Change in the Bourse Shoot and Fruit Growth due to the Gibberellins Paste in the Young Fruit of Niitaka Pear (유과기 지벨렐린 도포제 처리에 의한 '신고'배의 과대지와 과실생장 변화)

  • The object of this study was to evaluate the effect of gibberellins (GA) pasting on the bourse shoot, and not just on the fruit characteristics but also on the quality, of Niitaka pear (Pyrus pyryfolia L). The fruit stalk was treated with GA (control, 25mg/fruit stalk) 35 days after reaching full bloom. In the GA-treated tree, the occurrence of abnormal bourse shoot (52.5%) increased, and the spur bud and flower number decreased. The diameter of the GA-paste-treated fruit increased during the pear growth period with GA treatment. The diameter, length, and weight of the GA-paste-treated fruit increased at harvest time, but the hardness was lower than that of the control. The differences in soluble solid, acidity, and fruit color between the control and the GA-paste-treated fruit were not significant. Post-harvest, during the storage period, the hardness of the GA-paste-treated fruit was lower than that of the control, and its weight loss ratio was higher than that of the control on the $60^{th}$ days of storage.

Production of High-Level Polyphenol Powders from Young Grape Leaves (어린 포도 잎을 이용한 폴리페놀 고함유 분말 제조)

  • Polyphenols are naturally occurring antioxidants and play many important roles in human health. Grape leaves have not yet been reported to contain the antioxidants. In this study, the physicochemical properties of powders from young grape leaves, obtained after hot-air or vacuum freeze drying, were analyzed. Weight loss rangedfrom 76-80%, but no difference was found between samples prepared by hot-air orvacuum freeze drying. The L-, a-, and b- values of grape powder all fell upon hot-air drying. Quercetin, resveratrol, catechin, and epicatechin were detected in all cultivars. The contents of the four polyphenols were generally higher in powders obtained by vacuum freeze dryingthan in those prepared by hot-air drying, indicating that vacuum freeze drying is better for optimal preservation of polyphenols. None of three polyphenols assayed was found in some cultivars or after certain drying treatments. Based on physiochemical properties, our findings suggest that powders obtained after vacuum freeze drying are optimal for industrial use.

Study on the Improved Abrasion Resistance of Polycarbonate Substrate by UV-curable Organic/Inorganic Hybrid Coatings (자외선 경화형 유기/무기 복합코팅에 의한 폴리카보네이트의 내마모성 향상 연구)

  • Transparent, abrasion resistant coatings with 4~13 ${\mu}{\textrm}{m}$ thickness were prepared by spin-coating on polycarbonates with organic/inorganic hybrid solutions, followed by UV curing and heat treatment at 12$0^{\circ}C$ for 12 hours. The coating solutions were composed of inorganic phase and organic phase in 0:100, 20:80, 30:70, 50:50, 80:20 wt% ratios, respectively, mixed with photoinitiator, senaitizer and surfactant. The inorganic phase was formed by sol-gel reaction of TEOS and silane coupling agent MPTMS in 1 : 2 or 2 : 1 molar ratios, the organic phase consisted of difunctional urethane acrylate oligomeric resin, multifunctional acrylate TMPTA and HDDA in 4 : 3 : 3 wt% ratio. The coating systems were investigated by FT-IR, $^{29}$ Si-NMR spectra. In addition, TGA/DSC for thermal analysis and SEM, AFM observation for coated surface were examined. Gererally, the homogeneity of phases, the surface smoothness of coating and abrasion resistance were improved with the higher content of inorganic component. Namely, coating system with below 10 $\AA$ surface roughness and T$_{g}$ of 15$0^{\circ}C$ showed only 10% decrease in light transmittance after abrasion test, whereas uncoated polycarbonate substrate exhibited 46% decrease..

Effects of Different Carbon Dioxide and Oxygen Concentration on the Quality of Oriental Melon during CA Storage (참외 CA저장시 $CO_2$$O_2$농도에 따른 품질 변화)

  • Oriental melon was stored in controlled atmospheres(5, 10 or 15% CO$_2$in combination with 3 or 10% O$_2$) and air as control, and was analyzed half monthly during a 45-day storage for various quality Parameters. Oriental melon which stored in CA condition was lower in weight loss and decay rate than that stored in air. There was no significant difference in firmness by CO$_2$or O$_2$concentration during a month of storage. Fruits stored in 10% CO$_2$+3% O$_2$showed higher level of soluble solids, Vitamin C content and L value than other condition. And also comparatively lower endogenous ethanol accumulation and better state of quality was observed in that condition.

