Analysis of Genetic Diversity and Structure of Prunus salicina Lindl. Populations in Adjacent Area (자두나무(Prunus salicina Lindl.) 접경지역 집단의 유전 다양성 및 구조 분석)

  • 자두나무(Prunus salicina Lindl.)는 세계에서 5번째로 많이 생산되는 과실로, 한국에서 재배하는 자두나무의 기본종은 중국 양쯔강 유역에서 기원했다. 2016년 과수용 자두나무와는 다른 자두나무가 양구에서 발견되었다. 양구군, 인제군, 고성군 일대의 자두나무 개체군의 유전다양성 및 집단 구조 분석을 통해 본 자두나무 개체군의 유전다양성 및 개체군 구조를 확인하고자 했다. 과수용 재배종을 포함한 시료를 채취하여 GBS 분석을 진행했고 주성분 분석과 STRUCTURE 분석을 통해 개체군간 유전적 구조를 확인했다. 재배종의 유전형이 다른 개체군에서도 나타나는 것으로 보아 유의한 유전적 분화가 일어났다고 보기 힘들었다. 하지만 고성군 고진동계곡 등 DMZ에 인접한 집단이 분계도에서 재배종 개체군과 가장 유전적 거리가 있는 것으로 나타났다. 하지만 본 분석으로는 야생집단으로 발견도니 자두나무의 실체와 기원을 단정짓기에는 부족한 것으로 보이며 외군 추가 및 더 많은 시료를 확보하는 등 추가 조사와 분석이 필요하다.

A nationwide analysis of mammalian biodiversity hotspots in South Korea (전국단위의 포유류 생물다양성우수지역 분석 연구)

  • Hotspots are top sites in terms of species diversity as the most threatened and most diverse sites which have been used to select priority areas for reserves. The purpose of this paper is to identify biodiversity hotspots through analyzing nationwide spatial patterns of species richness and rarity of Korean mammals. Four endangered mammals and eleven common mammals were selected as target species. Environmental variables as model input data were consisted of topography, distance, and vegetation structure etc. and Maxent was used to develop species distribution models for target species. Species richness and rarity were used as index of biodiversity. The results of this study were as follows. Firstly, hotspots of species richness for endangered mammals were in high elevation and steep mountain areas. However, species richness for whole mammals were high in low elevation of mountains. Secondly, distribution pattern of species rarity for endangered mammals were similar as richness. However, hotspots of species rarity for whole mammals were a little different from species richness. Species rarity was high in both low and high elevation of mountain areas. This study will provide the useful information for a biodiversity assessment, a habitat conservation, a national ecological network plan, and the management of protected areas.

Genetic Diversity and Phylogenetic Relationship of Genus Phyllostachys by ISSR Markers (ISSR 분자 마커를 이용한 왕대속 대나무의 유전적 다양성 및 계통 관계)

  • Four taxa of the genus Phyllostachys were analysed with PCR-based molecular inter-simple sequence repeat (ISSR) determine markers to determine their phylogenetic relationships. Many species of this genus are regarded as economical and ecologically important in the world. With the nine primers screened, ISSR assay generated a total of 64 reproducible bands. Analysis of ISSR from individual plants of genus Phyllostachys resulted in 43 polymorphic bands with 70.49%. When species were grouped by four taxa, within group diversity was 0.092 (Hs), while among group diversify was 0.499 (Gst) on a per locus basis. The estimated gene flow (Nm) between the pairs of species was 0.559. Hence, we can expect weak or low gene flow among species. Total mean genetic diversity ($H_T$) for four taxa was 0.291. The ISSR data allowed us to resolve well-supported clades in the genus Phyllostachys. The four taxa of the genus Phyllostachys analyzed were distinctly related to a monophyletic.

Spatio-temporal Moth Diversity (Insecta: Lepidoptera) of Mt. Baegun-san, Gwangyang, Jeonnam (전라남도 광양시 백운산 나방 (곤충강: 나비목)의 시공간적 다양성 연구)

  • This study was aimed to investigate the elevational and seasonal variation of moth diversity of Mt. Baegun-san, Gwangyang-city, Jeonnam. Moth collecting was carried out using a UV light trap at three different elevational sites from September to October 2014 and from April to October 2015 (9 times) except winter. A total of 288 moth species and 1,444 individuals in 15 families were identified. Geometridae (97 species and 740 individuals) were the most dominant in the numbers of species and individuals, followed by Noctuidae (62 species, 193 individuals), Erebidae (52 species, 177 individuals) and Notodontidae (16 species, 111 individuals). Elevational change showed that the lower elevation, the higher number of species and individuals. Seasonal change of numbers of species and individuals showed the M-shaped pattern with June and August peaks. Compared to the previous records from historical literatures, 197 species were newly added and the total moth species richness of Mt. Baegun-san comprised 449 species in 20 families. The Chao 2 estimated number of species using the observed species including unpublished data set was 1,530 species that comprised about 43% of the Korean Lepidopteran fauna. This large number of estimated species requested the more extensive and long-term survey for better understanding of the total moth fauna in this area.

