Selection of Lactic Acid Bacteria suitable for Manufacture of Freeze-dried Coffee (동결건조커피 제조에 적합한 유산균 균주 선발)

  • Probiotic functional foods are known to have various functional effects such as intestinal regulation, modulation of immune system, reduction of allergies, and lowering of cholesterol. The purpose of this study was to select probiotic strain that is most suitable for freeze-dried coffee for the development of functional coffee products. The survival rate of probiotics, at drinking condition of coffee, at acid, at bile and after freeze-dried in coffee were measured on 1 strain isolated from commercial freeze-dried coffee, 8 strains used as fermented milk starter, 1 Bifidobacterium and 1 Bacillus coagulans. Bacillus coagulans showed the highest survival rate from $2.4{\times}10^7cfu/g$ to $2.0{\times}10^7cfu/g$ especially after freeze-drying. The results at drinking condition of coffee, at acid tolerance, at bile tolerance and at storage test showed significantly better survival rate of Bacillus coagulans than that of control (Lactobacillus casei). Especially, Bacillus coagulans showed 3.8-fold higher survival rate at acid tolerance (pH 1, 120 minutes) than control. Thus, the lactic acid-producing Bacillus coagulans is characterized as a probiotic strain suitable for functional coffee formulation and commercialization.

Effects of Soil Texture, Irrigation System, and Soil Ameliorators on the Cadmium Adsorption in Soil and Uptake in Rice Plant (토양의 카드뮴흡착과 수도체 카드뮴흡수에 미치는 토성, 물관리 및 개량제 효과)

  • This study was conducted to investigate the adsorptive characteristics with cadmium in different levels of organic matter and lime in soil. And in order to identify the effect of soil ameliorators on cadmium uptake in rice plants, compost and lime were treated. Plants were grown at two soil textures(sandy loam and clay loam) with irrigation water containing $0.01\;mg\;L^{-1}$ of cadmium and treatments of two irrigation systems(intermittent irrigation and continuous submersion). The adsorption capacity of cadmium by soil was increased in proportion to initial concentration of solution, and it was higher at clay soil compared to loamy soil. The adsorption rate of cadmium by soil was increased with increasing the concentration of organic matter and lime in soil, highly increased at the both organic matter and lime treatment. Soil pH was negatively correlated with the cadmium contents of the both shoot and brown rice while Eh was positively correlated with those. In the harvest season, cadmium contents in the both leaves and brown rice were lower in the clay sail plots than sandy soil plots, and the continuous submersion plots were lower than intermittent irrigation plots. Cadmium uptake was highly reduced at the compost and lime mixture plot compared to other treatments among the continuous submersion plots. The cadmium content of shoot was positively correlated with that of brown rice in the harvest season.

Microstructure and Properties of Organic-Inorganic Hybrids(PDMS/SiO$_2$) Through Variations in Sol-Gel Processing (졸-겔공정의 변수조절을 통해 제조된 유기-무기복합체 (PDMS/SiO$_2$)의 미세구조와 특성)

  • SiO$_2$ and PDMS/SiO$_2$ xerogels which are derived PDMS into TEOS have been synthesized by sol-gel process and controlled pore size and distribution through 2 step acid/base catalyzed processes using HCI and NH$_4$OH as a catalyst. In HCl catalyzed SiO$_2$ and PDMS/SiO$_2$ xerogels, pH and gellation time of xerogel were 2.3~2.5 and 12~13 days, respectively, and the shape of xerogel was identified to pellet type and column type. Under acidic condition of final reaction solution, the hydrolysis rate is accelerating, resulting in long gel times. The shape of xerogel is pellet type. In contrast, under less acidic condition, the condensation rate is accelerating, resulting in shorter gel times and the shape of xerogel is column type. The surface area and average Pore size were changed 400$\rightarrow$600($\m^2$/g) and 15$\rightarrow$28$\AA$, respectively, depending to the increase of the mole ratio of HCl/NH$_4$OH, and represented uniform pore size distribution. It is that all the alkoxide groups are hydrolyzed by HCl after the first step and the condensation rate is enhanced by NH$_4$OH. The regular backbone structures of silica are formed at low temperature and the uniform pores are produced by heat treatment.

Method Development for the Sample Preparation and Quantitative Analysis of Synthetic Colors in Foods (식품 중 식용타르색소의 시료별 전처리방법 확립 및 함량 분석)

  • Method for sample preparation and quantitative analysis of 19 permitted and non-permitted synthetic colors in foods was developed based on reversed-phase ion-pairing high performance liquid chromatography. For color extraction of samples, deionized water was added, and pH was appropriately adjusted with 1% ammonia water. Any undissolved matters were extracted with 50% ethanol or 70% methanol. Lipid in snacks was first removed using n-hexane with centrifugation, water was added to extract colors, followed by clean-up and concentration using Sep-Pak $C_{18}$ cartridge. Recovery efficiencies at known concentrations of 19 standard food colors spiked into foods were in 90.3-97.9% range far soft drink, 79.2-101.9% for candy, 84.1-103.4% for jelly, 86.4-100.8% for chewing gum, 83.5-103.4% for ice cream, and 78.5-95.6% for snack.

