The Characteristics of Carbon Dioxide Fixation by Chlorella sp. HA-1 in Semi-continuous Operation (반연속식 운전에서 Chlorella sp. HA-1의 이산화탄소 고정화 특성)

  • The microalgal, Chlorella sp. HA-1, had good $CO_2$ fixation efficiency compared to other algal strains at the same operating condition. In this study, Chorella sp. HA-1 showed similar tolerance both 10% and 20% $CO_2$ concentration. By optimization of the major operation variables such as pH, initial cell concentration, light intensity, the $CO_2$ fixation rate could be raised to a reasonably high value, 372 $gCO_2/m^2{\cdot}day$ in a 3 L internally illuminated photobioreactor. In order to maintain the $CO_2$ fixation rate for a long time, the method of semi-continuous operation was employed, in which dilution ratio was the controlling parameter. Starting with the dilution ratio of 0.5 with the increased increment of 0.1, the constant $CO_2$ fixation rate was obtained.

Studies on Ginger Mosaic Virus (생강모자이크바이러스병에 관한 연구)

  • A mosaic virus disease of ginger plant was investigated to determine its virus group on the basis of host range, physical and chemical properties, serological behavior and electron-microscopic morphology. The disease gave rise to yellowsih and dark-green mosaic on the leaves in the early stage and stunted all the leaves as well as rhizomes in the late stage. In the field about 43\% of the plants were observed to be diseased The disease was able to be artificially infected to the ginger plants by the sap and transmission as well as to 23 other species of plants which were known to be the CMV susceptible plants by the sap transmission; Chenopodium amaranticolar, Nicotiana tabaccum var. Havana, cow pea, cucumber, tomato,... etc. The dilution end point of the virus ranged $10^{-4}-10^{-5}$ and the thermal inactivation point $65-70^{\circ}C$. Serological test showed a positive reaction by a CMV antiserum. An electron microscopy of the purified virus showed that the virus particles were spherical with a diameter of $28-32m\mu$. Virus particles from the infected tissue were observed to be free or aggregated in the mesophyll tissue of artificially infected tobacco plant. The mosaic disease of ginger plants were conclusively suggested to the CMV group.

Culture Optimization for Bacillus lentimorbus G-74 by Using a Miscanthus purpurascens Juice Medium (억새즙액 배지를 이용한 Bacillus lentimorbus G-74 균주의 배양 최적화)

  • Miscanthus purpurascens Juice containing potassium (37,952 mg/L), nitrogen (14,000 mg/L), phosphorus (6,800 mg/L), magnesium (5,969 mg/L), calcium (5,910 mg/L), etc., was investigated to develop a novel meidum far the mass cultivation of useful microorganisms. For this research, we first isolated an antagonistic bacterium G-74 from soil, which showed strong growth inhibition against two phytopathogenic fungi, Rhizoctonia solani and Botrytis cinerea, and identified as Bacillus lentimorbus G-74 based on the morphological characteristics and MIDI analysis. Culture conditions for G-74 strain in the M. purpurascens juice medium were optimized. Dilution rate of the medium, temperature and initial pH for the optimum growth of G-74 strain were 30% (V/V), $35^{\circ}C$ and 5.0, respectively. It was found that additions of 2.0% (W/V) corn starch as a carbon source and 1.0% (W/V) yeast extract as a nitrogen source in this medium increased B. lentimorbus G-74 growth to 66% more efficient than Luria Bertani medium.

  • Severe damage can occur around deposition holes due to complex interaction of thermo-hydro-mechanical (THM) loading during the long term operation of high level radioactive waste repository. Many candidate sites for repository are located in crystalline rock mass, therefore mechanism of damage follows the form of brittle fracture and failure. This paper briefly introduces major outcomes from 15 years international collaborative project, DECOVALEX, and presents major study results for current ongoing benchmark test study from DECOVALEX-THMC, to evaluate the effect of THM loading to rock mass in excavation damaged zone (EDZ) near deposition holes. Through benchmark test model by simplifying THM loading to boundary loading obtained numerical results are compared, and discrete fracture interaction after up to 1 million years operation is discussed.

  • A new underground LNG storage concept in the rock mass has been developed by combining underground cavern construction and new ice-ring harrier technologies with the conventional cryogenic insulation system. Technical feasibility of the storage system has been verified through construction and operation of the pilot storage cavern and a full-scale project is expected to start in the near future. One of the most important issues in the LNG storage system is the operational efficiency of the storage to minimize heat loss during a long period of operation due to the cryogenic heat transfer. This paper presents several important results of heat transfer and coupled hydro-thermal analyses by a finite element code Temp/W and Seep/W. A series of heat transfer analyses for full-scale caverns were performed to determine design parameters such as boil-off gas ratio (BOR), insulation thickness and pillar width. The result of the coupled hydro-mechanical analysis showed that BOR for underground storage system remains at about 0.04 %/day during the early stage of the operation. This value could be even much lower when the discontinuities in the rock masses are taken into consideration.