Effect of biomass pyrolysis liquid on the quality characteristics of 'Xiangshui' pears (Pyrus ussuriensis) during storage (바이오매스 열분해액이 '향수'배의 저장 중 품질특성에 미치는 영향)

  • The quality characteristics of 'Xiangshui' pears (Pyrus ussuriensis) treated with different concentrations of biomass pyrolysis liquid (BPL) during storage at $25^{\circ}C$ were investigated. The weight of 'Xiangshui' pears treated with BPL declined at a slower rate than that of the control. The rot index of BPL-treated 'Xiangshui' pears decreased with increasing storage times, and treatment with 20-fold-diluted BPL resulted in the lowest rot index after storage for 12 days. The total acid content of 'Xiangshui' pears treated with 20-fold-diluted BPL was 0.19%, and was the highest after storage for 12 days. After storage for 12 days, the total sugar content of 'Xiangshui' pears treated with 20-fold-diluted BPL was 7.19%; this was significantly higher than that of the control, but not significantly different from that of pears treated with other BPL dilutions. The vitamin C content of 'Xiangshui' pears showed a decreased trend, and pears treated with 20-fold-diluted BPL had a vitamin C content of 2.21 mg/100 g after storage for 12 days and showed the least decline compared to other treatments. In addition, respiration in 'Xiangshui' pears was effectively inhibited by treatment with BPL. In conclusion, BPL treatment exerts a protective effect on the quality of 'Xiangshui' pears during storage, with 20-fold-diluted BPL being the most effective.

Effect of Postharvest CO2 Treatment and Packaging Materials on the Quality of 'Gonji-7ho' Oyster Mushroom (Pleurotus ostreatus) during Storage (수확후 CO2 처리 및 포장필름 종류가 '곤지-7호' 느타리버섯의 저장 중 품질에 미치는 영향)

  • 느타리버섯은 전남 장흥군에서 병재배로 재배된 '곤지 7호' 느타리버섯 품종을 $3^{\circ}C$에서 1일간 냉각시킨 후 실험에 사용하였다. 이산화탄소 처리는 10mm 두께의 아크릴로 제작한 밀폐 챔버($40{\times}70{\times}40cm$)를 이용하여 CO2가스를 주입하여 내부의 CO2 농도를 $30{\pm}1%$로 조정하여 3시간 동안 밀폐시켰으며 $3^{\circ}C$ 저온 저장고에서 수행하였다. 처리 후 포장필름은 3처리로 하여 PVC랩, 재배농가에서 이용하는 필름인 $20{\mu}m$ OPP필름, $30{\mu}m$ OPP필름을 재료로 하여 산소투과율을 $5,000cc/m2{\cdot}day{\cdot}atm$로 조정한 필름(OPP5)을 사용하였다. PVC랩 처리는 스티로폼 트레이에 옮긴 후 균일하게 한 겹으로 둘렀고, OPP와 OPP5는 봉지에 옮겨 봉지 입구 4cm 안쪽을 비닐접착기로 열접착하여 밀봉하였으며 $3^{\circ}C$에 저장하면서 필름내부의 기체조성, 경도, 색도, 이취, 종합선도 변화를 조사하였다. 포장된 느타리버섯의 포장재 내 이산화탄소 농도는 저장초기 0.03%에서 점차 증가하여 저장 3일 후 PVC랩은 3.5~3.9%, 나머지 처리는 18%이상으로 증가하였으며 OPP필름보다 OPP5필름의 경우 포장재 내 이산화탄소 농도가 더 낮게 유지되었다. 생체중 감소는 PVC랩에서 저장 26일 후 9.9% 이하로 육안으로 보이는 시들음이 관찰되었으며 나머지 처리들은 저장 26일 후까지 1.6% 이하의 중량감소율을 보였다. 느타리버섯 갓 경도는 유지되었으며 줄기표면의 황색도 $b^*$값은 PVC랩에서는 증가 경향을, OPP나 OPP5에서는 유지 경향을 보였다. 느타리버섯의 이취 및 전체적인 품질은 무처리 후 PVC랩 포장은 저장 6일 후에 이취가 상품성 한계로 발생하였고 OPP5 포장은 포장재 중 가장 이취발생이 늦게 발생하였으며 CO2 처리에 의해 지연되는 것을 알 수 있었다. 이런 결과를 종합하여 느타리버섯의 $3^{\circ}C$ 저장 중 품질유지기간은 각 PVC랩(6일) ${\rightarrow}$ 30% CO2+PVC랩(7일) ${\rightarrow}$ OPP(10일) ${\rightarrow}$ 30% CO2+OPP(17일) ${\rightarrow}$ OPP5(20일) ${\rightarrow}$ 30% CO2+OPP5(22일)로 조사되었다. CO2처리 후 산소투과율을 $5,000cc/m2{\cdot}day{\cdot}atm$으로 조정한 미세천공(OPP5)필름이 가장 높은 전체적 품질점수를 나타내었으며 상대적으로 낮은 이취발생, 갈변과 갓무름이 적어 높은 점수를 얻는데 영향을 준 것으로 보인다.

Cultural Characteristics and Fruiting-body Productivity of New Cultivar 'Hwadam' (Lentinula edodes) by Punching Treatments on the Sawdust Medium (톱밥배지 침공처리에 따른 표고 '화담' 의 균사배양 및 자실체 생산성)

  • The effect of punching treatment on bags containing sawdust medium for the stable production of fruiting bodies of Lentinula edodes 'Hwadam' was investigated. After spawn inoculation, the sawdust medium temperature and CO2 concentration reached 27.0-28.8℃ on the 23rd day of incubation, and 14.0-15.6% on the 16th day of incubation, respectively, decreasing thereafter. The O2 concentration showed an opposite pattern to the CO2 concentration. As the number of punching treatments on the medium increased during the incubation period, the lightness value decreased, and conversely, the color difference and weight loss rate increased. There was no difference in the cultivation period according to the punching treatment. As the punching treatment increased on the sawdust medium, the fruiting body yield increased; a three-time punching treatment produced the best yield with 605.7 g. Thus, proper punching treatment on a sawdust medium during the incubation period can increase the number of fruiting bodies and yield.

