Effects of Dietary Fe and Se Levels on Lipid Levels in Serum and Liver of Rats (철분과 셀레늄의 섭취수준이 흰쥐의 혈액과 간장의 지질수준에 미치는 영향)

  • The purpose of this study was to investigate the effect of dietary Fe and Se levels on lipid contents of serum and liver in growing rats fed adequate Fe(35ppm), medium-high Fe(70ppm), high Fe(140ppm), adequate Se(0.05ppm), and high Se(0.Sppm) for 6 weeks. Serum levels of Fe and Se increased as the levels of dietary Fe and Se were increased, respectively(P<0.05, p<0.05), Serum levels of total cholesterol and (LDL+VLDL)-cholesterol increased as the level of dietary Fe was increased (p<0.001, p<0.001), and were lower in high-Se group than those in adequate-Se group(p<0.001, P<0.001). Liver Fe level in High-Se group was lower than that in adequate-Se group(p<0.01) and liver Se level decreased as the dietary Fe level was increased(p<0.05). Liver levels of phospholipid and total lipid in high-Se group were lower than that in adequate-Se group(p<0.05 p<0.01). Liver levels of triglyceride and total lipid decreased as the level of dietary Fe was increased(p<0.05 p<0.01). From these results, it is considered that serum lipid contents are increased in case of Fe overload and decreased with adding Se. Therefore, it could be suggested that adequate Fe intake and Se addition are recommended for prevention of lipid increment in serum.

  • This study was conducted to investigate the effect of Zn and Fe levels on lipid metabolism and micromineral contents in high fat diet-induced obese rats. Male sprague-dawley rats weighing approximately 100g were fed a normal or high fat diet. After 10 weeks high fat diet, rats were fed different levels of Zn and Fe. Serum triacyglycerol and HDL-cholesterol were affected to Zn and Fe levels in normal group. The micromineral (Zn, Fe, Cu, Mn) contents in liver were significantly different in high fat diet group by Zn intake levels and in normal diet group by Fe intake levels. However, micromineral contents in kidneys were not affected to different levels of Fe in both obese and normal rats. There were no differences in spleens of obese rats fed high fat diet according to Zn and Fe intake levels. But in normal group, Mn content of spleen were exclusively affected to Zn levels.

  • 철분투여가 용혈성 빈혈에 미치는 영향을 알아보기 위하여 흰쥐의 사료내에 철분을 혼합급여 또는 근육내에 주사하면서 60일간 사육하고 체중은 10일 간격으로 측정하였으며 적혈구의 취약성 실험과 적혈구막 및 혈장내의 vitamin E 함량과 인지질의 함량을 비색방법으로 측정한 결과 다음과 같은 성적을 얻었다. 증체량 측정에서는 A군의 190g에 비하여 C군 155g, B군 142g, D군 112g으로 철분함량이 높은 군에서 낮은 증가를 보였으며 적혈구 취약성 실험 결과 5마리 암수 평균값이 각각 A군 7.63% 및 2.68%, C군 10.84% 및 16.75%, B군 25.54% 및 33.62%, D군 47.23% 및 38.35%로 철분함량에 비례하여 각 실험군 사이에 유의한 증가를 나타내었다. 또한 적혈구막의 vitamin E 및 인지질의 함량은 5마리 암수 평균값이 각각 A군에서 18.54mg/dl 및 17.43mg/dl 그리고 120.55mg/dl 및 113.94mg/dl, C군에서 14.99mg/dl 및 10.56mg/dl 그리고 108.54mg/dl 및 99.97mg/dl, B군에서 13.10mg/dl 및 11.02mg/dl 그리고 91.67mg/dl 및 87.45mg/dl, D군에서 6.86mg/dl 및 8.87mg/dl 그리고 79.78mg/dl 및 70.12mg/dl로서 철분 투여량에 비례적으로 감소하였다. 혈장내의 vitamin E 및 인지질의 함량은 A군, C군, B군, D군의 순으로 각각 함량이 2.51mg/dl 및 31mg/dl, 1.94mg/dl 및 21mg/dl, 1.37mg/dl 및 20mg/dl 그리고 1.37mg/dl 및 15mg/dl로 철분 투여량에 비례적으로 감소하는 경향을 나타내었다.

