Effects of Medicinal Herb Extracts on Osteoblast Differentiation and Osteoclast Formation (한약재 추출물의 조골세포 분화 및 파골세포 형성에 미치는 영향)

  • Bone is continuously remodeled by osteoblasts and osteoclasts. We investigated the effects of medicinal herbs, which act on bone metabolism. Fifteen kinds of medicinal herb extracts were screened for bone formation activity with osteoblastic cells, and MC3T3-E1 and bone resorption were screened with osteoclasts derived from mouse bone marrow macrophages. Among these samples, Actinidia polygama, Eucommia ulmoides Oliv., Schizonepeta tenuifolia, Sorbus commixta, and Zingiber officinale Rosc. extracts showed strong bone-forming activity accompanied with osteoblast proliferation and alkaline phosphatase activity. In addition, these extracts decreased tartrate-resistant acid phosphatase activity against osteoclast differentiation. The results indicate that these medicinal herb extracts can potentially prevent bone-related diseases such as osteoporosis by increasing osteoblast differentiation and reducing osteoclast activity.

Effect of Pine (Pinus densiflora) Needle Extracts on Synthesis of Collagen in Osteoblastic MC3T3-E1 Cells (적송잎 추출물이 조골세포의 collagen 합성에 미치는 영향)

  • Osteoporosis is a disease involving a decrease in bone mineral density and an increased risk of fractures. The MC3T3-E1 pre-osteoblastic cell line is a well-accepted model of osteogenesis in vitro. Pine needles have long been used as a traditional health-promoting medicinal food in Korea. In this study, MTT assay, the alkaline phosphatase (ALP) activity and collagen synthesis of osteoblast cells were investigated to determine the effects of pine needle extracts on cell proliferation and differentiation. Pine needle extracts were prepared using hexane, ethanol and water. The effects of the pine needle extracts were examined by comparing the results with those of commercial agents, such as proanthocyanidin. The MC3T3-E1 cells exposed to proanthocyanidin showed increased proliferation in a concentration-dependent manner. The cells exposed to the hexane extract showed a similar increase in proliferation to that observed with proanthocyanidin. The hexane extract showed the highest ALP activity. Moreover, a supplement of pine needle extracts induced collagen synthesis in MC3T3-E1 cells. The pine needle extract produced the highest level of collagen synthesis at concentrations of $10{\sim}50\;{\mu}g/ml$. These results indicate that pine needle extracts have an anabolic effect on bone by promoting osteoblastic differentiation, and may be used in the treatment of common metabolic bone diseases.

The Effects of Medicinal Herbs Extracts on Estrogen-like Activities and Osteoblast Proliferation and Differentiation (한약재 추출물의 에스트로겐 유사활성 및 조골세포 증식과 분화에 미치는 영향)

  • The purpose of this study was to investigate the effect of 3 types of medicinal herbs (Glycyrrhizae radix, Astragali radix and Dioscorea rhizoma) extracts on estrogen-like activities, proliferation and differentiation in osteoblast. Human breast cancer cell line MCF7 was transfected using an estrogen responsive luciferase reporter plasmid for measure the estrogen-like activity. Estrogen-like activities of extracts were in the range of 1.11~5.73 fold to that of negative control. The extract of G. radix showed the strongest estrogen-like activities. The estrogen-like activities of 50 and $500{\mu}g/ml$ extracts of G. radix were similar to that of $10^{-8}$ and $10^{-7}$ M standard solution ($17{\beta}-estradiol$), respectively. G. radix extract showed no cytotoxicity against osteoblast MC3T3-E1 cells at $1{\sim}1,000{\mu}g/ml$. The extract of A. radix showed no significant proliferation of osteoblast. However, the extract of G. radix and D. rhizome showed maximum 148% and 133% proliferation effects. The extract of G. radix also increased alkaline phosphatase activity and the maximum was 122% at $100{\mu}g/ml$ compared to that of control. The nodule formation by the method of the Alizarin red S staining increased compared to control. These results suggest that G. radix is able to perform the bone formation and prevent osteoporosis.

  • 생쥐 착상전 초기배아에서 insulin과 tumor necrosis factor $\alpha$ (TNF$\alpha$)에 의한 배아의 형태 발생, 세포증식, apoptosis 및 MAPK활성의 변화를 조사하였다. Insulin에 의해 형태발생 및 포배당 세포수가 증가되었으며 TNF $\alpha$ 처리시 유의하게 감소하였다. TNT$\alpha$ 전처리시 insulin에 의한 발생 및 세포수 증가 촉진효과가 상쇄되었으며 TNF$\alpha$는 배아내 caspase-3의 활성을 증가시켰다. Insulin은 단시간내에 포배에서 mitogen activated protein kinase (MAPK or Erk1/2)의 활성을 증가시켰다. TNF$\alpha$는 포배내 MAPK의 활성을 감소시켰다. Insulin 처리 전 TNF $\alpha$를 전처리한 경우 insulin에 의한 MAPK 활성의 증가가 상쇄되었다. 배발생을 촉진하는 인슐린 신호전달 과정은 MAPK cascade를 경유하며 TNF $\alpha$를 경유한 신호전달과 Crosstalk이 존재하는 것으로 사료된다.