Storage Quality of Ready-to-Eat Campbell Table Grapes as Affected by Active Modified Atmosphere Packaging (기체충진 포장조건에 따른 신선편이 캠벨 포도의 저장 중 품질변화)

  • The storage quality of ready-to-eat Campbell table grapes which were packaged under modified atmospheres was investigated in order to examine the effect of high $O_2$ and $CO_2$ on the fruit. Fresh table grapes with 10-15 berries were packed into polypropylene (PP) trays and were top-sealed with polythylene terephthalate/PP film. The initial gas compositions inside the packages were air, 20% $O_2$/10% $CO_2$/70% $N_2$, and 40% $O_2$/60% $N_2$. Sealed packages with low density polyethylene film bags and perforated PP trays were also used as a further treatment and control, respectively. The quality attributes were assessed during storage at $5^{\circ}C$ for 28 days. Fruit packaged in high $CO_2$ concentration showed the lowest viable cell counts of inherent microorganisms among all samples, although they suffered from severe off-flavors. High levels of $O_2$ significantly lowered flesh weight loss and maintained the flavor of grape. In an overall sensory aspect, the high $O_2$ and $CO_2$ packages exhibited greater scores than the air and control at the end of the storage period. Other quality attributes showed no significant differences among treatments. Results suggest that packaging with an appropriate combination of high $O_2$ and $CO_2$ can be used as an effective processing treatment for improvement of the storability of ready-to-eat table grapes.

Storage Quality of Sulhyang Strawberries as Affected by High O2 Atmosphere Packaging (고산소 환경기체조절 포장조건에 따른 설향 딸기의 저장 중 품질변화)

  • The storage quality of fresh Sulhyang strawberries packaged under modified atmospheres was investigated to examine the effect of high $O_2$ on the fruit. Fresh strawberries were packed into PP trays and top-sealed with PET/PP film. Initial gas compositions inside the packages were varied with air, 40% $O_2$/60% $N_2$, 60% $O_2$/40% $N_2$, and 80% $O_2$/20% $N_2$. Sealed packages in PE film bags with air and perforated PP trays were also used as another treatment and control, respectively. Quality attributes and viable cell counts of pathogenic bacteria were assessed during storage at $5^{\circ}C$ for 12 days. High $O_2$ concentration showed no significant effects on the physicochemical and microbial qualities of strawberries. Fruit packaged in PE film bags with 6-15% $O_2$ and 7-9% $CO_2$ during storage had the lowest viable cell counts of inherent microorganisms among the treatment samples. Growth of pathogenic bacteria was suppressed in perforated packages where molds occurred frequently. In an overall sensory aspect, the PE film packages exhibited higher scores than the others at the end of storage period. The experimental results suggested that gas-permeable film packaging with an appropriate combination of $O_2$ and $CO_2$ rather than gas-barrier tray packaging with an initially high $O_2$ concentration would be suitable for improving the storability of strawberries.

Chemical Effects on Head Loss across Containment Sump Strainer under Post-LOCA Environment (LOCA이후 환경에서 원자로건물집수조 여과기의 수두손실에 대한 화학적 영향)

  • A test apparatus has been fabricated to simulate chemical effect on head loss through a strainer in a pressurized water reactor (PWR) containment water pool after a loss of coolant accident (LOCA). Tests were conducted under condition of same ratio of strainer surface area to water volume between the test appratus and the containment sump. A series of tests have been performed to investigate the effects of spray, existence of calcium-silicate with tri-sodium phosphate (TSP), and composition of materials. The results showed that head loss across the chemical bed with even a small amount of calcium-silicate insulation instantaneously increased as soon as TSP was added to the test solution. Also, the head loss across the test screen is strongly affected by spray duration and is increased rapidly at the early stage, because of high dissolution and precipitation of aluminum and zinc. After passivation of aluminum and zinc by corrosion, the head loss increase is much slowed down and is mainly induced by materials such as calcium, silicon, and magnesium leached from NUKONTM and concrete. Furthermore, it is newly found that the spay buffer agent, tri-sodium phosphate, to form protective coating on the aluminum surface and reduce aluminum leaching is not effective for a large amount of aluminum and a long spray.

