Selection of rs2rs2titi Soybean Genotype with Yellow Seed Coat (rs2rs2titi 유전자형을 가진 노란 콩 계통 선발)

  • Soybean [Glycine max (L.) Merr.] seed is an important dietary source of protein, oil, carbohydrates, isoflavones, and other nutrients for humans and animals. But, antinutritional factors in the raw mature soybean are exist. Kunitz trypsin inhibitor (KTI) protein and stachyose are main antinutritional factors in soybean seed. The genetic removal of the antinutritional factors will improve the nutritional value of soybean seed. The objective of this research was to breed a new yellow soybean strains (rs2rs2titi genotype) with the traits of lacking of KTI protein and low content of stachyose. Breeding population was developed from the cross of "Jinyangkong" and 15G1 parents. Presence or absence of KTI protein was detected based on Western Blot technique. Content of stachyose in mature seed was detected by HPLC. Total four new strains (603-1, 603-2, 625, and 694) with KTI protein free and low content of stachyose were selected. Four strains (603-1, 603-2, 625, and 694) have yellow seed coat and hilum. Plant height of 603-1 strain was 65 cm and 100-seed weight was 29.2 g. Plant height of 603-2 strain was 66 cm and 100-seed weight was 26.2 g. Plant height of 625 strain was 64 cm and 100-seed weight was 27.1 g. Content of stachyose for four new strains was 3.0~3.50 g/kg. Four strains selected in this research will be used to improve new yellow soybean cultivar with KTI protein free, and low content of stachyose.

  • Soybean [Glycine max (L.) Merr.] is grown worldwide for its high protein and oil content. Anthocyanins from black soybean seed coats are known to have many pharmaceutical effects. Soybean cultivars with large seed sizes and black seed coats are needed by soybean farmers. However, antinutritional factors, like protein, stachyose, and Kunitz trypsin inhibitor (KTI) exist in raw mature soybeans. Genetic elimination or reduction of these components is needed in soybean breeding. The objective of this research was to develop new a soybean strain with black seed coats and green cotyledons that was KTI protein free and low in stachyose. Six parents were used. The presence or absence of KTI protein was detected using the Western blot technique. The content of stachyose in mature seeds was detected using HPLC. One new strain was selected from 11 $F_2$ plants with black seed coats and green cotyledons that lacked KTI protein. The new strain had black seed coats and green cotyledons and was KTI protein free and low in stachyose. The plant height of the new strain was 66 cm, and its 100-seed weight was 28.4 g. The stachyose content of the new strain was 2.59 g/kg. The new strain developed in this research will be used to develop new cultivars that are KTI protein free and low in stachyose.

'Hyowon6', a Mid-Late Maturing Rice Variety with High Quality (고품질 중만생 벼 품종 '효원6호')

  • Hyowon6, a mid-late maturing, good eating quality rice variety with multi-resistance, was developed by the rice breeding team of Pusan National University in 2013. This variety was derived from a cross between Hwayeong (as a multi-resistant parent) and Koshihikari (as a good eating quality parent). Selection by the pedigree breeding method was carried out until the $F_8$ generation. A promising line, JS14-12-36-8-5-3-1-1-1, was developed and designated as HY103 in 2013. This variety headed on August 22, which was 11 days later than that of Hwayeong. The culm length and panicle length of Hyowon6 was 85.7 cm and 21.1 cm, respectively. The number of panicles per hill was 14.5 and the number of grains per panicle was 101.7. The ratio of ripened grain was approximately 92% and the 1,000-grain weight was approximately 21.36 g for brown rice, which were similar to those of Hwayeong. Hyowon6 was moderately resistant to lodging and also to neck blast, leaf blight, and stripe virus. The glossiness value of Hyowon6 was 83, which was considerably higher than that of Hwayeong.

A New Small Redbean Cultivar 'Geomguseul' with Lodging Resistance and High-Antioxidant Activity (쓰러짐에 강하고 항산화 활성이 높은 팥 '검구슬')

  • A new small red bean cultivar, 'Geomguseul', was artificially crossed between 'Chilbo' and 'SA8412-3-1-4-3-3-2-3' in 1998, fixed excellent agronomic characters by pedigree breeding method, and selected for the further trials with the name of 'Miryang 10'. It was prominent and showed good result, such as high grain quality, lodging resistance, and high-antioxidant activity in the regional adaptation yield trials (RYT) for three years from 2009 to 2011 and released for the public consumption as the name of 'SA9801-24-4-3-1' in 2011. 'Geomguseul' has a semi-determinate growth habit, yellow flower, green embryonic axis, black seed coat, white hilum, and small spherical seed (12.2 grams per 100 seeds). The average yield of 'Geomguseul' was 2.01 MT/ha in the regional yield trials (RYT) carried out for three years from 2009 to 2011 which was a little higher than that (1.98 MT/ha) of the check variety, 'Chungju'.

