Postmortem Changes of Sterile Fish Muscle Inoculated with a Proteolytic Pseudomonas sp. (무균 어육에 단백질 분해세군 접종후의 저장중의 변화)

  • 사후 저장기간 중 어육내에 진행되는 단백질 분해에 어육 자체효소와 새균활동 중 어느 쪽이 기여하는 바 큰가를 분별하기 위하여 금방 죽인 볼낙류(Sebastodes)의 어육을 무균방법으로 잘라내어 4 OZ 유리용기에 넣고 플라스틱으로 된 나사뚜껑으로 밀봉한 후 감마선 조사하지 않는 군(무균육과 무군육에 단백질 분해성 Pseudomonas 를 접종한 것으로 세분)과 0.5과 2.0 Mrad 선량 조사한 군(무균육, 조사 전에 접종시킨 어육, 그리고 조사후에 접종시킨 어육으로 세분)으로 나누며 $0^{\circ}C$에서의 저장 중의 어육의 총균수, pH, 관능 상해, 그리고 0.5M KCl 및 $70\%$ ethanol 가용성 총질소와 Amino-N을 검측함으로써 어육 단백질 분해 과정을 조사하였다. 0.5 Mrad 감마선량은 무균 어육에 접종된 $10^6$ per gm Muscle Pseudomonas sp.를 완전 사멸시키는데 충분하였으므로 완전한 무균상태와 처음부터 단백질 분해성 세균활동이 활발한 상태에 놓여 있는 어육 내에 진행되는 단백질 분해과정을 비교할 수 있었다. 기름지지 않는 어류에 속하는 볼박 어육의 저장 중에 일어나는 pH의 상승, 관능상태의 저락, 그리고 단백질 분해에서 오는 총질소와 Amino-N의 축적은 모두 세균의 번식활동과 직접 관련되어 있으며, 완전 무균 어육의 단백질 분해는 전혀 검측되지 안했으므로 통상 저장중에 일어나는 어육 담백질 분해는 전부가 세균활동에 기인됨이 확실해 졌다. 감마선 조사 직후 단백질 분해성 세균을 접종시킨 어육을 조사치 않고 접종시킨 어육에 비교할 때, 전자의 경우 세균번식이 현저히 장해 받았으며, 이에 따라 PH 상승과 총질소 및 Amino-N 축적도 상대적으로 부진하였다. 이 방사선을 조사받은 어육에 접종된 단백질 분해성 세균활동의 장해는 조사선량에 비례함이 밝혀졌다. 관능상태로 판단할 때 0.5 Mrad선을 조사받은 어육은 조사 받지 않은 것보다 선도유지기간이 2배 이상으로 연장되었다. 그러나 2.0 Mrad 선량 조사받은 어육의 선도유지는 오히려 조사 받지 않은 것보다 못하였다. 이 고선량 조사로 인한 식품의 물리적 및 화학적 변화의 해독성은 접종시킨 세균 번식 장해현상으로 나타났으며, 이 현상은 조사식품의 안전성 검측을 위한 하나의 도구로 사용될 가능성을 말해준다.

Effects of Electron Beam Radiation on the Microflora and Sensory Characteristics of Pork Loin (전자선 조사에 의한 국내산 신선 돈육의 미생물학적 및 관능적 특성 변화)

  • The effects of electron-beam irradiation on the microbial and sensory qualities of pork loins were studied. The pork loins were aerobically packed in polyethylene films and exposed to absorbed doses of electron-beam at 0, 0.5, 1 and 2 kGy in ambient temperature. All samples were stored at $4{\pm}1^{\circ}C$. As an irradiation dose increased, decontaminating effects increased in terms of total microbial count, lactic acid bacteria and psychrotrophs during storage. All difference degrees in the triangle test were very low levels. In the descriptive analysis, Low dose irradiation did not affect aroma, off-flavor and acceptability in fresh and cooked meats.

  • Quality changes of filter-sterilized Yakju were examined by filtration of Yakju through membranes followed by storing at $25^{\circ}C$ for 50 days. To evaluate quality changes of filter-sterilized Yakju, pH, titratable acidity, turbidity, and viable cell numbers of total bacteria, lactic acid bacteria, and yeast were measured. Titratable acidity, turbidity and viable cell numbers of non-sterilized Yakju increased, but pH profile decreased during the storage. In filter-sterilized Yakju, titratable acidity and turbidity did not change, while viable cells of total bacteria, lactic acid bacteria, and yeast were not detected during the storage. Addition of chitosan at the concentration of 0.1% (w/v) decreased the viable cell numbers significantly showing similar pH and titratable acidity profiles with non-sterilized Yakju.

The Pilot-scale Treatability Studies of Co-Composting for the Remediation of Diesel Contaminated Soil during the Winter (동절기 유류 오염토양 복원을 위한 Co-composting기술의 현장 적용성 연구)

  • This study was conducted to check the applicability of pilot-scale co-composting for the remediation of diesel contaminated soil during the winter. Nutrients and microbes were added to enhance the efficiency of bioremediation and fermenting composts were also added to stimulate the microbial activities. As a result. the soil pile was kept at adequate temperature for the bioremediation during the test period of 30∼40 days and initial concentration(2,340mg TPH/kg dry soil) was reduced to 216mg TPH/kg dry soil (approximately 91% removal). During the initial 10∼30 days, it was found that the TPH concentration and the microbial population were rapidly reduced and increased. respectively. The co-composting technology studied can be effectively applied to remediate the diesel contaminated soil during the winter.

