Changes in Serum LH, FSH, Prolactin, Testosterone, GOT and GPT Concentrations in the Growing Male Pig (성장중(成長中)인 수퇘지에 있어서 혈청중(血淸中) LH, FSH, Prolactin, Testosterone, GOT 및 GPT의 농도변화(濃度變化))

  • The present study was carried out to invest igate on changing phases of the concentrations of serum LH, FSH, prolactin, testosterone, GOT and CPT in male pigs at various body weight. Six Duroc ${\times}$ Large White ${\times}$ Landrace boars were used to obtain serial blood samples at approximately 20kg body weight intervals from birth to 130kg body weight. The results obtained in this study were as follows: 1. Serum LH concentrations ranged between 2.89 and 4.25 mIU/ml from birth to 130kg body weight. FSH concentrations were below 1.25 mIU/ml under the limit of detection of the assay. 2. Serum prolactin concentrations were 8.05ng/ml at birth, between 3.07 and 4.25ng/ml from 15 to 50kg body weight, between 7.30 and 6.89 ng/ml from 70 to 90kg body weight and then declined to 4.10 ng/ml at 110kg body weight. 3. Serum testosterone concentrations were 0.42 ng/ml at birth, between 0.11 and 0.09 ng/ml from 15to 30kg body weight, and then increased to 1.15ng/ml at 50kg body weight and remained fairly constant thereafter. 4. Serum GOT concentrations were highest (130.75 karmen units/ml) at birth, and rapidly declined between 58.56 and 65.25 karmen units/ml from 15 to 70kg body weight and then increased 83.60 karmen units/ml at 90kg body weight. 5. Serum GOT concentrations were highest (63.25 karmen ullits/ml) at birth and rapidly declined between 27.25 and 30.75 karmen units/ml from 15 to 70kg body weight and then increased to 41.62 karmen units/ml at 90kg body weight.

Effect of Di(2-ethyl hexyl)phthalate(DEHP) on the Onset of Puberty in Female Rat (암컷 흰쥐의 사춘기 개시에 미치는 di(2-ethyl hexyl)phthalate(DEHP)의 효과)

  • Phthalates such as di(2-ethyl hexyl)phthalate(DEHP) are industrial chemicals with wide-ranging human exposures because of their use in plastics and other common consumer products. Consequently, their adverse effects as endocrine disruptor in the reproductive physiology of both laboratory rodents and human have been studied extensively. The present study was undertaken to examine whether prepubertal exposure to DEHP affects on the onset of puberty and the associated reproductive parameters such as hormone receptor expressions in female rats. DEHP(100mg/kg/day) was administered daily from postnatal day 25(PND 25) through the day when the first vaginal opening(VO) was observed, and the animals were sacrificed on the next day. Gross anatomy and weight of reproductive tissues were compared to test the DEHP's effects on the cell proliferation. Furthermore, histological studies were performed to assess the structural alterations in the tissues. Specific radioimmunoassay was carried out to measure serum LH levels. To determine the transcriptional changes in progesterone receptor(PR), total RNAs were extracted and applied to the semi-quantitative reverse transcription polymerase chain reaction(RT-PCR). As a result, delayed VO was shown in the DEHP group(PND $37.3{\pm}0.7$) compared to the control group(PND $35.3{\pm}0.7$; p<0.05). DEHP treatment significantly decreased the wet weight of ovaries and uteri compared to the control group(p<0.05). Interestingly, elevation of serum LH levels was shown in the DEHP group(p<0.05). Graafian follicles and corpora lutea were observed only in the ovaries from the control animals. Numerous primary, secondary follicles and small atretic follicles were observed in the ovaries from DEHP-treated animals. Similarly, hypotrophy of luminal and glandular uterine epithelium was found in the DEHP-treated group. These effects were probably due to the inhibitory effects of DEHP on the synthesis and secretion of estrogen from granulosa cells. In the semiquantitative RT-PCR studies, the transcriptional activities of PR in both ovary(p<0.05) and uterus(p<0.01) from DEHP-treated animals were significantly lower than those from the control animals. The present studies demonstrated that the acute exposure to DEHP during the critical period of prepubertal stage could inactivate the reproductive system resulting delayed puberty in female rats.

