The seasonal appearance and chemical composition characteristics of cladode of Opuntia humifusa (천년초 줄기의 수확 시기 별 외관 및 성분 품질 특성)

  • The seasonal appearance and the chemical composition characteristics of cladode of Opuntia humifusa were investigated in this study. The minor (horizontal) and major (vertical) axes, the width, and weight of O. humifusa cladode were compared for its appearance features. Moisture, crude protein, ash, fat, color, dietary fiber, mineral, and amino acids contents were measured for the comparison of their chemical composition characteristics. The cladode of O. humifusa harvested in summer showed higher values of minor and major axis, width, and weight than those harvested in winter. According to the results of Hunter color index, cladode of O. humifusa harvested in summer showed the highest lightness level (Hunter L value). In the meanwhile, cladode of O. humifusa harvested in spring showed the highest Hunter a (redness) and b (yellowness) values. Cladode of O. humifusa harvested in summer had the lowest crude protein and crude fat contents, while it showed the highest crude ash content. The total contents of moisture and dietary fiber were significantly greater than summer-harvested cladode of O. humifusa followed by spring- and winter-harvested cladode of O. humifusa. The major mineral content of all seasonal cladode of O. humifusa was $Ca^{2+}$. Glutamic acid and aspartic acid were the major amino acids in all seasonal samples. Contrary, the contents of total amino acids and free amino acids were the lowest in summer-harvested cladode of O. humifusa. Taken together, it was concluded that the appearance and chemical quality of cladode of O. humifusa was versatile depending on the harvesting season.

The Effects of Antimicrobial, Antioxidant, and Anticancer Properties of Opuntia humifusa Stems (천년초 줄기의 항균, 항산화 및 항암효과에 관한 연구)

  • This study was performed to investigate antimicrobial, antioxidant, and anticancer properties of Opuntia humifusa (OH) stems. OH stems were extracted with hexane, methanol, butanol and water. The methanol and hexane fraction exhibited strong antimicrobial activities on three strains of microbes, Rhodococcus equi, Staphylococcus aureus, and Clostridium perfringens. In the peroxynitrite scavenging effect ($ONOO^-$) of OH stems, the antioxidative activity of methanol, butanol and water fraction but not hexane fraction showed significant increases in a concentration-dependent manner. The DPPH radical scavenging activities of OH stems were high in the butanol fraction compared with other fractions. Anti-proliferation effects on the B16-F10, HepG2, HT29, and MCF-7 cell lines were significantly higher in the methanol and hexane fractions than in the water and butanol fractions at $100{\mu}g/mL$ concentration of extracts. These results suggest that OH stems can be used for the development of functional foods with biological activity.

Food Components of Different Parts of Cheonnyuncho (Opuntia humifusa) Harvested from Yeosu, Jeonnam in Korea (전남 여수 돌산지역에서 재배되는 천년초의 부위별 식품성분 분석)

  • This study was carried out to investigate the food components of the fruit, cladodes, and flowers of freezedried Cheonnyuncho harvested from Yeosu, Jeonnam in Korea. The major components of freeze-dried Cheonnyuncho in proximate composition were carbohydrates and crude ash. Ca, K, and Mg were the predominant minerals in Cheonnyuncho. Calcium content was higher in the fruit and cladodes than in the flowers. Two major amino acids, glutamic acid and aspartic acid, made up over 25% of the total amino acids in Cheonnyuncho. Palmitic acid and stearic acid were most abundant out of all the saturated fatty acids in Cheonnyuncho. The saturated fatty acid content of the fruit was higher than that of the flowers and cladodes. The major unsaturated fatty acid of Cheonnyuncho was oleic acid. The cladodes contained unusually high amounts of linoleic acid compared to the fruit and flowers. The major free sugar in the fruit was sucrose, whereas that of the cladodes and flowers was fructose. The total free sugar content was the highest in fruit, followed by cladodes. The most abundant organic acid in the fruits and cladodes was malic acid, while that of the flowers was citric acid. Vitamin A concentration was highest in the flowers whereas vitamin C concentration was highest in the fruit.