Insect Fauna of Urban Green Park in Daegu Metropolitan City, Korea (II) (대구 도심 녹지 공원의 곤충상에 관한연구(II))

  • This study was conducted to know the insect fauna on the 5 green Parks areas of Daegu Metropolitan city. Materials were collected by net sweeping and pitfall trap of cup. As the result, 100 species of 93 genera belonging to 61 families in 9 orders were surveyed, among them, Yeonam Park showed highest insect occurrence as 61 species belonging to 35 families of 8 orders. The other side, lowest site was Gukchae-Bosang memorial Park as 2 species from 2 orders. There was more species in broad-leaf tree area than mixture area of broadleaf and needle-leaf trees. Turf grass areas in the Parks showed low insect diversities.

Genotoxicity Study of Immature Green Persimmon Extract (풋감 주정 추출물의 유전독성 연구)

  • The persimmon is commonly cultivated in temperate regions of the world, including China, Korea, Japan, Brazil, Turkey, and Italy. In some Asian cultures, consumers are aware of the health claims related to the persimmon and its functional ingredients. The rich phytochemistry of the persimmon has opened new avenues of research on diet-based regimens designed to cure various ailments. This study was conducted to identify the genotoxicity of immature green persimmon (Diospyros kaki THUNB.) extract (DKA). The bacterial reverse mutation assay, the chromosomal aberration assay, and the mammalian micronucleus test were performed to determine the DKA genotoxicity. The result of the bacterial reverse mutation assay revealed that the DKA did not induce mutagenicity in Salmonella typhimurium TA98, TA100, TA1535, TA1537 and Escherichia coli WP2uvrA with or without metabolic activation of S9 mixture. The oral administration of DKA also caused no significant increase in the number of micronucleated polychromatic erythrocytes or in the mean ratio of polychromatic to total erythrocytes. In addition, DKA did not cause a significant chromosome aberration on CHL cells in the presence or absence of S9 activation. In conclusion, DKA could be considered as a reliable and safe functional food since no toxicity was found under the condition of this study.

Insect Fauna of Cemetery Area in Forest from Korea (산림 내 묘지 지역의 곤충상에 관한 연구)

  • This study was carried out to know the insect fauna of turf grass area in forest. In Korea, graves and public cemetery in mountain areas are usually covered by turf grass which were mainly examined. There were 86 species belonging to 77 genera of 39 families in 11 orders. Among them, site 3 had highest occurrence (38%), second was site 2 (36%) and in the last place was site 1 (26%). Hemiptera revealed best species composition (29.1%) and came Coleoptera (19.8%), Orthoptera (15.1%), Lepidoptera (12.8%) and Odonata (5.8%) etc. The reason why Hemiptera and Orthoptera are mainly found in these areas is because they usually feed on short plants that grow in there. Taxonomic species composition, occurrence ratio by site and species and comparison analysis were provided.

Effects of Acidification on the Species Compositions of Heterotrophic Bacterial Community in Microcosm (수계 종속영양세균 군집의 종조성에 미치는 산성화의 영향)

  • In an artificial pH-gradient hatch culture system, the author analyzed the effects of acidification on the species composition of heterotrophic bacteria. As the result of this study, it was found that the numbers of total bacteria were not affected by acidification and that the population size of heterotrophic bacteria decreased as pH became lower. The heterotrophic bacteria isolated from all of the pH gradient were 12 genera and 22 species. and among them, gram negative and gram positive bacteria were 64% and 36%, respectivcly. As pH decreased, the distribution rate of gram negative bacteria increased while that of gram positive bacteria decreased. Regarding to distrihution rate of genuses in each pH gradient, 13 genuses appeared at pH 7 while only 5 genuses appeared at pH 3. which means that the diversity of genera decrease as pH decreased. As a result of cluster analysis, diversity indices 01 species had ranges from 1.13 to 2.37, and decreased as pH decreased. In order to evaluate the diversity of different size samples, we analyzed the expected number of species appearance according to pH by rarefaction method. The statistical significance of species diversity was verified by the fact that the number decreased at lower pH.

Biodiversity research of mushrooms in hallasan mountain natural reserve (한라산천연보호구역 내 버섯 종 다양성 연구)

  • This study surveyed mushrooms as part of the Hallasan Natural Reserve Basic Academic Survey, which was conducted for the purposes of securing basic data on the Hallasan Mountain Natural Reserve; our study targeted the area between Seongpanak Trail and Donnaeko Trail from April to October 2018. A total of 392 species from 184 genera in 65 families were investigated. Among them, 187 mushrooms were wood-decaying species. The greatest diversity in mushroom species was recorded in July (206 species) at an altitude of 600-800 m above sea level. Among the mushrooms investigated, five were climate-sensitive biological indicator species and two were potential candidates for that classification. Of these, 163 species were biological resources that require approval for overseas export.

  • Copepods play an important role as a primary consumer in the various aquatic ecosystems. Most of marine benthic copepods belong to the Order Harpacticoida, and are known over 3,000 species in the world. Research on the biodiversity of the benthic copepods are important in the development of the new bio-resources, and the genetic resources. The importances and the prospects of biodiversity research on the mavins benthic copepods are discussed.
