Immunomodulatory Effect of Mesenchymal Stem Cell-Derived Exosomes in Lipopolysaccharide-Stimulated RAW 264.7 Cells (Lipopolysaccharide로 자극한 RAW 264.7 세포에서 성체줄기세포 유래 엑소좀(exosome)의 면역 조절 효과)

  • Mesenchymal stem cells (MSCs) are multipotent stem cells that can be differentiated into a variety of cell types, including adipocytes, osteoblasts, chondrocytes, β-pancreatic islet cells, and neuronal cells. MSCs have been reported to exhibit immunomodulatory effects in many diseases. Many studies have reported that MSCs have distinct roles in modulating inflammatory and immune responses by releasing bioactive molecules. Exosomes are cell-derived vesicles present in biological fluids, including the blood, urine, and cultured medium of cell cultures. In this study, we investigated the immunomodulatory effects of mouse adipose tissue-derived MSCs (mAD-MSCs), cultured medium (MSC-CM) of mAD-MSCs, and mAD-MSC-derived exosomes (MSC-Exo) on lipopolysaccharide (LPS)-induced RAW 264.7 cells. We observed that the expression levels of IL-1β, TNF-α, and IL-10 were significantly increased in LPS-stimulated RAW 264.7 cells compared to those in LPS-unstimulated RAW 264.7 cells. Additionally, these values were significantly (p < 0.05) decreased in mAD-MSCs-RAW 264.7 cell co-culture groups, MSC-CM-treated groups, and MSC-Exo-treated groups. MSCs can modulate the immune system in part by secreting cytokines and growth factors. We observed that immunomodulatory factors such as IL-1β, TNF-α, and IL-10 were secreted by mAD-MSCs under co-culturing conditions of mAD-MSCs with activated RAW 264.7 cells. In addition, mAD-MSC-derived exosomes exhibited similar immunomodulatory effects in activated RAW 264.7 cells. Therefore, our results suggest that mAD-MSCs have an immunomodulatory function through indirect contact.

Establishment and Characterization of Multipotent Germ Line Stem Cells (MGSCs) from Neonatal Mouse Testis (신생 생쥐 고환에서 기인한 다분화능 생식줄기세포주의 확립 및 특성 분석)

  • Objective: The aim of this study was to investigate whether multipotent germline stem cells (MGSCs) can be established from neonatal mouse testis. Methods: Various cells containing MGSCs were collected from neonatal testis of ICR mice and allocated to plates for in vitro culture. After 7 days in culture, the cells were passed to a fresh culture plate and continuously cultured. From the third or fourth passage, the presumed MGSCs were cultured and maintained on mitomycin C-inactivated STO feeder cells. The MGSCs were cultured in a condition where mouse embryonic stem cells (ESCs) are cultured. Characteristics of the MGSCs were evaluated by RT-PCR, immunocytochemistry, alkaline phosphatase activity, karyotyping, and transmission electron microscopy. Results: Two MGSCs lines were established from 9 pooled sets of neonatal testicular cells. MGSCs colonies were morphologically undistinguishable from ESCs colonies and both MGSC lines as well as ESCs expressed undifferentiated stem cell markers, such as Thy-1, Oct-4, Nanog, Sox2 and alkaline phosphatase. Fine structure of undifferentiated MGSCs were similar to those of ESCs and 60% of MGSCs (12/20) had normal karyotype at passage 10. They were able to form embryoid bodies (EBs) and MGSC-derived EBs expressed marker genes of three germ layers. Conclusion: We could establish the MGSCs from neonatal mouse testis and they were differentiated to multipotent lineages of three germ layers. Molecular characteristics of MGSCs were similar to those of ESCs. Our results suggest a possibility that multipotent stem cells derived from testis, the MGSCs, could replace the ESCs in biotechnology and regenerative medicine.

Cell Death Study in Embryonic Stem Cell-derived Neurons and Its Applications (배아줄기세포 유래 신경계세포에서의 세포사멸 연구와 그 응용)

  • Specific protocols to increase the differentiation of neuronal cells from embryonic stem (ES) cells have been well established, such as retinoic acid induction and lineage selection of neuronal cells. For the neuropathological studies, ES-derived neurons (ES neurons) must show normal physiological characteristics related to cell death and survival and should be maintained in vitro for a sufficient time to show insults-specific cell death without spontaneous death. When mouse ES cells were plated onto astrocytes monolayer after retinoic acid induction, most ES cells differentiated into neuronal cells, which were confirmed by the presence of specific neuronal markers, and the cultures were viable for at least four weeks. When these cultures were examined for vulnerability to glutamate excitotoxicity, ES neurons were vulnerable to excitotoxic insults mediated by agonist-specific receptors. The vulnerability to excitotoxic death increased with developmental age of ES neurons in vitro. Specific receptors for Neurotrophin and GDNF family ligands were present in ES neurons. GDNF and NT-3 could modulate the survival and excitotoxic vulnerability of ES neurons. The vulnerability and resistance to toxic insults, which are essential requirements of model culture systems for neuropathological studies, make ES neurons to a useful model culture system. Especially ES cell are highly amenable to genetic modification unlikely to primary neuronal cells, which will give us a chance to answer more complicated neurophysiological questions. Recently there was an outstanding attempt to explore the cellular toxicity using human ES cells (Schrattenholz & Klemm, 2007) and it suggested that ES cells could be a new model system for neurophysiological studies soon and go further a large-scale screening system for pharmacological compounds in the future.