Ferulic Acid Content of Barley and Wheat Grains and Head Blight Resistance (맥류 종자의 페룰산(ferulic acid) 함량과 붉은곰팡이 저항성)

  • In order to find if a ferulic acid (FA) can be used as a selection index in cereal breeding for resistance to head blight and mycotoxin production, we analyzed FA in the grains of 80 cultivars of barley, rice, and wheat. FA content ranged 1.66-2.77 mg/g in barley (n=20), 0.56-1.53 mg/g in wheat (n=40), and 0.91-2.13 mg/g in rice (n=20). Among these, 7 cultivars each of barley and wheat with different FA content were tested for head blight and mycotoxin production by 2 Fusarium graminearum and 2 F. asiaticum strains. Mean pathogenicity of the wheat cultivars was significantly less than that of barley with higher FA and among wheat cultivars, there was no correlation between FA content and pathogenicity. Mycotoxin production was also lower in the wheat than in the barley as pathogenicity. However, pathogenicity and toxins produced by F. asiaticum were negatively correlated with FA content in barley. These results indicate that FA is not a resistance factor to head blight by F. asiaticum and F. graminearum or its mycotoxin production in barley and wheat.

Origin and evolution of Korean ginseng revealed by genome sequence

  • Panax ginseng (Ginseng or Korean ginseng) is one of the most important medicinal herbs in the world. We made a high-quality whole genome sequence of P. ginseng using 'Chunpoong' cultivar, which is the first cultivar registered in Korea Seed and Variety Service (KSVS) with relatively similar genotypes and superior phenotypes, representing approximately 3 Gbp and 60,000 genes. Genome sequence analyses of P. ginseng and related speciesrevealed the origin of Korean ginseng and the ecological adaptation of 18 Panax species around the world. Korean ginseng and American ginseng (P. quinquefolius) are tetraploid species having 24 chromosome pairs, while the other 16 species are diploid species with 12 chromosome pairs. Panax and Aralia are the closest genera belonging to the Araliaceae family that diverged approximately 8 million years ago (MYA). All Panax species evolved as shade plants adapting to cool climates and low light conditions under the canopy of deep forests from Southeast Asia such as Vietnam to Northeast Asia such as Russia approximately 6 MYA. However, through recurrent ice ages and global warming, most diploid Panax species disappeared due to the freezing winter, while tetraploid P. ginseng may have appeared by allotetraploidization, which contributed to the adaptation to cold temperaturesin Northeast Asian countries including the Korea peninsula approximately 2 MYA. American ginseng evolved by the adaptation of P. ginseng in Northeast America after the intercontinental migration 1 MYA. Meanwhile, most of diploid Panax species survived in high-altitude mountains over 1,600 meters in Southeast Asia because they could not endure the hot temperature and freezing cold. The genome sequence provides good basisto unveil the origin and evolution of ginseng and also supports practical gene chips which is useful for breeding and the ginseng industry.

Seeding Rate and Days for Low-density Transplant Cultivation (드문모심기 적정 파종량 및 육묘일수 분석)

  • In order to develop a safe seeding technology for low-density transplantation, we analyzed the changes in seeding growth according to different seeding rates and seeding days. We detected changes in leaf number and shoot diameter on the 10th seeding day, and in plant height on the 20th seeding day according to seeding rate. The increase in shoot and leaf number, and shoot diameter per seeding day decreased with anincrease in seeding rate. On seeding day 20, the reductions in shoot and root dry weights according to seeding rate was lower compared with those recorded at seeding day 10. Shoot dry weight was significantly related to root dry weight, plant height, and shoot diameter. With an increase in seeding rate, seeding health decreased and there was an increase in the rate of seeding health per seeding day decreased. For low-density transplantation, the optimal number of seeds required for transplanting 3-5 plants per hill is 11,000, which is estimated to be approximately 280-300 g of seeds for 22 g brown rice. Furthermore, the appropriate number of seeding days is estimated to be 13 to 16 days, which corresponds to a small reduction in seeding health.

