Effect of Ethanol Extracts of Goat's Beard on Streptozotocin Induced Diabetic Symptoms and Oxidative Stress in Rats (삼나물 에탄올 추출물이 Streptozotocin으로 유발한 흰쥐의 당뇨증상과 산화적 스트레스에 미치는 영향)

  • The effects of goat's beard (Aruncus dioicus var ramtschaticus Hara) ethanol extracts (AD) on the streptozotocin-induced diabetic symptoms and oxidative stress in rats were assessed. Experimental plots were divided into normal controls (NC), diabetes controls (DM), diet with 0.3% AD after diabetes induction (SA), and diet with 0.6% AD after diabetes induction (SB), then fed for 5 weeks. In the SA and SB groups as compared with the DM group, diabetic symptoms including weight loss, increase in feed and water intake, and enlargement of the liver and kidney were improved. The levels of blood glucose and serum fructosamine were reduced by $17.9{\sim}27.2%$ and $25.6{\sim}32.6%$, respectively. The activities of alanine aminotransferase, aspartate aminotransferase and the levels of triglycerides, total cholesterol, and LDL-cholesterol in the serum were reduced by $25.6{\sim}30.3$, $42.37{\sim}55.51$, $26.85{\sim}30.44$ and $37.29{\sim}39.11%$, respectively, whereas the level of HDL-cholesterol was increased by $37.29{\sim}39.11%$. Hepatic total (T) and O type (O) activities of xanthine oxidoreductase, O/T ratio(%) and level of lipid peroxide were significantly decreased, while the level of glutathione was increased, and also the activities of superoxide dismutase and glutathione S-transferase were increased by $56.84{\sim}94.90%$ and $57.14{\sim}68.92%$, respectively. In the above results, it was noted that AD has an antidiabetic effect which alleviated hyperglycemia and the AD reduced and/or prevented the tissue damage caused by diabetes yia the inhibition of reactive oxygen species (ROS) generating systems concurrent with an increase in ROS scavenging.

Areal Distribution Ratios of Constituent Rocks with Geologic Ages and Rock Types by GIS in the Gyeongsangbug-Do and Daegu Areas (GIS에 의한 경북-대구지역 구성암류의 지질시대별 및 암종별 분포율)

  • On the ArcGIS 9.2 program in Gyeongsangbug-Do and Daegu areas, distribution ratios of rock types and geologic ages were obtained from the 1 : 250,000 scaled digital geologic and geomorphic maps. The obtained distribution ratios here will be used the geologic information data for industrialization and development planning of rock resources. The Gyeongsangbug-Do area consists of 86 rock types that can be divided into 10 large groups in geologic age. Their geologic distribution ratios show the decreasing in the order of Cretaceous, Precambrian, Jurassic, Quaternary, Age-unknown and Tertiary, all of which occupy the prevailing ratio of 96.30% in the area. Of which, sixteen rock types are somewhat dominant ones (64.04%). They are of Precambrian Yulri group and granite gneiss of the Yeongnam metamorphic complex and biotite gneiss of the Sobaegsan metamorphic complex, Age-unknown granite, Jurassic granite, Cretaceous Gasongdong and Dogyedong formations of the Yeongyang sub-basin, Nagdong and Chunsan formations and intermediate-basic volcanics of Euiseong sub-basin, Jinju and Jindong formations and andesite-andesitic tuff of Milyang sub-basin, and hornblende granite, and Quaternary alluvium. They show relatively narrow ranges of 2.07-6.53% in geologic distribution in exception of Jurassic granite showing 13.14%. And the rest 70 rock types appear to very narrow range between 0.01 and 1.94 %. On the other hand, twelve rock types are developed in the Daegu area. Their geologic ages appear to be classified into Cretaceous and Quaternary occupying 86.05% and 11.39%, respectively. Seven rock types take possession of 94.04% among the all rocks. The major rock types are Jinju formation of the Sindong group, Chilgog, Haman and Jindong formations of the Hayang group, andesite and andesitic tuff, hornblende granite and Quaternary alluvium. With exception of andesite and andesitic tuff of 37.40%, the types show slightly wide range of 3.25-17.39%, which apparently differ trends from that of Gyeongsangbug-Do area. And the rest of rock types have narrow ranges of 0.22-1.81% in the Daegu area.

