Isolation and Identification of Antimicrobial Active Substance from Sophora flavescens Ait. (고삼으로부터 항균활성 물질의 분리 및 구조 동정)

  • The ethanol extract and its chloroform fraction of Sophora flavescens Ait. exhibited growth inhibition on some food poisoning bacteria. The minimum inhibitory concentration of the above extracts were $50{\sim}500\;ppm$ and below 50 ppm Listeria monocytogenes (ATCC 19111, 19112, 19113, 19114, 15313). By silica gel column chromatography twice, antimicrobial active compound S-10-6 was isolated from chloroform fraction of Sophora flavescens Ait. The fraction S-10-6 showed strong growth inhibition at 10 ppm on 5 strains of L. monocytogenes, Bacillus subtilis ATCC 14593 and Staphylococcus aureus KFCC 11764 but Esherichia coli ATCC 25922 was not inhibited at 100 ppm and also confirmed bactericidal effect at 30 and 50 ppm on 5 strains of L. monocytogenes. The antimicrobial compound S-10-6 was identified as kushenol F, a kind of flavanone compound, by EI/MS, $^1H-NMR\;and\;^{13}C-NMR$.

Obovatol Extracted from Magnolia Obovata Inhibits Inflammation Mediator Generation and Prostate Carcinoma PC-3, LNCap Cell Growth Through Induction of Apoptotic Cell Death via Inactivation of $NF-{\kappa}B$ (후박에서 추출한 Obovatol의 염증매개 생성의 억제와 세포자멸사를 통한 Human Prostate Carcinoma PC-3 및 LNCap 세포의 세포증식에 대한 영향)

  • 목적 : 후박(厚朴)(Magnolia Obovata)에서 추출한 낮은 농도의 Obovatol 약침액의 RAW264.7 세포에서 LPS로 유발된 염증,$TNF-{\alpha}$로 유발된 human Prostate carcinoma PC-3 및 LNCap 세포의 세포증식에 대한 영향과 그 기전을 살펴보고자 하였다. 방법 : RAW264.7 세포에서 LPS로 염증을 유발하고 낮은 농도의 Obovatol 약침액을 처리한 후 cell viability, NO 생성량,iNOS와 COX-2의 발현, $NF-{\kappa}B$활성,전사능력을 관찰하기 위해 MTT assay, NO determination assay, western blot analysis, EMSA, luciferase activity assay를 시행하였고,LNCap, PC-3 세포에 $TNF-{\alpha}$로 증식을 유도하고 낮은 농도의 Obovatol 약침액을 처리한 후 cell growth, apoptosis 및 apoptosis와 연관된 $NF-{\kappa}B$의 활성 변화를 관찰하기 위해 WST-1, Cell morphogy test, DAPI staining and TUNEL assay, EMSA, luciferase activity assay를 시행하였다. 결과 : 1. RAW264.7 세포에서 낮은 농도의 Obovatol 약침액 처리는 $NF-{\kappa}B$의 활성 및 전사능력을 낮추고 iNOS와 COX-2의 발현과 NO 생성을 감소시켜 LPS로 유발된 염증을 억제하였다. 2. LNCap, PC-3 세포에서 낮은 농도의 Obovatol 약침액 처리는 $NF-{\kappa}B$의 활성을 낮추어 세포자별사를 촉진함으로써 $TNF-{\alpha}$로 유발된 암세포의 성장을 억제하였다. 결론 : 이상의 결과는 낮은 농도의 Obovatol 약침액이 항염 및 인간 전립선암세포주인 PC-3, LNCap에 대한 증식억제 효과가 있음을 입증한 것이며,향후 이를 바탕으로한 생체 연구에서의 긍정적인 결과는 Obovatol 약침액이 만성염증성 질환 및 전립선암의 예방과 치료에 대한 효과적인 치료제 개발에 초석이 될 것으로 기대된다.