Effect of Feeding Lactobacillus reuteri to Broiler on Growing Performance, Intestinal Microflora and Environmental Factor (lactobacillus reuteri의 급여가 육계의 성장 특성, 장내미생물 변화, 혈청 성상 및 사육환경에 미치는 영향)

  • A feeding trial was carried out to evaluate the effects of feeding Lactobacillus reuteri culture(LRC) on the performance, nutrients digestibility, intestinal microflora, serum metabolites, ammonia generation and litter dampness in broiler chicks. Four hundred eighty, one day old male chicks were fed into none, 0.1, 0.2 and 0.4% of LRC supplementation fer seven weeks. Basal diets excluded antibiotics contained ME 3,100, 3,100 kcal/kg, and CP 22.0, 20.0% for starter and grower, respectively. Weight gain of chicks fed LRC was significantly higher than no supplemental group in overall period(P<0.05). Feed intake was the highest in the 0.1% LRC, but not statistically different from other treatments. Feed conversion showed no significance among treatments. Viable Lactobacillus spp. number of chicks fed 0.2 and 0.4% LRC was significantly higher in cecum at seven weeks of age compared to the none(P<0.05). The tendency of anaerobes number was similar to Lactobacillus spp in ileum and cecum. Total number of E. coli and Salmonella were no difference in all treatments. In serum metabolites, feeding LRC increased triglyceride, and inorganic phosphorus, but no different total protein, albumin, total cholesterol, glucose, blood urea nitrogen and Ca. Nutrients digestibility improved significantly in 0.4% LRC compared to that of none(P<0.05). Fecal NH$_3$, gas generation was greatly decreased in the LRC supplemental groups(P<0.05). Moisture contents of bedding was also significantly decreased in LRC feeding group. It was concluded from the present study that feeding Lactobacillus reuteri culture improved the growth performance and nutrients digestibility of broiler chicks and minimize the fecal noxious gas emission.

Effect of Different Stocking Densities in Plastic Wired-Floor House on Performance and Uniformity of Korean Native Commercial Ducks (고상식 오리사에서 다양한 사육밀도가 토종 실용오리의 생산성 및 균일도에 미치는 영향)

  • This study investigated the effects of various stocking densities on the growth performance of Korean native ducks. Twelve hundred day-old ducklings were reared for 50 days in a duck house (windowless, plastic wire-floor pen). Two weeks later, 852 of these ducks (350±30 g) were selected for at least 80% uniformity per stocking treatment (six treatments, four replications/treatment, depending on a stocking density of 4~9 birds m-2). Experimental diets were corn-soybean-based and broiler ducks were grouped based on age [0~21 days of age (CP 21%, ME 2,900 kcal/kg) and 21~50 days of age (CP 17%, ME 3,100 kcal/kg)]. Body weight (BW) did not significantly differ among different aged treatment groups (P>0.05); however, uniformity decreased with higher stocking densities (P<0.05). Feed intake and feed conversion ratio of ducks aged 42~50 days significantly decreased in the T4-6 treatments compared with the T1-3 treatments (P<0.05). In all experimental periods, BW gain was not significantly different among treatments, but feed intake decreased significantly as the stocking density increased (P<0.05), and feed conversion ratio was significantly different among six treatments with stocking density (P<0.05). In conclusion, we found that the stocking density of Korean native ducks in a plastic wire-floor pen was approximately 7 birds m-2 in consideration of productivity and uniformity.