Effects of Sasa coreana Nakai on the Lipid Compositions of Serum in High Cholesterol Diet Rats (신의대(Sasa coreana Nakai) 추출물이 고콜레스테롤 식이 투여 흰쥐의 혈청 지질에 미치는 영향)

  • This study was performed to investigate the effects of Sineui-Dae (Sasa coreana Nakai, a kind of bamboo) extracts on the improvement of serum lipid composition by using rats fed a hypercholesterol diet for 4 weeks. The experiment animals were administered with the following diets; high cholesterol diet group (HC diet) as a control and three supplemented groups with high cholesterol diets (HCW, HCM or HCH diet). Three kinds of extracts were prepared by orderly extraction with hexane, methanol, and water. We measured free cholesterol, cholesteryl ester, total cholesterol LDL- and HDL-cholesterol triglyceride and phospholipid in the serum of rats in three experimental and control groups. There were no significant differences in body weights and feed intakes between the HC control group and the extract supplemented groups. The levels of total cholesterol and LDL-cholesterol in the serum of extract supplemented groups were lower than that of the group fed with HC diet only. Serum HDL-cholesterol levet which is known as an antiatherosclerosis factor, was higher in all groups supplemented with bamboo extract by 17.2-21.9% compared to the HC control group. All groups supplemented with bamboo extracts showed the lowering effect of atherogenic index compared to the HC control group (HC group: 2.96${\pm}$0.08, HCM group: 1.48${\pm}$0.02, HCW group: 1.69${\pm}$0.04, HCH group: 1.84${\pm}$0.01). Furthermore, serum triglyceride and phospholipid decreased significantly in the HCM diet compared to the HC control diet. These results suggest that Sineui-Dae bamboo extract, especially methanol extract, has improving effects on hyperlipidemia of rats fed a high cholesterol diet.

Effect of Medicinal Herb Composites on Antioxidative and Cognition-Enhancing Activities in Rats (생약복합물이 흰쥐의 체내에서 항산화 및 인지개선활성에 미치는 영향)

  • The purpose of this experiment was designed to investigate the effects of medicinal herbs (MH) extracts on dementia induced by trimethyltin chloride (TMT) in rats. Six-week-old male Sprague-Dawley rats were randomly divided into five groups; normal group (group 1), control group (group 2), MH extracts group (250, 500 mg/kg) (group 3, group 4) and positive control group (tacrine group, group 5). In the control group to induce dementia, a 2.5 mg/kg of TMT intraperitoneal injection was used for 14 days (1 per day) in the rats. In the MH extracts group 250 mg/kg and 500 mg/kg of MH extracts were medicated in an oral inoculation for 20 days (1 per day). After 30 minutes, a 2.5 mg/kg of TMT intraperitoneal injection, which causes dementia, was used for 14 days (1 per day). In the positive control group (Tacrine group) 10 mg/kg of Tacrine, the dementia treatment, was medicated in an oral inoculation. After 30 mintues, 1 mg/kg of TMT intraperitoneal injection, which causes dementia, was used for 14 days (1 per day). The present author observed the passive avoidance performance test, and memory ability test (Y maze test), the values of MDA, acetlycholinesterase (AchE) activity in the brain and antioxidant enzyme in serum. MH extracts significantly improved memory of AD model rats in the Y-maze test, and also significantly improved memory of AD model rats in the passive avoidance test. MH extracts significantly reduced AChE activity, and significantly increased the SOD level, but not catalase and MDA. From the results above, MH extracts is thought to be effective in the improvement of antioxidant enzymes and memory ability.

