  • Agrobacterium tumefaciens의 연구는 유전공학 시대를 맞이하여 많은 연구자들의 커다란 주목을 끌고있다. 식물세포내에 외부 유전자를 도입시키는, 확실히 믿을 수 있는 vector로 등장된 때문이다. 원래 이세균은 식물 줄기나 뿌리에 암종을 유발시키므로서 암발성 원인 구명 연구로 흥미를 끌게 되었다. 연구결과는 암발생 예방및 치료에 목적을 둠은 덩연할 것이다. 많은 약제가 시험되었으나 별로 진전을 보지 못하던중 비 원인성인 Agrobacterium radiobacter, strain 84에 의한 생물학적 방제의 성공으로 유일한 방제법을 갖게되었다. 뒤이어 암종발생 기작도 밝혀졌다. Agrobacterium의 세계는 온통 유전공학 기술로 채워져 있다. 암종발생에서 방제원리에 이르기까지 수없이 먼 옛날부터 이미 익혀오던 DNA 조작기술이었던가\ulcorner 암종을 유발시키는 agrocin84 plasmid를 갖는 비병원성 Agrobacterium을 찾아 생물학적 방제법을 확립하였다. 그후 병원성 Agrobacterium은 이에 대하여 어떻게 살아남을 것인가\ulcorner 실로 놀라운 일이라 아니할 수 있을까\ulcorner 이 병원성 Agrobacterium은 비 병원성 Agrobacterium 속에 있는 agrocin 84 plasmid을 탈취하여 자신이 agrocin84를 생성분비하며 암종 유발을 계소하여 간다. 아니면 비병원성 Agrobacterium이 병원성 Agrobacterium에게 agrocin 84 plasmid를 넘겨주었을까\ulcorner 왜 넘겨주었을까\ulcorner 공존을 위하여서일까\ulcorner 우리의 유전공학 기술은 이것을 막아줄수 있을까\ulcorner 생물학적 방제의 재성공을 위하여 논제의 연구는 왜 필요했던가\ulcorner 그 전후를 여기에 서술해 본다.닭이며 또한 제한된 지면에서 충분히 고찰하기는 어렵다. 우리나라에서 자주 거론되는 백신 및 종류에 국한하여 그 문제점과 앞으로의 전망을 고찰해 보기로 한다.ocking electrode를 제작하여 복합고분자 전해질과의 계면저항을 측정하였다.nm (1.2921eV)는 acceptor-bound exciton 인 I1(AO,X) 이고, 964.6nm(1.2853eV)는 donor-acceptor pair(DAP) 발광, 1341.9nm (0.9239eV)는 self activated(SA)에 기인하는 광발광 봉우리로 고찰되었다.가 높을수록 방출전류가 시간에 따라 급격히 감소하였다. 각 duty비에서 방출전류의 양이 1/2로 감소하는 시점을 에미터의 수명으로 볼 때 duty비 대 에미터 수명관계를 구해 높은 duty비에서 전계방출을 시킴으로써 실제의 구동조건인 낮은 duty비에서의 수명을 단시간에 예측할 수 있었다. 단속적으로 일어난 것으로 생각된다.리 폐 관류는 정맥주입 방법에 비해 고농도의 cisplatin 투여로 인한 다른 장기에서의 농도 증가 없이 폐 조직에 약 50배 정도의 고농도 cisplatin을 투여할 수 있었으며, 또한 분리 폐 관류 시 cisplatin에 의한 직접적 폐 독성은 발견되지 않았다이 낮았으나 통계학적 의의는 없었다[10.0%(4/40) : 8.2%(20/244), p>0.05]. 결론: 비디오흉강경술에서 재발을 낮추기 위해 수술시 폐야 전체를 관찰하여 존재하는 폐기포를 놓치지 않는 것이 중요하며, 폐기포를 확인하지 못한 경우와 이차성 자연기흉에 대해서는 흉막유착술에 더 세심한 주의가 필요하다는 것을 확인하였다. 비디오흉강경수술은 통증이 적고, 입원기간이 짧고, 사회로의 복귀가 빠르며, 고위험군에 적용할 수 있고, 무엇보다도 미용상의 이점이 크다는 면에서 자연기흉에 대해 유용한 치료방법임에는