  • 병윈성 비브리오균의 병원인자에는 hemolysin, protease, phospholipase A2, siderophore 외에도 콜레라균 만이 생산하는 cholera toxin 등이 있다. 이 중에서도 대부분의 병원성 비브리오균에서 생성되는 대표적인 인자는 hemolysin과 protease로 알려져 있다.   • Growth characteristics of sourdough lactic acid bacteria was investigated to obtain basic informations for sourdough starter. The optimum temperature and pH on bacterial growth and lactic acid production of Lactobacillus brevis UC-22 in sourdough broth were 35'E and around pH 5.5, respectively. And the optimum concentrations of the carbohydrate sources added to the broths was 2% maltose. The acidity significantly increased during growth by Lactobacillus brevis UC-22 fur 18 hours while pH significantly decreased during growth.

Anticancer and Antioxidant Effects of Solvent Extracts from Dried Onion with Different Drying Methods (건조방법에 따른 건조 양파 추출물의 항암 및 항산화 효과)

  • We investigated to determine the inhibitory effects of solvent extracts from dried onion on growth of cancer cell lines (HT-1080 human fibrosarcoma and HT-29 human colon cancer cells) and $H_{2}O_{2}$-induced oxidative stress. Two different drying methods, low temperature vacuum dryer and freeze dryer, were employed to dry onion. Inhibitory effects of acetone with methylene chloride (A+M) and methanol (MeOH) extracts from onion by two drying methods on the growth of HT-1080 and HT-29 cancer cells increased in a dose dependent manner (p<0.05) and the higher inhibitory effect was shown in onion extracts dried by low temperature vacuum dryer. The treatments of hexane, 85% aq. methanol, butanol and water fractions significantly inhibited the growth of both cancer cells (p<0.05) and onion fractions dried by freeze dryer showed a higher inhibitory effect compared with those dried by low temperature vacuum dryer. In order to determine a protective effect on H2O2-induced oxidative stress, DCHF-DA (dichlorodihydrofluorescin diacetate) assay was conducted. All fractions including crude extracts of dried onion appeared to significantly reduce the levels of intracellular reactive oxygen species (ROS) (p<0.05). Higher antioxidant effect was observed in onions dried by the low temperature vacuum dryer method. These results indicate that the low temperature vacuum dryer is useful to dry and produce onion powder.

The Glucosinolate and Sulforaphane Contents of Land Race Radish and Wild Race Radish Extracts and Their Inhibititory Effects on Cancer Cell Lines (재래종 무와 갯무 추출물의 암세포주 증식 저해 활성 및 Glucosinolate와 Sulforaphane의 함량)

  • The inhibitory effects of land race radish (LRR) and wild race radish (WRR) extracts on cancer cell lines were investigated. A and their glucosinolate and sulforaphane contents were analyzed. The anticancer activitiesy of the LRR and WRR extracts on the breast cancer cell line MCF-7 were determined by a CCK (cell counting kit) assay, in which WWR showed higher inhibition rates than LRR. The sulforaphane content of WRR was higher than that of LRR. In the lung cancer cell line, A-549, WRR showed higher inhibition rates and a higher total glucosinolate content than LRR. The glucosinolate contents of the radishes were analyzed by the Pd-quicktest method, showing that WRR contained more glucosinolate than LRR in both the trunk and root. In conclusion, these results indicate that wild race radish could be used for the quality improvement of radishes.

Effect of Solvent Extracts from Sargassum hemiphyllum on Inhibition of Growth of Human Cancer Cell Lines and Antioxidant Activity (짝잎모자반(Sargassum hemiphyllum)의 암세포주 증식 억제 및 항산화 효과)

  • This study was carried out to determine the inhibitory effects of solvent extracts from Sargassum hemiphyllum on growth of cancer cell lines (AGS human gastric adenocarcinoma and HT-29 human colon cancer cells) and production of lipid peroxides. Inhibitory effects of acetone with methylene chloride extract from S. hemiphyllum on the growth of AGS and HT-29 cancer cells were increased as dose dependent patterns (p<0.05). The methanol extract was more effective on inhibition of growth of AGS. The treatments of hexane, 85% aq. methanol, butanol and water fractions significantly inhibited the growth of cancer cells (p<0.05) and the inhibitory effect was stronger in HT-29. In DCFH-DA (dichlorodihydrofluorescin diacetate) assay, acetone with methylene chloride and methanol extracts showed a stronger inhibitory effect on the production of cellular lipid peroxides (p<0.05) compared with the butanol and hexane fractions. These results indicate that the consumption of S. hemiphyllum may be recommended as a potent functional food for preventing cellular oxidation and cancer.

Apoptotic Effects of Curcumin and EGCG via Akt-p53 Signaling Pathway in HCT116 Colon Cancer Cells (HCT116 대장암 세포에서 Akt-p53 신호경로를 통한 커큐민과 EGCG의 apoptosis 효과)

  • p53 is tumor suppressor gene that regulates apoptosis such as caspase-dependent and p21-mediated signaling pathways. PI3K/Akt is known to be over-activated in cancer cells. Akt activates many survival-related signals such as mTOR and COX-2. Inactivation of Akt would result in non-inhibition of p53 as well as induced apoptosis. In this study, we showed that curcumin and EGCG activate p53 via inhibition of the Akt signaling pathway. Treatments using curcumin and EGCG in different concentrations for 24 hr and 48 hr inhibited proliferation of HCT116 colon cancer cells and increased apoptotic cell death. Also, our data showed that curcumin and EGCG increased the p53 expression and decreased the p-Akt. Treatment of LY294002 (Akt inhibitor) resulted in decreased cell proliferation of cancer cells, while LY294002 treated with curcumin or EGCG showed a greater decrease of cell proliferation. In addition, inhibition of Akt induced p53 activation in HCT116 colon cancer cells. These results suggest that curcumin and EGCG induce apoptosis by inhibiting Akt and increase p53 in HCT116 colon cancer cells.