  • By modified method yellow corvenia(called $Y_3$) was prepared with $4\%$ salt, $4\%$ KCl, $6\%$ sorbitol, $0.5\%$ lactic acid and $4\%$ alcohol extract of red pepper to improve the quality of fermented sea food. In this study, changes of volatile compounds and fatty acid composition obtained from modified fermented yellow corvenia($Y_3$) were experimented during fermentation, comparing with conventional fermented yellow corvenia(called $Y_1,\;20\%$ of salt contents). Total lipid of yellow corvenia was composed of $78.1\%$ of neutral lipid, $21.2\%$ of phospholipid and $0.7\%$ of glycolipid. And monoeonoic acid was held $37.4\%$ of fatty acid composition of total lipid and saturated fatty acid ($34.8\%$), polyenoic acid ($27.7\%$) were followed. Saturated fatty acid($C_{14:0},\;C_{16:0},\;C_{18:0}$) in $Y_1,\;Y_3$ increased, polyenoic acid ($C_{22:6}\;C_{22:5}\;C_{20:5}$) decreased while monoenoic acid($C_{16:1}\;C_{18:1}$) in those was little fluctuated during fermentation. Thirty-three kinds of volatile component in whole volatile compounds obtained from $Y_1,\;Y_3$ at 90 days fermentation were identified, and composed of some hydrocarbons (8 kinds), alcohols (7 kinds), acids (6 kinds), aldehydes(4 kinds), sulfides(2 kinds), ketones (2 kinds), one of phenol and 3 kinds of the other components. Among the whole volatile compounds 2-ethoxy ethanol and was held $79.35\%$ in $Y_3$, whereas nonadecane was held $75.85\%$ in $Y_1$. During fermentation 8 kinds of volatile acids, 5 kinds of amines and 9 kinds of carbonyl compounds were also detected. Those volatile acid such as acetic acid, isovaleric acid, n-caproic acid, n-butyric acid were the major portion of total volatile acids in $Y_3$ at 90 days fermentation. Meanwhile, carbonyl compounds such as ethanal, 2-butanone and butanal were the major ones, while TMA held the most part of volatile amines in $Y_3$ during fermentation. From the result of sniff test, the components which are believed to contribute to the characteristic flavor of fermented product $Y_1,\;Y_3$ are deduced to be volatile acid, carbonyl compounds and amines in order. Conclusively, there was little difference in composition of volatile components, but merely a little difference in content of those between $Y_3$ and $Y_1$.

Gamma Irradiation Effects on Conchospores of Porphyra Species 2. The Effects of High Gamma Irradiation on Germination and Growth of Conchospores of Two Varieties (김의 각포자에 대한 r-선의 조사효과 2. 두 품종의 각포자의 발아생장에 미치는 고선량 r-선의 조사효과)

  • For the fundamental studies of radiation breeding in edible marine algae, the biological effects on conchospores of Porphyra species by gamma-irradiation were examined. Two varieties, Keun-cham-gim (Porphyra tenera Kjell. form tamatsuensis Miura) and Saga No.5, were chosen for this study, and their conchospores after r-irradiation($5.0{\sim}20.0$ KR) were cultured for 50 days. The results obtained were summarized as follows. 1. Gamma-irradiation in less than the dose of 20KR did not affect germination of conchospores, and almost all spores grew into two cells germ in 24 hours after irradiation, but withering germs were gradually increased in number according to higher exposure within 5 days old culture. 2. High irradiation caused the induction of giant cells, abnormal useless growth of hold-fast, lumpish thalli and callus-like lumpy tissues. 3. The liberation of neutral spores from young germs and carpospores from mature thalli were observed on the frond exposed at $10{\sim}20$ KR irradiation. All spores were normal in division and its size. 4. The best irradiation effect on growth of Keun-cham-gim was observed at 10 KR dose, whose growth-rates were $140\%$ in wet weight and $108\%$ in mean frond area, but only $48\%$ was recorded in wet weight at 20 KR exposure. Saga No.5 were in contrast with Keun-cham-gim, and their most growth-rate was $400\%$ in wet weight ($258\%$ in frond area) at 10 KR irradiation and the worst was $20\%$ at the dose of 20 KR. 5. The withering phenomenon to death by treatment of gamma-ray presented substantial difference between two varieties. Survival rate compared with control in Keun-cham-gim was $70.7\%$ at 20 KR, but that in Saga No.5 recorded $47.0\%$ at same dose. 6. Synthesizing the results of high and low r-irradiation, it was suggested tat high r-irradiation in more than 5.0 KR inhibited conspicuously the growth of germs derived from conchospores, and about half of them withered at 15.0 KR dose or more.