  • This study was carried out to investigate effects of iron content on the quality of soy sauce, bean paste and red pepper paste, and to elucidate the origin of iron and change of the contents during production processes. For the first step, the iron contents in commercial soy sauce and changes of the contents during brewing process were determined. The results obtained were as follows. 1, Iron contents of raw materials were 108 ppm in soy bean, 133ppm in defatted soy bean, 79 ppm in wheat, 5 ppm in sodium chloride, 58 ppm in seed koji, 300-2000 ppm in spore of Aspergillus oryzae, 240 ppm in wheat gluten, 20 ppm in sodium carbonate (above figures were of dry weight basis), 6 ppm in hydrochloric acid, 18 ppm in caramel and 0.3ppm in brewing water respectively. 2, Iron contents in koji were 200-240 ppm (as dry weight basis) and increased, more or less, in progress of koji-making period. 3. Iron contents in the mashes during fermentation were 40 rpm after 1 month, 43-47 ppm after 3 months and 49-62ppm after 6 months. 4. In chemical soy sauce, the iron content was 159 ppm after hydrolysis of wheat gluten with hydrochloric acid, and 184 ppm after neutralization. 5. Higher iron contents were detected both in fermented and chemical soy sauce when the concentration of total nitrogen increased, but the levels were higher in chemical soy sauce than in fermented one at the same concentration of total nitrogen. 6. In the case of fermented soy sauce, the iron content in the filtrate was decreased by press-filtration, but no significant change was found between before and after heat-sterilization. 7. Iron contents in commercial soy sauce were varied with the producers, however, the average value was 62.7 ppm as calculated as 1.0 percent of total nitrogen. And the average level of iron in home-made soy sauce produced by conventional method was 37.68 ppm.

Effect of Dietary Calcium Levels on Iron Utilization in Female Rat (칼슘의 섭취수준이 암쥐의 체내 철분이용에 미치는 영향)

  • The effect of dietary calcium levels, 50%, 100%, and 200% of requirement, on iron utilization was evaluated in 30 Sprague-Dawley female rats by use of balance study for 3 weeks. In the results of this study, there were no significant difference in feed intake, body weight gain, hemoglobin level, hematocrit, calcium and iron levels in serum and tissues across the groups supplemented different calcium levels. Calcium content in kidney of high-calcium group was significantly higher than that of other groups. Urinary and fecal calcium excretions increased as the level of dietary calcium was increased. With increasing levels of dietary calcium, daily calcium retention was accelerated, but daily calcium retention rate was diminished. Iron intake was significantly higher in adequate-calcium group than that in low-calcium or high-calcium group. Urinary and fecal iron excretions were significantly lower in low-calcium group than those in adequate-calcium or high-calcium group. Apparent retention and retention rate estimated by intake, urinary and fecal excretions of iron were significantly lower in high-calcium group t]lan those in low-calcium or adequate-calcium group. These results suggest that taking dietary calcium supplements reduce the absorption of dietary iron.

Effects of Feeding Ferritin Gene Transferred Yeast (Saccharomyces serevisiae) on Performance, Iron Concentration in Organs and Egg of Chickens (Ferritin 유전자 전이 효모(Saccharomyces serevisiae)의 급여가 닭의 생산성, 장기 및 계란의 철분함량에 미치는 영향)

  • Three experiments were conducted to investigate the effect of feeding yeast accumulated transgenic ferritin(FRT, Saccharomyces cerevisiae) as a probiotic on the performance, iron contents in the liver, spleen, bone and yolk of laying hens and broiler chicks. Effects of feeding FRT were compared with that of feeding wild-type yeast(W0) and yeast grown on 20 mM ferric citrate-added medium (W20). In Expt 1, to investigate the effect of feeding yeast (control, W0 FRT) on performance and iron content of organs of broiler chicks which were fed basal diet supplemented with 75mg/kg iron(Fe75) or not (Fe0), three hundred sixty one-day-old male broiler chicks were fed a corn-sov based diet for five weeks. Weight gain, feed intake and feed conversion were measured weekly. In Expt 2, fifteen 33-week-old ISA Brown laying hens were placed in individual cages and were fed control, W0 and FRT diets for Four weeks. In Expt 3, twenty four 45-week-old ISA Brown laying hens were placed in individual cages and were fed a basal diet for a week. Then, experimental diets (control, W0, W20, FRT) were fed for three weeks. Iron contents in the liver, heart, spleen and tibia were determined at the end of all experiments. Iron content in yolk was measured weekly (expt 2, 3). The level of yeast added and iron concentration of FRT were $1{\times}10^8$cfu/kg diet and 500 mg/kg cell (DM) respectively in Expt 3, yeast was supplemented at $2{\times}10^{10}$cfu/kg diet and the iron content of FRT was 1000mg/kg cell (DM). In Expt 1. birds fed Fe75 showed significantly higher weight gain compared with Fe0 (P<0.05). However, weight gain and feed intake of birds fed FRT was significantly lower than control (P<0.05). In Expt 2, the iron content of the liver was decreased in the FRT treatment (P<0.05). In Expt 3, iron concentration of the liver and spleen tended to be increased by feeding FRt. However, the iron content of the tibia tended to be decreased in the FRT treatment. These results suggest that feeding FRT as a probiotic cannot improve performance and iron content in organs of broiler chicks and laying hens.

