Vie investigated the clarification effects of foxtail millet wine, a traditional wine in Cheju island, by centrifugation, protease treatment and ultrafiltration(UF). It was difficult to remove completely cloudy substances in foxtail millet wine only by centrifugation. With protease concentration of $5{\times}10^{-4}%(w/v)\;at\;35^{\circ}C$, for 2 hrs, foxtail millet wine was clarified effectively. Papain and bromelain appeared similar effects to the clarification of wine, but ficin was inferior to those. Ultrafiltration with pore size $100K\;and\;0.22{\mu}m$ membranes had appeared better clarification effects than the best result of enzyme treatment, and then was considered as a simple and economic method.

The effects of chitosans with two different molecular weights (150 and 37 kDa) on the clarification of persimmon vinegar were investigated. The coagulated solids increased with increasing chitosan concentrations. Chitosan treatment had no effect on pH and acidity. Turbidity, browning, contents of tannin and soluble solids decreased with increasing chitosan concentrations. An increase in chitosan concentration resulted in an increase in color L* value and a decrease in a* and b* values. Reduction in turbidity and tannin contents somewhat differed depending on the molecular weights of chitosan. In sensory evaluation, chitosan treatment did not affect color and flavor over all concentrations tested. However, astringent taste was noticeably strong by treatment with 500mg/L chitosan, resulting in low overall acceptability. Therefore, the most effective clarification of persimmon vinegar was achieved by treatment with 400mg/L chitosan, irrespective of molecular weight.

Persimmon vinegar was clarified with 400mg/L chitosans of two different molecular weights (MW 150 and 37 kDa), and its quality changes were investigated during storage at room temperature for 6 months. No significant changes in pH, acidity, and tannin content were observed. However, turbidity and browning slightly increased while protein content slightly decreased. Soluble solids content slightly decreased when treated with high MW chitosan. Color L* value decreased while a* and b* values increased with storage periods. The major organic acid in the persimmon vinegar after 6 months storage was acetic acid with minor lactic, malic, tartaric, galacturonic, and succinic acids. Overall, the quality of persimmon vinegar clarified by chitosan treatment, irrespective of molecular weight, was more stable without noticeable changes during storage than that of control group without chitosan treatment.
Among the strains isolated form the various sources, the strain AC-12 producing a powerful pectinase was selected by the extensive screening test. The selected strain was indentified and its toxicity investigated. The conditions of the pectinase production, the characteristics of the purified enzyme and the clarification effect on the apple juice were studied. 1. The selected strain AC-12 was identified by the classification method of paper and fennel and named as Aspergillus sp. AC-12. 2. As a result of the breeding test of the white mouse, no toxicity was found from this enzyme. 3. The yield of pectinase in the medium of defatted rice bran was much better than that in the medium of wheat bran. 4. The optimum conditions for the culture of the strain in the medium of defatted rice bran were that the cultural time was 72hrs, the amount of water to be added about 80%, temperature $30{\sim}35^{\circ}C$ and pH $3.0{\sim}5.0$. 5. The yield of pectinase was slightly increased by the addition of pectin to the medium of defatted rice bran and by the addition of pectin, $NaNO_3$ and $K_2HPO_4$ to the medium of wheat bran, respectively. 6. The optimum conditions for the enzyme activity were pH $3.0{\sim}4.0$ and temperature $40{\sim}50^{\circ}C$. The enzyme was stable below $40^{\circ}C$ and pH $2.0{\sim}8.0$, respectively. But above $50^{\circ}C,$ this enzyme was abruptly inactivated. The activity was slightly increased by the addition of $MnSO_4\;and\;CuSO_4.$ 7. It was regarded that the opimum temperature for the clarification of the apple juice was $40{\sim}50^{\circ}C$, the optimum pH 3.0 and the optimun concentration of the enzyme 0.1%, and the apple juice was almost clarified by the reaction at $45^{\circ}C$ for 60 minutes.
These experiments were carried out to obtain useful yeast for Korean grape wine brewing. The 213 strains of yeasts were isolated from grapes sampled in Daejeon city and suburbs and then 3 strains among the isolated yeasts were selected as useful strains and identified. The results obtained as follows; 1. The 3 strains of W-49, W-50 and W-127 selected as useful yeasts were clarified to grape wine mash rapidly. 2. In the mashes clarified after storage of 45 days, the value of optical density of the case of W-50 strain was highest and the color was very thick, compared with the case of another strains. 3. In the pannel test of the grape wines, the case of W-49 and W-50 strains were best, the case of the W-127 strain and control yeast, Hb were similar each other as next order. 4. Among 3 strains selected as useful yeasts it were identified that the strains of W-49, W-50 selected as useful strains were Saccharomyces cerevisiae and the strain of W-127 was Saccharinyces pretoriencis by the Ladder manual. 5. All of these useful yeasts were appeared red color on TT C stain.