Effect of Korean Red Ginseng Powder on the Lipid Concentrations and Tissue Lipid Peroxidation in the Rats Fed High Fat Diet (고지방급여 흰쥐의 혈청과 간의 지질 농도 및 조직 과산화지질 농도에 미치는 홍삼분말의 영향)

  • Effect of Korean red ginseng (KRG) on the level of serum and liver lipids and lipid peroxidation was investigated in the rats fed high fat diet. Content of serum total cholesterol was significantly decreased (P<0.05) in KRG I group and KRG II group. Content of HDL-cholesterol was significantly increased by 69.75% and 39.15% in KRG I and KRG II group compared to control group, respectively. Atherogenic index (hi) was also significantly decreased by 74.76% and 37.38% in KRG I and KRG II groups compared to control group, respectively. Serum triglyceride content was significantly decreased (p<0.05) in only KRG II group. Antioxidative activity of KRG on the lipid peroxidation of serum and tissues in rats was also studied in vivo by measuring the formation of thiobarbituric acid reactive substances (TBARS). Contents of TBARS in the serum of both KRG groups were significantly decreased (p<0.05) and that of nonheme iron in serum was significantly increased (p<0.05) in a dose-dependent manner, which suggested that lipid peroxidation contents are inversely correlated with serum nonheme iron content. Content of TBARS in liver was significantly decreased (p<0.05) in KRG I and KRG II groups, without any influence in other tissues. Content of TBIARS in liver microsomal fractions stimulated by Fe$^{2+}$/ascorbate was significantly decreased (p<0.05) in KRG I and KRG II groups, whereas this observation did not occur in liver mitochondrial fractions. When the effect of KRG on TBARS content in the liver fractions of homogenates, microsomes, and mitochodria stimulated by Fe$^{2+}$/ascorbate was tested in vitro experimental model, TBARS of liver three fractions was significantly decreased at 6 mg/mL KRG compared with those of control. These results suggested that KRG powder have hypocholesterolemic effect as well as antioxidative effect in the serum and liver of the rats fed high fat diet.

Food Components of Aldaegu(Salted and Dried Cod) (알대구의 식품성분(食品成分))

  • This study was attempted to evaluate the chemical components of Aldaegu, Gadus macrocephalus, which consumed as a salted and dried product popularly in Korea. The contents of such compounds as amino acids, nucleotides and their related compounds, fatty acids and minerals were analysed. The content of total free amino acids was 814.9 mg% on dry basis and the major amino acids were tyrosine, alanine, histidine, leucine, phenylalanine, lysine and valine in a decreasing order. These amino acids were resulted as 45% of total free amino acids in Aldaegu. In the nucleotides and their related compounds, inosine and hypoxanthine were the only compounds detected. The overall content of nucleotide and their related nitrogenous compounds, free amino acid-N., ammonia-N., creatine and creatinine-N., and betaine-N., was 78.6% of extractable nitrogen. Ammonia-N was the most abundant of other nitrogenous compounds, resulting 36.6% of extractable nitrogen from the sample. In the fatty acid composition of total lipid and phospholipid, polyenoic fatty acid was abundant holding about 48% respectively. However, higher amounts of saturated fatty acids(39.8%) were found in glycolipid, and the predominant fatty acids in quantity were palmitic $acid(C_{16:0})$, stearic acid $(C_{18:0})$, oleic $acid(C_{18:1})$, docosahexaenoic $acid(C_{22:6})$, eicosapentaenoic $acid(C_{20:5})$ and eicosatetraenoic $acid(C_{20:4})$. Aldaegu contained 27,430 ppm of sodium and the trace amount of lead and cadmium was detected, which gave a good safety in the food sanitation aspects. It was presumed that free amino acids, ammonia, hypoxanthine, betaine, creatine and creatinine, and minerals play an important role for the characteristics of taste and flavor of Aldaegu.