Inhibition of Neurogenesis of Subventricular Zone Neural Stem Cells by 5-ethynyl-2'-deoxyuridine (EdU) (5-ethynyl-2'-deoxyuridine (EdU)에 의한 뇌실하 영역 신경줄기세포의 신경 세포로의 분화 억제)

  • In the subventricular zone (SVZ) and the subgranular zone of the brain, neurogenesis occurs throughout one's lifespan. Neural stem cells (NSCs) in these regions divide to maintain their stem cell pools as well as differentiate into neurons and glial cells. To monitor cell division, a thymidine analogue such as 5-ethynyl-2'-deoxyuridine (EdU) has been used. In some cases, EdU was applied to label newly born neurons. Here, we report about the effects of EdU on the proliferation and differentiation of NSCs cultured from mouse SVZ. First, when NSCs were cultured in a proliferation medium containing EdU for 24 hr, they did not generate any neurons under the following differentiation conditions. When EdU was applied to the proliferating NSCs for 1 hr prior to differentiation, neurogenesis was still substantially reduced. Second, EdU decreased cell proliferation of NSCs in dose- and time-dependent manners. Finally, EdU inhibited differentiation into oligodendrocyte lineage, while the number of glial fibrillary acidic protein (GFAP)-positive astrocytes increased. To our knowledge, these findings are the first to show the effects of EdU on the differentiation of SVZ NSCs and suggest that cell division is necessary for differentiation into neurons and oligodendrocytes.

Chondrogenic Differentiation of Human Mesenchymal Stem Cells on a Patterned Polymer Surface (패턴된 폴리머를 이용한 중간엽줄기세포의 연골 분화)

  • Mesenchymal stem cells (MSCs) are an attractive tool in tissue engineering as they have the required potential to treat injured articular cartilage. UV-exposed DTOPV (S-triazine bridged p-phenylene vinylene) is a biocompatible and fluorescent polymer with a hydrophilic surface. Previous studies have demonstrated that the surface wettability and hydrophilicity play critical roles in regulating cell adhesion and proliferation. The objective of this study was to improve the potential of in vitro MSC differentiation into Chondrocytes using DTOPV. MSCs were cultured on two different substrates: (1) tissue culture polystyrene (TCPS) as a reference and (2) UV-exposed and patterned DTOPV films. Chondrogenesis of MSCs was induced for two weeks on TCPS and DTOPV in the presence of an induction medium containing transforming growth factor (TGF)-${\beta}3$. Interestingly, the MSCs on TCPS adhered and spread, while those on DTOPV tended to form aggregates within several days. The cells cultured on DTOPV for two weeks had a round morphology, with stronger Safranine O staining of the extracellular matrix than that of the cells cultured on TCPS. Also, Type II collagen gene was significantly expressed in cells induced on DTOPV. These results indicate that chondrogenic differentiation of MSCs proceeds more rapidly on DTOPV than on TCPS. Therefore, in cartilage tissue engineering, DTOPV could be used to induce effective chondrogenic differentiation of MSCs.

Experiment of Response with Voltage for Stem Cell Regeneration Treatment (줄기세포재생 치료를 위한 배지의 전압 반응 실험)

  • In this paper, the electrical values of resistance, current, and voltage condition of reactivity is analyzed by applying the direct current (DC) voltage in medium for stem cell regeneration treatment. The voltage response by medium is related to electrical stimulation in the process of induction of differentiation for stem cell and the differentiation condition can be checked depending on the response of voltage condition. If the voltage level is lower in reacting response of a medium, the stem cell stimulation condition is stable, and if the voltage changing level is higher, the stem cell stimulation condition is unstable and a considerable loss will be resulted in the differentiation process. In this research, the optimization of electrical stimulation condition is expected for possible stem cell regeneration treatment.