This study was conducted to evaluate the effect of ultrafiltration (UF) process variables on permeate flux and membrane resistance and to clarify apple vinegar for quality improvement. Apple vinegar was clarified in a laboratory ultrafiltration system with hollow fiber membrane made of polysulfone and MWCO 30,000 and 10,000. The permeate flux increased with the increase of flow rate and the optimum pressure was $1.5\;kgf/cm^2$ in this system. The turbidity of clarified apple vinegar treated UF largely decreased. pH and acidity of treated samples showed the same level as those of untreated apple vinegar. The permeate flux continuously declined while the fouling material accumulated on the membrane as the operation time increased. Resistance decreased with lower pressure, which could be explained by expansion of pore size at lower pressure and minor compaction of the polarized layer at lower pressure.
The effect of gelatin concentration, pH of fruit juice and other factors for the enzymatic clarification of fruit juice by the use of polygalacturonase was studied. The proper addition of gelatin was much effected on clarification by polygalacturonase and optimum concentrations of gelatin for the enzymatic clarification of Zugaru juice, Magnolia Gold juice, Golden Delicious juice, Jonagold juice, Jonathan juice, Campbell Early juice were 0.04, 0.01% 0.02%, 0.06% 0.01%, and 0.04%, respectively. Optimum pH for the clarification by the use of polygalacturonase and gelatin was $3.2{\sim}3.5$ contrary to optimum pH 4.8 of polygalacturonase for the hydrolysis of pectic acid. At the reaction temperature of $45^{\circ}C$ and polygalacturonase concentration of 0.01mg/ml, fruit juices were completely clarified by optimum addition of gelatin for 60min.

Optimum extraction condition and clarification process were investigated to manufacture liquid extract tea using Phellinus linteus. Carbohydrates content in the raw Phellinus linteus was 80.9%. Major minerals were K, Ca, Fe and Na, but vitamin B$_1$, B$_2$, and C were not detected. The best extraction condition was 5% raw material at 10$0^{\circ}C$ within 3 hr. To clarify the extract, three methods of filtration with depth filter pad, centrifugation and addition of several filter aids were studied. Filtration with depth filter pad and centrifugation were highly effective on the clarification and the changes in the turbidity for 7 weeks at 4$0^{\circ}C$ were not occurred. The optimum clarification condition was centrifugation above 6,000 rpm or filtration with 8 ${\mu}{\textrm}{m}$ depth filter pad.

This study was carried out to observe the change of oriental melon juice quality during manufacture by heating condition and enzyme treatment. To use over-production oriental melon effectively, oriental melon juice was prepared with change of heating temperature and heating time. The heating conditions for minimum filtration time were 98.57$^{\circ}C$ and 11.29 min. The optimum conditions Predicted for separation ratio of clear solution, filtration time and browning color intensity of oriental melon juice were 98$^{\circ}C$, 13 min. In the pectin-degrading enzyme treatment test, turbidity showed minimum value in 4.40 mg%(pectin-degrading enzyme content), 39.72 mg% (gelatin content) and 24.09 min (treatment time).Browning color intensity showed minimum value in 9.33 mg% (pectin-degrading enzyme content),44.70 mg% (gelatin content) and 115.56 min (treatment time). The optimum conditions predicted for turbidity and browning color intensity of oriental melon concentration Juice were 6.6 mg% (pectin-degrading enzyme content), 33.6 mg% (gelatin content) and 70 min (treatment time).
Penicillium sp. CB-20 was selected for strong polygalacturonase activity among various strains of molds found in soil. The optimum pH for the enzyme activity was 5.0 and optimum temperature was 4$0^{\circ}C$. The enzyme was relatively stable in acidic condition and unstable by heat treatment. The activation energy, Km and V$_{max}$ for the polygalacturonase were 2.499 Kcal/mol, 2.13$\times$10$^{-2}$mol/l, and 104.17 $\mu$mol/min. The activity of polygalacturonase was inhibited by Ag$^{+}$, Cu$^{++}$, Pb$^{++}$, Fe$^{+++}$, $Ca^{++}$, Na$^+$, Mn$^{++}$. The enzyme can be inactivated by the treatment ethylenediamintetra acetic acid, 2,4-dinitrophenol and $H_2O$$_2$. The results indicate the possible involvement of histidine, chelate and terminal amino group as active site. The enzyme was endo-type polygalacturonase.
