A Study on the Differentiation of the Implantation Sites in the Pseudopregnant Rat Uterus (가임신 흰쥐 자궁조직의 착상부위분화에 관한 연구)

  • 포유류 배아의 착상기전을 규명할 목적의 일환으로 자궁내막조직의 탈락막 형성과 분화에 미치는 난소 스테로이드 호르몬의 영향과 인위적 자극등의 영향을 조사하였다. 가임신을 유도시킨 흰쥐와 정상임신군의 자궁내막조직을 착상부위와 비 착상부위로 분리하여 자궁내막조직의 분화에 지표가 되는 Alkaline phosphatase(ALPase)의 활성을 측정하였다. 가임신군에서 인위적 자극(trauma)을 가한 자궁이 자극을 받지않은(control) 자궁에 비해 Progesterone(P)만을, 또는 progesterone과 estradiol(E + P)을 동시 처리받은 실험군에서 ALPase 활성이 유의한 차이(p<0.01)로 높게 나타났다. 정상임신군에서는 착상시기인 임신 제6일군의 착상부위에서 I처리군과 P처리군의 ALPase활성이 비착상 부위에 비해 유의한 차이(P < 0.05)로 크게 나타나며, 특히 임신 제6일군에서는 착상, 비착상 부위의 P처리군의 활성이 Intact군보다 유의하게(P<0.05) 높아졌다. 정상임신 제 9일 Intact군의 ALPase활성은 임신 제3일, 6일의 Intact 군의 활성에 비하여 착상(3, 6일 :P<0.01), 비 착상부위 (3일 :P<0.05, 6일 :P<0.02)에서 다같이 유의한 차이로 높아졌다. 본 연구결과에서 배아가 분화, 발생해 감에따라 자궁내막조직 분화도 활발해지고, 착상부위 분화는 착상시기부터 현저해지며, 특히 P호르몬의 영향이 크다는 것과 배아 아닌 인위적 자극으로도 탈락막 형성 유도가 가능하다는 것을 확인할 수 있었다.

  • Time course monitoring of FCW/DCW ratio, production of intra and extracellular protein, and peroxidase activity were performed during suspension culture of Taxus chinensis cells. The observed FCW/DCW ratio was 12 at day 14, which was the lowest value during cultivation, and the specific protein production, based on dry cell weight, was also the lowest at day 14, which showed 4.3 mg/g DCW. The pattern of POD activity was similar to that of protein production. The results in this report were obtained using actively growing cells in flasks, therefore it is possible to use those results to control the process and indicate the stresses imposed on cells during large-scale cultivation.

Web Program for Laboratory Animal Group Separation Based on Biological Characteristics (생체지표를 활용한 웹기반의 실험동물 군(郡) 분리 프로그램)

  • The laboratory animal group separation is dividing animal population into subgroups, which have similar average and standard deviation values among the subgroups, based on the biological characteristics such as body weight, glucose level in blood, etc. Although group separation is very important and initial step in experimental design, it needs a labor intensive process for researchers because of making similar average and standard deviation values among the subgroups using the raw biological characteristics. To reduce the labor cost and increase the efficiency of animal grouping, we developed a web program named as laboratory animal group separation (LAGS) program. This LAGS uses biological characteristics of population, number of group, and the number of elements per each subgroup as input data. The LAGS automatically separates the population into each subgroup that has similar statistical data such as average and standard deviation values among subgroups. It also provides researchers with the extraordinary data generated in the process of grouping and the final grouping results by graphical display. Through our LAGS, researchers can validate and confirm results of laboratory animal group separation by just a few mouse clicks.

  • 간척지는 바다와 접하는 하구언, 개벌로 이루어진 해안 등을 농공상업용지로 개간한 토지로서 간척지 이용에 있어 가장 우선적으로 용수공급 방안 수립이 고려되어야 한다. 해안유출지하수(submarine groundwater discharge)는 지하수 담수체가 존재하는 마지막 장소로 염해가 없는 청정수질 용수이다. 해안유출지하수 발생 구간을 탐지하기 위해 실측자료의 시공간적 한계를 극복할 수 있는 인공위성 영상을 활용한 원격탐사 기법을 이용하여 광역규모의 다양한 자료를 이용하고자 한다. MODIS (Moderate-Resolution Imaging Spectroradiometer)는 지구 생물권 활동에 관한 자료를 제공하는 미항공우주국 Terra EOS (Earth Observation system)위성의 주센서로 해양, 육상과 대기 분야에 적용이 가능한 다목적 센서이다. MODIS는 36개의 밴드를 이용하여 대기, 지표, 해양 관련 다양한 정보들을 제공하고 있다. 본 연구에서는 간척지의 효율적인 용수공급을 위한 해안유출지하수의 최적 개발 및 이용을 위해 MODIS MOD11 product 지표면온도(Land Surface Temperature; LST), MODIS MOD13 product 식생지수(Normalized Difference Vegetation Index; NDVI), 기상청의 지중온도와 실측자료를 이용하여 새만금 간척지를 대상으로 해안유출지하수 발생 구간을 탐지하고자 한다.

