Extraction of${\beta}-carotene$ from Carrot by Supercritical Carbon Dioxide (초임계이산화탄소에 의한 당근 중의 ${\beta}-carotene$ 추출)

  • ${\beta}-carotene$ was extracted from freeze-dried carrot by supercritical carbon dioxide $(SC-CO_2)$ and mixtures of $CO_2$ doped with ethanol or methanol as a cosolvent at temperatures of 40 to $60^{\circ}C$ and pressures of 138 to 276 bar. Solubility of ${\beta}-carotene$ in $SC-CO_2$ increased with the increase of extraction pressure and the decrease of extraction temperature. The highest solubility observed was $4.90\;{\mu}g/g\;CO_2\;for\;{\alpha}-carotene\;and\;0.604\;{\mu}g/g\;CO_2$ for ${\alpha}-carotene\;at\;40^{\circ}C$ and 276 bar. Addition of ethanol increased the solubility being the largest increase of 82% using a mixture of $CO_2$ and 17.4% ethanol. $SC-CO_2$ extraction can be used to selectively obtain natural carotenoids, free of solvent residuals, which can be used as food additives.

  • Brown rice oil contains palmitic acid, linolenic acid, linoleic acid, oleic acid, stearic acid, tocopherol, squalene, etc. The oil including essential fatty acids was extracted from the domestic brown rice bran using supercritical carbon dioxide(SCC) process, and the extracts were analyzed with GC-MSD. The extraction amount of brown rice oil was dependent upon the operating pressure and temperature, and the fatty acid composition of oil was varied with the reduced density(${\rho}_{\gamma}$) of the SCC. About 70~80% of brown rice oil was extracted in 4 hours. Especially, squalene which was not found in solvent extract phase was identified in supercritical fluid extraction phase only.

Studies on the Recovery of Triglyceride from Used Shortening by Supercritical Fluid Extraction (초임계유체 추출에 의한 폐식용유의 재활용에 대한 기초연구)

  • Supercritical fluid extraction was applied to recycling triglyceride from used shortening. Used shortening and its fractions were analyzed with high performance size exclusion chromatography for their composition in triglycerides, polymer and low molecular weight compounds. Conjugated diene value and color of the fractions were also measured with a UV spectrophotometer and a colorimeter, respectively. Pressure and temperature ranges employed were $15{\sim}30$ MPa and $40{\sim}60^{\circ}C$, respectively. Concentration of fat in supercritical (SC) $CO_2$ ranged from $0.3\;X\;10^{-3}{\sim}7.4\;X10^{-3}(g\;fat/g\;CO_2)$. An exponential relation between concentration of fat in SC $CO_2$ and density was observed. Color of the extracts was light yellow which was very close to that of the fresh shortening. Low molecular weight compounds were preferentially concentrated in the initial fraction, while polymer was extracted in the final fraction. Conjugated diene value of the initial fractions was clearly lower than that of feed. It increased sharply as the polymer content in the fraction became significantly large.

A Comparative Study on the Compositions of Hwangryeonhaedok-tang's Essential Oils Obtained by Supercritical Carbon Dioxide Extraction and Hydrodistillation Methods

  • 초임계추출법과 수증기증류법을 이용하여 황련해독탕의 정유성분을 추출하여 성분 패턴을 비교하였다. 이때 초임 계추출조건은 압력 200 atm, $45^{\circ}C$ 그리고 추출시간 25분 이었다. 이때의 추출된 성분의 93.9%인 37개의 성분을 가스크로마토그래피/질량분석기로 확인하였다. 주요성분으로는 tetradecenoic acid (11.7%), Vanillin (5.9%), dl-Limonene (5.5%) 및 Eicosane (4.6%)으로 나타났다. 수증기증류법으로 추출한 정유에서는 34개 성분을 확인할수 있었다. 주요성분으로는 tetradecenoic acid (8.9%), Vanillin (5.8%)및 Eicosane (4.7%)를 확인 할 수 있었다. 또한 구강내의 12균주를 이용하여 항균효과를 측정하여 최소엑제농도(MIC)와 사멸농도(MBC) 0.025 - 12.8 mg/ml와 0.05 - 12.8 mg/ml 각각 나타났다.

Purification of Paclitaxel and Its Derivatives by Supercritical Fluid Chromatography(SFC) (초임계 유체 크로마토그래피(SFC) 방법에 의한 Paclitaxel 및 그 유도체의 분리 정제)

  • Studies were carried out to elucidate the effects of pressure, temperature and mobile phase composition on supercritical $CO_2$ chromatographic separations of paclitaxel, baccatin III, 10-deacetyl baccatin III, 7-epi-10-deacetyltaxol, cephalomanine, and 10-deacetyltaxol. High resolutions of paclitaxel, 10-deacetylbaccatin III, 10-deacetyltaxol were observed with optimized pressure, temperature, and mobile phase composition. The highest resolution between paclitaxel and 10-deacetylbaccain III was observed at 275 kg/$\textrm{cm}^2$, $40^{\circ}C$ with the mobile phase composition of gradient mixture of 3.9-3.6 mL/min $CO_2$, 0.1-0.4 mL/min methanol for 20 min. Resolutions of baccatin III, capalomannine, and 7-epi-10-deacetyltaxol were found to be low in this study. On-line coupled SFE/SFC process was applied to isolate paclitaxel from yew tree powder. As a consequence, paclitaxel with a purity of 95% was obtained with a recovery yield of 38%.