Use of Freshness Indicator for Determination of Freshness and Quality Change of Tofu During Storage (신선도표시계를 이용한 두부의 저장 중 신선도 측정 및 품질 변화)

  • To determine freshness and quality change of tofu during storage, we manufactured a freshness indicator and monitored the surface pH, volatile basic nitrogen (VBN), thiobarbituric acid reacted substance (TBARS), total bacterial counts, and sensory evaluation. Tofu had a change in color of the freshness indicator after storage of 8 days at $4^{\circ}C$, and its pH and total bacterial counts reached 5.6 and 7.63 log CFU/g, respectively. VBN and TBARS values increased and reached the decay point at the time of color change of the freshness indicator. Sensory evaluation also indicated that samples were unacceptable by off-odor and decrease of firmness at day 8 of storage. These results suggest that a freshness indicator should be useful in determining expiration date of tofu products during marketing by indicating the microbial safety as well as sensory change.

Retardation of Kimchi Fermentation by the Extracts of Allium tuberosum and Growth Inhibition of Related Microorganisms (부추추출물의 김치발효 지연 및 관련 미생물 증식억제)

  • The effect of retarding the fermentation of Kimchi by the extract of leek(Allium tuberosum) were tested by measuring the changes in pH, acidity and total cell number as well as number of microorganisms involved in Kimchi fermentation such as Lactobacillus, Leuconostoc, Pediococcus, Streptococcus and yeasts. The changes of pH and acidity of Kimchi stored at $25^{\circ}C$ indicated that the shelf-life of Kimchi with leek extract was retarded by 1.5 days compared with Kimchi without leek extract. Growth of Lactobacillus, Leuconostoc, Pediococcus and yeasts in Kimchi were remarkably inhibited by adding the leek extract at the initial and the 1st day of fermentation. This result suggested the methanol extract of leek can be sucessfully used for the extension of shelf-life of Kimchi.

Inactivation of Microorganisms and Browning Enzymes in Angelica keiskei Juice Using High Hydrostatic Pressure (초고압을 이용한 신선초 녹즙의 살균 및 갈색화 효소의 불활성화)

  • Effects of high hydrostatic pressure on microorganisms and browning enzymes in Angelica keiskei juice were investigated using response surface methodology. The optimum process condition for maximum reduction of total aerobes was $5700\;kg_f/cm^2$ (558.6 MPa) pressure and 7.16 min process time, and 3.44 log cycle reduction of total aerobes was predicted at the optimum condition. E. coli, initially $8.8{\times}10^3\;CFU/ml$, was completely inactivated by high hydrostatic pressure at all process conditions ($3800{\sim}6700\;kg_f/cm^2\;pressure;\;3{\sim}17\;min\;process\;time$). Polyphenol oxidase and peroxidase were partly inactivated by the high hydrostatic pressure. It was also indicated that inactivation of microorganisms and browning enzymes by hydrostatic pressure is dependent on pressure rather than process time.

Effects of Propolis Treatment on the Quality of Dried Squid (Propolis 의 처리가 마른 오징어의 품질에 미치는 영향)

  • Effect of propolis treatment on the quality of dried squid were investigated. Dried squid containing 29% of moisture content were prepared and treated with 0.25% and 0.5% of propolis. Its total microbial count, TBA value and browning degree were assayed during 30 days. Total microbial count of dried squid treated with propolis were $72{\sim}80\;cells/g$, compared with control showing 25,000 cells/g. TBA values of dried squid were by $0.1{\sim}0.2$ lower than control. Browning degree was not different between treatment and control, but browning degree of propolis treatment showed lower than control through colorimetric analysis. These results suggested that quality of dried squid containing higher moisture could be improved by propolis treatment.

  • An attempt was made in this study to investigate the possibility of processing squid along with pork into an intermediate moisture food, as an acceptable new item in Korea. To obtain a palatable and low cost product, portions of mixture consisting of equal amounts of ground squid and pork were cooked in soy sauce for 20 min, soaked in glycerol for 20 min at $80^{\circ}C$, and then air dried for 90 min at $60^{\circ}C$. During storage for 42 days in saturated humidity incubator at $37^{\circ}C$, the total microbial counts of the products packaged in polyethylene (0.05 mm), polypropylene (0.05 mm) and plastic laminated aluminum foil were decreased from 230 per gram to 40, 20 and 10 per gram respectively. In another long range storage test of 6 months, the results paralleled the above trends. Even though the TBA value of the samples increased during the storage, it was not so serious as to damage on the organoleptic quality of this product. The results indicate that stored product was as acceptable as the freshly prepared one.

  • To develop natural food preservative, antimicrobial activity of Pinus densiflora Sieb. et Zucc. (PD) ethanol extract against pathogens were investigated. The growth of pathogens such as Staphylococcus aureus ATCC 29737, Bacillus subtils KCTC 1021, Bacillus megaterium KCTC 3007 and Vibrio parahaemolyticus ATCC 17802 was inhibited as much as 4 to 6 log cycle in tryptic soy broth containing 40mg/㎖ of PD extract but Salmonalla typhimurium ATCC 14028 and Pseudomonas fluorescens ATCC 21541 were not inhibited. The total, gram negative and psychrotrophic bacterial count were inhibited in minced pork containing 40mg/㎖ of PD extract for 20 days at 4$\^{C}$. The sensory quality of hamburger patty such as taste, flavor and overall acceptability were not decreased by the addition of 40mg/㎖ PD(p<0.05).