Effect of Broccoli Sprouts on Cholesterol-lowering and Anti-obesity Effects in Rats Fed High Fat Diet (브로콜리 싹 에탄올 추출물이 고지방식이를 급여한 흰쥐의 콜레스테롤 저하 및 비만 억제효과에 미치는 영향)

  • This study was conducted to investigate the cholesterol lowering and anti-obesity effects of an ethanol extract of broccoli sprouts (BS) in rats fed high fat diet. Male Sprague-Dawley rats weighing $150{\sim}155g$, were divided into 6 groups; a normal diet group (ND), a high fat diet group (HFD), a normal diet and BS with 200 mg/kg treated group (ND-BSL), a normal diet and BS with 400 mg/kg treated group (ND-BSH), a high fat diet and BS with 200 mg/kg treated group (HFD-BSL), and a high fat diet and BS with 400 mg/kg treated group (HFD-BSH). The body weight gain and mesenteric adipose tissue weight were increased by high fat diet, but gradually decreased to the corresponding level of ND group after administration of BS extract. The liver and epididymal adipose tissue weights of HFD group were the highest among the six groups, although the difference was not significant. Food intake was lower in high fat diet groups compared with normal diet groups. The serum ALT and AST activities that were elevated by the high fat diet were significantly decreased after BS administration. Levels of serum total cholesterol, LDL-cholesterol, triglyceride and the atherogenic index tended to be decrease in the BS administered groups compared with HFD group. However, HDL-cholesterol level in serum decreased in HFD group and markedly increased in BS administered groups. There were no differences in the contents of serum triglyceride, LDL-cholesterol and HDL-cholesterol between normal diet groups. Levels of total cholesterol and triglyceride in liver and adipose tissues were also lower in BS administrated groups than in HFD group. The activities of heparin-releasable lipoprotein lipase (HR-LPL) and total-extractable LPL (TE-LPL) in adipose tissue were increased in HFD group compared with the BS administered groups, but those of the ND-BSL group and ND-BSH group were similar to ND group. These results suggest that BS ethanol extract may exert cholesterol-lowering effect and potentially reduce lipid storage.

Clinical Characteristics of Pulmonary Tuberculosis Presenting Prolonged Fever Despite Primary Short-Course Anti-tuberculosis Treatment (1차 항결핵약제 치료 후에도 지속적으로 발열을 보인 폐결핵 환자의 임상상)

  • Background : Usually fever subsides within one week in over 90% of pulmonary tuberculosis (TBp) patients after the start of short-course anti-tuberculosis therapy, but occasionally it persists over two weeks after treatment. When the fever persists, drug resistance, combined infection, or drug fever, and so on, are considered as an etiology and, in some cases, drugs are changed. But inadvertent discontinuation of a short-course regimen inevitably will extend the duration of treatment, and the treatment completion may be delayed. This study was performed to investigate the causes of prolonged fever (PF) and to identify the predictors of PF in drug-susceptible TBp patients in Korea. Method : Five hundred-ninety-eight patients, who were admitted to Asan Medical Center from January 1996 to March 1999, diagnosed with TBp and prescribed short-course, anti-tuberculosis treatment, were reviewed. PF was defined as having fever over two weeks despite treatment. The causes of PF were analyzed. Drug-sus-ceptible TBp patients who presented no causes for PF, except turberculosis itself, were selected(n=22), and they were compared with those who had no fever at diagnosis (n=22) and those who had fever at diagnosis, which had subsided within two weeks after treatment (n=22). Clinical, laboratory, and radiological parameters were compared among the three groups. Results : Twenty-eight (4.8%) of 598 patients showed PF over two weeks despite short-course treatment. The causes of PF were drug fever (n=2), multi-drug resistant tuberculosis (n=3), disseminated Mycobacterium kansasii infection (n=1), and drug-susceptible tuberculosis itself (n=22). The patients with PF had more risk factors for tuberculosis, long duration of symptoms before treatment, night sweats, weight long, numerous acid fast bacilli on sputum smear, anemia, hyponatremia, hypoalbuminemia, over three lung cavity numbers and extensive infiltration, indicating that they had prolonged and extensive lung diseases. Conclusion : The main cause of PF in TBp despite short-course regimen seems to be drug-susceptible but extensive disease in Korea. Any changes to the drug regiment provided for TBp patients with prolonged fever despite treatment should be carefully considered.