A Study on Physiochemical Characteristics of Xanthoceras sorbifolia Seeds Oil (문관나무 종자유의 이화학적 특성 분석)

  • This study investigated the physiochemical characteristics of Xanthoceras sorbifolia seed oil. Xanthoceras sorbifolia seed oil was extracted by supercritical fluid extraction (420 atm, $50^{\circ}C$), hexane extraction and heat-pressed extraction ($160^{\circ}C$, $180^{\circ}C$). Acid values and peroxide values were evaluated, as well as the degree of lipid oxidation. The heat-pressed ($160^{\circ}C$) extraction gave a $53.5{\pm}2.5%$ higher yield of oil, compared with the other extraction methods. The acid values from the super critical fluid extraction were the highest, while peroxide values were highest from the heat-pressed extraction at $160^{\circ}C$ (3.10 meq/kg). The contents of linolenic acid and oleic acid were 38.63~41.13% and 26.29~26.85%, respectively. Contents of stigmasterol and ${\beta}$-sitosterol were 6.01~6.49 mg/100 g and 58.19~59.85 mg/100 g, respectively. These results indicate that Xanthoceras sorbifolia seed oil can possibly serve as new edible oils.

Desorption Characteristics of Grinding Oil from Swarf by using Supercritical Carbon Dioxide (초임계 이산화탄소를 이용한 스와프로부터 연마유 탈착 특성)

  • The recovery of stainless steel fiber by removing cutting oil from grinding swarf, which is classified as specified wastes, was investigated. Swarf loaded with grinding oil was regenerated by supercritical carbon dioxide. And, the effects of temperature(313.15K-323.15K), pressure(10MPa-30MPa) on regeneration efficiency were studied. Regeneration effiency was increased as the pressure was increased. Also, at the same pressure, the experiments at higher temperature were more efficient for regeneration. The experiment results was predicted by applying a one-parameter mathematical model assuming linear desorption kinetics. The predicted value showed good agreement with experimental data.

  • The purpose of this study was to find an optimum extraction condition of acanthoside-D from acanthopanax cortex with supercritical carbon dioxide as a solvent. In this effort, effects of the extraction conditions including pressure, temperature and presence or absence of a cosolvent on the extraction efficiency were investigated. The ethanol, water or 50% methanol was used as a cosolvent whilst the operating pressure ranged from 200 bar to 300 bar. The acanthoside-D concentrations were determined by means of HPLC equipped with a UV detector. From the results, it was observed that increase of higher pressure led to the higher extraction efficiency. Further, water was found to be the best cosolvent among the entrainers tested.

Supercritical Carbon Dioxide Extraction of Beef and Pork for Low Lipid Sausage Manufacturing (저지방 소시지 제조를 위한 쇠고기와 돼지고기의 초임계 이산화탄소 추출)

  • Lipid and cholesterol were extracted from beef and pork by the supercritical carbon dioxide $(SC-CO_2)$ for the manufacturing of low-lipid and low-cholesterol sausage. The ranges of extraction temperature and pressure were from 35 to $55^{\circ}C$ and from 103 to 375 bar, respectively. $SC-CO_2$ extraction yield of beef lipid increased as extraction pressure increased and/or extraction temperature decreased, while extraction temperature was more influential on the cholesterol extraction than pressure condition. When lipid and cholesterol of freeze-dried beef with varied moisture contents were extracted, their solubilities increased as the moisture content reduced. The extraction of lipid and cholesterol from pork was shown the same tendency as the beef. The chunk type of beef shape was more suitable for the lipid and cholesterol extraction than the powder type of beef. The color of meat after $SC-CO_2$ extraction was lighter than the raw freeze dried meat because of the extraction of color pigments. After $SC-CO_2$ extraction, beef and pork were rehydrated and mixed with raw beef and pork containing lipid and cholesterol. Their mixing ratio up to 50 : 50 did not affect physical properties of the sausage compared with the control sausage.

Extraction of Glabridin from Licorice Using Supercritical Carbon Dioxide (초임계 이산화탄소를 이용한 감초 중의 glabridin 추출)

  • The purpose of this study is to investigate the feasibility of a cosolvent-modified supercritical $CO_2\;(scCO_2)$ extraction technique for the production of licorice extracts with high levels of glabridin. The effects of various parameters such as the type and amount of modifiers, extraction temperature ($40{\sim}80^{\circ}C$) and pressure ($10{\sim}50.0\;MPa$) on the extraction efficiency were examined at a fixed flow rate of 1 mL/min. The organic solvent extraction with pure methanol was also conducted for a quantitative comparison with the $scCO_2$ extraction. The recovery of glabridin from licorice was found to be extremely small for pure $scCO_2$. However, the addition of modifiers such as ethanol and acetone to $scCO_2$ resulted in a significant improvement in the recovery of glabridin. The recovery of glabridin was observed to increase with pressure at a constant temperature. Furthermore, the purity of the glabridin obtained from the $scCO_2$ extraction was higher compared with the organic solvent extraction.