Protective Effects of 5-Androstendiol (5-AED) on Radiation-induced Intestinal Injury (방사선에 의한 장점막 손상에 대한 5-Androstenediol의 보호효과)

  • Purpose: We examined the radioprotective effects of 5-androstendiol (5-AED), a natural hormone produced in the reticularis of the adrenal cortex, as a result of intestinal damage in gamma-irradiated C3H/HeN mice. Materials and Methods: Thirty mice (C3H/HeN) were divided into three groups; 1) non-irradiated control group, 2) irradiated group, and 3) 5-AED-treated group prior to irradiation. Next, 5-AED (50 mg/kg per body weight) was subcutaneously injected 24 hours before irradiation. The mice were whole-body irradiated with 10 Gy for the histological examination of jejunal crypt survival and the determination of the villus morphology including crypt depth, crypt size, number of villi, villus height, and length of basal lamina, as well as 5 Gy for the detection of apoptosis. Results: The 5-AED pre-treated group significantly increased the survival of the jejunal crypt, compared to irradiation controls (p<0.05 vs. irradiation controls at 3.5 days after 10 Gy). The evaluation of morphological changes revealed that the administration of 5-AED reduced the radiation-induced intestinal damages such as villus shortening and increased length of the basal lamina of enterocytes (p<0.05 vs irradiation controls on 3.5 day after 10 Gy, respectively). The administration of 5-AED decreased the radiation-induced apoptosis in the intestinal crypt, with no significant difference between the vehicle and 5-AED at 12 hours after 5 Gy. Conclusion: The results of this study suggest that the administration of 5-AED has a protective effect on intestinal damage induced by $\gamma$-irradiation. In turn, these results suggest that 5-AED could be a useful candidate for radioprotection against intestinal mucosal injury following irradiation.

Role of Group II Phospholipase $A_2$ in the Pulmonary Oxidative Stress of the Acute Lung Injury Induced by Gut Ischemia-Reperfusion (장의 허혈-재관류로 유도된 급성 폐손상에서 산화성 스트레스에 관여하는 group II phospholipase $A_2$의 역할)

  • Background: The various pathogeneses of acute respiratory distress syndrome have been suggested but not established yet. In the present study, the role of group II phospholipase $A_2$($PLA_2$) in the pathogenesis of gut ischemia-reperfusion(I/R) induced acute lung injury (ALI), especially in the pulmonary oxidative stress with infiltration of neutrophils was investigated. Material and Method: To induce ALI, reperfusion of mesentery was done for 120 min after clamping of superior mesenteric artery for 60 min in Sprague-Dawley rats that weighed about 300g. To exmaine the role of group II $PLA_2$ in ALI, especially endothelial injury associated with the action of neutrophils, lung myeloperoxidase activity, lung leak index, bronchoalveolar lavage fluid protein were measured, and pulmonary $PLA_2$ activity changes in gut I/R were also measured. The role of group II $PLA_2$in the neutrophilic generation of free radicals was assessed by inhibiting group II $PLA_2$ with rutin, manoalide and scalaradial. Furthermore, to verify the oxidative stress in the lung, histologic and free radical detecting cytochemical electron microscopy were done. Result: After reperfusion, ALI was developed with accumulation of neutrophils in the lung, which was confirmed by the increase of myeloperoxidase activity, lung leak index and bronchoalveolar lavage protein (p<0.001). The pulmonary and intestinal group II $PLA_2$ activities significantly increased after gut I/R which were reversed by rutin(p<0.001). In vitro, cytochrome-c reduction assay denoted the inhibitory effects of rutin, scalaradial and manoalide on the production of free radicals from isolated human neutrophils. Histologically, neutrophilic accumulation and pericapillary edema in the lung after gut I/R was detected by light microscopy which was suppressed by rutin. In $CeCl_3$ cytochemical electron microscopy, the increased production of hydrogen peroxide in the lung after gut I/R was confirmed and also the production of hydrogen peroxide was decreased by rutin. Conclusion: On the basis of these experimental results, the inhibition of group II $PLA_2$ seemed to mitigate gut I/R-induced ALI by suppressing the production of free radicals from the infiltrated neutrophils. Collectively, group II $PLA_2$ seems to play a crucial role in gut I/R-induced ALI by neutrophilic oxidative stress.

