Changes in Allergenicity of Porcine Serum Albumin by Microwave, Sonication, and High Hydrostatic Pressure (Microwave, 초음파 및 초고압 처리에 의한 돼지 혈청 알부민의 항원성 변화)

  • Even though pork have frequently induecd allergic reactions in Korea, few papers have been published on pork allergy. This study was carried out to investigate the changes in allergenicity of porcine serum albumin (PSA) by microwave, sonication, and high hydrostatic pressure (HHP). The binding ability of p-IgG to PSA treated with microwave (1,5, or 10 min) directly decreased with increasing treatment time. Particularly, the binding ability of PSA treated 10 min was about 30%. Immunoblotting assay with p-IgG showed that band of PSA treated microwave directly disappeared at 5 and 10 min. However, the binding ability of PSA was not changed by the microwave treatment without heat. Also the reduction of allergenicity by sonication or HHP treatment was not found. In conclusion. allergenicity of PSA treated with microwave directly decreased with increasing time, therefore these results may be used for development of hypoallergenic pork.

Microbial and Quality Changes during Storage of Raw Oyster Treated with High Hydrostatic Pressure (초고압 처리한 생굴의 저장 중 미생물수 및 품질 변화)

  • Raw oysters were treated at $10^{\circ}C$ and $22^{\circ}C/350$ Mpa/15 min, and microbial counts and quality were measured during storage of 14 days at $10^{\circ}C$. Total viable cell count (TVCC) in untreated oyster increased greatly during storage from starting inoculum of $1.6\times10^2\;CFU/mL$, and reached to $5.6\times10^2\;CFU/mL$ after 4 days of storage. TVCC of the pressure-treated was about $10^1\;CFU/mL$ right after high hydrostatic pressure treatment and increased slowly during storage, and about $10^3\;CFU/mL$ even after 7 days of storage. Lactic acid bacteria count (LABC) in the untreated was increased greatly during storage from starting inoculum of $3.3\times10^3\;CFU/mL$ at 3 days of storage and $7.2\times10^4\;CFU/mL$ after 4 days of storage. LABC in the pressure-treated was detected only after 5 days of storage, and about $10^2\;CFU/mL$ after 8 days of storage. The pH of the untreated was 6.19 and decreased gradually during storage, and 5.83 after 4 days of storage. The pH of the pressure-treated showed little change during storage, and 6.07, 6.03 and 5.82 after storage of 4, 8 and 14 days, respectively. Volatile basic nitrogen (VBN) in the untreated was 16.8 mg%, and maintained almost constant until 1 day of storage, and then increased suddenly, and 30.1 mg% after 4 days of storage. VBN of the pressure-treated stayed unchanged during storage, and about 20 and 23 mg% even after 4 and 8 days of storage, respectively. Hunter $L^*,\;a^*\;and\;b^*$ values were increased until 2 days of storage and then showed no change during storage. Demerit score was the lowest in the thawed raw oyster, and then in the increasing order of the pressure-treated (4 day and 8 day storage) and the untreated (4 day storage).

Quality Changes of Jujube Wine by Hydrostatic Pressure and Freezing Treatment during Storage (초고압 및 냉동처리에 의한 대추술의 저장 중 품질변화)

  • Jujube wine was treated with hydrostatic pressure (500 MPa, 5 min) or freezing ($-20^{\circ}C$, 3 days and thawed $20^{\circ}C$, 4 hr) and their microbial counts, physicochemical properties, and sensory characteristics were investigated during storage at $35^{\circ}C$ for 60 days. Microorganisms in pressure-treated jujube wine were not detected during the whole period and chemical compositions as well as color were slightly changed. Sensory quality was significantly preserved until 20 days without increasing sweet aroma and taste. In freezing-treated wine, bacterial counts were decreased after 10 days and remained below 10 CFU/mL while lactic acid bacteria and yeast were not detected. Changes of chemical composition were smaller than those of the untreated wine but bigger than those of the pressure-treated or heat-treated wine. Instrumental color was changed after $10{\sim}20$ days resulting from the increase of L value and the reduction of a value. Sensory quality was significantly similar with the fresh wine for 10 days, suggesting that pressure treatment would be the most effective sterilization method to improve the shelf life of jujube wine whereas freezing treatment would be insufficient.