Effects of nitric oxide on the proliferation and differentiation of human periodontal ligament cells (산화질소가 인간 치주인대세포의 증식과 분화에 미치는 영향)

  • Objective: This study evaluated the effects of nitric oxide (NO) on the proliferation and differentiation of human periodontal ligament cells involved in orthodontic tooth movement. Methods: A range of concentrations of sodium nitroprusside (SNP), a NO donor, were administered to samples of human periodontal ligament cells, followed by measurement of cell viability, alkaline phosphatase (ALP) activity, and expression of osteonectin and bone sialoprotein. Results: Cell viability, ALP activity and the expression of osteonectin and bone sialoprotein were increased in human periodontal ligament cells treated with SNP concentrations of < 0.2 mM compared with controls, but were decreased with SNP concentrations of > 1.0 mM. Conclusion: NO has a biphasic effect on proliferation and differentiation in human periodontal ligament cells, with a stimulatory effect at low concentrations and an inhibitory effect at high concentrations.

Effect of Extracts from Root Bark of Ulmus parvifolia on Inhibition of Growth and DNA Synthesis of Human Cancer Cells. (느릅나무 근피 추출물에 의한 인체 암세포 증식 및 DNA 합성 억제효과)

  • Growth and DNA synthesis inhibitory effects of extracts from root bark of Ulmus parvifolia on MG-63 human osteosarcoma cells, HT-29 human colon cancer cells and K-562 leukemia cancer cells were studied. The root bark extract of Ulmus parvifolia was extracted with methanol, hot water and juice. The methanol extract showed the highest inhibitory effect on growth of MG-63, HT-29 and K-562 cancer cells by >85%. The treatment of hot water and juice extracts from root bark of Ulmus parvifolia also inhibited growth of the above cancer cells with increasing concentration. DNA synthesis of MG-63 and HT-29 cancer cells was significantly inhibited by adding methanol, hot water and juice extracts from root bark of Ulmus parvifolia with increasing concentration, showing that the inhibitory effect of growth was more effective on HT-29 cancer cells. These results suggest that the methanol extract from root bark of Ulmus parvifolia may have specific active com-pounds on anticancer effect. The hot water extract also showed a strong inhibitory effect on growth of cancer cells, indicating that the active compounds may be stable to heat.

Effect of Spices on the Growth of Pathogenic Bacteria (향신료가 식중독세균의 증식에 미치는 영향)

  • The sensitivity of various pathogenic bacteria (Aeromonas hydrophila, Estherichia coli O157:H7, Listeria monocytogenes, Staphylococcus aureus 196E, Salmonella typhimurium and Vibrio parahaemolyticus) to the spices, allspice, clove, oregano, and thyme, was tested. Tryptic soy broth (TSB) containing 0∼2% (w/v) of spices was inoculated with 10sup 5/∼10$\^$6/ cells/$m\ell$ of each bacterium and incubated at 35$^{\circ}C$ for 24 hr. The growth of pathogenic bacteria was inhibited with increasing concentrations of spices in the culture broth. At 2% spice concentration, Gram positive bacteria were more sensitive than Gram negative bacteria with the exception of V. parahaemolyticus. Clove had the highest antibacterial activity, followed by allspice and oregano. At the concentration of 0.3%, clove inhibited the growth of all strains tested. Kanagawa-positive strain of V. parahaemolyticus displayed the highest sensitivity to clove and allspice. Thyme was the least effective for growth inhibition, while 1% clove killed all pathogens tested.

Immunomodulatory Effects of Polysaccharides Derived from Persimmon Leaves on Cyclophosphamide-induced Immunosuppressed Mice (감잎 유래 다당체가 면역억제 마우스의 면역조절에 미치는 영향)

  • We prepared 2 different crude polysaccharides from persimmon leaves, by hot water (PLW-0) and pectinase digestion (PLE-0), respectively. PLW-0 and PLE-0 showed similar sugar compositions and contained 11 different sugars, including rarely observed sugars such as 2-methyl-fucose, 2-methyl-xylose, apiose, and aceric acid. However, the uronic acid content of PLE-0 was lower than that of PLW-0, because of pectinase treatment. In normal mice, administration of PLW-0 and PLE-0 increased the spleen index and splenocyte proliferation. The effect of PLE-0 on the spleen index and splenocyte proliferation was greater than that of PLW-0. We subsequently assessed the immunomodulatory activities of PLW-0 and PLE-0 on cyclophosphamide (CY)-induced immunosuppressed mice. We revealed that mice treated with PLW- 0 or PLE-0 showed increased splenocyte proliferation, NK cell activity, and white blood cell numbers. Taken together, our results indicate that PLW-0 and PLE-0 can enhance immune function in normal mice and modulate CY-induced immune suppression.

