Quinone Reductase Inductive Activities of Various Organs of Pepper (고추 기관별 항산화 활성과 quinone reductase 유도활성)

  • We analyzed antioxidant and quinone reductase (QR) inductive activities of various organs of pepper for utilizing by-product of them. Peppers were separated into fruits, roots, stems, and leaves and extracted with methanol for the analysis. As a result, pepper leaves showed higher phenol content than other organs. Using the DPPH assay, there was not considerably different activity depending on pepper organs, but pepper leaves showed significantly higher antioxidant activity using the ABTS assay. In FTC and TBA assay, stems and leaves showed significantly higher lipidperoxidation inhibitory activity. In QR inductive assay, pepper tissues showed different QR inductive activity: leaves>roots>>stems>fruits. In addition, pepper leaves showed highest antiproliferation activity on hepa1c1c7 among pepper tissues in $50-200\;{\mu}g/mL$. These results indicate that pepper leaves have high potential to be a good functional food material due to high QR inductive and antioxidant activities.

Antioxidant Effects of Phenolic Compounds Isolated from Deffated Perilla Seed Flour (탈지들깨박에서 분리한 페놀화합물의 항산화효과)

  • The free, ester and insoluble bound phenolic acids in the extracts from defatted perilla seed flour were isolated and their antioxidative activities were evaluated in comparison with commercial synthetic antioxidants. Total phenolic content of the perilla seed was 0.75% as chlorogenic acid. Each percent ratio of the content of free, ester, and insoluble bound phenolic acid to total phenolic content was 87.5, 7.5 and 5.0% respectively. Chlorogenic acid was identified as a major phenolic acid and a small amount of caffeic acid was also identified in the free phenolic acid extract, but they were not found in soluble ester and insoluble bound phenolic extracts by two dimensional paper chromatography. Each type phenolic extract from 30g of deffated perilla flour showed antioxidant activity similar to that of BHT (0.02%, w/w) in 200g of soybean oil substrate inspite of the difference of each phenolic content.

Increasement of antioxidative activity in Codonopsis lanceolata adventitious root treated by Methyl jasmonate and salicylic acid (Methyl Jasmonate 및 Salicylic Acid 처리에 의한 더덕(Codonopsis lanceolata) 부정근의 항산화 활성 증가)

  • Traditionally, Codonopsis lanceolata root have been used as a source of natural heath food. This study was initiated to investigate the impacts of methyl jasmonate (MeJA) and salicylic acid (SA) on adventitious growth C. lanceolata, the production of secondary metabolites, such as flavonoids, total phenolic compound, antioxidative activity (DPPH). The highest phenolics content was observed in treatment of 20 uM MeJA (74.53 mg/g). The content of total flavonoids followed the similar pattern as that of total phenolics, showing 38.45 mg/g of C. lanceolata treated by 20 uM MeJA. The DPPH scavenging activity was 24.2 ($IC_{50}$) of C. lanceolata treated by 20 uM MeJA. These results provide useful information for enhancing biological properties of cultural roots of C. lanceolata.
