Mutagenesis of of Hypsizygus marmoreus by Gamma Ray Irradiation (감마방사선 조사에 의한 느티만가닥버섯의 변이)

  • This study was performed to develop new Hypsizygus marmoreus cultivars that have enhanced functional materials and improved physiological characteristics with mutagenesis by gamma ray irradiation. Protoplasts of H. marmoreus brown strain HYM-056 were irradiated by gamma ray for mutagenesis, and then 2,000 clones of mutants were randomly selected and the fruiting bodies were induced by bottle culture. Among them, 157 isolates with fast-growing, heavy and many fruiting body-producing were selected. The isolates were cultured in plastic bottle containing rice bran, barley hulls and fir sawdust to form the fruiting bodies. About 100 days after inoculation, characteristic of fruiting bodies were investigated. The isolates were divided into 6 groups based on color, shape and size of pileus, and length, diameter, number and weight of stipe. In addition, the genetic variation of the isolates was analyzed by URP-PCR fingerprinting.

The Cultural Characteristics of the Fruit Body Formation by Two Selected Strains of Winter Mushroom(Flammulina velutipes) Collected from Korea (한국산(韓國産) 팽나무버섯에서 선발(選拔)된 두 계통(系統)에 대한 자실체배양상(子實體培養上)의 특성(特性))

  • The cultural characteristics of fruit bodies formation in the two selected strains(brown and yellow strain) of winter mushroom collected in Korea were investigated. The effects of various environmental conditions and physicochemical pretreatments were evaluated by the character of fruit bodies. There was no differences between two strains in non-selectivity of media, optimum temperature($11^{\circ}C{\sim}15^{\circ}C$), relative humidity(90%), and illumination(60 lux). In the application of alternated temperature (the best condition : $5^{\circ}C{\cdot}10^{\circ}C$ and $5^{\circ}C{\cdot}15^{\circ}C$), ultraviolet ray(30~50 seconds), electricity(5 seconds), thiamine hydrochloride(0.05~0.1%), urea, and IAA as a pretreatments, there was no differences between two strains. The brown strain had larger pileus and longer stipes than these of the yellow strain, while the yellow strain yielded more weight and number of the fruit body formation than the brown strain.

Plant Regeneration via Organogenesis from Leaf Explant Culture of Taraxacum coreanum Nakai (흰민들레 잎조직으로 부터 기관형성을 통한 식물체 재생)

  • Plant Regeneration via organogenesis from leaf disk of Korean dandelion was investigated. Leaf disk cultured on MS medium with various combinations of BA (0-4 mg/L) and 2,4-D (0-1 mg/L). Shoot regeneration from leaf explant was observed after 3 weeks of culture. The highest shoot regeneration frequency from leaf disk was obtained with 2 mg/L BA. To analyze the effect of leaf age along shoot formation, we measured number of shoots per explant, shooting rate, fresh and dry weight of leaf explant. The highest number of shoots (11.5) per explant were obtained leaf from 7 weeks old plantlets after seed germination. The regenerated shoots were transferred in 1/2 MS medium with 0.5 mg/L NAA for root formation. Regeneratied plantlets thought organogenesis were growing to whole plants in the pots with acclimation.

Component analysis and immuno-stimulating activity of Sparassis crispa stipe (꽃송이버섯 기부의 성분분석 및 면역활성)

  • This study was carried out to investigate the component and immunoregulatory effects of Sparassis crispa stipe. Herein, S. crispa was divided into the pileus and stipe to compare their ingredients (${\beta}-glucan$), antioxidant activity (in vitro), and the immunoregulatory function (cytokines, leukocytes, and spleen weight). The ${\beta}-glucan$ content in each part showed about 1.8 times higher content in the stipe than that in the pileus. The stipe also showed a higher total phenol content and antioxidant activity than the pileus. The cytokines $TNF-{\alpha}$, IL-2, and IL-10 have adjusted in the S. crispa extract-injected groups. In addition, the number of leukocytes was also significantly elevated in the rats administered with the S. crispa stipe extract. These results suggest that the stipe of S. crispa has great potential as an ingredient in functional foods.

Method for Increasing Stability by Reducing the Motion of a Lightweight Floating Body (경량 부유체의 운동 저감으로 안정성 증가방법에 관한 연구)

  • Demand for leisure facilities such as mooring facilities for berthing leisure vessels and floating pensions based on floating bodies is increasing owing to the rapid growth of the population and related industries for marine leisure activities. Owing to its relatively light weight as a fluid, inclination is easily generated by waves and surcharges flowing to the coast, resulting in frequent safety accidents because of the low stability. As a solution to this problem, a motion reduction device for floating bodies is proposed in this study. The device (motion reduction device based on the air pressure dif erence) was attached to a floating body and the effect was analyzed by comparing the results with those of a floating body without motion reduction. The effect analysis was further analyzed using a computer analysis test, and the method for increasing the stability of the floating body was studied, and its the effect was verified. Based on the analysis of the test results, the stability of the floating body increased with a motion damping device is higher than that of the floating body without a motion reducing device as the wave momentum reduces, owing to the air pressure difference. Therefore it was concluded that the use of such a device for reducing motion a floating body is useful not only for non-powered ships but also for powered and semi-submersible ships, and further research should be conducted by applying it to various fields.