Characteristics of iron powder formulation produced from porcine blood by enzymatic treatment (효소 처리한 돈혈 활용 철분분말제제 특성)

  • In this study, enzyme (thermoase) hydrolysis was applied to the porcine blood order to increase the iron content and solubility. It was confirmed that content of iron was increase up to 158.11 mg/100 g porcine powders after 0.2% thermoase treatment at $60^{\circ}C$ during 4 hr. The solubility of porcine blood powders was higher than other enzyme (various protease), temperature, reaction time. This optimized conditions were also worked to the in vitro iron bioavailability rate increasement, the bioavailability of hydolyzed porcine powders was 3-fold higher than that of an iron supplement on the market. These results indicate the possibility of porcine blood powder in iron supplements market as natural material. Also utilizing of reduced porcine blood will be possible to improve environmental issues.

A Refining of Natural Diatomite and Synthesis of SiC Powder (규조토 정제 및 탄화규소 분말합성)

  • For high value-added applications of natural blue diatomite, the physical refining process and synthesis of SiC from refined diatomite were investigated. Approximately 30 percent Fe ($Fe_2O_3$) in raw blue diatomite was removed by a particle sieve separation process; the Fe composition for 325 mesh down powder was approximately 2 percent. Although a wet and/or dry magnetic separation process had some influence on the separation and/or refining of Fe composition, the Fe composition in the non-magnetic by-product was approximately 2 percent. Water leaching separation was effective in removing the Fe composition; approximately 40 percent of the Fe in raw blue diatomite was removed. The synthesis of ${\beta}$-SiC by a carbothermal reduction of the $SiO_2$ in the refined diatomite using carbon (graphite, carbon black), the effects of an acid-treatment on removing the Fe, and the specific surface area for the synthesized powder were also investigated. The impurities were mostly eliminated and the specific surface area was increased to $52.5m^2/g$.

Manufacturing of Iron Binding Peptide Using Sericin Hydrolysate and Its Bioavailability in Iron Deficient Rat (실크 세리신 단백질을 이용한 유기 철분제의 제조 및 철분 결핍쥐에서의 생물학적 유용성)

  • Silk sericin protein was hydrolyzed by seven proteolytic enzymes to examine the effectiveness of the hydrolysates to bind iron. The amino acid nitrogen contents of hydrolysates by Flavourzyme were higher than the others enzymes, and its iron binding capacity showed dose-dependent increase. The bioavailability of iron binding peptide from sericin hydolysates was investigated in iron-deficient rats. Three-week-old male rats were fed iron-deficient diet for three weeks. Rats were divided into four groups (DD: no treated group on iron deficient diet, DD+HI: heme-iron treated group, DD+OI: sericin-Fe, and DD+II: inorganic iron ($FeSO_4$) treated group, and then iron supplemented by injection for one week. After oral administration for one week, the iron contents of serum and liver were significantly higher in DD+OI ($4.2\;{\mu}g/mL$ and $80.1\;{\mu}g/mL$) and DD+HI ($3.2\;{\mu}g/mL$ and $70.6\;{\mu}g/mL$) than DD ($2.0\;{\mu}g/mL$ and $47.9\;{\mu}g/mL$). Hemoglobin content of treated groups was significantly higher than DD, but the significant difference among groups was not shown. Aspartate aminotransferase (AST) and alanine aminotransferase (ALT) levels did not show any significant difference among all groups. Binding iron to peptide from sericin hydolysates seems to improve its bioavailability and to hasten the cure of iron deficiency in experimental rat.