Flowering and Inflorescence Development Characteristics of Korean Mint Affected by Different Photoperiods (광주기에 따른 배초향의 개화 및 화서 발달 특성)

  • The Korean mint (Agastache rugosa Kuntze) is used as a leaf vegetable. Therefore, it is important to improve the quality and quantity of leaves by maintaining vegetative growth. When the development of a plant is switched from vegetative to reproductive growth, leaf development may be lowered, leading to a decrease in marketability. Therefore, this study was conducted to investigate the effect of the photoperiod on the flowering characteristics and inflorescence development of the Korean mint to select an appropriate photoperiod for leaf production. The seeds were sown in 105-cell trays on 29 November 2021, and planted in each pot on 27 December 2021, when four main leaves appeared. After planting, the light/dark period was adjusted to 10/14hrs, 12/12hrs, 14/10hrs, 16/8hrs. After 19 days of planting, the first flowering occurred in 10/14hrs, which had a short photoperiod, and the 12/12hrs had 20 days for flowering, and other treatments had 21 days. In addition, it was confirmed that the Korean mint has the characteristics of facultative quantitative short-day plant, through the increase in the development of inflorescences as the light period is shorter than the dark period. As for the growth of Korean mint, the longer the photoperiod, the higher values were shown with leaf length, number of leaves, fresh weight, dry weight, and leaf area of the shoot. Therefore, it is considered that 14 h or more light period is suitable for increasing the productivity of Korean mint as a leafy vegetable.

Morphological Development of Eggs, Larvae and Juveniles of the Clidoderma asperrimum (Cypriniformes: Pleuronectidae) (줄가자미 Clidoderma asperrimum의 난발생 및 자치어 형태발달)

  • Egg development and morphological change of larvae and juvenile of the Roughscale sole, Clidoderma asperrimum, were investigated in the present study. Adult fishes were collected on the East Sea, Korea, from 2017 to 2018 and reared in a circular water tank (Ø 6×1 m) at water temperature of 12.8±1.9℃. Fertilized eggs ranged from 1.42 to 1.59 mm (mean 1.51±0.04 mm, n=50) in diameter. The eggs were spherical in shape, transparent, floating and colorless. The egg yolk was separated from the egg membrane 60 mins post-fertilization (PF), and an embryo was formed in 62 hrs PF. More than 50% of the eggs hatched within 144 hrs PF in the range of 10.2~11℃(mean 10.8℃). The size of the newly hatched larvae were 4.22~4.64 mm (mean 4.53±0.16 mm) in total length (TL), their mouth and anus were not open yet. At 10 days after hatching, the preflexion larvae reached 5.88~6.62 mm (mean 6.31±0.33 mm) in TL, and the yolk absorption was completed and the mouth began to open. At 55 days after hatching the larvae reached to flexion larvae stage and they were 10.4~13.3 mm (mean 12.7±1.3 mm) in TL, and the tip of notochord was bent upward. At 120 days after hatching the larvae reached to juvenile stage and they were 35.3~40.5 mm (mean 39.5±2.4 mm) in TL. Their all fins had completed set of the fin-rays (D. 79~94: A. 63~75) and the juveniles adopted a benthic life.

Analysis of Growth Characteristics and Major Components of Angelica gigas Root under Flower Colors (참당귀 꽃 색에 따른 생육특성 및 뿌리의 주성분 함량 분석)

  • The aim of this study was to provide basic data for breeding new varieties of Angelica gigas Nakai by identifying the growth characteristics and useful components of its below- and above-ground parts. The basic varieties expressing dark purple (N79-A), light blue-pink (62-C), and white (NN115-C) flowers were identified and collected in the cultivation area, and their seeds were cultured for use as experimental materials. Qualitative growth characteristics of the above-ground parts were then examined. Purple coloration was evident throughout the entire plant body in the N79-A group, whereas the 62-C and NN115-C groups displayed yellow-green coloration. In terms of quantitative growth characteristics, the 62-C group displayed significantly lower above- and below-ground growth than the other two groups. Levels of nodakenin and decursinol-angelate were high in the NN115-C group (0.88 ± 0.13%) and N79-A group (2.56 ± 0.12%). However, the overall results were not statistically significant. The results could provide a foundation for breeding new varieties of Angelica gigas Nakai, which are used predominantly for medicinal purposes and have low bolting and high yield potential.