Volatile Flavor Components in Various Edible Portions of Angelica keiskei Koidz (신선초의 식용부위별 향기성분)

  • Volatile flavor components in whole edible portion, stem and leaf of fresh angelica (Angelica keiskei Koidz) were extracted by SDE (simultaneous steam distillation and extraction) method using the mixture of n-pentane and diethylether (1:1, v/v) as an extract solvent and analyzed by GC-FID and GC/MS. Identification of the volatile flavor components in aroma concentrate was mostly based on the RI of GC and mass spectrum of GC/MS. Twenty five hydrocarbons, 15 alcohols, 3 aldehydes, 6 esters, 2 ketones and 1 acid were identified in the whole edible portion of angelica. Twenty hydrocarbons, 13 alcohols, 4 esters and 1 acid were identified in the stem sample of angelica. Nineteen hydrocarbons, 11 alcohols, 4 aldehydes, 6 esters, 2 ketones and 1 acid were identified in the leaf sample of angelica. ${\gamma}-Terpinene$, germacrene B, ${\delta}-3-carene$, cis-3-hexen-1-ol, ${\gamma}-muurolene$ and ${\gamma}-elemene$ were the main components in each edible portions of angelica. The terpenoid compounds in volatile flavor components identified from whole edible portion, stem and leaf samples were confirmed as 75.76%, 86.42% and 78.21%, respectively. These results suggest that terpenoid compounds have a great effect on the flavor characteristics of angelica.

Uplift Capacity of Pipe Foundation for Single-span Greenhouse (단동 온실용 파이프 기초의 인발저항력 검토)

  • In order to provide design data support for reducing gale damage of single-span greenhouses, this paper experimentally evaluated the uplift capacity of a rafter pipe and continuous pipe foundation (anti-disaster standard), usually used for single-span greenhouses according to compaction ratio, embedded depth, and soil texture. In the reclaimed soil (Silt loam) and the farmland soil (Sandy loam), the ultimate uplift capacities of rafter pipe were 72.8kgf and 60.7kgf, respectively, and those of continuous pipe foundation were 452.7kgf and 450.3kgf, respectively at an embedded depth of 50cm and compaction rate of 85% (the hardest ground condition). The results showed that the ultimate uplift capacity of continuous pipe foundation was significantly improved at more than 6 times that of the rafter pipe. The soil texture considered in this paper had a sand content of 35%~59% and a silt content of 39%~58%, and it was shown that the ultimate uplift capacity did not have a significant difference depending on soil texture, and these results show that installing the rafter pipe and continuous pipe foundation while maintaining appropriate compaction conditions can give an advantage in securing stability in the farmland of greenhouses without significantly being influenced by soil texture. Based on the results of this paper, it was determined that maintaining a compaction rate above 75% for the continuous pipe foundation and above 85% for the rafter pipe was advantageous for securing stability in greenhouses. Especially when continuous pipe foundation of anti-disaster standard was applied, it was determined to be significantly advantageous in acquiring stability in greenhouses to prevent climate disaster.

  • 연구 목적: 본 연구는 인간배아줄기세포 동결에 minimum volume cooling (MVC) 초자화 동결방법이 유용하게 이용될 수 있는지의 여부를 조사하고자 실시하였다. 연구재료 및 방법: 인간배아줄기세포 콜로니는 0.05% collagenase 처리와 기계적 처리에 의해 작은 clumps로 자른 다음 동결 방법에 따른 효율을 비교 검토하고자, i) 대략 40-50개의 clumps를 10% DMSO가 들어있는 동결액에 $5{\sim}10$분간 처리하여 1ml cryo-vial에 넣고 slow-cooling용 cryo-module에 장착하고 -80C에서 overnight한 후 $LN_2$에 침지하여 완만동결을 실시하거나 ii) 10% ethylene glycol (EG)이 들어 있는 동결액에서 5$\sim$10분 처리하고 30% EG과 0.5 mol sucrose가 들어 있는 동결액에서 30초간 처리하여 본 연구를 위해 개발된 MVC straw에 10 clumps씩 적재한 다음 $4{\sim}5$ MVC straw를 $LN_2$가 들어있는 cryo-vial에 넣어 MVC 초자화동결을 실시하였다. 융해 후 생존율을 조사하였고 배아줄기세포의 특성을 유지하고 있는지의 여부를 조사하였다. 결 과: 융해 후, 인간배아줄기세포의 생존율은 완만동결을 실시했던 군 (20.0%) 보다 MVC 초자화동결을 실시했던 군에서 (76.0%) 유의하게 높게 나타났다. 또한 회복과정에서 완만동결을 실시했던 군에서는 세포성장이 매우 더디고 안 좋은 반면, MVC 초자화동결을 실시했던 군은 배아줄기세포의 증식이 동결을 실시하지 않은 세포와 같은 상태로 2주 이후부터 빠르게 전환되고 회복되는 것을 확인할 수 있었다. 이와 더불어 MVC 초자화동결-융해 후 회복된 배아줄기세포에서 정상 핵형, alkaline phosphatase acitivity, SSEA-4와 TRA-1-60 염색 및 Oct-4 발현을 확인하였으며 체외분화의 특성도 확인하였다. 결 론: 새로이 개발된 MVC 초자화동결을 이용하면 인간배아줄기세포는 고유의 특성을 잃지 않고 성공적으로 동결될 수 있다.