Effect of High Pressure Processing on the Rancidity of Yeonhaeju Soybean (Bazaz) Powder during Storage (초고압처리에 의한 연해주 대두분말의 저장기한 중의 산패도 변화)

  • Changes of rancidity of soybean powder from Yeonhaeju (Bazaz) during storage were evaluated by TBA, and compared with those of Korean soybean (Baektae). Lipoxygenase (LOX) in the soybean powder was inactivated by high pressure processing (HPP) to increase the stability of soybean powder during storage. The level of inactivation of LOX was measured by spectrophotometer at 234 nm. HPP decreased the activity of LOX in the soybean powder of Baektae, compared to that of the control (i.e., soybean powder without HPP treatment) of Baektae, while TBA values of both HPP treated Baektae and the control were increased up to 24 days of storage. However, in case of Bazas, both LOX activity TBA values decreased after HPP treatment, compared to those of controls. The antioxidant compounds in both soybeans were measured and quantitatively evaluated by on-line $ABTS^{+}$ assay. Based on the trolox equivalent (TE) value at the retention time 38.2 and 40.1 min, the antioxidant components in Bazaz were higher than that of Baektae. It might indicate that relatively lower TBA values of HPP treated Bazaz was due to lower LOX activity as well as higher antioxidant compounds in the species.

Comparison of Immuno-Modulatory Regulatory Activities of Rubus coreanus Miquel by Ultra High Pressure Extracts Process (초고압 공정에 의한 복분자의 면역조절효능 비교)

  • This study was performed to investigate the enhancement of anticancer activities and immuno modulatary activities from R. coreanus. by ultra high pressure extracts process. The cytotoxicity on human kidney cell (HEK293) was showed below 19.5% in adding 1.0 $mg/m{\ell}$ concentration. The anticancer activity was increased over 10% by high pressure processing in AGS and A549 cells. The immune cell growth using human immune B and T cells was improved by the high pressure extracts of Rubus coreanus in adding 1.0 $mg/m{\ell}$ concentration. The secretion of two kinds of cytokine, the IL-6 and $TNF-{\alpha}$ from human immune B and T cells were also enhanced in adding extracts by high pressure process of R. coreanus. The ultra high pressure extraction technique showed high efficiency in extracting of bioactive compound. The ultra high pressure technique could be used combined with other technique to improve the extracting rate and extracting efficiency.

Structural Changes and Inactivation of Saccharomyces cerevisiae in Grape Juice Induced by High Hydrostatic Pressure (초고압에 의한 포도주스의 Saccharomyces cerevisiae 구조적 변화와 사멸효과)

  • 포도는 전세계에서 널리 소비되는 과실로 포도 과피에 존재하는 천연색소인 flavonoid는 혈중 콜레스테롤 함량 저하, 항알러지성, 항암성, 항바이러스성, 항염성의 생리적 기능이 있다고 알려져 있다. 최근에 들어와 이들 과실주스 가공에 열처리를 최소화하는 살균방법으로 자연 그대로의 영양성분, 맛과 향기 개선을 위한 초고압 처리에 관한 연구가 폭넓게 이루어지고 있다. 본 연구는 주스에서 문제가 되고 있는 ethanolic spoilage 균주인 S. cerevisiae의 초고압 살균 효과와 세포 구조적 형태를 연구하였다. 1.2$\times$$10^{6}$ cfu/ml의 S. cerevisiae를 포도주스에 접종하고 24시간 배양하여 멸균한 high barrier주머니에 20m1씩 넣고 2$0^{\circ}C$ 에서 200-600 MPa 조건으로 0-20분 동안 초고압 장치로 실시하였다. 생균수는 YM agar로 poured 방법으로 실시하였으며 200 MPa에서 5, 10, 15, 20분 후의 생균수는 각각 2.2$\times$$10^{7}$ , 4.5$\times$$10^4$, 2.8$\times$$10^4$, 9.8$\times$$10^3$, 9.5$\times$$10^3$cfu/ml로 tailing 현상을 관찰하였고, 400 MPa에서 5분 후 급격하게 감소하였다. S. cerevisiae의 사멸속도는 초고압 처리가 높을수록 증가했으며 세포 손상도는 압력과 처리시간이 길수록 증가하였다. 이들 조건에 따른 효모 세포의 구조적 관찰을 scanning electron microscopy와 electron microscopy로 하였다. S. cerevisiae 세포는 압력에 의한 pinhole, surface roughening을 발견하였고, 세포 내부의 세포질, 액포, 핵 손상과 세포질 물질들이 압력에 의하여 세포벽으로 이동하여 내부가 비어있는 현상을 관찰하였다.

Textural Characteristics and Bacterial Reduction in Glutinous Rice and Brown Rice Pretreated with Ultra-High Pressure (초고압 전처리한 찹쌀과 현미의 텍스처 특성과 세균 저감 효과)

  • Ultra-high pressure (UHP) was applied to glutinous rice (Dongjin) and brown rice (Shindongjin) to determine its effects on the textural characteristics of cooked rice and bacterial reduction in uncooked rice. Hardness and stickiness of the cooked glutinous and brown rice pretreated with 500 MPa for 5 min were significantly attenuated compared to that of the control rice (p<0.05). After the UHP treatment, particles of the glutinous and brown rice were observed to be finer when cross sections of the rice were analyzed using field emission scanning electron microscope. The total bacterial count in the brown rice treated with 600 MPa for 5 min was reduced from $10^7CFU/g$ to $10^4CFU/g$, while the number of thermoduric bacteria in the brown rice was not significantly different after the UHP treatment. These results suggest that UHP may be an effective method for improving texture of dried rice and to reduce bacterial count in raw rice.