Experimental Study of the Oven Dried Soil Weight Measurement Using Singularity Analysis (특이점 분석에 의한 노건조된 흙의 건조무게 측정에 관한 실험적 연구)

  • The dry unit weight of the soil can be obtained by measuring the weight of the oven dried soil. According to the Korean Industrial Standards (KS F), oven dried soil at 110±5℃ continuously increases in weight when exposed to air. However, there is no regulation on the weight measurement time of oven dried soil, making it difficult to accurately measure the dry weight. In this study, a method to easily determine the dry weight was presented through the analysis of the weight singularity that appears during weight measurement. The weight singularity represents the smallest value among the measured weights, and is the weight at which the effect of moisture absorption in the air of the sample is minimized. In the course of the experiment, a container was selected using a statistical analysis method, and a photograph of the soil samples were presented using an optical microscope. In addition, the temperature of the weight singularity was measured using a non-contact infrared thermometer. As a result of analyzing the weight singularity of six types of soil, including Jumunjin sand, Naeseong stream sand, Yecheon weathered granite soil, Jeju sand, Sabkha sand, and Ulleung sand, the weight singularity of oven dried sample appeared between 8 and 27 seconds after weight measurement, and the temperature ranged from 103.4 to 108.13℃. The weight decrease rate of the singularity was 0.0066 to 0.0085% depending on the soil samples.

Comparison of Mycelium Cultivation and Fruiting Body Characteristics of Lentinula edodes According to the Sawdust Media Nutrients and Inoculation and Cultivation Conditions (표고 톱밥배지 영양원, 접종 및 배양조건에 따른 균사배양 및 자실체 특성 비교)

  • The cultivation conditions of shiitake mushrooms (Lentinula edodes) influence the production and quality of fruiting bodies. We conducted this study to improve the productivity and quality of shiitake mushrooms by modifying the cultivation conditions. Two types of spawns (sawdust and liquid spawn) were used, and corn flour was used as a nutritional source for the sawdust medium. A blue light-emitting diode (LED; 300 lux) was also used instead of a white LED during the incubation period. Sanbaekhyang was used as the experimental variety. When using corn flour, the mycelial growth rate increased 1.1 to 2.7 times the growth rate of the control up to 21 days of incubation, and the weight loss rate of the media was also higher. Mushroom productivity increased 1.2 times when the liquid spawn was used compared to when the sawdust spawn was used, and the blue LED also increased fruiting body production by 1.1 times compared to the white LED. Mushroom productivity increased when the liquid spawn was used, and the blue LED also increased fruiting body production. Fruiting body weight and the size of the cap were greater when sawdust spawn was used. The fruiting body weight and the stipe diameter were greater when the blue LED was used. Taste analysis showed that the saltiness increased when corn flour was used, and the sourness increased when the blue LED was used.

Effects of Flavonoid from Rhus verniciflua on Testosterone Secretion by Rat Leydig Cells In Vitro (옻나무 유래 Flavonoid 처리가 흰쥐 Leydig 세포의 체외배양에서 Testosterone 분비에 미치는 영향)

  • This study was performed to report a direct dose dependent stimulatory effect of the Flavonoid(F) on basal testosterone secretion and a dose dependent effect on LH induced testosterone production by Leydig cell of matured rats in vitro culture. F was obtained kom the Rhus vernicifua through aceton extraction and silica gel adsorption column chromatography. Leydig cells (1$\times$10$^{6}$ cells/well) from 12 weeks old rats were incubated with or without F(0, 20, 40, 80, 160 ng) or insulin-like growth factor-I(IGF-I) in the presence or absence of LH(10, 100ng). 1. The maximal stimulatory concentrations of testosterone in culture media were showed at 24hr of culture. but these testosterone level were decreased at 36 hr of culture. 2. Flavonoid(80ng) were significantly(P < 0.05) increased testosterone production compared with control groups for 12 hr culture. 3. Testosterone secretion by Leydig cells stimulated with LH(10, 100ng) for 6 hr and 12hr culture compared with 3 hr culture. 4. LH 10 ng augmented testosterone were increased by addition of F 40 ng for 12 hr culture. 5. F(0 and 40 ng) also enhanced LH 10 ng stimulated testosterone for 3 hr Leydig cells culture. 6. Addition of IGF-I 100 ng to the culture medium for 6 hr were increased the concentration of testosterone by Leydig cells stimulated with 100 ng LH. These results indicate that Flavonoid has a direct stimulatory effect on basal testosterone secretion in rat Leydig cells, and also modulates LH mediated testosterone. Therefore, Flavonoid may act as a modulator on gonadal development or gonadal steroidogenesis in direct or indirect.