  • 한강 자연성 회복은 사회환경을 고려한 한강의 생태적 기능회복과 복원, 자연의 복원력을 기반으로 한 사람이 공생 공존하는 새로운 관계로의 전환점을 의미하고 있다. 본 연구에서는 한강 유역권 산·학·연 관계자 100명을 대상으로 설문조사를 실시하여 한강 자연성 회복 비전과 추진전략을 도출한 결과, '맑은 물이 흐르는 강'(1순위), '더불어 사는 강'(2순위), '생명이 숨쉬는 강'(3순위), '살아 움직이는 강'(4순위)으로 정책방향을 수립할 수 있었다. 또한 설문조사의 결과를 바탕으로 AHP 및 델파이 분석을 통해 한강 상류, 중류(서울권, 경기권), 하류별 자연성 회복 평가지표간 가중치를 산정하여 주요 추진과제를 선정하였다. 상류유역에서는 비점오염원 관리를 통한 환경오염 감소, 중류 유역에서는 도심하천의 수질관리강화 및 물 순환 체계 개선, 하류 유역에서는 생물다양성 확보 및 상·하류 지역 협력을 통한 상수원 수질 개선의 방안 필요의 결과가 도출되었다. 본 연구를 통하여 한강의 하천관리정책은 이수·치수 중심의 하천조성에서 하천생태복원 및 강과 사람과의 공존과 공생이 조화를 이룬 하천조성으로 관점이 변화되었음을 확인할 수 있었으며, 가중치 평가를 통한 최적 조합의 하천 관리방안이 도출되었다.

Selection of Biodiversity Indicators for a National Assessment in Korea (국내 생물다양성 평가를 위한 지표 선정)

  • This study was conducted to select indicators for assessing national biodiversity. For this purpose, 140 biodiversity-related indicators were identified as a result of inventorying biodiversity-related indicators used in Korea and abroad, and when these indicators were applied to the pressure, status, and response indicator system, it was found that status indicators accounted for the largest number of indicators, with 29 pressure, 59 status, and 44 response. We also categorized the status indicators into genes, species, habitat, function, and quality, and found that species and habitat indicators accounted for the majority. Pressure indicators were categorized into direct exploitation, pollution, alien species, climate change, and habitat change. As a result, it was found that direct exploitation and pollution accounted for most of the pressure indicators. In addition, this study used internationally used indicator selection criteria to establish criteria for selecting domestic biodiversity assessment indicators. Using this list of indicators and indicator selection criteria, we evaluated the prioritization of domestically applicable biodiversity indicators through relevant expert consultations. 1) Vegetation class, 2) Land cover indicators, and 3) Change of protected area ranked highly. In fact, these indicators have been used in many studies due to the availability of assessable data. However, most of the highly scored indicators are based on ecosystem area, and further consideration of ecosystem functions and components(species) is needed.

Cone Characteristics and Seed Quality among Harvest Times in the Clonal Seed Orchard of Larix kaempferi (낙엽송 클론 채종원에서 구과 채취시기에 따른 구과특성 및 종자품질)

  • Harvest time is one of the most important determining factors of seed quality, especially for species that produce seeds over irregular and long-term periods, such as Larix kaempferi. A cone collection plan must be established to increase seed production efficiency and stable mass production. We investigated seed qualities such as seed efficiency, germination rate, and T50 (germination speed), with 7 or 8 cone collection times at a clonal seed orchard of L. kaempferi in Chungju between 2021 and 2022. A multivariate analysis was then performed for the collected data. In early August, decreases in the moisture contents and browning of cones were observed. These were followed by a decrease in germination rate, with a peak at the end of September, but no clear trend was observed. The later the cones were harvested, the better the seed vigor (T50). However, the seed yield and efficiency decreased owing to increases in seed scattering and the number of insect-damaged seeds. As a result, the optimal time of seed harvest for the seed orchard was in early August. To produce uniform seedlings, insect damage must be reduced through timely control and harvest cones in early September. This shows that the degree of browning and moisture content of cones can be used as indicators of the timing of cone collection in L. kaempferi seed orchards.

Relationships between Air Pollution by SO2 and Soluble Sulphur Contents in the Leaves and Bark pH in Urban Forest Trees (도심지역(都心地域)의 아황산(亞黃酸) 가스에 의(依)한 대기오염(大氣汚染)과 수목내(樹木內) 엽(燁)의 수용성(水溶性) 유황(硫黃) 함량(含量) 및 수피산도(樹皮酸度)와의 관계(關係))