Functional Properties of Hydrolysates Prepared from Agar Treated with Organic Acids (한천 유기산 가수분해물의 생리활성)

  • This study was concerning various physiological activities of agar hydrolysates. All agar hydrolysates showed strong antimicrobial activity against Bacillus subtilis and Bacillus cereus. Also, the agar hydrolysates prepared at the temperature of $110^{\circ}C$ or $120^{\circ}C$ showed antimicrobial activity against St. aureus and E. coli. Among the agar hydrolysates, several hydrolysates treated with citrate or malate at $110^{\circ}C$ or $120^{\circ}C$ conditions showed tyrosinase activity inhibition, and their inhibition rates of tyrosinase activity were about 80%. Some tested samples treated with 0.5% organic acid at $100^{\circ}C$ or $110^{\circ}C$ inhibited the growth of cancer cell. Two agar hydrolysates prepared with 0.5% citrate and lactate at $110^{\circ}C$ for $180^{\circ}C$ min had relatively high cancer cell growth inhibition among the tested samples. The agar hydrolysates treated with citrate and lactate at $110^{\circ}C$ for 180 min obtained the main peaks of six and seven from Sephadex G-15 column chromatography. Among the main peaks, the cancer cell growth inhibition of C-3 and L-3 fractions were higher than that of other fractions.

Physiological Activities of Commercial Instant Curry Powders and Individual Spices (시판 instant curry 및 curry 사용원료의 생리활성)

  • Physiological activities of hot water extracts of 10 commercial instant curry powders and 6 spices, were investigated. All spice extracts except ginger showed significant antioxidant activities on the autoxidation of linoleic curry acid (p<0.01). Antioxidant activities of clove and fennel were significantly higher than ${\alpha}-tocopherol$, instant curry powders, and other spices, Red pepper $(52.8{\pm}2.13%)$, clove, and coriander showed significant inhibitory activities against angiotensin-I-converting enzyme (p<0.001). Cytotoxic effects of instant curry powder and spices against human cancer cell lines were examined through MTT assay. Black pepper $(29.31{\pm}2.21%\;cytotoxic\;rate)$ and cardamon $(19.41{\pm}3.92%)$ were effective against MCF-7 (p<0.01), Clove $(42.92{\pm}5.57%)$ against HeLa (p<0.01). Ginger $(34.21{\pm}1.11%)$, cardamon, and black pepper against A172 (p<0.001), garlic $(82.88{\pm}0.53%)$ against SN12C (p<0.001), garlic $(71.63{\pm}0.38%)$, red pepper, ginger, fenugreek, SPC, cumin, and MPC against SNU-638 (p<0.001), and cassia $(82.84{\pm}16.92%)$ against A549 (p<0.001).

Bone Marrow Cell Proliferation Activity through Intestinal Immune System by the Components of Atractylodes lancea DC. (창출 성분의 장관면역 자극을 통한 골수세포 증식활성)

