The effect of Natural Antimicrobial Agent (천연 물질의 항균 효과)

  • Manuka oil named New Zealand’s tea tree oil is oil-soluble and comes firom nature. Manuka oil and its extract $\alpha$-pinene, Oxy’less clear, R-limonene which is one of the component of Citron extracted from Grapefruit seed and Citrex were used to estimate the antimicrobial activity and to improve the capability of antiseptic. Disk diffusion method was used to measure the antimicrobial activity. Escherichia coli which is gram-negative bacteria and Staphylococcus aureus which is gram-positive bacteria were used as strain. The antimicrobial activity of Manuka oil and $\alpha$-pinene for Escherichia coli, Staphylococcus aureus was similar when the concentration of Manuka oil and $\alpha$-pinene are 10u/paper disk. However, antimi-crobial activity of Manuka oil fDr Escherichia coli, Staphylococcus aurem was better than that of $\alpha$-pinene when the concentration of Manuka oil and $\alpha$-pinene was low. Antimicrobial activity of Oxy’less clear is better than that of propyl para hydroxybenzoate(PPHB), R-limonene at all the concentration and is similar to that of $\alpha$-pinene. Characterization of Natural Antiseptic System Utilized Propolis and Herb Essential Oil (프로폴리스와 허브에센셜오일을 이용한 천연방부제형의 특성)

  • The cosmetic and toiletries are necessary health care & household for common life. However we need antiseptic which is effecting harmlessly to the human body. There are propolis, Lavender, Lemon, essential oil in the natural antiseptic materials. This work proceeded design Natural-antiseptic system with three materials as above-mentioned. Natural-antiseptic system was accomplished with propolis (2%), Lavender essential oil (0.3%), Lemon essential oil (0.3%) safety out of Polysorbate 20 (0.5%), Polysorbate 80 (0.5%), PEG (60) hydrogenated castor oil (0.45%), ethanol (5%). The antimicrobial test was experimented on E. coli and Bacillus stearothermophilus. In this antimicrobial test, we found that the effect of antisepsis against E. coli and Bacillus stearothermophilus with propolis 0.3%, Lavender essential oil 0.045% and Lemon essential oil 0.045% was improved. Therefore could expect Natural-antiseptic system product for moisturizing skin toner for face, nourishing essence and wet tissue for clean other things.

A Study on the Use of Human Antibacterial Peptide LL-37-derived FK-13 as a Cosmetic Preservative (인간 항균펩타이드인 LL-37 유래의 FK-13의 화장품보존제로 활용에 대한 연구)

  • Here, we conducted the study on the possibility of using FK-13, a short analog of human-derived antibacterial peptide LL-37, as a cosmetic preservative to discover a natural cosmetic preservative that is safe for human body. For the purpose, FK-13 composed of 13 amino acids was synthesized by solid-phase peptide synthesis, and purified using reversed phase-high performance liquid chromatography (RP-HPLC). The purity and molecular weight were confirmed by liquid chromatography-mass spectrometry (LC-MS) analysis. FK-13 showed high antimicrobial activity on the three gram-positive bacteria (Staphylococcus aureus, Bacillus subtilis, and Staphylococcus epidermidis), the three gram-negative bacteria (Escherichia coli, Salmonella typhimurium, and Pseudomonas aeruginosa), and also even the fungus Candida glabrata. FK-13 had a broad spectrum of antibacterial activity, showing a suitability as a cosmetic preservative. In addition, FK-13 showed high thermostability and higher antibacterial activity in a comparative test with existing natural herbal cosmetic and chemical preservatives. Therefore, as FK-13 is a safe material and has high antibacterial activity at a low concentration, it is likely to be applied as a peptide natural cosmetic preservative that can replace existing chemical preservatives.