  • This study was carried out to investigate relationships between atmospheric $SO_2$ concentration and both soluble sulphur contents in the tree leaves and bark pH to test the possibility of using them as bioindicators for air pollution. Mt. Kwanak, Mt. Nam, Mt. Bukhan (located in Seoul) and Mt. Paldal (located in Suwon, Kyonggi-do) as polluted areas and Pyongchang, Kwangwon-do as an unpolluted area were selected for this study. Soluble sulphur contents in the leaves and hark pH of two tree species (Pines densiflora S. et Z. and Quercus mongolica Fisch.) were analyzed in May, August, and October, 1990 by $BaSO_4$ precipitation method and pH measurement, respectively. In both species, concentration of soluble sulphur in the leaves increased with increasing concentration of atmospheric $SO_2$ (correlation coefficient : 0.52). Soluble sulphur contents in the 2-year-old needles of Pines densiflora (0.170%) and current year leaves of Quercus mongolica (0.081%) in Mt. Nam in the center of Seoul were higher than those in unpolluted Pyongchang area (0.023% and 0.034%, respectively). Bark pH decreased with increasing atmospheric $SO_2$ concentration (correlation coefficient : -0.52). Bark pH of P. densiflora (pH 3.42) and Q. mongolica (pH 3.63) in Mt. Nam were lower than those in Pyongchang area (pH 3.94 and pH 4.93, respectively). Both soluble S content in the leaves and bark pH were recognized as suitable bioindicators for air pollution by $SO_2$. Especially, bark pH showed more sensitive response to air pollution by $SO_2$ than soluble S concentration in the leaves. The lowest concentration of soluble sulphur and the highest bark pH in August were considered to be due to heavy rain during the rainy season. Soluble S content in the leaves and bark pH were not significantly different at 5% level between the two species in polluted areas.

Estimation of Soil Cooling Load in the Root Zone of Greenhouses (온실내 근권부의 지중냉각부하 추정)

  • Root zone cooling, such as soil or nutrient solution cooling, is less expensive than air cooling in the whole greenhouse and is effective in promoting root activity, improving water absorption rate, decreasing plant temperature, and reducing high temperature stress. The heat transfer of a soil cooling system in a plastic greenhouse was analyzed to estimate cooling loads. The thermal conductivity of soil, calculated by measured heat fluxes in the soil, showed the positive correlation with the soil water content. It ranged from 0.83 to 0.96 W.m$^{[-10]}$ .$^{\circ}C$$^{[-10]}$ at 19 to 36% of soil water contents. As the indoor solar radiation increased, the temperature difference between soil surface and indoor air linearly increased. At 300 to 800 W.m$^{-2}$ of indoor solar radiations, the soil surface temperature rose from 3.5 to 7.$0^{\circ}C$ in bare ground and 1.0 to 2.5$^{\circ}C$ under the canopy. Cooling loads in the root zone soil were estimated with solar radiation, soil water content, and temperature difference between air and soil. At 300 to 600 W.m$^{-2}$ of indoor solar radiations and 20 to 40% of soil water contents,46 to 59 W.m$^{-2}$ of soil cooling loads are required to maintain the temperature difference of 1$0^{\circ}C$ between indoor air and root zone soil.

Spatial Analysis of Ecological Characteristics for Benthic Macroinvertebrate Community Structure in Lake Hoengseong Region (횡성호 일대 저서성 대형무척추동물 군집구조의 생태적 특성 분석)

  • Benthic macroinvertebrates were investigated in Hoeongseong Lake region from March to October 2010, Korea. Macroinvertebrate communities, composition of the functional feeding groups, habitiat oriented groups and the biological water quality were assessed above and below Hoeongseong Dam in the lake region. Six sites, two (St.1~2) above the lake, two (St. 3~4) into the lake, and two (St. 5~6) below the dam, were selected for quantitative (Surber sampler $30cm{\times}30cm$, mesh size 0.2 mm) and qualitative (Hand net) samplings of benthic macroinvertebrates. As a result, a total of 83 species belonging to 43 families, 17 orders, 8 classes, and 5 phyla were recognized. The EPT-group (50 spp. : 60.24%) that is major taxa or EPT-group plus Diptera (61 spp. : 73.49%) occupied most of benthic macroinvertebrates community. Based on quantitative sampling, the number of benthic maroinvertebrates above the lake was 2,399 individuals including 54 species, 28 families, 11 orders, 4 classes, and 4 phyla, whereas 510 individuals including 16 species, 12 families, 7 orders, 4 classes, and 3 phyla in the lake and 626 individuals including 62 species, 33 families, 13 orders, 6 classes, and 4 phyla below the dam were collected respectively. Dominance index was the highest, with 0.82-0.93 ($0.87{\pm}0.05$) in Hoeongseong lake (St. 3-4), diversity index was 3.04-3.16 ($3.10{\pm}0.06$), evenness index was 0.79-0.85 ($0.82{\pm}0.03$), and richness index was 7.27-8.52 ($7.90{\pm}0.63$), which were relatively higher below the dam sites. In the functional feeding groups, collector-gatherers and collector-filterers were the highest in the whole sites, and predators (Micronecta sedula) was appeared highly in the lake. Moreover, swimmers, burrowers, and clingers were considerably occupied in all collecting sites. The result of the DCA, similarity analysis, and MRPP were well reflective of the composition of lake and stream macroinvertebrates. ESB indicate that the lake sites were evaluated heavily polluted under priority improvement waters. Also, Semisulcospira gottschei, Ecdyonurus kibunensis, Epeorus pellucidus, Rhoenanthus coreanus, Stenelmis sp., and Cheumatopsyche brevilineata are considered as an indicator species above and below Hoeongseong Lake, whereas Macrobrachium nipponense and Micronecta sedula are indicated in the lake.