Effects of Copper and Zinc Supplementation on Growth Performance, Nutrient Digestibility, Meat and Carcass Characteristics in Finishing Pigs (구리 및 아연의 수준별 급여가 비육돈의 생산성, 영양소 소화율, 육질 및 도체 특성에 미치는 영향)

  • This study was conducted to investigate the effects of copper and zinc supplementation on growth performance, nutrient digestibility, and meat and carcass characteristics in finishing pigs. A total of 72 $(Landrace{\times}Yorkshire{\times}Duroc)$ pigs (58.47 kg initial BW) were assigned to 6 treatments in a $2{\times}3$ factorial design $(Zn\;levels{\times}Cu\;levels)$. The sources of zinc and copper were Zn-methionine chelate and Cu-methionine chelate, respectively. Zinc levels used were 80 and 120 ppm and copper levels used were 10, 30 and 60 ppm. Throughout the entire experimental period, the average daily gain (ADG) and average daily feed intake (ADFI) were not significantly affected by Cu or Zn levels, or their relative levels. The G:F ratio was significantly affected by the relative levels of Cu and Zn (p<0.05), specifically at 30 ppm Cu and 120 ppm Zn. Dry matter digestibility was significantly affected by the levels of Cu (p<0.02), Zn (p<0.01) and the relative levels of each (p<0.04), in particular at 30 ppm Cu and 120 ppm Zn. Nitrogen digestibility was significantly affected by Zn levels (p<0.01) and the combination of 30 ppm Cu and 120 ppm Zn (p<0.03). The $L^*-value$, shear force, cooking loss and pH were not significantly affected by Cu levels, Zn levels or their combination. The $a^*- (p<0.04)\;and\;b^*- values (p<0.01)$ were significantly affected by Zn levels at 80 ppm. The Water hoding capacity was significantly affected by Cu and Zn in combination (p<0.01) at 10 ppm Cu and 120ppm Zn. The carcass weight, backfat thickness and carcass grade were not significantly affected by Cu levels, Zn levels or their relative levels. The carcass percentage was significantly affected by the combination (p<0.04) of 30ppm Cu and 120ppm Zn. In conclusion, dietary supplementation of Cu and Zn at 30 and 120 ppm, respectively, is effective for feed efficiency, nutrient digestibility and carcass percentage, while at the levels of Cu at 10 ppm and Zn at 120 ppm have effects on WHC.

Effects of the Dosing Regimen of Tissue-type Plasminogen Activator on Blood Coagulation System in Experimental Pulmonary Embolism (실험적 폐색전증에서 조직형플라스미노겐활성체의 투여방법이 혈액응고기전에 미치는 영향)

  • Background: As a physiologic plasminogen activator, tissue-type plasminogen activator (t-PA) could induce effective thrombolysis in massive pulmonary embolism, without the risk of systemic hemorrhage. However, therapeutic doses of t-PA has been associated with systemic lytic state, and fibrin selectivity may be influenced by the dosing regimen of t-PA. To investigate the effects of duration of t-PA infusion on blood coagulation system, we performed this study. Method: In a canine model of pulmonary embolism, which was induced by injection of autologous blood clots, we administered equal doses of t-PA (1 mg/kg) over 15 minutes in $t-PA_{15}$ group, over 180 minutes in $t-PA_{180}$ group, and only saline in control group. Then serial blood samplings were made to check complete blood count, prothrombin time, activated partial thromboplastin time, thrombin time, fibrin, plasminogen, ${\alpha}_2$-antiplasmin, coagulation factor V and VIII, and fibrin(ogen) degradation products. Results: 1) In all 3 groups, complete blood count showed same changes. Hemoglobin, hematocrit and platelet count decreased, but WBC count increased. 2) Prothrombin time, activated partial thromboplastin time, and thrombin time were prolonged during 15-60 minutes after t-PA administration in $t-PA_{15}$ group, and from 30 minutes through 180 minutes after administration in $t-PA_{180}$ gorup. 3) Fibrin, ${\alpha}_2$-antiplasmin, and cogulation factor V and VIII decreased in both $t-PA_{15}$ and $t-PA_{180}$ group, but returned to basal levels earlier in $t-PA_{15}$ group. 4) Fibrin(ogen) degradation products increased after pulmonary embolism in all groups, and further increased in both $t-PA_{15}$ and $t-PA_{180}$ groups after t-PA infusion. But more pronounced increment was noted in $t-PA_{180}$ gorup. Conclusion: In pulmonary embolism, the shorter (15 minutes) infusion of t-PA would have less risk of systemic hemorrhage than the longer (180 minutes) infusion when the doses is equal. And, this suggests that manipulating the duration of t-PA infusion can reduce the risk of major bleeding.