Comparison of Push-out Bond Strengths According to Relining Procedure and Cement Type on Fiber Post (Fiber post의 Relining 방법과 시멘트 유형에 따른 Push-out Bond Strength의 비교)

  • When restoring endodontically treated teeth is the mismatch between fiber post size and post space diameter, the resin cement layer is excessively thick in post space and voids are likely to form in it, thus predisposing to de-bonding. The method to overcome this problem is to reline the fiber post with composite resin. This individual anatomic post improves the adaptation of post to root walls and decreases the resin cement thickness. The purpose of this in vivo study was to evaluate the push-out bond strength of fiber post according to relining procedure and luting agents type used for simplicity of clinical procedure. Forty-two extracted teeth were divides into six groups.(n=7) A1: relined fiber post cemented with Luxacore/all-bons 2, A2: non-relined fiber post cemented with Luxacore/all-bond2, B1: relinind fiber post cemented with Calibra/XP-bond, B2: non-relined fiber post cemented with Calibra/XP-bond, C1: relined fiber post cemented with RelyX Unicem, C2: non-relined fiber post cemented with RelyX Unicem Push-out bond strength was affected by interaction between relining procedure and luting agent type. Relined fiber post presented higher push-out bond strength value than non-relined fiber post and statically significant differences(p<0.05) Cementation with RelyX Unicem showed significantly higher bond strength than other luting agents(p<0.05).

Molecular Phylogeny of Korean-type Coliphages and American-type Coliphages Determined by a RAPD Analysis (RAPD 분석법에 의한 한국형 대장균파아지와 미국형 대장균파아지의 분자적 계통분류)

  • RAPD-PCR was applied to identify the phylogenetic relationship between isolated Korean-type coliphages ($\phi$C1, $\phi$C2, $\phi$C3 and $\phi$C4) and well-known American coliphages ($\phi$T2, $\phi$T4, $\phi$T5, $\phi$T7 and ${\phi}{\lambda}$). Subsequently, a computer analysis was carried out with the results of RAPD-PCR. As a result, 9 individuals were divided into five groups. The Korean-type coliphages formed a single cluster which showed very high genetic similarity but the American-type coliphages revealed very low genetic similarity among them. In particular, the $\phi$T2와 $\phi$T4 (T$_{even}$ phages) made one sub-cluster among American coliphages, and they were very distant from $\phi$T5, $\phi$T7 and ${\phi}{\lambda}$. However, ${\phi}{\lambda}$ made a cluster with the Korean-type coliphages that we isolated. The genome size of Korean-type coliphages was ranged from 25,000 bp to 35,000 bp. Among them, the genome of $\phi$C2 was the smallest and that of $\phi$C1 was the biggest, while others were in the middle of the size.

Automatic Building Modeling Method Using Planar Analysis of Point Clouds from Unmanned Aerial Vehicles (무인항공기에서 생성된 포인트 클라우드의 평면성 분석을 통한 자동 건물 모델 생성 기법)

  • In this paper, we propose a method to separate the ground and building areas and generate building models automatically through planarity analysis using UAV (Unmanned Aerial Vehicle) based point cloud. In this study, proposed method includes five steps. In the first step, the planes of the point cloud were extracted by analyzing the planarity of the input point cloud. In the second step, the extracted planes were analyzed to find a plane corresponding to the ground surface. Then, the points corresponding to the plane were removed from the point cloud. In the third step, we generate ortho-projected image from the point cloud ground surface removed. In the fourth step, the outline of each object was extracted from the ortho-projected image. Then, the non-building area was removed using the area, area / length ratio. Finally, the building's outer points were constructed using the building's ground height and the building's height. Then, 3D building models were created. In order to verify the proposed method, we used point clouds made using the UAV images. Through experiments, we confirmed that the 3D models of the building were generated automatically.