Search for the activity measurement of radionuclides I-131 (131I을 이용한 방사능 측정에 관한 연구)

  • Iodine is one of important nuclides to be checked for radiation exposure after nuclear power facility accidents. After Chernobyl accident, it was observed that there is a greater amount of organic iodine in the atmosphere than inorganic iodine. In this study, we not only varied the amount of sample being exposed to $^{131}I$ and the duration of exposure to $^{131}I$ but also diluted the sample in distilled water and mixed the sample in kelp and liquid $^{131}I$ to measure and analyze the radiation detection levels. We concluded that the radiation levels were not high enough to be harmful to human body. The radiation from $^{131}I$ decreased over time, and we calculated the half life at 7-9 days. We found that the radiation from any sample containing $^{131}I$ was halved by up to 7days.

산소 플라즈마로 표면 개질 된 Si-DLC 필름의 젖음각 거동

  • DLC 필름은 바이오 적합성, 특히 생체 적합성이 뛰어나기 때문에 바이오 코팅분야에서 널리 이용된다. 많은 연구 결과에 의하면 세포와 장기 등이 바이오 재료 표면에 적절히 접합할 수 있도록, 재료 표면을 산소나 질소를 이용하여 플라즈마 처리로 초친수성 표면으로 개질하고 있다. 하지만, 시간이 지남에 따라서 친수성 표면은 점차 재료의 표면 처리 전의 성질인 소수성을 회복하게 된다. D실제 생체에 적용하기 위해서 이러한 시효 효과에 대한 정확한 평가가 이루어져야 한다. 따라서 산소와 질소 플라즈마 처리 후의 친수성 성질이 소수성 성질로 변해가는 거동을 조사하는게 중요하다. 13.56 MHz의 plasma assisted chemical vapor deposition (PACVD) 법을 이용하여 DLC와 Si-DLC를 500 ${\mu}m$ 두께의 P-type 실리콘(100) 기판에 증착하였다. 박막 증착 과정에 사용한 기체는 벤젠과 희석된 silane이 사용되었다(SiH4/H2=10:90). 박막 증착은 -400 V의 바이어스 전압을 인가하였으며, 이때 증착 압력은 1.33Pa으로 일정하게 유지하여, 두께 $0.55{\pm}0.01{\mu}m$로 증착하였다. X-ray Photoelectron Spectroscopy (XPS) 법을 이용하여 실리콘 함량을 측정하였으며, 증착 된 Si-DLC의 실리콘 함량은 0~4.88 at. %였다. 이후에 질소와 산소 플라즈마를 이용하여 챔버 압력을 1.33 Pa로 유지하여, -400 V의 바이어스 전압을 인가하여 10분간 표면 처리를 하였다. 표면 처리된 DLC와 Si-DLC 표면 위에서의 물방울(water droplet)의 젖음각을 20일간 측정하였다. 플라즈마 표면 처리 된 모든 시편에서 초기 젖음각은 $10{\sim}20^{\circ}$의 친수성 성질을 보였지만, 점차 젖음각이 상승하여 산소 플라즈마 처리 된 Si-DLC를 제외하고는 5일이 지나면서 거의 소수성 표면으로 회복되었다. 산소 플라즈마 처리 된 Si-DLC의 경우, 젖음 각 측정 기간(20일) 동안 $15^{\circ}$ 미만의 친수성 성질을 유지하였다.

Behaviour of Lightweight Concrete Slab Reinforced with GFRP Bars under Concentrated Load (집중하중을 받는 GFRP 보강근 경량콘크리트 슬래브의 거동)

  • This paper is a preliminary study to apply the lightweight concrete slabs reinforced with GFRP (glass fiber reinforced polymer) bars to the bridge deck slabs or some other concrete structures. So, some different behaviors between the conventional steel reinforced concrete slab and the lightweight concrete slab reinforced with GFRP bars were investigated. For this purpose, a number of slabs were constructed and then the three point bending test and numerical analysis for these slabs were performed. The flexural test results showed that the lightweight concrete slabs reinforced with GFRP bars were failed by the shear failure due to the over-reinforced design. The weight and failure load of the GFRP bar reinforced lightweight concrete slabs were 72% and 58% of the steel reinforced concrete slab with the same dimensions, respectively. Results of the numerical analysis for these slabs using a commercial program, midas FEA, showed that the load-deflection curve could be simulated well until the shear failure of the slabs, but the applied loads and the deflections continuously increased even beyond the shear failure loads.