Co-author network for convergent research pattern analysis in stem cell sector (줄기세포분야 융합연구형태 분석을 위한 공저자 네트워크)

  • This study was carried out to confirm a convergent research pattern and researchers' role in stem cell sector by social network analysis. Articles were extracted from 1996 to 2012 in PubMed, 515 authors of 270 embryonic stem cell and induced pluripotent stem cell articles and 1,515 authors of 580 adult stem cell and mesenchymal stem cell articles. Degree(D) and betweenness(B) centrality was measured and co-author network was generated for researcher's role. As a result, Core researcher and Intermediary researcher was identified in co-author network. Core researcher had high D. centrality, otherwise high B. centrality or not. Intermediary researcher for convergent research had high B. centrality and low D. centrality. Conclusively, co-author network will be used as objective data not only to find core researchers in subject area for improving achievement but also to select experts for research project evaluation.

Establishment of Functional Cells for Vascular Defect Disease from Human Embryonic Stem Cell via Region Sorting Depending on Cell Volume (세포 크기 차이를 이용한 유세포 분석을 통한 인간배아줄기세포 유래 기능성 혈관세포의 확립)

  • Human embryonic stem cells have been highlighted as a valuable cellular source in the regenerative medicine field, due to their pluripotency. However, there is the challenge of the establishment of specific functional cell type forms of undifferentiated human embryonic stem cells (hESC). To establish and purify functional cell types from hESCs, we differentiated undifferentiated hESCs into vascular lineage cells and sorted the specific cell population from the whole cell population, depending on their cell volume, and compared them with the non-sorted cell population. We observed that about 10% of the PECAM positive population existed in the VEGF induced differentiating human embryoid body (hEB), and differentiated hEBs were made into single cells for cell transplantation. After making single cells, we performed cell sorting using a fluorescence-activated cell sorter (FACs), according to their cell volume on the basis of FSC region gating, and compared their therapeutic capacity with the non-sorted cell population through cell transplantation into hindlimb ischemic disease model mice. 4 Weeks after cell transplantation, the recovery rate of blood perfusion reached 54% and 17% in the FSC regions of sorted cells- and non-sorted cells, respectively. This result suggests that derivation of a functional cell population from hESCs can be performed through cell sorting on the basis of cell volume after preliminary differentiation induction. This approach may then greatly contribute to overcoming the limitations of marker sorting.

Different Potential of Hematopoietic Differentiation in Two Distinct Mouse Embryonic Stem Cells (두 개의 다른 마우스 배아줄기세포의 차별적인 조혈세포 분화능)

  • Embryonic stem(ES) cells have tremendous potential as a cell source for cell-based therapies. Realization of that potential will depend on our ability to understand and manipulate the factors that influence cell fate decision and to develop methods for getting enough cell numbers for clinical applications. Hematopoiesis has been widely studied, and hematopoietic differentiation from ES cells is a good model to study lineage commitment. In this study, we investigated stemness and compared the efficiency of hematopoietic differentiation using two different mouse embryonic stem cell lines TC-1 and B6-1. Although the two cell lines showed known stem cell properties with minor differences, the embryoid body formation efficiency in methylcellulose was much higher in TC-1 than B6-1. When measured potentials of hematopoietic differentiation using functional(colony-forming cell) and phenotypic(specific marker expression) assays, we found that TC-1 can differentiate into hematopoietic cells in methylcellulose culture but B6-1 cannot. These results imply that we can improve the efficiency of hematopoietic cell differentiation by selection of proper cell lines and this may be also applied in the differentiation of human embryonic stem cells.

Mouse Embryonic Stem Cell Uptakes of Buforin 2 and pEP-1 Conjugated with EGFP (생쥐 배아 줄기세포의 Buforin 2 및 pEP-1에 결합된 EGFP의 세포 내 수송)

  • Differentiation of cells can be induced through modulation of endogenous regulators using exogenous factors. Useful transfection systems to transport a specific exogenous regulator into cell have been tried but still there are many obstacles to overcome. In this study, we examined the transfection efficiency of cell permeable peptides (CPPs) in mouse embryonic stem cell under the various conditions. To identify the CPP-mediated translocation of a protein, we employed recombinant CPP-enhanced green fluorescent protein (EGFP). Viability of R1 cells was different between experimental groups depending on the kind of CPP and the concentration of CPP-EGFP. Translocation of CPP-EGFPs into the R1 cells was not detected until 30 min after CPP-EGFPs treatment in all groups. After 1 hr, translocation of pEP-1-EGFP-N was detected, but it could not be detected in the other group. Transfection of pEP-1EGFP-N was independent on its concentration. The time course did not show saturation even after 24 hr in pEP-1-EGFP-N. These results showed that the permeability depended on the kind of CPP and the location of His-tag in the case of examined CPPs, and did not need biological energy. On summary, the efficiency of transfection of CPP-EGFP depends on the CPP sequences but the culture time is not a key factor in transfection for the mouse embryonic stem cell. For the future studies to improve the efficiency of translocation of protein into embryonic stem cells, it is needed to develop modified CPP or mediator. The studies would be very useful to induce the differentiation of embryonic stem cells.