Adhesion-induced generation of oxygen free radical from human alveolar macrophages and its mechanisms (폐포대식세포의 부착에 의한 산소유리기 분비능 활성화 및 그 기전)

  • Background : Neutrophils or monocytes separated in vitro by the adherence to plastic surface are known to be activated by surface adherence itself and subsequent experimental data might be altered by surface adherence. In the process of surface adherence, adhesion molecules have a clear role in intracellular signal pathway of cellular activation. Human alveolar macrophages(HAM) are frequently purified by the adherence procedure after bronchoalveolar lavage. But the experimental data of many reports about alveolar macrophages have ignored the possibility of adhesion-induced cellular activation. Method : Bronchoalveolar lavage was performed in the person whose lung of either side was confirmed to be normal by chest CT. With the measurement of hydrogen peroxide release from adherent HAM to plastic surface and non-adherent HAM with or without additional stimulation of phorbol myristate acetate(PMA) or N-formyl-methionyl-leucyl-phenylalanine (fMLP), we observed the effect of the adherence to plastic surface. We also evaluated the effect of various biological surfaces on adhesion-induced activation of HAM. Then, to define the intracellular pathway of signal transduction, pretreatment with cycloheximide, pertussis toxin and anti-CD11/CD18 monoclonal antibody was done and we measured hydrogen peroxide in the culture supernatant of HAM. Results : 1) The adherence itself to plastic surface directly stimulated hydrogen peroxide release from human alveolar macrophages and chemical stimuli such as phorbol myristate acetate(PMA) or N-formyl-methionyl-leucyl-phenylalanine(fMLP) colud not increase hydrogen peroxide release in these adherent macrophages which is already activated. 2) PMA activated human alveolar macrophages irrespective of the state of adhesion. However, fMLP stimulated the release of hydrogen peroxide from the adherent macrophages, but not from the non-adherent macrophages. 3) HAM adherent to A549 cell(type II alveolar epithelium-like human cell line) monolayer released more hydrogen peroxide in response to both PMA and fMLP. This adherence-dependent effect of fMLP was blocked by pretreatment of macrophages with cycloheximide, pertussis toxin and anti-CD18 monoclonal antibody, Conclusion : These results suggest that the stimulatory effect of PMA and fMLP can not be found in adherent macrophage because of the activation of human alveolar macrophage by the adherence to plastic surface and the cells adhered to biologic surface such as alveolar epithelial cells are appropriately responsive to these stimuli. It is also likely that the effect of fMLP on the adherent macrophage requires new protein synthesis via G protein pathway and is dependent on the adhesion between alveolar macrophages and alveolar epithelial cells by virtue of CD11/CD18 adhesion molecules.

A Reflectance Normalization Via BRDF Model for the Korean Vegetation using MODIS 250m Data (한반도 식생에 대한 MODIS 250m 자료의 BRDF 효과에 대한 반사도 정규화)

  • The land surface parameters should be determined with sufficient accuracy, because these play an important role in climate change near the ground. As the surface reflectance presents strong anisotropy, off-nadir viewing results a strong dependency of observations on the Sun - target - sensor geometry. They contribute to the random noise which is produced by surface angular effects. The principal objective of the study is to provide a database of accurate surface reflectance eliminated the angular effects from MODIS 250m reflective channel data over Korea. The MODIS (Moderate Resolution Imaging Spectroradiometer) sensor has provided visible and near infrared channel reflectance at 250m resolution on a daily basis. The successive analytic processing steps were firstly performed on a per-pixel basis to remove cloudy pixels. And for the geometric distortion, the correction process were performed by the nearest neighbor resampling using 2nd-order polynomial obtained from the geolocation information of MODIS Data set. In order to correct the surface anisotropy effects, this paper attempted the semiempirical kernel-driven Bi- directional Reflectance Distribution Function(BRDF) model. The algorithm yields an inversion of the kernel-driven model to the angular components, such as viewing zenith angle, solar zenith angle, viewing azimuth angle, solar azimuth angle from reflectance observed by satellite. First we consider sets of the model observations comprised with a 31-day period to perform the BRDF model. In the next step, Nadir view reflectance normalization is carried out through the modification of the angular components, separated by BRDF model for each spectral band and each pixel. Modeled reflectance values show a good agreement with measured reflectance values and their RMSE(Root Mean Square Error) was totally about 0.01(maximum=0.03). Finally, we provide a normalized surface reflectance database consisted of 36 images for 2001 over Korea.