  • Of hot-water extracts prepared from 10 herbal components of Sip-Jeon-Dae-Bo-Tang, Atractylodes lancea DC. (ALR) and Panax ginseng C.A. Meyer (PG) showed the most potent bone marrow cell proliferation activity through intestinal immune system whereas other extracts did not have the activity except for Astragalus membranacues Bunge (ASR) and Angelica acutiloba Kitagawa (AR) having low activity. Especially, ALR had the potent activity irrespective of classes of ALR, a place of production and the condition of breeding. In addition, we found that hot-water extract from Atractylodes lancea DC rhizomes (ALR-0) contributed mainly to Peyer's patch cells mediated-hematopoietic response of Sip-Jeon-Dae-Bo-Tang. ALR-0 was further fractionated into MeOH-soluble fraction (ALR-1), MeOH-insoluble and EtOH-soluble fraction (ALR-2), and the crude polysaccharide fraction (ALR-3). Among these fractions, only ALR-3 showed potent stimulating activity for proliferation of bone marrow cells mediated by Peyer's patch cells, dose-dependently. In treatments of ALR-3 with $NaIO_4,\;NaClO_2$ and pronase, all significantly reduced the intestinal immune system modulating activity of ALR-3, and the activity of ALR-3 was much affected by $NaIO_4$ oxidation particularly. These results reveal that macromolecules, such as polysaccharide, rather than low-molecular-weight substances, are the potent intestinal immune system modulating compound of ALR.

Effects on Proliferation of Human Lung Cells induced by Fermented Silkworm thorn Fruits Based Kombucha (꾸지뽕-콤부차가 인체 폐세포 및 폐암세포의 증식에 미치는 영향)

  • 본 실험은 콤부차의 발효시 에너지원으로 첨가되는 당(sucrose) 대신 꾸지뽕(Cudrania tricuspidata Bureau; silkworm thorn) 과일 발효액을 첨가하여 꾸지뽕-콤부차의 기능성을 구명하고자 하였다. 대조구인 콤부차는 끓여서 식힌 물 900mL에 홍차 2.4g을 넣어 한시간 동안 추출한 후 초기당도가 10°Bx가 되도록 sucrose를 첨가하였고, 처리구는 sucrose대신 꾸지뽕 과일 무게 대 sucrose의 비율을 1 대 0.9의 비율로 조제하여 발효시킨 꾸지뽕 발효액(당도 50°Bx)을 10°Bx가 되도록 희석하여 첨가하였다. 여기에 발효균인 SCOBY를 첨가한 후 실온에 3주간 보관하면서 1주일 간격으로 시료를 채취하여 총폴리페놀성 화합물 및 카테킨류 함량, 항산화 활성 및 인체 정상 폐세포주인 MRC-5와 폐암세포주인 A549의 세포 증식에 미치는 영향을 구명하였다. 발효 3주 동안 채취한 꾸지뽕-콤부차를 MRC-5 세포에 처리하였을 때 발효 2주까지는 꾸지뽕-콤부차가 대조구에 비해 약 10~30% 세포 증식효과를 보였고 발표 3주째에는 유사한 증식효과를 보였다. 폐암세포주 A549에 처리시에는 발효 2주째 대조구에 비해 낮은 증식율을 보였으나 그 차이는 크지 않았다. 이 결과는 꾸지뽕-콤부차가 인체 폐세포 증식을 촉진하나 폐암세포의 증식을 크게 억제하지는 않음을 의미한다. 총폴리페놀성화합물 함량은 대조구의 경우 발효기간이 경과함에 따라 증가하는 반면 꾸지뽕-콤부차는 조제직후 대조구에 비해 유의적으로 높은 함량을 보이다 서서히 감소하였는데 발효 2주째 대조구와 유사한 수준에 도달하였으며 3주째에는 대조구에 비해 낮은 함량을 보였다. 카테킨류(Epigallocatechin, Epigallocatechin gallate, 그리고 Epicatechin gallate, epicatechin)는 총 페놀성화합물과는 반대의 경향을 보였는데, 발표 2주까지는 꾸지뽕-콤부차의 함량이 유의적으로 높았다가 발표 3주째 크게 낮아졌다. 활성산소 제거능은 발효 2주째까지는 대조구에 비해 낮았으나 3주째 유의적으로 높아져 꾸지뽕-콤부차의 항산화활성은 카테킨류 함량에 비례함을 알 수 있었다. 기능성분 함량과 MRC-5 증식에 관한 상관분석시 총풀리페놀함량이 세포증식에 정의 상관관계를 나타내었다.