Proximate, Free Sugar, Amino Acid, Dietary Fiber and Saponin Composition of Agngelica Keiskei Koidz (신선초(Agngelica Keiskei Koidz)의 일반성분, 유리당, 아미노산, 식이 섬유 및 사포닌 조성)

  • To accept basic data of utilizing of Agngelica Keiskei Koidz as a raw material of industrial products, major chemical components were investigated. Comparing proximate composition of leaf and steam of Agngelica Keiskei, leaf contained higher crude protein, crude fat and crude ash, but lower moisture and crude fiber. The content of vitamin C in leaf and steam were 75mg% and 59mg%, respectively. The major free sugars were fructose and glucose and sucrose was also detected in a small amount. Total amino acid contents of leaf and steam were 1737.4mg%, 319.7mg%, respectively. Although the amino acid compositions of leaf and steam were different, threonine, histidine, leucine, glutamic acid and glycine were the major components. The major free amino acids were histidine, alanine, leucine, threonine and arginine, but were percent in a trace amount. The contents of total dietary fiber(TDF) in leaf and steam were 31.89 and 43.37% on dry basis, respectively. The content of saponin in leaf and steam were 535.51mg% and 463.09mg%, respectively.

Anti-diabetic and Anti-oxidative Effects of Opuntia humifusa Cladodes (천년초 선인장 줄기의 항당뇨 및 항산화 효과)

  • To investigate the anti-diabetic and antioxidant effects of Opuntia humifusa cladodes, O. humifusa cladodes powder was extracted with 75% ethanol and fractionated with various solvents. Among the extracts fractionated with various solvents, ${\alpha}$-amylase and ${\alpha}$-glucosidase inhibitory activities of O. humifusa cladodes were highest ($89.94{\pm}1.15%$ and $29.01{\pm}3.03%$, respectively) in the ethyl acetate fraction. Further, total phenolic and flavonoid contents of the ethyl acetate fraction were the highest ($196.02{\pm}5.26$ and $114.00{\pm}10.03{\mu}g/mg$, respectively). DPPH and ABTS radical scavenging activities increased according to the concentration of O. humifusa cladodes extract, and those of the ethyl acetate fraction were the highest. Ferric reducing antioxidant powers of chloroform and the ethyl acetate fraction were higher than those of other fractions. Overall, the ethyl acetate fraction of O. humifusa cladodes showed the highest anti-diabetic and antioxidant effects. Results indicate that O. humifusa cladodes powder has potential as a useful ingredient with anti-diabetic and antioxidant effects.

Chemical compositions and antioxidant activities of Cheonnyuncho (Opuntia humifusa) stems and fruit (천년초의 성분특성과 항산화 활성)

  • The purpose of this study was to investigate the chemical composition of the freeze-dried stems and fruit of the cactus Cheonnyuncho. The analysis showed that powdered stems have the highest fat content (1.91%) and the powdered fruits have he highest protein content (2.62%). The K content of the fruits higher than that of the stems, while the Ca, Mg, Na and P contents of the stems were higher than those of the fruits. Both the stems and fruits powders contained high levels of the amino acids glutamic acid and aspartic acid. The free sugars such as sucrose, fructose, and glucose were detected in both the stems and fruits. The 75% ethanol (EtOH) extract showed a relatively high antioxidative activity compared to those of the water and 75% methanol (MeOH) extracts. Furthermore, the 75% EtOH extract of the stem powder exhibited a total polyphenol content of 3.60 g/100 g, and a total flavonoid content of 2.00 g/100 g. The antioxidant activities of the stem and fruit powder extracts, measured in DPPH radical scavenging experiments, were higher than that of the control group.

Hepatoprotective Effect of Cheonnyuncho (Opuntia humifusa) Extract in Rats Treated Carbon Tetrachloride (천년초(Opuntia humifusa) 추물물의 사염화탄소를 처치한 흰쥐에서의 간보호 효과)

  • Effect of Cheonnyuncho extract on the liver injury of rats treated carbon tetrachloride $(CCl_4)$ was studied. Cheonnyuncho extract was administerd at dose of 0.5 and 1 g/kg/day, p.o. for 2 weeks. $CCl_4$ was treated at dose of $0.5\;mL/kg$, i.p. 3 hours later from the last pretreatment of Cheonnyuncho extract. Administration of Cheonnyuncho extract at a dose of 1 g/kg decreased serum AST, ALT and ALP activities by 36, 41, and 22% respectively compared to $CCl_4$ treatment group. Increased lipid peroxidation and decreased SOD and GST activities were also recovered by pretreatment of Chonnyuncho extract in liver of rats. These results suggest that Cheonnyuncho extract has hepatoprotective effect against liver injury.