The Actual State and Transformation of Major Garden Ornaments in Changdeokgung Palace during the Modern and Contemporary Period (근현대기 창덕궁 내 주요 점경물의 실상과 변형)

  • This study investigated the actual state and transformation of the major garden ornaments in Changdeokgung Palace(昌德宮) in the modern and contemporary period, focusing on Nakseonjae(樂善齋), Juhamnu(宙合樓), Jondeokjeong(尊德亭), and Daebodanji (大報壇址). This study can be used as useful data for establishing the restoration and maintenance plan of the garden ornaments in Changdeokgung Palace, and the main results of the study can be summarized as follows. First, according to a photo taken by the Czech Vráz, in 1901, a total of six garden ornaments, including a stone pond and odd-shaped stones, were located in the backyard of Nakseonjae. Since liberation, arbitrary relocation of garden ornaments has frequently occurred, and in the process, two odd-shaped stones, originally located on the first floor of the terraced flower bed, have been transferred to the backyard of Gyeonghungak(景薰閣). Second, unlike the late Joseon Dynasty when 「Donggwoldo(東闕圖)」 was produced, odd-shaped stones were arranged symmetrically in the backyard of Juhamnu in the early 1900s. It was a traditional style garden ornament with similar appearance, size, and design. However, all the odd-shaped stones in the backyard of Juhaumnu were relocated to other places in the 1970s and 1980s. One is located at the rear of Aeryeonjeong(愛蓮亭) through the entrance of Bingcheon(氷泉) and the vicinity of Geumcheongyo(錦川橋), and the other remains in front of the Yeongyeongdang Jangnakmun(演慶堂 長樂門). Third, among the garden ornaments located in the area of Jondeokjeong in the past, one odd-shaped stone is now relocated around the stone bridge near the pavilion and the Yeonghwadang(暎花堂) with its components separated. The bondstone near Yeonghwadang was relocated in 1990 for the purpose of installing an imitation of Angbuilgu(仰釜日晷). Another odd-shaped stone has been relocated to the front door of the Secret Garden(後苑), and now it is difficult to grasp the location. Fourth, the two bondstones remaining in the Daebodanji were actually building materials that were used as the foundation stone for the entrance pillars of the Yi Royal Office Building(李王職廳舍) during the Japanese colonial period. After liberation, the Yi Royal Office Building was reorganized into Imperial Estate Administration Office(皇室財産事務總局), and when the office building was burned down in 1960, the stone statues and foundation stones placed on the stylobate were relocated to the Daebodanji. The bondstone at Daebodanji is a representative example of construction materiasl being mistaken for gaeden ornaments.

Roles of the Insulin-like Growth Factor System in the Reproductive Function;Uterine Connection (Insulin-like Growth Factor Systems의 생식기능에서의 역할;자궁편)