Quality Changes during Storage of Kochujang Treated with Heat and High Hydrostatic Pressure (열과 초고압으로 처리한 고추장의 저장 중 품질 변화)

  • Effects of high pressure and thermal pasteurization on the survival of microorganisms and quality changes of kochujang during 120 days of storage at 37$^{\circ}C$ were investigated. Viable cell counts were 1.43$\times$10$^{6}$ CFU/g in heat-treated, and 1.56$\times$10$^3$ CFU/g in pressure-treated, and decreased up to 3 log cycle, compared with 3.78$\times$10$^{6}$ CFU/g in the untreated kochujang. Viable cell counts decreased by the storage period at 37$^{\circ}C$. Viable cell counts decreased up to 2 log cycle from 3.78$\times$10$^{6}$ to 5.43$\times$10$^4$ CFU/g in the untreated kochujang, 4 log cycle from 1.43$\times$10$^{6}$ to 3.10$\times$10$^2$ CFU/g in heat-treated after 120 days of storage, while those in pressure-treated were not detected after 90 days from the initial stage of 1.56$\times$10$^3$ CFU/g. pH decreased significantly by the storage time. Titratable acidity increased significantly during storage, and pressure-treated kochujang showed lower values than heat-treated. Amino nitrogen content decreased significantly during storage, and pressure-treated kochujang showed higher values than heat-treated and lower values than the untreated. There were no significant changes in reducing sugar and ethanol content regardless of the treatment condition and the storage period. Hunter L, a and b values decreased significantly during storage. In the untreated kochujang, the changes in color accelerated compared with heat and pressure-treated.

Changes of SDS-PAGE Pattern and Allergenicity of BSA and BGG in Beef Extract Treated with Heat and High Pressure (물리적 처리에 따른 우육추출물중의 BSA와 BGG단백질의 SDS-PAGE패턴 및 항원성의 변화)

  • In our previous report, we indicated that not only BSA but also BGG played an important role in the allergenicity of beef. In this study, the effect of heat or high-pressure treatments to beef extract on the SDS-PAGE patterns was examined. The antigenicity of each treated samples was also investigated by Western blots assay with the sera of BGG-positive beef allergic patients. The BGG band and its antigenicity slightly disappeared but not generally in $100^{\circ}C$ group, indicating $100^{\circ}C$ treatment is not sufficient to totally eliminate the antigenicity of beef allergens. Compared with BGG band, BSA band significantly disappeared in SDS-PAGE with $100^{\circ}C$ treatment, indicating BSA is more heat- sensitive than BGG. When the beef extract was heated at $120^{\circ}C$, not only BSA but also BGG bands was largely disappeared in both SDS-PAGE and Western blots. High pressure (HP) treatment even at 600 MPa did not affect SDS-PAGE and Western blots pattern of BSA. On the contrary, BGG treated with HP showed visible changes in SDS-PAGE. 600 MPa treatment significantly reduced the antigencity. Interestingly, these behaviors of BGG were not found in the same experiments with pure BGG treated with HP. From these results, it was speculated that some kinds of proteolytic enzymes in beef extracts were involved in the BGG molecular degradation by HP treatment. The aging experiments of beef extracts treated with HP supported this hypothesis. Further studies are needed to clarify the function and working mechanism of enzymes associated with BGG degradation in beef extracts by HP treatment.

Changes in Quality of Low Salt Fermented Anchovy by High Hydrostatic Pressure Treatment (초고압처리에 의한 저염 멸치젓의 품질 변화)

  • Effects of ultra-high pressure treatment on viable cell count and quality characteristics of low salt fermented anchovy under different operating conditions such as pressure$(200{\sim}500\;MPa)$, temperature$(20{\sim}50^{\circ}C)$ and treatment time$(5{\sim}20\;min)$ were investigated. Viable cell count decreased gradually with the increase of pressure and suddenly at 400 MPa. It also decreased by seven folds at $50^{\circ}C$ and logarithmically with the increase of treatment time. Peroxide value increased with the increase of pressure, temperature and treatment time, and temperature played a major role. Thiobarbituric acid value was higher by two folds in samples treated than in the untreated regardless of any conditions investigated. Volatile basic nitrogen was almost the same in all samples except the one at $50^{\circ}C$. The sample treated at greater than $30^{\circ}C$ under high hydrostatic pressure indicated higher value in amino nitrogen. Treatment at $20^{\circ}C/300$ MPa/15 min showed greater reductions in viable cell counts, remaining better quality of low salt fermented anchovy.