Enhanced Antimicrobial Activities and Physicochemical Characteristics of Isoliquiritigenin Encapsulated in Hydroxypropyl-β-Cyclodextrin (아이소리퀴리티게닌을 담지한 2-하이드록시프로필-베타사이클로덱스트린의 물리화학적 특성 및 항균활성 연구)

  • Isoliquiritigenin (ILG) is a hydrophobic component in licorice and has a variety of pharmaceutical and biological activities. In this study, we prepared an isoliquiritigenin-hydroxypropyl-${\beta}$-cyclodextrin (ILG/HP-${\beta}$-CD) complex by freeze-drying method to enhance its water solubility. The complex was characterized by phase solubility studies, DSC, SEM, and 1H NMR. Antimicrobial activities against Staphylococcus aureus (S. aureus) and Escherichia coli (E. coli) were evaluated by broth dilution method. The results showed that the stoichiometry of ILG/HP-${\beta}$-CD complex was 1 : 1. The antimicrobial activity of ILG/HP-${\beta}$-CD complex was higher than that of using free ILG against S. aureus and E. coli. Therefore, we suggest that ILG/HP-${\beta}$-CD complex may be used as a natural antiseptic and could potentially replace synthetic preservatives in cosmetic and food industries.

Antimicrobial Activity, Quantification and Bactericidal Activities of Licorice Active Ingredients (감초 성분의 항균활성, 정량 및 방부력에 관한 연구)

  • The present study was aimed at investigating the antimicrobial activities of licorice's active ingredients. Four samples of licorice ingredients (glycyrrhizin, liquiritin, liquiritigenin, and isoliquiritigenin) were evaluated for their antimicrobial activities against six skin microorganisms. The bioassay applied for determining the antimicrobial effects employed a disc diffusion assay, the minimum inhibitory concentration, and the challenge test. The ingredients showed antibacterial activities. Especially, isoliquiritigenin has significant antimicrobial activities against two Gram-positive (Bacillus subtilis, Propionobacterium acnes) and two Gramnegative (Escherichia coli, Pseudomonas aeruginosa) bacteria. These samples had much higher antimicrobial activities than synthetic preservatives. Our results reveal that liquiritigenin and isoliquiritigenin could be useful compounds for the development of antibacterial agents for the preservation of cosmetics and foods. The two flavonoids, liquiritigenin and isoliquiritigenin, sourced from Korea, China, Uzbekistan, were quantified using HPLC. The results demonstrated that Korean licorice has two flavonoids (liquiritigenin, isoliquiritigenin) in much higher quantities than was observed in the licorice obtained from the two other countries. Thus, isoliquiritigenin and Korean licorice extract represent new candidates for antimicrobial agents.

Antioxidant, Antiaging and Antimicrobial Effects of Ethanolic Extract and Ethyl Acetate Fraction from Eclipta prostrata (한련초 에탄올 추출물과 에틸아세테이트 분획물의 항산화, 항노화 및 항균 효과)

  • An annual plant, Eclipta prostrata (Linn) is a member of the Asteraceae plant family and inhabited in tropical or subtropical regions of the world. Through many previous researches, E. prostrata has been extensively studied for its hepatoprotective effect, antivenom potential against viper venom, antioxidant, hair-growth, wound-healing efficacy and so on. In this study, for better understanding of the potential of E. prostrata as skin protectant, we conducted the experiments evaluating the antioxidant and antiaging efficacy. To this end, 50% ethanolic extract of E. prostrata and its ethyl acetate fraction were prepared and investigated. For the evaluation of antioxidant capacity of the samples, $FSC_{50}$ and $OSC_{50}$ were estimated. As a result, $OSC_{50}$ of ethyl acetate fraction was 2.7 times superior to $OSC_{50}$ of L-ascorbic acid, a well known antioxidant agent. Futhermore E. prostrata showed notable reactive oxygen species (ROS) scavenging effect and protective effect against $H_2O_2$ in the celluar level as well. Especially, in the $^1O_2$ induced hemolysis test, $64{\mu}g/mL$ of ethyl acetate fraction showed greater than 6 times increased retardation effect compare to control which means E. prostrata has remarkable antioxidant capacity. To validate the antiaging effect of the samples, we conducted elastase inhibition assay using elastase solution extracted from human skin fibroblasts, Hs68. As a result, $16{\mu}g/mL$ of each sample showed 6.8% and 14.0% of elastase inhibition respectively. Finally, antimicrobial activity of E. prostrata was assessed to validate the possibility as alternative preservative. From the result, ethyl acetate fraction showed oustanding antimicrobial activity as of methyl paraben, a well known chemical preservative. In conclusion, these results suggest that E. prostrata can be used as natural skin protectant or preservative as natural ingredient in food or cosmetics industry.