Effects of YK-209 Mulberry Leaves on Disaccharidase Activites of Small Intestine and Blood Glucose-Lowering in Streptozotocin-Induced Diabetic Rats (YK-209뽕잎이 Streptozotocin 유발 당뇨쥐 소장의 이당류분해 효소 활성과 혈당강하에 미치는 영향)

  • The purpose of this study was investigated the effects of YK-209 mulberry leaves on disaccharidase activites of small intestine and blood glucose-lowering in diabetic rats induced with streptozotocin (STZ). Male Sprague-Dawley rats weighing 100$\pm$10 g were randomly assigned to one normal and four STZ-induced diabetic groups; YK-209 0% mulberry leaves diet (DM group),0.1% YK-209 mulberry loaves diet (DM-0.1Y group),0.2% YK-209mulberry leaves diet (DM -0.2Y group), and 0.4% YK-209 mulberry leaves diet (DM -0.4Y group). Diabetes was induced by intravenous injection of 55 mg/kg body weight of STZ in sodium citrate buffer (pH 4.3) via tail vein after 3 weeks feeding of experimental diets. Rats were sacrificed at the 9th day of diabetic states. The functional ingredients in the mulberry leaves, the 1-deoxynojirimycin (DNJ) contents of YK-209 mulberry leaves was higher than those of the Cheongil mulberry leaves. ${\gamma}$ -Aminobutyric acid (GABA) and rutin contents of YK-209 mulberry leaves were 1.3 and 1.4 times higher than those of the Cheongil mulberry leaves, respectively, and vitamin C contents of YK-209 mulberry leaves were also higher than those of the Cheongil mulberry leaves. Intestine index was increased in all diabetic groups, compared with normal group but not significantly different among all diadetic groups. Level of blood glucose was decreased in diadetic rats by supplementation YK-209 mulberry leaves. The disaccharidase activities in proximal part of intestine such as maltase, sucrase, and lactase in YK-209 mulberry leaves supplementation groups were significantly lower than those of DM group, In conclusion, this research indicated that the functional ingredients of YK 209 mulberry leaves were higher than those of the Cheongil leaveses, and YK-209 mulberry leaves has the hypoglycemic effect in STZ-induced diabetic rats.

Effect of Iron Supplementation on Mineral Utilization in Rats (철 보충이 흰쥐의 무기질 이용에 미치는 영향)

  • The purpose of this study was to investigate the effect of iron supplementation on utilizations of various minerals, Ca, Mg, Na, K, Fe, Cu, Zn. Intakes, serum levels, and excretions of these minerals were analyzed and compared in rats fed diet with 100, 200, 400% of iron requirement (Fe, 2Fe, 4Fe groups) for 6 weeks. The feed intake, body weight gain and feed efficiency ratio were not significantly different among the groups. Serum Fe of 2Fe group was significantly lower than other two groups, and serum Zn of 4Fe group was highest. Mineral intakes were not significantly different among three groups. With iron supplementation, urinary excretions of Ca, Fe, Cu, Zn were elevated, but fecal excretions of minerals were not significantly different. And the daily retentions of Na and Fe were significantly elevated, but apparent absorbabilities of minerals were not significantly different. In summary, according to iron supplementation, mineral absorbabilities were not different but urinary excretions of some minerals were increased. Therefore, it could be suggested that adequate iron intake and well-balanced diet are more desirable than nutrient supplementation for mineral balance.