  • It has been known for a long time that gonadotropins and steroid hormones play a pivotal role in a series of reproductive biological phenomena including the maturation of ovarian follicles and oocytes, ovulation and implantation, maintenance of pregnancy and fetal growth & development, parturition and mammary development and lactation. Recent investigations, however, have elucidated that in addition to these classic hormones, multiple growth factors also are involved in these phenomena. Most growth factors in reproductive organs mediate the actions of gonadotropins and steroid hormones or synergize with them in an autocrine/paracrine manner. The insulin-like growth factor(IGF) system, which is one of the most actively investigated areas lately in the reproductive organs, has been found to have important roles in a wide gamut of reproductive phenomena. In the present communication, published literature pertaining to the intrauterine IGF system will be reviewed preceded by general information of the IGF system. The IGF family comprises of IGF-I & IGF-II ligands, two types of IGF receptors and six classes of IGF-binding proteins(IGFBPs) that are known to date. IGF-I and IGF-II peptides, which are structurally homologous to proinsulin, possess the insulin-like activity including the stimulatory effect of glucose and amino acid transport. Besides, IGFs as mitogens stimulate cell division, and also play a role in cellular differentiation and functions in a variety of cell lines. IGFs are expressed mainly in the liver and messenchymal cells, and act on almost all types of tissues in an autocrine/paracrine as well as endocrine mode. There are two types of IGF receptors. Type I IGF receptors, which are tyrosine kinase receptors having high-affinity for IGF-I and IGF-II, mediate almost all the IGF actions that are described above. Type II IGF receptors or IGF-II/mannose-6-phosphate receptors have two distinct binding sites; the IGF-II binding site exhibits a high affinity only for IGF-II. The principal role of the type II IGF receptor is to destroy IGF-II by targeting the ligand to the lysosome. IGFs in biological fluids are mostly bound to IGFBP. IGFBPs, in general, are IGF storage/carrier proteins or modulators of IGF actions; however, as for distinct roles for individual IGFBPs, only limited information is available. IGFBPs inhibit IGF actions under most in vitro situations, seemingly because affinities of IGFBPs for IGFs are greater than those of IGF receptors. How IGF is released from IGFBP to reach IGF receptors is not known; however, various IGFBP protease activities that are present in blood and interstitial fluids are believed to play an important role in the process of IGF release from the IGFBP. According to latest reports, there is evidence that under certain in vitro circumstances, IGFBP-1, -3, -5 have their own biological activities independent of the IGF. This may add another dimension of complexity of the already complicated IGF system. Messenger ribonucleic acids and proteins of the IGF family members are expressed in the uterine tissue and conceptus of the primates, rodents and farm animals to play important roles in growth and development of the uterus and fetus. Expression of the uterine IGF system is regulated by gonadal hormones and local regulatory substances with temporal and spatial specificities. Locally expressed IGFs and IGFBPs act on the uterine tissue in an autocrine/paracrine manner, or are secreted into the uterine lumen to participate in conceptus growth and development. Conceptus also expresses the IGF system beginning from the peri-implantation period. When an IGF family member is expressed in the conceptus, however, is determined by the presence or absence of maternally inherited mRNAs, genetic programming of the conceptus itself and an interaction with the maternal tissue. The site of IGF action also follows temporal (physiological status) and spatial specificities. These facts that expression of the IGF system is temporally and spatially regulated support indirectly a hypothesis that IGFs play a role in conceptus growth and development. Uterine and conceptus-derived IGFs stimulate cell division and differentiation, glucose and amino acid transport, general protein synthesis and the biosynthesis of mammotropic hormones including placental lactogen and prolactin, and also play a role in steroidogenesis. The suggested role for IGFs in conceptus growth and development has been proven by the result of IGF-I, IGF-II or IGF receptor gene disruption(targeting) of murine embryos by the homologous recombination technique. Mice carrying a null mutation for IGF-I and/or IGF-II or type I IGF receptor undergo delayed prenatal and postnatal growth and development with 30-60% normal weights at birth. Moreover, mice lacking the type I IGF receptor or IGF-I plus IGF-II die soon after birth. Intrauterine IGFBPs generally are believed to sequester IGF ligands within the uterus or to play a role of negative regulators of IGF actions by inhibiting IGF binding to cognate receptors. However, when it is taken into account that IGFBP-1 is expressed and secreted in primate uteri in amounts assessedly far exceeding those of local IGFs and that IGFBP-1 is one of the major secretory proteins of the primate decidua, the possibility that this IGFBP may have its own biological activity independent of IGF cannot be excluded. Evidently, elucidating the exact role of each IGFBP is an essential step into understanding the whole IGF system. As such, further research in this area is awaited with a lot of anticipation and attention.