Comparative Study on the Antimicrobial Activity of Glycyrrhiza uralensis and Glycyrrhiza glabra Extracts with Various Countries of Origin as Natural Antiseptics (원산지별 감초 추출물의 항균 활성 비교 및 천연방부제로써의 효능 연구)

  • The aim of this study was to evaluate the antimicrobial activities of Glycyrrhiza uralensis and Glycyrrhiza glabra extracts with various countries of origin. Three samples of licorice with various origins (Korea, China, and Uzbekistan) were evaluated for their antimicrobial activities against six skin microflora. The bioassay applied for determining the antimicrobial effects included the disc diffusion assay, minimum inhibitory concentration, and challenge test. The ethyl acetate fractions of G. uralensis and G. glabra extracts showed significant antimicrobial activities against two gram-positive (Bacillus subtilis, Propionibacterium acnes) and two gram-negative (Escherichia coli, Pseudomonas aeruginosa) bacteria. These samples had much more intensive antimicrobial activities than synthetic preservatives on B. subtilis, P. acnes, and P. aeruginosa, especially. Korean licorice showed the highest antimicrobial activity amongst the samples tested. In view of the observed inhibitory features of these G. uralensis and G. glabra extracts, it is suggested that they could be used as natural antiseptics against bacterial contamination in cosmetics and foods, instead of the common synthetic preservatives currently employed.

국내 자생 물봉선속(Impatiens L.)의 항산화활성 및 생리활성물질 함량 차이 비교

  • 물봉선속(Impatiens L.)은 일년생 또는 다년생 초본으로 우리나라에 5-7종이 분포한다. 물봉선의 화장품용 항산화제 및 천연 방부제로서의 유용성이 밝혀졌으며, 최근 항염, 항산화 등 생리활성에 대한 연구가 국내 자생하는 물봉선 및 노랑물봉선을 대상으로 수행된 바 있으나 이하 분류군에 대한 연구는 미비한 실정이다. 본 연구에서는 국내 자생하는 물봉선속 분류군들의 항산화활성과 생리 활성물질의 함량을 분석하고 지역 간 분류군별 차이를 확인하고자 하였다. 따라서 국내 자생하는 물봉선속 분류군들의 항산화활성을 검정하기 위하여 DPPH, ABTS, TPC, TFC 4가지 방법을 이용하여 분석하였다. 국내 자생하는 물봉선속 5분류군 가야물봉선(Impatiens atrosanguinea (Nakai) B.U.Oh & Y.P.Hong), 노랑물봉선(Impatiens noli-tangere L), 물봉선(Impatiens textorii Miq), 미색물봉선(Impatiens noli-tangere var. pallescens Nakai), 처진물봉선(Impatiens furcillata Hemsl)이 12개 지역에서 수집되었으며, 잎 추출물(70% 에탄올)에 대해 분석되었다. 물봉선속 분류군들의 잎 추출물의 DPPH 라디칼 소거 활성 검정 결과 가야물봉선(4.91 ± 3.00 mgAAE/g)이 가장 높았고 처진물봉선(1.77 ± 0.55 mgAAE/g)이 가장 낮았으며, ABTS의 경우 가야물봉선(3.14 ± 1.35 mgAAE/g)로 가장 높았고 미색물봉선(1.87 ± 0.16 mgAAE/g)이 가장 낮았다. TPC의 경우 미색물봉선(5.48 ± 1.05 ugGAE)이 가장 높았고 노랑물봉선(2.78 ± 1.98 ugGAE)이 가장 낮았으며, TFC의 경우 물봉선(0.70 ± 0.20 ugGAE/g)이 가장 높았고 노랑물봉선(0.45 ± 0.08 ugGAE/g)이 가장 낮게 나타났다. 수집지역별로는 각각 DPPH와 ABTS의 경우 노랑물봉선, TPC의 경우 가야물봉선, 노랑물봉선, 물봉선, TFC의 경우 처진물봉선이 지역별 차이를 보였다. 이번 연구 결과를 토대로 국내 자생하는 물봉선속 분류군 별 항산화활성과 생리활성물질 차이를 확인할 수 있었고 추후 유용 소재로써의 이용과 우수 개체선발에 도움이 될 것으로 사료 된다.