무세미 조제시스템 개발

  • 현재 유통되고 있는 일반백미의 경우 소비자가 밥을 짓기 위해서는 쌀을 씻어야하는 번거로움 뿐만아니라 쌀 중량의 약 15배의 물이 소요되고, 또한 이때 발생된 쌀뜨물이 수질오염의 원인이 되기 때문에 무세미가공 및 보급 필요성이 점차 높아지고 있다. 무세미 가공분야의 선진국인 일본의 경우 ‘92년부터 습식 무세미조제기가 실용화되기 시작, 현재 사다께 등 6개사에서 기계장치를 생산보급 하고 있으며, 무세미의 유통도 일반화되어가고 있는 추세이다. 국내의 경우 무세미조제기는 근래에서야 습식 무세미 조제설비가 국산화되어 보급초기 단계 있으며, 무세미의 안정적 가공 및 보급을 위해서는 앞으로 많은 연구가 이루어져야 할 것으로 생각된다. 특히 습식의 경우 쌀이 물에 접촉하는 시간이 길 경우 품질저하의 우려가 크고, 쌀중량의 1.4배에 해당하는 물이 가공과정에 필요하기 때문에 물사용량을 억제할수 있는 가공기술의 개발 필요성이 커지고 있다. 따라서 본 연구에서는 물사용량을 최소화 할 수 있는 무세미 조제시스템을 개발하고자 하였으며, 주요결과를 요약하면 다음과 같다. 시작기는 연마 및 공기세척부, 정전기 세척부, 미세가수세척부로 구성하여 쌀이 단계적으로 세척될 수 있도록 제작되었다. 성능시험 결과, 각 세척공정별 세척수의 탁도 감소효과는 연마 및 공기크리닝부에서 22.67ppm, 정전기 크리닝부에서 8.33ppm, 미세가수세척부에서 17.34 ppm이 감소되는 것으로 나타났다. 특히 정전기세척부의 탁도감소 효과는 다른 두공정에 비해 작지만 제거가 쉽지 않은 미세한 쌀겨들을 제거하여 탁도를 개선을 시켰기 때문에 미세가수세척장치에서 가수량을 적게 사용하는데 기여한 것으로 판단된다. 가공시 적정 탁도를 확보할 수 있는 가수량은 430cc/kg로 기존의 습식에 비해 약 69%정도 세척수 절감효과가 있었다. 이때 미세가수세척부의 원통스크린 회전수는 108, 205rpm범위가 적정한 것으로 나타났다. 쌀의 품위는 탁도가 가공전 97.33ppm(일반백미)에서 가공후 최대 48.00ppm으로 낮아졌으며, 백도도 가공전 36.80에서 42.80으로 향상되어 씻지 않고도 밥을 지을수 있는 무세미 가공이 가능하였다. 이밖에 쇄립률은 가공전 5.30%에서 7.37%로 다소 증가하는 것으로 나타났고, 함수율은 가공전 15.60%에서 15.80%로 약 0.2%가 증가하였으나 기존의 연구결과에 비춰볼 때 문제가 되지 않을 것으로 여겨진다.

Cost Analysis of Ocean Outfall and Tertiary Treatment Processes in Suyong Sewage Treatment Plant (수영 하수처리장 방류수의 해중 방류법과 3차 처리시설 설치시 비용 비교 분석)

  • Sewage has been almost treated by secondary treatment process. Secondary-treated effluent of sewage treatment plant caused the pollution of nearby beach. Nitrogen(N) and Phosphorus(P) in effluent water have caused many problems on estuary, such as red tide, eutrophication and aquatic toxicity. Therefore, the effective nitrogen and phosphorus removal from sewage treatment plants is necessary to prevent those pollution problems. However, little sewage treatment plant in Korea is effectively being operated for the removal of the nutrients. This study is analyzed for the effectiveness of cost when tertiary treatment process and Ocean Outfall are applied for the water quality of Suyong Bay After secondary treatment process, the effluent was discharged from the seabed in the depth of 32m of 4000m offshore. Pollutant concentration is decreased as much as the 180 times after the result of initial dilution, so that environmental protection requirement of Suyong Bay can satisfied. This Ocean Outfall process can save the 2.6~3.5 times as much as the cost of construction and operation for tertiary treatment process running over a 20 year.