The Effect of Acetylcholine on the Intracellular $Ca^{2+}$ Increase of the Mouse Early 2-cell Embryos (생쥐 초기 2-세포 배의 세포내 칼슘 증가에 미치는 Acetylcholine의 영향)

  • Many studies have shown that the development of mouse early 2-cell embryos in vitro is related with the intracellular $Ca^{2+}$ changes. In ICR strain mouse, the development of embryos arrests at early 2-cell stage, but the arrested early 2-cell embryos can be rescued by the addition of $Ca^{2+}$-related materials. Acetylcholine (ACh) increases intracellular $Ca^{2+}$ concentration ([$Ca^{2+}$]i) via the mAChR-PLC-IP3 pathway in mouse oocytes. We examined whether ACh rescues 2-cell block in mouse. In early 2-cell embryos, ACh increased [$Ca^{2+}$]i in a dose-dependent manner (p<0.001), and had an effect on rescue of 2-cell block and embryonic development. To identify the signal pathway involved in ACh-induced rescue of 2-cell block, we first applied an agonist of ACh receptor (AChR). Like ACh, carbachol increased intracellular $Ca^{2+}$ concentration ([$Ca^{2+}$]i) and atropine, an antagonist of ACh receptor, blocked the ACh-induced $Ca^{2+}$ increase. In $Ca^{2+}$-free medium, ACh also increased [$Ca^{2+}$]i, indicating that $Ca^{2+}$ increased by ACh is mainly released from the intracellular $Ca^{2+}$ store. The ACh-induced $Ca^{2+}$ increase was blocked by PLC inhibitor (U73122), ryanodine receptor (RyR) antagonist (dantrolene), and CaM KII inhibitor (KN-93), but not by IP3R antagonists (xestospongin C). These results show that ACh increases intracellular $Ca^{2+}$ concentration via mAChR/PLC/RyR, and this contributes to the rescue of 2-cell block.

  • Systemic acquired resistance (SAR) is a form of systemic immunity that prevents secondary infections of distal uninfected parts of plants by related or unrelated pathogens. SAR is mediated by several SAR-inducing chemicals or mobile signals that accumulate after pathogen infection. Several chemicals that move systemically have already been identified as SAR-inducing factors, despite the fact that the early mobile signal remains unclear. These chemicals can be transported into either the apoplastic or symplastic compartments. Many of the chemicals associated with SAR remain unknown in terms of their transport routes. There is recent evidence that azelaic acid (AzA) and glycerol-3-phosphate (G3P) are transported via plasmodesmata (PD) channels, which regulate the symplastic route. In contrast, salicylic acid (SA) is preferentially transported from pathogen-infected to uninfected parts via the apoplast. The pH gradient and SA deprotonation lead to apoplastic accumulation of SA before it accumulates in the cytosol. Moreover, there is evidence that the mobility of SA over a long distance is crucial for SAR and that the partitioning of SA into the symplast and cuticles is controlled by transpiration. Further research has shown that a portion of the total SA in leaves is partitioned into cuticular waxes. The purpose of this review is to discuss the role of SAR-inducing chemicals and the regulation of transport in SAR.

Physico-chemical and Sensory Characteristics of Cooked Sausage Substituted with KCl or MgCl2 for NaCl (KCl 또는 MgCl2의 NaCl 대체 소시지의 이화학적 및 관능적 특성)

  • This study was carried out to investigate changes in physicochemical and sensory properties of cooked sausages replaced sodium chloride (NaCl) to potassium chloride (KCl) or magnesium chloride ($MgCl_2$) during storage for 30 days under $4^{\circ}C$. All sausages were prepared with different combination of salts as follow; CTL (1.5% NaCl), KCL (0.9% NaCl+0.6% KCl), MCL (0.9% NaCl+0.6% $MgCl_2$), KML (0.9% NaCl+0.3% KCl+0.3% $MgCl_2$) and PST (1.5% PanSalt). Among sausages moisture content in KML was the highest (p<0.05). Lightness and redness in CTL were lower than those of other treatments, but MCL and KML containing $MgCl_2$ showed higher CIE $L^*$ and $a^*$ values than CTL. The pH in CTL was the highest during storage, however, no significant difference was determined between two treatments, MCL and KML (p>0.05). Crude fat content and water holding capacity (WHC), hardness and cohesiveness of MCL sausages were higher than those of CTL. In sensory characteristics of cooked sausages, saltness in MCL was the lowest during 10 and 20 days of storage (p<0.05). Yellowness in PST was lower than other treatmeants. Gumminess and chewiness of texture property of sausages from MCL and KML were higher than CTL. The results indicate that the replacement of NaCl by KCl improved texture, but meat color was not improved as expected. In contrast, the replacement of NaCl by $MgCl_2$ enhanced color, texture and WHC, whereas partial replacement of NaCl by $MgCl_2$ must reduce bitter taste as compared to sausages manufactured with a NaCl only. Therefore, $MgCl_2$ may be a salt replacing NaCl in cooked pork sausages.