Antimicrobial Activities of Licorice Extracts from Various Countries of Origin according to Extraction Conditions (원산지별 감초추출물의 추출 조건별 항균활성)

  • In this study, Glycyrrhiza uralensis and Glycyrrhiza glabra extracts, with their countries of origin as Korea (Jecheon), Uzbekistan and China, were prepared under various extraction conditions. There were 8 extraction conditions which the licorice were subjected to, and all conditions had different extraction solvents, temperatures and times. Antimicrobial activity on skin flora was evaluated comparatively by a disc diffusion assay, broth macrodilution assay, and kill time curve assay. Based on the antimicrobial activity of their extract confirmed by disc diffusion assay, we established optimal extraction conditions. The Korean licorice extract (85% ethanol, $40^{\circ}C$, 12 h) showed the best activity amongst the samples examined. In particular, its antimicrobial activity against Propionibacterium acnes was the highest. Minimum inhibitory concentrations (MIC) and minimum bactericidal concentrations (MBC) of the licorice extracts revealed that the Korean licorice ($156{\mu}g/ml$ and $1,250{\mu}g/ml$) had better antimicrobial activity than that of the Uzbekistani licorice ($625{\mu}g/ml$ and $2,500{\mu}g/ml$) and the Chinese licorice ($625{\mu}g/ml$ and $5,000{\mu}g/ml$). Taken together, it was shown that Korean licorice extracted in group F (85% ethanol, $40^{\circ}C$, 12 h) had the highest antimicrobial activity amongst the licorices from the other countries of origin. These results also suggest that the optimal extraction conditions are 85% ethanol, $40^{\circ}C$, 12 h, and that licorice has a potential application as a natural preservative in cosmetics products, thereby replacing synthetic preservatives.

Antioxidative and Antibacterial Activities of Artemisia princeps Pampanini Extracts (사자발쑥 추출물의 항산화 및 항균 활성)

  • In the present study, the antioxidative and antibacterial activities of Artemisia princeps Pampanini (A. princeps Pamp.) extract were investigated. The ethyl acetate fraction of A. princeps Pamp. showed the most prominent free radical (1,1-diphenyl-2-picrylhydrazyl, DPPH) scavenging activity ($FSC_{50}=12.27{\mu}g/mL$). Reactive oxygen species (ROS) scavenging activities ($OSC_{50}$) of A. princeps Pamp. extract on $Fe^{3+}-EDTA/H_2O_2$ systems were investigated using a luminol-dependent chemiluminescence assay. The ethyl acetate fraction of the extract ($OSC_{50}=0.33{\mu}g/mL$) had a 5 times greater ROS scavenging activity than L-ascorbic acid ($1.50{\mu}g/mL$), known as a water soluble antioxidant. The cellular protective effects of fractions of A. princeps Pamp. on the rose-bengal sensitized photohemolysis of human erythrocytes were examined. The aglycone fraction of extracts suppressed photohemolysis in a concentration dependent manner. The inhibitory effects of A. princeps Pamp. extract on tyrosinase were investigated to assess their whitening efficiency. The ethyl acetate fraction demonstrated a 7 times higher tyrosinase inhibitory effect ($IC_{50}=29.20{\mu}g/mL$) than albutin, known as a whitening agent. The antibacterial activity of ethyl acetate fractions against various normal skin flora were measured. The results showed that the antibacterial activity of the fraction was the highest on Staphylococcus aureus, Bacillus subtilis, and Propionibacterium acnes. Antioxidant substances were isolated and purified from the ethyl acetate fractions. Eupatilin and jaceosidin were identified. These results indicate that the extract/fractions of A. princeps Pamp. can function as antioxidant and/or antibacterial agents for the skin.