Seismic Retrofit of Welded Steel Moment Connections Considering the Presence of Composite Floor Slabs (바닥슬래브를 고려한 용접철골모멘트접합부의 내진보강)

  • In the 1994 Northridge earthquake, connection damage initiated from the beam bottom flange was prevalent. The presence of a concrete slab and resulting composite action was speculated as one of the critical causes of the prevalent bottom flange fracture. In this study, four seismic retrofit schemes are proposed in order to salvage welded steel moment connections with composite floor slabs in existing steel moment frames. Because top flange modification of existing beams is not feasible due to the presence of a concrete floor slab, three schemes of bottom flange modification by using welded triangular or straight haunches or RBS(reduced beam section), and beam web strengthening by attaching heavy shear tab were cyclically tested and analyzed. Test results of this study show that haunch and web-strengthened specimens can eliminate the detrimental effect caused by composite action and ensure excellent connection plastic rotation exceeding 5% rad. Design recommendations for each retrofit scheme together with supplemental numerical studies are also presented.

A Soil Mechanical Study for a Practical Application to Forest Road Construction (임도설계(林道設計)에의 응용(應用)을 위한 흙의 토질(土質) 역학적(力學的) 특성(特性))

  • This study was carried out to discuss how soils in the area planned for a forest road construction can be mechanically tested and practically applied. For this, 16 soil test samples from 8 plots(2 samples per plot) were used. The major tests are focused on unit weight before and after cut, water content, liquid and plastic limits, sieve and hydrometer analysis etc. The total unit weight(${\rho}_t$) before and after cut are $1.69g/cm^3$ and $1.19g/cm^3$, respectively. Their water contents are 21.0% and 20.5%. The coefficient of uniformity U and coefficient of curvature C obtained from sieve and hydrometer analysis are 125 and 0.42, which mean generally not well graded. On the soil classification by USCS, SM(silty sand or silt-sand mixed soil)is a Key soil, but it seems to be not good for fill material. From the standard proctor test are resulted $1.40{\pm}0.065g/cm^3$ for the unit weight(${\rho}$) in the nature and $1.88{\pm}0.049g/cm^3$ for the optimum proctor unit weight(${\rho}pr$) each. With this to say, it is necessary more powerful compaction work at earth filling, with which this soil reachs enough the ${\rho}pr$, and more earth.

Effect of Aggregate Size on the Shear Capacity of Lightweight Concrete Continuous Beams (경량콘크리트 연속보의 전단내력에 대한 골재크기의 영향)

  • Twenty-four beam specimens were tested to examine the effect of the maximum aggregate size on the shear behavior of lightweight concrete continuous beams. The maximum aggregate size varied from 4 mm to 19 mm and shear span-to-depth ratio was 2.5 and 0.6 in each all-lightweight, sand-lightweight and normal weight concrete groups. The ratio of the normalized shear capacity of lightweight concrete beams to that of the company normal weight concrete beams was also compared with the modification factor specified in ACI 318-05 for lightweight concrete. The microphotograph showed that some unsplitted aggregates were observed in the failure planes of lightweight concrete beams, which contributed to the enhancement of the shear capacity of lightweight concrete beams. As a result, the normalized shear capacity of lightweight concrete continuous beams increased with the increase of the maximum aggregate size, though the increasing rate was lower than that of normal weight concrete continuous beams. The modification factor specified in ACI 318-05 was generally unconservative in the continuous lightweight concrete beams, showing an increase of the unconservatism with the increase of the maximum aggregate size. In addition, the conservatism of the shear provisions of ACI 318-05 was lower in lightweight concrete beams than in normal weight concrete beams.

Determination of Structural Lightweight Concrete Mix Proportion for Floating Concrete Structures (콘크리트 부유구조체 적용을 위한 구조용 경량콘크리트의 최적배합비 선정)

  • This study aims to provide information for the design and use of structural lightweight concrete (SLWC) for floating concrete structures in a marine environment. An experimental program was set up and comprehensive experimental campaign were carried out to determine SLWC mix proportions that can satisfy specified concrete strength, density, and slump values all of them were determined from previous research. Comparisons with previous SLWC mix designs that have been utilized for actual floating concrete structures were made. Key aspects needed to be considered regarding to the use of SLWC for floating marine concrete structures were discussed.

Cultural Characteristics and Morphological Comparison of the Wild Mushroom Lentinula edodes Cultivated on Sawdust Substrate (표고 야생형 계통의 배양 및 형태적 특성 비교)

  • The cultivation characteristics of 26 wild strains of Lentinula edodes were investigated for their use as breeding material. Strains NIFoS 68, 136, 1521, 1651, and 2064 showed an above average mycelial growth on potato dextrose agar at 10, 20, and $30^{\circ}C$. NIFoS 411 showed the lowest mycelial growth at $20^{\circ}C$, but the highest growth at $30^{\circ}C$. The rate of weight loss of L. edodes cultivated on sawdust (2 kg) ranged from 13.5 to 47.5%, with the highest rates showed by NIFoS 50 (47.5%), NIFoS 128 (34.5%), and NIFoS 54 (34.4%). Fruiting bodies were produced in nearly all (24/26) strains and productivity ranged from 3 g to 446 g/2 kg medium. Temperature was not significantly correlated with mushroom production or mycelial growth. Larger weight loss correlated strongly with fruit yield. In terms of production, NIFoS 50 (446 g), NIFoS 952 (435 g), and NIFoS 53 (421 g) were useful as breeding material. The NIFoS 667 strain was superior in terms of morphology. NIFoS 670 showed the characteristic yellowish-brown color of fruiting bodies.
